Contents of Total Mercury and Methylmercury in Deep-sea Fish, Tuna, Billfish and Fishery Products (심해성 어류, 다랑어류, 새치류 및 어류가공품의 총수은 및 메틸수은 함량)

  • The aim of this study was to determine total mercury and methyl mercury level in fishes (Deep-sea Fish, Tuna, Billfish) and fishery products. A total of 101 fishes and 44 fishery products samples collected from commercial market in Gyeonggi-do. Total mercury were analysed by mercury analyzer and methyl mercury were analyzed by gas chromatography with electron capture detector. In the fishes, total mercury was detected in all samples and methylmercury was detected in 92 samples of them. The detection rate of methylmercury was 91.1% in fishes. The mean concentration (mg/kg) of total mercury and methylmercury were $1.968{\pm}0.505/0.496{\pm}0.057$ for Billfish, $0.665{\pm}0.091/0.252{\pm}0.033$ for Deep-sea Fish and $0.577{\pm}0.085/0.218{\pm}0.025$ for Tuna, respectively. The Swordfish contains the highest level of total mercury (1.968 mg/kg) and methylmercury (0.496 mg/kg). In Mabled rockfish, the ratio of methylmercury's contents about total mercury's contents was the highest as 66.5%. In case of fishery products, frozens made of 100% of raw material contained the highest level of total mercury and methyl mercury. The weekly intake of total mercury and methylmercury was calculated in 4.72% and 5.24% of Provisional Tolerable Weekly Intake (PTWI) respectively. This study showed that the weekly intake of methylmercury from Deep-sea Fish, Tuna and Billfish was less than the PTWI recommended by the Joint FAO/WHO expert committee on food additives.

First Report on Isolation of Salmonella Enteritidis from Eggs at Grocery Stores in Korea

  • Salmonella Enteritidis is responsible for causing foodborne diseases upon consumption of egg products. While cases of S. Enteritidis isolation from eggs have been reported in other countries, no such cases have previously been reported in Korea. In this study, we report the first isolation and identification of S. Enteritidis from domestically distributed eggs in Korea. Eggs were collected from eight countrywide grocery stores during different seasons between 2011 and 2012. Egg contents and washing solution of egg shells were incubated in buffered peptone water, and the enriched broth was further enriched in tetrathionate broth and Rappaport-Vassiliadis. The secondary enriched broth was streaked on xylose lysine desoxycholate agar. The suspected colonies were confirmed to S. Enteritidis by a biochemical test, serotyping, and PCR test. Genetic relatedness among the isolates was analyzed using Diversilab Salmonella kit. Three strains of S. Enteritidis were isolated from egg contents and egg shells collected from grocery stores of the Eumseong-city in the fall of 2011. All three stains showed resistance to chloramphenicol, streptomycin, nalidixic acid, and ampicillin by the disk diffusion method. In addition, the isolates showed more than 99% DNA homology, indicating that they were presumably identical strains. Therefore, there is a requirement to monitor and control against S. Enteritidis from eggs in Korea.

Quantitative Determination of Ascaridole, Carvacrol and p-Cymene in the Biopesticides Products Derived from Chenopodium ambrosioides L. Extracts by Gas Chromatography

  • BACKGROUND: The commercial biopesticides containing Chenopodium ambrosioides L. extracts which have been registered as one of the ingredients of the commercial biopesticide by the organic agriculture materials, and have been widely used in Republic of Korea. However, the quantitative analysis method of the active substances for the commercial biopesticides containing C. ambrosioides L. extract has not been conducted.METHODS AND RESULTS: To analyze the quantitative analysis of ascaridole, carvacrol, and p-cymene as active substances of C. ambrosioides L. extract, hydrophilic lipophilic balance cartridge was used for solid phase extraction. The active substances were analyzed by the gas chromatography with flame ionization detector. The limit of quantitation values of ascaridole, carvacrol, and p-cymene were 10, 5, and 2 mg/L, and the recovery rates were 96.3, 84.0, and 82.5% in liquid products and 98.3, 99.1, and 97.3% in solid products, respectively. The total content of ascaridole, carvacrol, and p-cymene in the commercial biopesticides was ranged from 0.08 to 12.75%.CONCLUSION: From these results, this method was suitable for the quantitative analysis of the active substances of commercial biopesticides containing C. ambrosioides L. extract.

Molecular Characterization of an Isolate of Bean Common Mosaic Virus First Identified in Gardenia Using Metatranscriptome and Small RNA Sequencing

  • Gardenia (Gardenia jasminoides) is a popular and economically vital plant known for its ornamental and medicinal properties. Despite its widespread cultivation, there has been no documentation of plant viruses on gardenia yet. In the present study, gardenia leaves exhibiting symptoms of plant viral diseases were sampled and sequenced by both metatranscriptome and small RNA sequencing. As a consequence, bean common mosaic virus (BCMV) was identified in gardenia for the first time and named BCMV-gardenia. The full genome sequence of BCMV-gardenia is 10,054 nucleotides (nt) in length (excluding the poly (A) at the 3' termini), encoding a large polyprotein of 3,222 amino acids. Sequence analysis showed that the N-termini of the polyprotein encoded by BCMV-gardenia is less conserved when compared to other BCMV isolates, whereas the C-termini is the most conserved. Maximum likelihood phylogenetic analysis showed that BCMVgardenia was clustered closely with other BCMV isolates identified outside the leguminous plants. Our results indicated that the majority of BCMV-gardenia virus-derived small interfering RNAs (vsiRNAs) were 21 nt and 22 nt, with 21 nt being more abundant. The first nucleotide at the 5' termini of vsiRNAs derived from BCMV-gardenia preferred U and A. The ratio of vsiRNAs derived from sense (51.1%) and antisense (48.9%) strands is approaching, and the distribution of vsiRNAs along the viral genome is generally even, with some hot spots forming in local regions. Our findings could provide new insights into the diversity, evolution, and host expansion of BCMV and contribute to the prevention and treatment of this virus.

  • The vitalization of Agro-industrial complex could be an alternative to enhance farmers off-farm income and overcome FTA's impacts on the import of agricultural products. However, it is evaluated the governmental policy on the Agro-industrial complex has not been successfully done as expected. The Agro-industrial complex started to be composed in 1984 and the 427 Agro-industrial complex have been composed nationwide by 2011. Meanwhile, three Agro-industrial complex where 61 factories are running have been composed and two Agro-industrial complex are being under construction in Seocheongun as of 2011. It is investigated that there are constraints in the government supporting systems for Agro-industrial complex. They are; Aged and poor infrastructure facilities, Weak marketing competition, Scattered supporting agencies, and Poor loan system, etc. In this respect, the policy of supporting system was suggested and recommended to vitalize Agro-industrial complex in the study. They are; Better loan system, Improving aged facilities, Marketing support, Assisting technology, Suppling labor power, and Forming factory-network, etc.

The fecal microbiota composition of boar Duroc, Yorkshire, Landrace and Hampshire pigs

  • Objective: To investigate the effect of host genetics on gut microbial diversity, we performed a structural survey of the fecal microbiota of four purebred boar pig lines: Duroc, Landrace, Hampshire, and Yorkshire. Methods: The V3-V4 regions of the 16S rRNA genes were amplified and sequenced. Results: A total of 783 operational taxonomic units were shared by all breeds, whereas others were breed-specific. Firmicutes and Bacteroidetes dominated the majority of the fecal microbiota; Clostridia, Bacilli, and Bacteroidia were the major classes. Nine predominant genera were observed in all breeds and eight of them can produce short-chain fatty acids. Some bacteria can secrete cellulase to aid fiber digestion by the host. Butyric, isobutyric, valeric, and isovaleric acid levels were highest in Landrace pigs, whereas acetic and propionic acid were highest in the Hampshire breed. Heatmap was used to revealed breed-specific bacteria. Principal coordinate analysis of fecal bacteria revealed that the Landrace and Yorkshire breeds had high similarity and were clearly separated from the Duroc and Hampshire breeds. Conclusion: Overall, this study is the first time to compare the fecal microbiomes of four breeds of boar pig by high-throughput sequencing and to use Spearman's rank correlation to analyze competition and cooperation among the core bacteria.

A Study of Current Status on Pesticide Residues in Commercial Dried Agricultural Products (서울시 유통 건조농산물 중의 농약잔류 실태 연구)

  • The purpose of this study was to investigate 253 kinds of pesticide residues in 58 commercial dried agricultural products in Seoul. The determinations of the pesticide residues were performed using multiresidue methods and were carried out by a gas chromatography-nitrogen phosphorus detector (GC-NPD), an electron capture detector ($GC-{\mu}ECD$), a mass spectrometry detector (GC-MSD) and high performance liquid chromatography-ultraviolet detector (HPLC-UV), and a fluorescence detector (HPLC-FLD). The pesticide residue detection rate in the commercial dried agricultural products was 24.1% (14 of 58 samples). Twelve pesticide residues without maximum residue limits (MRLs) were detected. In the vegetable groups, the frequency of pesticide residues was found to be in the increasing order of dried fruiting vegetables > dried leafy vegetables > dried stalk and stem vegetables. The pesticides used on dried red pepper in the dried fruiting vegetables were varied (7 kinds) and numerous (4 of 8 samples). The pesticide types detected in the commercial dried agricultural products were in the order of pyrethroid > organochloride > organophosphorus and insecticide > fungicide > herbicide ${\cdot}$ nematicide. The primary pyrethroid pesticide detected was cypermethrin. According to the producing areas of products, large numbers of pesticide residues were found in the order of Korea, China, North Korea, USA, and Vietnam.

  • The purpose of this study is to provide basic information on exporter's coordination methods from the attitude of agro-food exporters. The implications are; Firstly, most of exporters believe that the agro-food export raises farmer's competitiveness rather than it does the farmhouse's income increase. Secondly, The stabilization of the farm-gain income by exporting agricultural products are most important in order to reinforce the cohesion between farmers and exporters. Thirdly, the exporters intend to participate in establishing and operating a large-scale producers' organization and marketing company under export polices. So, it is necessary that the government drive to coordinate the exporters and producers.

The Survey on Pesticide Residues in Vegetables Collected In Seoul (서울지역 유통 채소류의 잔류농약 조사)

  • The characteristics of pesticide residues by multiresidue analysis method using GC, HPLC and GC-MSD were examined for 18,069 numbers of 91 kinds of vegetables in Seoul from 2007 to 2009. Detection rates of pesticide residues were 11.2% in 2007, 8.5% in 2008 and 12.0% in 2009, respectively, and the excess rates of Maximum Residue Limits (MRLs) showed a declining tendency as 4.6% in 2007, 2.8% in 2008 and 2.1% in 2009, respectively. The pesticides exceeding MRLs were 43 ingredients in 2007, 30 ingredients in 2008 and 36 ingredients in 2009, respectively. The frequencies of pesticides exceeding MRLs were 71 of azoxystrobin in 2007, 29 of procymidone in 2008, 24 of endosulfan in 2009, respectively. The excess rate of mustard green for MRLs in comparison with sample numbers was the highest among vegetables analyzed more than 10 cases each year. As a result of comparing annually pesticide residues about pesticide ingredients showed high frequency rates for pesticide detection, there was a statistical significance for the detection quantities of diazinon, endosulfan, paclobutrazol and procymidone.

Global Trade Networks of Agro-Food and the Status and Prospects in Korea (농식품 무역의 글로벌 네트워크와 한국의 위상)

  • In this paper, using trade data of 57 HS 4 digit based agricultural and food(agro-food) products among 47 countries during 2005 to 2014, the international competitiveness and trade structure have been analyzed from the context of global networks employing the methods of social network analysis. Firstly, the differences in the network structure by agricultural products have been revealed. The number of disconnected groups was significantly lager in order of vegetables, fruits and processed foods. Secondly, the differences in the community structure by agricultural and food products have been also revealed. That is to say, for some commodities, the community structure has been changed dynamically, on the other hand, there are some agricultural products that have not changed its community structure despite the increasing trends of trade volume. Thirdly, even though the international competitiveness of Korea's agricultural products was still very limited in the sense that only 26 items have been included in the top3 network of 57 agricultural products, there has been possibilities of the increasing patterns of the competitiveness.
