Phosphorylation of Akt Mediates Anti-Inflammatory Activity of 1-p-Coumaroyl ${\beta}$-D-Glucoside Against Lipopolysaccharide-Induced Inflammation in RAW264.7 Cells

  • Hydroxycinnamic acids have been reported to possess numerous pharmacological activities such as antioxidant, anti-inflammatory, and anti-tumor properties. However, the biological activity of 1-p-coumaroyl ${\beta}$-D-glucoside (CG), a glucose ester derivative of p-coumaric acid, has not been clearly examined. The objective of this study is to elucidate the anti-inflammatory action of CG in lipopolysaccharide (LPS)-stimulated RAW 264.7 macrophage cells. In the present study, CG significantly suppressed LPS-induced excessive production of pro-inflammatory mediators such as nitric oxide (NO) and $PGE_2$ and the protein expression of iNOS and COX-2. CG also inhibited LPS-induced secretion of pro-inflammatory cytokines, IL-$1{\beta}$ and TNF-${\alpha}$. In addition, CG significantly suppressed LPS-induced degradation of $I{\kappa}B$. To elucidate the underlying mechanism by which CG exerts its anti-inflammatory action, involvement of various signaling pathways were examined. CG exhibited significantly increased Akt phosphorylation in a concentration-dependent manner, although MAPKs such as Erk, JNK, and p38 appeared not to be involved. Furthermore, inhibition of Akt/PI3K signaling pathway with wortmannin significantly, albeit not completely, abolished CG-induced Akt phosphorylation and anti-inflammatory actions. Taken together, the present study demonstrates that Akt signaling pathway might play a major role in CG-mediated anti-inflammatory activity in LPS-stimulated RAW264.7 macrophage cells.

Critical role of protein L-isoaspartyl methyltransferase in basic fibroblast growth factor-mediated neuronal cell differentiation

  • We aimed to study the role of protein L-isoaspartyl methyltransferase (PIMT) in neuronal differentiation using basic fibroblast growth factor (bFGF)-induced neuronal differentiation, characterized by cell-body shrinkage, long neurite outgrowth, and expression of neuronal differentiation markers light and medium neurofilaments (NF). The bFGF-mediated neuronal differentiation of PC12 cells was induced through activation of mitogen-activated protein kinase (MAPK) signaling molecules [MAPK kinase 1/2 (MEK1/2), extracellular signal-regulated kinase 1/2 (ERK1/2), and p90RSK], and phosphatidylinositide 3-kinase (PI3K)/Akt signaling molecules PI3Kp110β, PI3Kp110γ, Akt, and mTOR. Inhibitors (adenosine dialdehyde and S-adenosylhomocysteine) of protein methylation suppressed bFGF-mediated neuronal differentiation of PC12 cells. PIMT-eficiency caused by PIMT-specific siRNA inhibited neuronal differentiation of PC12 cells by suppressing phosphorylation of MEK1/2 and ERK1/2 in the MAPK signaling pathway and Akt and mTOR in the PI3K/Akt signaling pathway. Therefore, these results suggested that PIMT was critical for bFGF-mediated neuronal differentiation of PC12 cells and regulated the MAPK and Akt signaling pathways.

혈관내피세포에 부착하는 THP-1에 대한 발효마늘추출물의 효과 (Effect of Fermented Garlic Extract on Adhesion of THP-1 Cells to Bovine Aortic Endothelial Cells)

  • 마늘은 전통적으로 cycloalliin을 포함한 기능성 식품으로 널리 알려져 있다. 본 연구를 통해 혈관계에서 발효마늘추출물이 어떤 효능을 갖는지 알아보았다. 먼저, 발효마늘추출물이 내피세포의 단핵구 부착을 억제하는 것을 관찰하였다. 농도 변화에 따른 실험결과, 발효마늘추출물을 0.1 또는 $1{\mu}g/ml$ 농도로 전처리 하면, LPS에 의하여 촉발되는 소 대동맥 내피세포에 달라붙는 THP-1의 부착이 억제되는 것을 확인하였다. 단핵구의 내피세포 부착이 염증반응 초기단계에 진행되는 과학적 근거를 고려하면 이와 같은 결과는 발효마늘추출물이 혈관 염증을 저해한다고 볼 수 있다. 또한 세포신호전달물질의 활성변화를 관찰한 결과, 발효마늘추출물이 소 대동맥 내피세포에서 eNOS와 Akt의 활성을 유도함을 알았다. eNOS의 인산화 반응은 발효마늘추출물을 처리한지 10분 후에 최대로 상승되었으며, 이와 유사하게 Akt도 발효마늘추출물을 처리하고 10분 후에서 인산화 반응이 최대로 상승되었다. 산화질소 양 변화 측정에서도 발효마늘추출물은 산화질소의 생성을 상승시켰다. 한편, 산화질소 생성을 저해하는 억제제 처리는 발효마늘추출물에 의해 나타나는 세포부착 저해능력을 감소시켰다. eNOS와 Akt를 매개로 하는 LPS 유도 세포 부착 정도가 발효마늘추출물에 의하여 억제된다는 것은 산화질소가 세포부착에 중요 매개물임을 의미한다. 결론적으로, 발효마늘추출물은 염증관련 질환을 예방하는 데에 중요한 역할을 한다고 할 수 있다.

나노방출제어시스템을 이용하여 trichloroacetic acid와 epidermal growth factor의 순차적 방출을 적용한 인간치은섬유아세포의 세포생존 관련 유전자 연구분석 (Analysis of cell survival genes in human gingival fibroblasts after sequential release of trichloroacetic acid and epidermal growth factor using the nano-controlled release system)

  • 목적: 본 연구에서는 구강연조직재생에서 인간치은섬유아세포에 hydrophobically modified glycol chitosan (HGC) 기반 나노방출제어시스템을 적용 시 PI3K-AKT 신호전달의 세포생존 연관 유전자를 통해 trichloroacetic acid (TCA)와 epidermal growth factor (EGF)의 영향을 확인하고자 하였다. 연구 재료 및 방법: TCA와 EGF를 방출하는 나노방출제어시스템을 제작하였다. 실험군은 3가지 군으로 나누었다; 대조군(CON), TCA-담지형 나노방출제어시스템 투여군(EXP1), TCA-와 EGF- 담지형 나노방출제어시스템 투여군(EXP2). 인간치은섬유아세포 배양 후 PI3K-AKT 신호전달에 연관된 총 29개 유전자를 실시간 중합효소연쇄반응으로 분석했다. 일요인 분산분석 및 다중회귀분석이 사용되었다. 결과: 세포생존 관련 유전자들은 EXP1과 EXP2에서 상향조절되었다. 다중회귀분석에서는 ITGB1이 AKT1의 발현에 가장 영향력 있는 요소로 결정되었다. 결론: HGC기반 나노방출제어시스템을 통한 TCA와 EGF의 적용은 세포생존에 관한 신호전달을 상향 조절시킬 수 있다.

Iris Nertschinskia Ethanol Extract Differentially Induces Cytotoxicity in Human Breast Cancer Cells Depending on AKT1/2 Activity

  • Recently, we reported that an ethanol extract of Iris nertschinskia induces p53-dependent apoptosis in the MCF7 human breast cancer cell line. However, the detailed mechanisms were not fully explored. Here, we demonstrate another aspect of the activity of I. nertschinskia in breast cancer cells. We compared the response to an ethanol extract of I. nertschinskia in two different human breast cancer cell lines, Hs578Tand MDA-MB231, respectively with relatively low and high AKT1/2 activity by trypan blue exclusion assay and FACS analysis. Knockdown of endogenous AKT1 or AKT2 in breast cancer cells by RNA interference determined the sensitivity to I. nertschinskia ethanol extract compared to control cells. The I. nertschinskia ethanol extract induced cell death in a manner that depended on the level of phosphorylated AKT1/2 protein and was associated with a significant increase in the sub-G1 cell population, indicative of apoptosis. Our results indicate that an ethanol extract of I. nertschinskia differentially induces cell death in breast cancer cells depending on their level of phosphorylated AKT1/2.

진생베리 열수 추출물의 다당체 분해 효소와 인슐린 신호전달 분자 PTP1B와 AKT1에 미치는 효과 (Effects of Ginseng Berry Water Extract on the Polysaccharide Hydrolysis of Extracellular Enzymes and Intracellular PTP1B and AKT1)

  • 인삼은 건강에 효과적인 약초라고 알려져 왔다. 인삼의 열매인 진생베리는 인삼의 주성분과 비슷한 ginsenoside, saponin, polyphenol, polyacetylene, alkaloid 등의 성분을 포함한다. 본 연구의 목적은 인삼과 같이 진생베리 열수 추출물(Ginseng berry water extract, GBE)이 당뇨와 연관된 세포 외 효소와 인슐린 신호전달 경로에 있는 분자의 발현에 어떤 효과를 가지고 있는지를 조사하였다. ${\alpha}$-Amylase와 ${\alpha}$-glucosidase는 섭취한 다당분자를 분해하여 포도당을 생성함으로 항당뇨 약물개발의 표적 효소이다. GBE에 의한 두 효소의 활성 억제능을 in vitro에서 측정하였으나 최고 $1,000{\mu}g/ml$ 농도에서도 효소활성 억제능이 나타나지 않았다. 인슐린 신호전달 경로의 영향을 확인하기 위해서 HepG2 세포에서 GBE에 의한 인슐린 신호전달 경로의 주요 단백질인 protein-tyrosine phosphatase 1B (PTP1B)와 Akt1의 발현수준 변화를 Western blot 방법으로 조사하였다. 이때 인슐린에 의한 이들 분자의 변화에 GBE가 영향을 주는 것으로 나타났다. PTP1B는 인슐린에 의해 증가된 발현량이 저농도의 GBE이 의해 더욱 증가하였으나, $200{\mu}g/ml$ 농도의 GBE에 의해서는 다소 감소하는 것으로 나타났다. 또한, Akt1도 인슐린에 의해 증가된 발현량이 GBE 농도에 따라 감소하는 것으로 나타났다.

인슐린 매개성 Microsomal Epoxide Hydrolase의 발현증가에서 Akt의 역할 (Role of Akt in Insulin-Mediated Increase in Expression of Microsomal Epoxide Hydrolase)

  • The present study examines the effect of dominant-negative Akt on the insulin-mediated microsomal epoxide hydrolase (mEH) induction in rat hepatocytes. We also assessed the role of insulin in the expression of soluble epoxide hydrrolase (sEH). Insulin increased mEH levels and the enzyme activities, whereas sEH protein expression was unaffected by insulin. The specific PI3K inhibitors or p70 S6 kinase inhibitor ameliorated the insulin-mediated increase in mEH protein levels. Infection with adenovirus expressing dominant-negative and kinase-dead mutant of Akt1 effectively inhibited the insulin-mediated increase in mEH expression and mEH activity. These results suggest that mEH and sEH are differentially regulated by insulin and PI3K/Akt/p70S6K are active in the insulin-mediated regulation of mEH expression.

유전적 고혈압 발병에 대한 Calcineurin 및 PKB/Akt의 연관성 (Involvement of calcineurin and PKB/Akt in development of hereditary hypertension)

  • Severe hypertension (>180 mmHg) develops in spontaneously hypertensive rats (SHR) after 12 wk-old; however, it is not clear whether what kinds of molecular mechanism leads to altered cardiac performance following developmental stages in SHR. Also, although the effect of calcineurin (Cn) to promote cardiomyocyte hypertrophy in vivo and in vitro is established, its overall necessity as a hypertrophic mediator is currently an area of ongoing debate. Thus, we have examined i) body weight and blood pressure, ii) differences of expression and distribution of signaling molecules such as Cn, protein kinase B/Akt (PKB/Akt), and extracellular signal-regulated kinase (ERK) between SHR and their age-matched control Wistar-Kyoto (WKY) rats following developmental stages. In 16 wk-old SHR compared with WKY, 2-dimentional echocardiography showed cardiac enlargement and hypertrophy of left ventricle, significantly. Taken together, we suggest that Cn is associated with hereditary cardiac hypertrophy, the process being related to the molecular signaling mechanisms involving PKB/Akt and ERK.

Induction of Autophagy by Rosa acicularis Leaves Extracts in RAW264.7 Cells

  • Autophagy contributes to enhancing the immune system (innate and adaptive immune system) against foreign pathogens. Autophagy of macrophages is used as a major indicator for developing vaccine adjuvants to increase the adaptive immune response. In this study, RAL increased the production of immunostimulatory mediators and phagocytotic activity in RAW264.7 cells. RAL increased p62/SQSTM1 expression. Inhibition of TLR4, JNK, and PI3K/AKT blocked RAL-mediated increase of p62/SQSTM1. RAL activated JNK and PI3K/AKT signaling. RAL-mediated activation of JNK and PI3K/AKT signaling was reversed by TLR4 inhibition. Taken together, it is believed that RAL-mediated autophagy may be dependent on activating via TLR4-dependent activation of JNK and PI3K/AKT signaling in macrophages.

Korean Red Ginseng inhibits apoptosis in neuroblastoma cells via estrogen receptor ${\beta}$-mediated phosphatidylinositol-3 kinase/Akt signaling

  • Background: Ginseng has been shown to exert antistress effects both in vitro and in vivo. However, the effects of ginseng on stress in brain cells are not well understood. This study investigated how Korean Red Ginseng (KRG) controls hydrogen peroxide-induced apoptosis via regulation of phosphatidylinositol-3 kinase (PI3K)/Akt and estrogen receptor (ER)-${\beta}$ signaling. Methods: Human neuroblastoma SK-N-SH cells were pretreated with KRG and subsequently exposed to $H_2O_2$. The ability of KRG to inhibit oxidative stress-induced apoptosis was assessed in MTT cytotoxicity assays. Apoptotic protein expression was examined byWestern blot analysis. The roles of ER-${\beta}$, PI3K, and p-Akt signaling in KRG regulation of apoptosis were studied using small interfering RNAs and/or target antagonists. Results: Pretreating SK-N-SH cells with KRG decreased expression of the proapoptotic proteins p-p53 and caspase-3, but increased expression of the antiapoptotic protein BCL2. KRG pretreatment was also associated with increased ER-${\beta}$, PI3K, and p-Akt expression. Conversely, ER-${\beta}$ inhibition with small interfering RNA or inhibitor treatment increased p-p53 and caspase-3 levels, but decreased BCL2, PI3K, and p-Akt expression. Moreover, inhibition of PI3K/Akt signaling diminished p-p53 and caspase-3 levels, but increased BCL2 expression. Conclusion: Collectively, the data indicate that KRG represses oxidative stress-induced apoptosis by enhancing PI3K/Akt signaling via upregulation of ER-${\beta}$ expression.
