Tenderness Survey of Branded Hanwoo Beef - 2007: Assessment of Warner-Bratzler Shear for Hanwoo Beef by Quality Grade and Subprimal Cuts (브랜드 한우고기 연도 조사 - 2007 : 육질등급 및 소분할 부위별 전단력 평가)

  • Eight subprimal cuts purchased from the branded Hanwoo beef of 3 quality grades ($1^{++}$, $1^+$, 1) at 13 stores were evaluated the tenderness using Warner-Bratzler shear (WBS). The beef frequency ratio (%) depending on postmortem aging periods were investigated. The 37.5 (quality grade $1^{++}$), 45.8 (quality grade $1^+$), and 36.4% (quality grade $1^{++}$) of branded beef were aged for < 7 d, 26.6 (quality grade $1^{++}$), 47.2 (quality grade $1^+$) and 36.4% (quality grade 1) were aged for $7{\sim}13$ d, and 31.3 (quality grade $1^+$), 4.2 (quality grade $1^+$) and 25.8% (quality grade $1^+$) were aged for 14 to 20 d. The temperature of cold room in stores was ranged average 2.27 to $2.42^{\circ}C$. WBS values for ansimsal (tenderloin), witdngsimsal (ribeye), cheggtsal (shortloin), doganisal (knuckle) and moongchisatae (hind shank) from branded Hanwoo beef of quality grade $1^{++}$ were tender than those from branded Hanwoo beef of quality grade 1 (p<0.05). WBS values for ansimsal (tenderloin) were 2.56 (quality grade $1^{++}$), 2.76 (quality grade $1^+$) and 3.10 kg (quality grade 1), respectively, and those for doganisal (knuckle, quality grade $1^{++}$), hongdukesal (eye of round, quality grade $1^+$) and bosupsal (top sirloin, quality grade 1) were 4.76, 4.96 and 5.66kg, respectively (p<0.05). The frequency ratio (%) of WBS < 3.9 kg in the all subprimal cuts from branded Hanwoo beef of quality grade $1^{++}$ were 100 [ansimsal (tenderloin) and cheggtsal (shortloin)], 87.5 [witdngsimsal (ribeye)] and 62.5% [bosupsal (top sirloin)], whereas that of WBS > 4.6 kg were 50.0% [hongdukesal (eye of round) and doganisal (knuckle)]. The frequency ratio of WBS < 3.9 kg in the an subprimal cuts of quality grade $1^+$ were 100 [ansimsal (tenderloin) and witdngsimsal (ribeye)] and 44.4% [cheggtsal (shortloin) and gurisal (chuck tender)], whereas that of WBS > 4.6 kg were 66.7 [hongdukesal (eye of round)], 55.6 [doganisal (knuckle)] and 44.4% [bosupsal (top sirloin)]. The frequency ratio (%) of WBS < 3.9 kg in the all subprimal cuts of quality grade 1 were 88.9 [ansimsal (tenderloin)], 62.5 [cheggtsal (shortloin)] and 44.4% [witdngsimsal (ribeye)], whereas that of WBS > 4.6 kg were 100.0 [doganisal (knuckle)] 62.7 [hongdukesal (eye of round)], 62.5 [gurisal (chuck tender)] and 55.6% [moongchisatae (hind shank)]. From these results, subprimal cuts from branded Hanwoo beef were marketed with short aging periods and high frequency ratio (%) of WBS > 4.6 kg.

  • Experiments were performed to determine age-associated changes in ornithine decarboxylase (ODC) gene and Ha-ras cellular oncogene expression in tissues of female rats. In the kidney, ODC mRNA levels did not show age-associated changes, while ODC enzyme activities were decreased with advancing age from 3 to 10 months. These results suggest that post-transcriptional mechanism (s) are involved in the age-dependent decrease in renal ODC enzyme activity. In addition, we found no correlation between testosterone-induced renal ODC expression and DNA methylation pattern. Ha-ras mRNA levels in brain decreased as animals aged from 3 to 6 months, while renal Ha-ras mRNA levels were not influenced by age. Results demonstrate the age-dependent expression of Ha-ras in a tissue-specific manner.

A Study on the Factors Affecting Bone Mineral Density in Adult Women - Based on the Mothers of Elementary School Students- (일부 초등학생의 어머니를 대상으로 한 성인 여성의 골밀도에 영향을 미치는 요인에 관한 연구)

  • The bone mineral density(vertebrae Ll-4, femoral neck, Ward's triangle and trochanter) of 160 healthy adult women aged 29-45 was measured and general characteristics, anthropomentric measurements, and dietary intake were also studied to determine the main factors affecting the bone mineral density(BMD) of adult women. The nutrient intake of the subjects was adequate to RDA level except energy, Ca, Fe, and vit A. The BMD of vertebrae L24 showed significant positive association with nutrient intake such as animal protein, P, Ca(animal) and Fe(animal). The BMD of the femoral neck correlated significantly with anthropometric measurements such as height, weight, waist circumference, hip circumference, body fat and body mass index. The BMD of both sites(vertebrae L24 and femoral neck) were significantly related to vertebrae L24 was daily Ca intake and that of femoral neck was daily energy expenditure. In conclusion, adequate nutrient intake, especially protein, P, Ca and Fe as well as increases in physical activity were suggested to prevent the loss of bone mineral density in adult women.

  • One hundred and twenty indigenous pigeons of either sexes of different age groups were sacrificed in different seasons to assess the interactions of sex, age and seasons on the proximate composition and gross energy values of fresh boneless meat. The dry matter and protein contents decreased but the fat, ash and energy contents increased linearly as the birds aged. Seasons and age had significant influences on either of these two performance traits. Meat of growing pigeons of both sexes was found to contain high protein and low fat throughout the year.

Effects of dry aging on physicochemical properties of beef cattle loins (건조숙성에 따른 육우 등심의 이화학적 특성에 미치는 영향)

  • This study investigated the quality change in dry aged beef cattle loins (temperature: $3{\pm}1$, lative humidity: $75{\pm}5%$, period: 14 days), compared to non-aged beef cattle loins (control). The water content of dry aged loins was lower than that of the control (p<0.001). The fat content, protein content, pH value, and redness in dry aged loins were higher than in the control. Moreover, dry aging significantly improved the water holding capacity and cooking yield of beef cattle loin (p<0.001). The shear force of dry aged loins was significantly lower than in the control (p<0.01). The flavor, tenderness, juiciness, and overall acceptability of dry aged loins were higher than in the control according to sensory evaluation. Therefore, dry aging can improve the quality properties of beef cattle loins.

Nutrition survey of the Aged on Jeju Island (제주지역 고령자 영양실태 조사연구)

  • The purpose of this survey was to investigate the nutrient and food intake and food habits of the aged Jeju island. The survey was conducted to 102 persons, 50 persons aged 65 to 74 years and 52 persons aged 75 years over from July 5 to 25, 1981. The result obtained was summarized as follows; 1. Average nutrient intake per day Calorie intake was 1408 ㎉(which was 64% of RDA) for male and was 1343 ㎉ (84%) for female aged 65 to 74 years. For male aged 75 years over calorie intake was 1316㎉ (60%) and for female was 1292㎉ (81%). According to the intake of calorie, the ratio of carbohydrate, protein, fat, were average 74.8%, 15.1%, 10.1%. Protein intake was insufficient quantiatively, but the proportion of animal protein to total protein intake were 26.1 to 33.5%. Other nutrient intake, such as Ca, vitamin A and C were lower than the recommended dietary allowance. 2. Food intake In per capita food consumption were 69.1g to 744.7g and average 320.1g of cereal (125.1g of rice and 160.5g of barley) and 63.9g of fishes. Total food consumption and vegetables were diminished by older age. 3. Correlation assessment The correlation coefficient between residental stus lived with family, high activity, good health, and nutrient intake were significant (P<0.05). Poor teeth had influence on less nutrient intake, but was not significant. 4. Food preferences Food liked over 70% of subjects were persimmon, fishes, leef, tomato, and boiled rice with barley, etc. owing to tender food, economical reasons, seasonal influence, and food habits. Food disliked over 20% of subjects were instant noodle, small sardin, boiled rice, and ice-crea, etc. The majority of subjects had never had the processing food, such as ham, sausage, butter and margarine, etc.

  • CCAAT/enhancer binding proteins(C/EBP) are a group of transcription factors expressed during preadipocyte differentiation. In the C/EBPs, C/EBPa plays an important role in lipid deposition and adipocyte differentiation. In this studies, we report the identification, characterization, and expression of a Hanwoo CIEBP$\alpha$ The Hanwoo C/EBP$\alpha$DNA includes a 1059 bp open reading frame encoding a protein of 353 amino acids. The CIEBPa amino acid sequences of the Hanwoo show strong conservation with the corresponding sequences reported in other species. The distribution of C/EBP$\alpha$ mRNA in various tissues of Hanwoo aged 12 months were investigated using Northern blotting analysis. The highest expression was detected in adipose tissue and more lower expression was detected in colon and lung. We also identified expression of C/EBPa mRNA in Hanwoo sirloin and adipose tissue aged 12, 26, and 30 months by real-time RT-PCR. The higest expression were detected at 26 months in the sirloin and at 12 and 26 months in the adipose tissue.

Ginsenosides attenuate the 3-nitropropioic acid-induced rat striatal degeneration in an age-dependent manner

  • The number of reporting the effects on ginseng's physiological, pharmacological, and behavioral effects has been increased every year. Major active components of Panax ginseng, are the ginsenosides, which are mainly triterpenoid dammarane derivatives. 3-Nitropropionic acid (3-NP) is blown to induce cellular energy deficit and oxidative stress related neurotoxicity via an irreversible inhibition of the mitochondrial enzyme succinate dehydrogenase (SDH). Intraperitoneal injection of 3-NP produces striatal degeneration. Aged animals was more vulnerable to 3-NP than young animal. We used three different ages of 5-, 8-, and 26-week-old rats. 3-NP alone treatment induced striatal lesion and increased lesion volume with age-dependent manner in 5-, 8-, and 26-week-old rats by $30.2{\pm}5.8$, $v$, and $51.3{\pm}8.4mm^3$, respectively. However, pretreatment of GTS (100 mg/kg/day) before 3-NP reduced striatal lesion in 5-,8-, and 26-week-old rats by $3.15{\pm}6.1$, $8.89{\pm}1.9$, and $27.3{\pm}5.6mm^3$, respectively. Pretreatment of GTS also significantly increased survival rate in 5-week-old rats (3-NP alone: GTS +3-NP = $40.4{\pm}6.3$: $72.5{\pm}9.5\%$) than 8-week-old rats (3-NP alone: GTS + 3-NP : $13.5{\pm}5.2\%$ : $45.1{\pm}3.1\%$). In 26-week-old rats, 3-NP alone treated group died on day 18, whereas GTS +3-NP-treated group prolonged lifespan to 30 days. Thus, pretreatment of GTS before administration of 3-NP extended lifespan in all ages. The present results indicate that aged animals are more vulnerable to 3-NP and GTS pretreatment protected 3-NP-induced striatal damage in different ages of animals.

Marination and Physicochemical Characteristics of Vacuum-aged Duck Breast Meat

  • We investigated marinade absorption and physicochemical characteristics of vacuum-aged duck breasts that were halved and individually vacuum-packed for chiller aging at $4^{\circ}C$ for 14 d. One half was marinated for 0, 7, or 14 d, while the second half was used as a control. Marinade absorption, cooking loss, cooking yield, texture profile, pH, color, protein solubility, and thiobarbituric acid reactive substances (TBARS) values were evaluated, and protein sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) was performed. Marinade absorption and pH did not vary significantly after 14 d of aging. Marination increased the pH, color ($a^*$ and $b^*$) values, and cooking yield and reduced cooking loss. TBARS values significantly increased with aging time, but were significantly reduced by marination. Myofibril and total protein solubility increased with aging and marination, while SDS-PAGE showed protein degradation. Hence, aging and marination can be used simultaneously to improve physicochemical quality and cooking yield of vacuum-aged duck breast.

Age Prediction in the Chickens Using Telomere Quantity by Quantitative Fluorescence In situ Hybridization Technique

  • Telomeres are special structures at the ends of eukaryotic chromosomes. Vertebrate telomeres consist of tandem repeats of conserved TTAGGG sequence and associated proteins. Birds are interesting models for molecular studies on aging and cellular senescence because of their slow aging rates and longer life spans for their body size. In this longitudinal study, we explored the possibility of using telomeres as an age-marker to predict age in Single Comb White Leghorn layer chickens. We quantified the relative amount of telomeric DNA in isolated peripheral blood lymphocytes by the Quantitative Fluorescence in situ Hybridization technique on interphase nuclei (IQ FISH) using telomere-specific DNA probes. We found that the amount of telomeric DNA (ATD) reduced significantly with an increase in chronological age of the chicken. Especially, the telomere shortening rates are greatly increased in growing individuals compared to laying and old-aged individuals. Therefore, using the ATD values obtained by IQ FISH we established the possibility of age prediction in chickens based on the telomere theory of aging. By regression analysis of the ATD values at each age interval, we formulated an equation to predict the age of chickens. In conclusion, the telomeric DNA values by IQ FISH analyses can be used as an effective age-marker in predicting the chronological age of chickens. The study has implications in the breeding and population genetics of poultry, especially the reproductive potential.
