버섯균사체 배양물로부터 면역증진 기능성 소재 개발

  • This study relates to low and medium molecular weight isoflavone-${\beta}$-D-glucan produced by submerged liquid culture of Agaricus blazei, a method of producing the isoflavone-B-D-glucan using autolysis enzyme of Agaricus blazei mycelia, and use of the isoflavone-B-D-glucan for anti-cancer and immunoenhancing effect. In acordance with one aspect of the present study, it deals with a method of producing isoflavone-${\beta}$-D-glucan, which comprises the followings; 1) culturing and separating mushroom mycelia, 2) producing low-medium molecular weight isoflavone-${\beta}$-D-glucan from high molecular weight one. The cytotoxicity on human cnacer cell line (Caco-2, MCF-7), the expression of Cyclin D, Bcl-2, Bax protein, p21 protein, p53 protein in MCF-7 cells assessed by SDS-PAGE and immunoblotting, and other immuno related factor such as TNF-a and IL-1B activities were examined. Structural identification of isoflavone-${\beta}$-D-glucan which shoed cytotoxicity against cancer cell and immunoenhancing effects was carried by separation with DEAE-cellulose column chromatography, TLC, HPLC, IR, NMR, Clinical test for the cancer patients (n=119) for 6 month was carried out, and immunoenhancing factors(NK cell number, ratio of T4/T8) were checked. We concluded the identified isoflavone-${\beta}$-D-glucan has immuno enhancing effects and could be useful for cancer chemoprevention.

Characteristics of a new Agaricus bisporus cultivar 'Jinhyang' (양송이 신품종 '진향'의 특성)

  • The main characteristics of a new cultivar 'Jinhyang', a cross of a collection of monokaryotic strains of the brown button mushroom, were examined. 'Jinhyang' was not significantly different from the control cultivar 'Dahyang' in mycelial culture and cultivation period. The cap of 'Jinhyang' was thicker and darker in color than that of 'Dahyang'. The hardness and individual weight of the fruiting body of 'Jinhyang' were slightly higher than those of 'Dahyang'; however, the differences were not significant. The yield of 'Jinhyang' was 14.1 kg/m2, which was 8% higher than that of 'Dahyang'. Therefore, it is expected that it will be possible to supply this cultivar growers in the future.

Plant-growth promoting traits of bacterial strains isolated from button mushroom (Agaricus bisporus) media

  • A diverse group of plant-growth promoting bacteria were isolated in button mushroom (Agaricus bisporus) media to investigate the plant-growth promoting traits of compounds including indole acetic acid (IAA), ammonia, 1-aminocyclopropane-1-carboxylic acid deaminase, siderophore, and hydrogen cyanide. Twenty-one bacterial strains showing positive effects for all the test traits were selected and classified to confirm bacterial diversity in the media habitat. Plant-growth promoting traits of the isolates were also assessed. All strains produced IAA ranging from 20 ㎍/mL to 250 ㎍/mL. Most of the isolates produced more than 80% siderophore. Four strains (Pantoea sp., PSB-08, Bacillus sp., PSB-13, Pseudomonas sp., PSB-17, and Enterobacter sp., PSB-21) showed outstanding performances for all the tested traits. In a bioassay of these four strains using mung bean plant, the best growth performances (23.16 cm, 22.98 cm, 2.27 g/plant, and 1.83 g/plant for shoot length, root length, shoot dry weight, and root dry weight, respectively) were obtained from the plants co-inoculated with Bacillus sp., PSB-13. The resultant data indicate that button mushroom media have got a diverse group of bacteria with plant growth promoting abilities. Thus, the media could be a good recycling resource for using to an effective bio-fertilizer.

Fourteen Unrecorded Species of Agaricales Underw. (Agaricomycetes, Basidiomycota) from the Republic of Korea

  • Agaricales species form pileate-stipitate fruiting bodies and play important roles in maintaining the terrestrial ecosystem as decomposers, symbionts, and pathogens. Approximately 23,000 Agaricales species have been known worldwide, and 937 species have been recorded in the Republic of Korea. However, most of them were identified solely based on morphological characteristics that often led to misidentifications. The specimens collected from 2018 to 2020 in the Republic of Korea were identified based on phylogenetic analysis of the internal transcribed spacer (ITS) sequences. Their identities were confirmed by microscopic characteristics. As a result, 14 Agaricales species were discovered for the first time in the Republic of Korea. They belonged to nine genera: Agaricus, Calocybe, Cortinarius, Hygrocybe, Inocybe, Lepista, Leucoagaricus, Marasmius, and Psathyrella. Detailed macroscopic and microscopic descriptions were provided to help distinguish these species. The morphological and molecular data provided in this study will serve as reliable references for the identification of Agaricales species.

Ozone Effects on the Mushrooms Yields (버섯 수량성에 미치는 오존의 영향)

  • The objective of this study was to determine mushrooms yields using ozone. The results of this study showed as follows; 1. According to ozone treatment on the Pleurotus ostreatus growth length of stipe became short, diameter of stipe and pileus yields of fruiting body increase 7% than the control treatments. 2. According to ozone treatment on the Agaricus bisporus growth, length of stipe became short, diameter of stipe and pileus became large, yields of fruiting body had increase 4% than the contol treatment. 3. According to ozone treatment on the Flamulina velutipes growth, day required for primordial formation after inoculation(DPI) became late for 2 days.

Occurrence and control of mushroom flies during Agaricus bisporus cultivation in Chungnam, Korea (충남지역 양송이버섯 재배지 버섯파리 발생과 방제 실태)

  • Mushroom flies are one of the most serious threats to mushroom production and cultivation in Korea, and they cause significant losses in yield and quality. In this study, we investigated the occurrence and control of mushroom flies in farms where Agaricus bisporus was cultivated in Chungnam. The cultivation period was more than 7 years in the 32 mushroom farms examined; about 28% were environment-friendly cultivation farms, and 72% were conventional cultivation farms. Mushroom flies were a major pest in about 84% of the examined farms. Most of the mushroom flies were sciarids and phorids (71% and 24%, respectively). The adult flies were found throughout the year, but their occurrence was the highest in September when the temperature increased. In general, the occurrence of mushroom flies in the examined farms was high; however, insect nets and post-crop steaming programs used as basic control methods were not implemented properly. Our findings show that integrated pest management of mushroom flies is urgently required for A. bisporus cultivation in Korea.

Browning of Minimally Processed Mushrooms (Agaricus bisporus Sing.) as Affected by Picking Season and Postharvest Holding Time (수확시기 및 가공전 보관기간이 신선 편이가공 양송이의 갈변에 미치는 영향)

  • This study was conducted to compare development of browning in minimally processed mushrooms (Agaricus bisporus Sing.) by picking season and postharvest holding time of the raw material. 'White' type of the mushrooms were harvested and minimally processed immediately after harvest or holding at 2$^{\circ}C$ for 7 days after picking in 4 seasons. The processed mushrooms were kept at 5$^{\circ}C$, and changes in surface color and content of total phenolic compounds of the mushrooms were measured. Respiration rate of the whole mushrooms was ranged from 9.55 to 17.93 mL C0$_{2}$/kg$\cdot$ hr, and the rate was low in mushrooms harvested in summer and autumn compared with those picked in spring and winter. Hunter L value of the mushrooms decreased rapidly just after slicing, especially in summer-harvest mushrooms. The mushrooms processed with the raw materials stored at 2$^{\circ}C$ for 7 days after harvest showed the lowest 'L' value among the sliced mushrooms prepared at different holding time. Consequently, summer-harvest mushrooms and the mushrooms kept at 2$^{\circ}C$ for more than 7 days after harvest would be unsuitable for minimally processing due to rapid browning.

Inhibitory Activity of Edible Mushrooms on the Tissue Thromboplastin (Tissue Factor) (조직 혈액응고인자에 대한 식용버섯류의 저해활성)

  • Tissue thromboplastin (tissue factor), a membrane bound glycoprotein is an important initiating factor in blood coagulation cascade, which leads to the formation of thrombin by activating both factor X and IX. Activation of blood coagulation by TF is essential for blood injury, and stimulates the blood coagulation in myocardial infarction, cancer and blood coagulatory diseases. High density lipoprotein, apolipoprotein A-II were known to be biological TF inhibitors. Recently, studies on search for TF inhibitors from natural products have been active in Korea. Among the edible mushrooms screened for inhibitory activities on the TF, Lentinus edodes showed the most strong activity, followd by Agaricus bisporus and Ganoderma lucidium. And the fractionation of the above mushrooms with the chloroform ($CHCl_3$) and ethylacetate (EtOAc) was done and evaluated for the inhibitory activities on TF. In Ganoderma lucidium, $CHCl_3$ fraction and $H_2O$ layer were not active, but EtOAc fraction exhibited a strong inhibitory activity on TF and the $IC_{50}$ value was $1.07{\times}10^{-4}\;g$. In the case of Agaricus bisporus, there were no inhibitory activities on the TF in all of the fractions. $CHCl_3$ fraction and $H_2O$ layer of Lentinus edodes did not show inhibition on the TF but EtOAc fraction showed strong inhibition on the TF, and the $IC_{50}$ value was $7.70{\times}10^{-4}\;g$.

Antimutagenic Effects of Enzymatic Browning Reaction Products of polyphenol Compounds by polyphenoloxidase derived from Mushroom(Agaricus bisporus) (양송이 유래 Polyphenoloxidase에 의한 Polyphenol 화합물의 효소적 갈변생성물의 돌연변이 억제효과)

  • The antimutagenic effects of enzymatic browning reaction products (MEBRPs) of polyphenol compounds (catechol, homocatechol, hydroxyhydroquinone, pyrogallol) by enzyme extracted from mushroom (Agaricus bisporus) were demonstrated through spore rec-assay using B. subtilis $H17(rec^+)$ and $M45(rec^-)$, Ames test using S. typhimurium TA98 and TA100 and SOS chromotest using E. coli PQ37/plasmid pKM101. In spore rec-assay, the MEBRPs showed antimutagenic effects by decreasing of the inhibition zone induced by MNNG. In Ames test with S-9mix in both TA98 and TA100, all of MEBRPs showed strong antimutagenic effects of about 21 to 99% against mutation by $B({\alpha})P$ and Trp-P-1, as adding $300\;{\mu}l$ of the MEBRPs. In SOS chromotest, MEBRPs showed antimutagenic effects by inhibiting the SOS-inducing function induced by 4NQO and MMC, as increasing in concentration of the MEBRPs. But they did not showed mutagenicity in these bacterial assays.

Characteristics of Crude Protein-bound Polysaccharide from Agaricus blasei Murill by Extraction and Precipitation Conditions and Its Antitumor Effect (추출 및 침전조건에 따른 아가리쿠스버섯에서 분리한 조단백다당류의 특성 및 암세포 증식억제 효과)

  • Efficient extraction method of crude protein-bound polysaccharide (CPBP) from Agaricus blasei Murill was established. CPBP yields by ultrasonic and hot water extractions were 13.0 and 7.8%, respectively. Pressure extraction for 3 hr gave the highest ${\beta}-glucan$ content; no significant difference was observed between 2 and 3 hr extraction. Four volumes added ethanol gave the highest yields of CPBP and ${\beta}-glucan$ contents at 10.89 and 35.97%, respectively. Decomposition temperature of CPBP was $240-365^{\circ}C$, showing relatively good thermal stability. In SRB (sulforhodamine B) assay, CPBP treatment at $1,000\;{\mu}g/mL$ for 72 hr inhibited proliferations to A549, MCF-7, and AGS cancer cells by 43.9, 21.4, and 32.5%, respectively.
