Fruit-body Production of Ganoderma neo-japonicum by Sawdust Cultivation (톱밥재배에 의한 자흑색불로초(Ganoderma neo-japonicum)의 자실체 발생)

  • Ganoderma neo-japonicum, which is also known as black lingshi mushroom and medicinal mushroom. Present experiments were conducted to determine the possibility of artificial culture with oak sawdust of G. neo-japonicum. The duration of mycelial growth and days of pinhead formation of oak sawdust bag (2.4 kg) were 28~35 days and 25~29 days, respectively. The yield of mushroom fresh fruitbody was 135~157 g.

The Effect of Purchasing Factors of Environment-Friendly Agricultural Products on Consumer Attitude and Purchasing Intention (친환경농산물의 구매요인이 소비자태도와 구매의도에 미치는 영향)

  • The study tested the moderating effect of 215 women living in Busan to analyze the purchase intention (awareness, health interests, environmental interests) of environment-friendly agricultural products and it's the causal relationship between consumer attitudes and purchasing intention. The results confirmed that all purchase factors for environment-friendly agricultural products had significant causality on consumer attitudes. Significant causality of consumer attitudes on purchase intention was also confirmed. The moderating effect of the number of purchases was analyzed to only significantly affect consumer attitudes on purchase intention and consumer attitudes according to number of purchases were analyzed to vary depending on the intensity of purchase. Information presented in this study means that consumers who are aware of environment-friendly agricultural products, interested in health, and interested in protecting the environment exhibit positive attitudes on environment-friendly agricultural products as well as higher purchase intentions. Therefore, if government and local organizations strengthen the promotion of environment-friendly agricultural products, there may be an acceleration of the consumption of environment-friendly agricultural products, followed by an increase in the income of farmers as well as an increase in the number of farmers who cultivate environment-friendly agricultural products. This may ultimately aid in the conservation of the natural environment of Korea and enhance the health and well-being of the public.

Usability Evaluation Procedure for the Agricultural Equipments in Local Agricultural Task Environment

  • Objective: to improve agricultural working environment using tools and equipments. Background: most existing tools currently used, in fact, are discrepant from human factor guidelines so that they are still exposed to the musculoskeletal disease. Method: the provided tools and equipments are mostly designed considering human factor engineering or usability engineering in a way to avoid musculoskeletal disease. Results: propose the top-down procedure and usability engineering steps to evaluate them for the system design suitability. Conclusion: apply them conveniently into rural environment to help farmers working environment.

Induction of pathogenicity mutants from Elsinoe fawcettii, the causal fungus of citrus scab by genetic transformation

  • Genetic transformation carried out to induce the pathogenicity mutants from the two isolates, Elsinoe fawcettii R-34 and MUD of citrus scab fungus to hygromycin resistant by transferring plasmides (pUCATPH) that contain hygB gene. We produced protoplast for transformation by using of combinations of available enzymes including ${\beta}$-D-glucanase, ${\beta}$ -glucuronidase, Iyticase and driselase. The protoplasts regenerated at 64 $\mu\textrm{g}$/ml of hygromycin B but not 128 $\mu\textrm{g}$ in sensitivity test to identify the concentration of useful marker for the selection of transformants. Approximately 1200 and 67 hygromycin resistant isolates from strain R-34 and strain MUD, respectively, were isolated on PDA added with 200 $\mu\textrm{g}$ /ml of hygromycun B. Fifty seven and 4 of hygromycin resistant isolates from strain R-34 and MUD, respectively, did not produce necrotic lesions on the leaf in detached-leaf assay. Finally, 9 isolates were isolated from strain R-34, and these Isolates produced non or very few symptoms on seedlings of citrus in greenhouse pathogenicity test. And it's very interesting that some isolates produced melanose-like symptom on very young leaves which it was not typical symptom and somtimes produced on only expanded leaf.

Cultural Characteristics and Fruitbody Formation of Phellinus gilvus (Phellinus gilvus의 배양적 특성과 자실체 형성)

  • For artificial cultivation of Phellinus gilvus (Schw.) Pat we have conducted a study on cultural characteristics and condition of fruitbody formation. The optimum temperature was about $30^{\circ}C$ at pH $6.0{\sim}7.0$ for mycelial growth. Optimum sawdust media were oak sawdust+willow sawdust(5:5, V/V), oak sawdust+willow sawdust+rice bran (4.5:4.5:1, V/V) and oak sawdust+pine sawdust+rice bran(4.5:4.5:1, V/V) and, the spawn incubation period was about $25{\sim}26$ days. Mycelial growth in the inner portion of oak log was 200 mm after 60 days and duration for the first fruitbody primordia were formed about 90 days after inoculation.
