Effects of adaptation time and inclusion level of sugar beet pulp on nutrient digestibility and evaluation of ileal amino acid digestibility in pigs

  • Objective: Two experiments were conducted to determine the effects of adaptation time and inclusion level of sugar beet pulp (SBP) on nutrient digestibility and to evaluate the ileal amino acid digestibility of SBP fed to pigs. Methods: In Exp. 1, thirty-six crossbred barrows ($85.0{\pm}2.1kg$) were allotted to 6 diets in a completely randomized design with six replicates per diet. Diets included a corn-soybean meal diet and 5 test diets containing 14.6%, 24.4%, 34.2%, 43.9%, or 53.7% SBP, respectively. The adaptation time consisted 7, 14, 21, or 28 d consecutively for each pig followed by 5 d for fecal collection. Feces were collected from d 8 to 13, d 15 to 20, d 22 to 27, and d 29 to 34, respectively. In Exp. 2, six pigs ($35.1{\pm}1.7kg$) with T-cannulas at the terminal ileum were fed to 3 diets in a replicated $3{\times}3$ Latin square design with 3 periods and 2 replicate pigs per diet. Each period consisted 5 d for diet adaptation followed by 2 d for digesta collection. Results: The digestible energy (DE) value and the apparent total tract digestibility (ATTD) of gross energy (GE), dry matter (DM), ash, and organic matter in diets linearly decreased (p<0.05) as the adaptation time increased or as the dietary SBP increased, while the ATTD of neutral detergent fibre and acid detergent fibre in diets linearly increased (p<0.01) as the dietary SBP increased. The DE value and the ATTD of GE and crude protein (CP) in SBP linearly increased (p<0.05) as the adaptation time increased, while the ATTD of CP in SBP linearly decreased (p<0.01) as the inclusion level increased. The standardized ileal digestibility of Lys, Met, Thr, and Trp in SBP was 37.03%, 51.62%, 40.68%, and 46.22%, respectively. Conclusion: The results of this study indicated that the ATTD of energy and nutrients were decreased as inclusion rate of SBP increased.

Effects of inclusion level and adaptation period on nutrient digestibility and digestible energy of wheat bran in growing-finishing pigs

  • Objective: This experiment was to evaluate the effect of different inclusion levels and adaptation periods on digestible energy (DE) and the apparent total tract digestibility (ATTD) of chemical constituents in diets supplemented with wheat bran. Methods: Thirty-six crossbred barrows with an initial body weight of $85.0{\pm}2.1kg$ were allotted to 6 diets in a completely randomized block design with 6 pigs per diet. Diets included a cornsoybean basal diet and 5 additional diets which were formulated by replacing corn and soybean meal in control diet with 15%, 25%, 35%, 45%, or 55% wheat bran. The experiment lasted for 34 d, and feces were collected from d 8 to 13, 15 to 20, 22 to 27, and 29 to 34 respectively. Results: The results showed no interaction effects between inclusion level and adaptation period on the concentration of DE and the ATTD of gross energy (GE) and crude protein (CP) in wheat bran. The DE value and ATTD of GE in wheat bran decreased (p<0.05) significantly as the inclusion level of wheat bran increased, but no difference in the ATTD of CP was observed. The ATTD of CP in wheat bran increased (p<0.10) significantly as the adaptation period for pigs was prolonged. In addition, the concentration of DE and the ATTD of GE in wheat bran decreased linearly (p<0.05) when pigs were fed either an increased level of wheat bran or given an increased adaptation period to the diets. Conclusion: Wheat bran showed a negative effect on the concentration of DE and ATTD of GE and CP as the inclusion level increased. A longer adaptation period can gradually increase the DE value and ATTD of GE and CP in wheat bran, and at least 14 to 21 d of adaptation might be recommended for growing-finishing pigs fed the high-fiber diets with wheat bran.

동부 르완다 쌀 농업인의 기후변화에 대한 적응 방법 결정 요인 (Determinant Factors of Rice Farmers' Selection of Adaptation Methods to Climate Change in Eastern Rwanda)

  • The negative impact of climate change on the agricultural sector is rapidly increasing, and it is urgent to prepare policies at the government level to mitigate it. In the case of Rwanda's agricultural sector, which lacks the government's budget and farmers' capital, efficient and effective policy implementation is of paramount importance. To this end, rather than establishing related policies in the public sector from the top down, it is necessary to establish a bottom-up customized policy that is reflected in policy establishment by identifying the characteristics and behaviors of farmers who actually participate in adaptation activities. In this study, the effects of farmers' characteristics and farmers' perception status/adaptation status to climate change on the selection of adaptation methods for climate change were analyzed. 357 rice farmers randomly selected from Eastern Rwanda were surveyed to explore the information related to farmers' perception to climate change and adaptation methods as well as basic information of the farm. Research shows that the probability of selecting a variety of adaptation methods rather than not responding to climate change increases the younger the age, the higher the education level, and the easier access to climate information and credit. As a policy proposals, it is judged that public support such as strengthening agricultural technology support services, including more detailed guidance for elderly and low-educated farmers, and improving access to farm loan services by agricultural financial institutions is needed. In addition, it is necessary to adjust the planting time and cultivation method, provide timely information related to climate change, and provide crop variety improvement services to farmers.

Stress Tolerance of Bifidobacterium infantis ATCC 27920 to Mild-heat Adaptation

  • Two-dimensional gel electrophoresis (2-DE) was employed to assess the thermo-tolerance characteristics of Bifrdobacterium infantis ATCC 27920 to mild heat adaptation. When exposed to various heat levels, pH, and hydrogen peroxide ($H_2O_2$) stress conditions, B. infantis ATCC 27920 exhibited high level of stress resistance. Under mild-heat treatment ($46^{\circ}C$), no significant change in viability level was observed after 2 hr. Interestingly, improved viability was observed in mild-heat adapted ($46^{\circ}C$ for 1 hr) cultures exposed to $55^{\circ}C$, in comparison to control experiments. Viability was not affected by pH, bile, and $H_2O_2$ stress conditions. 2-DE analysis revealed those mild-heat adaptation up-regulated 4 proteins and down-regulated 3 proteins. Among these protein spots, isopropyhnalate dehydratase (leuD), glycosyltransferase (glgA), and ribosomal protein L5 (rp1E) were identified by matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALD1-TOF/MS).

Comparative analysis of liver transcriptome reveals adaptive responses to hypoxia environmental condition in Tibetan chicken

  • Objective: Tibetan chickens, which have unique adaptations to extreme high-altitude environments, exhibit phenotypic and physiological characteristics that are distinct from those of lowland chickens. However, the mechanisms underlying hypoxic adaptation in the liver of chickens remain unknown. Methods: RNA-sequencing (RNA-Seq) technology was used to assess the differentially expressed genes (DEGs) involved in hypoxia adaptation in highland chickens (native Tibetan chicken [HT]) and lowland chickens (Langshan chicken [LS], Beijing You chicken [BJ], Qingyuan Partridge chicken [QY], and Chahua chicken [CH]). Results: A total of 352 co-DEGs were specifically screened between HT and four native lowland chicken breeds. Gene ontology and Kyoto encyclopedia of genes and genomes enrichment analyses indicated that these co-DEGs were widely involved in lipid metabolism processes, such as the peroxisome proliferator-activated receptors (PPAR) signaling pathway, fatty acid degradation, fatty acid metabolism and fatty acid biosynthesis. To further determine the relationship from the 352 co-DEGs, protein-protein interaction network was carried out and identified eight genes (ACSL1, CPT1A, ACOX1, PPARC1A, SCD, ACSBG2, ACACA, and FASN) as the potential regulating genes that are responsible for the altitude difference between the HT and other four lowland chicken breeds. Conclusion: This study provides novel insights into the molecular mechanisms regulating hypoxia adaptation via lipid metabolism in Tibetan chickens and other highland animals.

Effects of corn particle size on energy and nutrient digestibility in diets fed to young pigs and adult sows

  • Objective: This research was carried out to investigate the effects of corn particle size on the apparent total tract digestibility (ATTD) of energy and nutrients fed to pigs at four different growth stages and therefore to provide basis for better application of corn in pig feeds. Methods: Eighteen weanling piglets, 18 growing barrows, 24 gestating sows and 24 lactating sows were used in this study. Within each stage, pigs were allotted to 1 of 3 or 4 corn-soybean meal diets which were formulated with different corn particle size in a completely randomized design with 6 replicate pigs per diet. Each stage lasted for 19 days, including 7 days for cages adaptation, 7 days for adaptation to diets and followed by 5 days for total collection of feces and urine. Results: For nursery and growing stages, the results showed that digestible energy content and ATTD of gross energy (GE), dry matter (DM), neutral detergent fiber (NDF), and acid detergent fiber (ADF) was increased (p<0.05) as the corn particle size reduced. Meanwhile, the metabolizable energy content and ATTD of crude protein (CP) tended to increase. For gestating sows, no differences were found in the ATTD of nutrients among dietary treatments. As for lactating sows, there were linear and quadratic increases (p<0.05) in the ATTD of DM, GE, NDF as the corn being finer milled. Quadratic response in ATTD of ADF and CP (p<0.05) were observed as sows fed with four different diets. Conclusion: Reducing corn particle size can increase digestibility of nutrients fed to young pigs and lactating sows. No effects were observed in present experiment when gestating sows were fed with different particle sized corn.

Differential Embryo Development among Tibetan Chicken, DRW and Shouguang Chicken Exposed to Chronic Hypoxia

  • Avian embryos at high altitude are independent of maternal protection against hypoxia, which is contrary to mammals. It is well known that chronic hypoxic exposure at key points can significantly impact on avian development. Tibetan Chicken, a Chinese indigenous breed, living in Tibetan areas with an altitude of 2.2 to 4.1 thousand meters, has an adaptive mechanism to hypoxia. In the present study, fertilized eggs of Tibetan Chicken were incubated under 13% and 21% oxygen concentration. Two lowland chicken breeds, Shouguang Chicken, an indigenous chicken breed in Shandong Province of China, and Dwarf Recessive White Chicken, an imported breed in Beijing, were used as control groups. The embryo mass and some organs such as brain, heart, liver, stomach and eye weight in the three species were measured at Hamburger-Hamilton stage 39, 41, 43 and 45 under hypoxic and normal conditions. The results showed that in hypoxia Tibetan Chicken significantly differed from the two lowland chicken breeds in embryo mass at Hamburger-Hamilton stage 41, 43 and 45 (p<0.01). In particular, Dwarf Recessive White Chicken and Shouguang Chicken showed retarded growth in hypoxic incubation (p<0.01), whereas Tibetan Chicken showed no significant difference between hypoxic and normal conditions (p>0.05). In addition, heart and the other organs showed different susceptibility to hypoxia at the studied stages. In conclusion, chronic hypoxia induced a change in the embryo development of the three different species and Tibetan Chicken showed adaptation to hypoxia. Of note, the embryo developmental physiology of Tibetan Chicken in response to hypoxia will shed light on the process of physiological acclimation or evolutionary adaptation as well as the study of clinical disease.

Complete genome sequencing and comparative genomic analysis of Lactobacillus acidophilus C5 as a potential canine probiotics

  • Lactobacillus acidophilus is a gram-positive, microaerophilic, and acidophilic bacterial species. L. acidophilus strains in the gastrointestinal tracts of humans and other animals have been profiled, but strains found in the canine gut have not been studied yet. Our study helps in understanding the genetic features of the L. acidophilus C5 strain found in the canine gut, determining its adaptive features evolved to survive in the canine gut environment, and in elucidating its probiotic functions. To examine the canine L. acidophilus C5 genome, we isolated the C5 strain from a Korean dog and sequenced it using PacBio SMRT sequencing technology. A comparative genomic approach was used to assess genetic relationships between C5 and six other strains and study the distinguishing features related to different hosts. We found that most genes in the C5 strain were related to carbohydrate transport and metabolism. The pan-genome of seven L. acidophilus strains contained 2,254 gene families, and the core genome contained 1,726 gene families. The phylogenetic tree of the core genes in the canine L. acidophilus C5 strain was very close to that of two strains (DSM20079 and NCFM) from humans. We identified 30 evolutionarily accelerated genes in the L. acidophilus C5 strain in the ratio of non-synonymous to synonymous substitutions (dN/dS) analysis. Five of these thirty genes were associated with carbohydrate transport and metabolism. This study provides insights into genetic features and adaptations of the L. acidophilus C5 strain to survive the canine intestinal environment. It also suggests that the evolution of the L. acidophilus genome is closely related to the host's evolutionary adaptation process.

Metabolomics reveals potential plateau adaptability by regulating inflammatory response and oxidative stress-related metabolism and energy metabolism pathways in yak

  • Species are facing strong selection pressures to adapt to inhospitable high-altitude environments. Yaks are a valuable species and an iconic symbol of the Qinghai-Tibet Plateau. Extensive studies of high-altitude adaptation have been conducted, but few have focused on metabolism. In the present study, we determined the differences in the serum metabolomics between yaks and the closely related species of low-altitude yellow cattle and dairy cows. We generated high-quality metabolite profiling data for 36 samples derived from the three species, and a clear separation trend was obtained between yaks and the other animals from principal component analysis. In addition, we identified a total of 63 differentially expressed metabolites among the three species. Functional analysis revealed that differentially expressed metabolites were related to the innate immune activation, oxidative stress-related metabolism, and energy metabolism in yaks, which indicates the important roles of metabolites in high-altitude adaptation in yaks. The results provide new insights into the mechanism of adaptation or acclimatization to high-altitude environments in yaks and hypoxia-related diseases in humans.

Effect of culturing media on biocontrol ability and physiological state of Burkholderia gladioli strain B543.

  • Long-term repeated culturing of biocontrol agents on a certain medium often results in reduced biocontrol efficacy and altered physiology. Effect of culturing media on biocontrol ability and physiological state of Burkholderia gladioli strain B543 was investigated. Over 20 times repeated cultivation of B. giadioli strain B543 on Kings B medium or nutrient agar medium showed improved biological control of cucumber damping-off caused by Pythium ultimum, while one time cultivation on KB or NA did not. The repeated cultivation also induced the physiological changes of the biocontrol agent such as antifungal activity and the production of protease and siderophore. Our result indicates that adaptation to proper culturing medium can alter biocontrol ability and must consider in optimizing the use of biocontrol agents.
