The temperature change due to focused ultrasound in dissipative acoustic medium is very important because it provides us much information. To measure the temperature change inside of the dissipative acoustic medium non-invasively, we adopt a temperature sensitive film which has thermochromic particles with critical temperature of $30^{\circ}C$. As a dissipative acoustic medium, agar layer is chosen in the study. The temperature change due to the ultrasound was measured depending on the concentration of the sugar in the agar layer. The color change on the film due to the ultrasound was investigated when the concentration of sugar was from 25% to 40%. As the result, there were rapid increases of discolored area on the film within 2~5 second after the ultrasound driving and the increasing rates decreased after the period. To compare the simulation results were also shown. However in the simulated result, the discolored areas linearly increased from start to 10 seconds. The reason of the differences between the experimental results and simulated ones is that the change of thermal conductivity and heat capacity of the medium were not considered in the simulation program.
The antifungal activity of crude methanolic extract and fractions from Yucca smalliana Fern. leaves, roots and flowers were investigated in vitro against a panel of plant pathogenic fungi. The minimal inhibitory concentration(MIC) was determined by an agar dilution method. Preliminary liquid culture and agar plate assays showed that the growth of Fu sarium oxysporum, Phytophthora capsici, Rhizoctonia solani and Botrytis cinerea were inhibited by Y. smalliana extracts. The extracts from flowers and leaves showed antifungal activity of 64.0% and 34.0% against F. oxysporum, 66.0% and 62.0% against P. capsici, and 27.0% and 41.0% against B. cinerea, respectively. The methanolic extract from Y. smallina leaves in distilled water was fractionated using solvents of increasing polarity: hexane, ethyl acetate and butanol. These fractions had a broad spectrum of antifungal activity, found to reside entirely in the butanol and aqueous fraction. The aqueous fraction showed inhibition rate of 60.0, 67.8, 84.6 and 58.3% against F. oxysporum, R. solani, C. gloeosporioides, and B. cinerea, respectively, and the butganol fracgtion showed 36.0, 46.0, 66.1 and 58.3%, respectively. Phenolics(e.g. flavonoids, steroids and terpenoids) were observed in the thin layer profile of the different fractions. Leave extract showed a prominent antioxidant activity totally scavenging the free radical of DPPH at a concentration of 1 mg/ml.
A capillary method was used to evaluate the properties of mass transfer process and diffusion coefficients in the gel food. Amaranth dye was selected as a diffusant material to visualize the degree of diffusion procedure easily. After contacting cylinder containing agar gel with amaramth dye solution for some hours, the gel was cut to five segments by 0.5 cm in length. The diffusant concentration from the segments were measured by the spectrophotometer at 523 nm. Prediction models for the diffused quantities in gel food were established by the regression program of SPSS package program. Generally, diffusion coefficient can be calculated by Fick's second law, however, it will be determined by using numerical analysis method more easily. Finally the diffusion coefficients in this research were calculated by arithmetic mean of the measured values. As raising gel agent concentration, the mean diffusion coefficient tended to decrease because the obstruction effect came to become significant.

This experiment was conducted to determine factors affecting callus induction of Vernal alfalfa. Growth regulators, basal medium, medium pH, explant age, and agar concentration for callus induction were investigated. The results obtained were as follows: For the callus induction, 2-5 mg/l 2,4-D alone was found to be most effective on callus induction. Cytokinins did not have positive effect on the callus induction, and even the more cytokinin added induced the less callus. Callus yield was much higher in B5 or SH medium than in any other media. The calli induced in PC and MS media were more friable than those induced in other media. The medium pH of 5.8 gave the best response of callus induction. At higher than pH 7.0, callus induction was inhibited severely. The effective seedling age for callus induction was around 9 days. In agar concentration, 0.5 % (W/V) was suitable for callus induction and it was severely depressed at above 1 %. Callus induction was not influenced by day length or illumination. Calli cultured under 1618 hour lightldark cycle became more compact and green than those cultured under the dark.
Conidial germination of Septoria glycines Hemmi, brown spot fungus of soybean, was studied by slide germination test. Poor conidial germination of S. glycines was observed on sterile distilled water, but potato dextrose agar(PDA) and distilled water floated with soybean leaf disc furnished a satisfactory medium for conidial germination. Exogenous supply of carbon source was essential for conidial germination, while phosphorous and potassium were not evident as that for carbon. Soluble starch was the most suitable as a carbon source for conidial germination and followed by D-glucose, D-galactose and lactose in that order. Maximum germination was attained in the $5\times10^{-2}mol$. concentration of glucose. Germination was decreased with increment of conidial concentration and was almost completely suppressed in the density of 10,000 conidia per $mm^2$. It suggested existing a self-inhibitor(s). Non-washed conidia germinated more than washed conidia and this was obvious when the conidia density was over $2\times10^3$ conidia per $mm^2$ on the dry agar block.
Magaldrate. an antiacid was synthesized by reacting magnesium oxide, aluminum sulfate, and dried aluminum hydroxide gel. The optimum synthesis conditions based on the yield of t he product were established by applying Box-Wilson experimental design. It was found that the optimum synthesis conditions of Magaldrate were as follows: Reaction temperature; 61~$85{\circ}C$, concentration of two reactants. Mgo and $Al(OH)_3$: 16~19.8%, molar concentration ratio of two reactants, [MgO]/[$Al(OH)_3$]; 4.2~5.0, temperature of washing water; 36~$41^{\circ}C$ and drying temperature of the product: 76~$80^{\circ}C$. Magaldrate was synthesized under the optimum synthesis conditions and identified by analyzing the chemical composition, and by differential scanning calorimetry and X-ray diffraction method. The Magaldrate sample synthesis under these conditions was used to prepare 15.6% Magaldrate original suspension which was utilized to make 13% Magaldrate suspension dispered in various concentrations of eight types of suspending agents. The acid-neutralizing capacity of 13% Magaldrate suspension dispersed in 0.25% suspending agents was examined by Rosset-Rice method. The maximum pH was reached within 1 minute in all suspension tested, and duration maintained between pH 3~5 was decreased in the order of Na alginate Na silicate(meta) Veegum HV pectin agar>Na>CMC>xanthan gum>bentonite. It was found that the hysteresis loop area was increased with temperature in the case of Riopan Plus and the addition of agar, whereas the area was decreased with temperature in the case of the addition of Na alginate and xanthan gum. 13% Magaldrate suspension tends to sediment by the addition of bentonite.

$Gynura$$segetum$ (Lour.) Merr. ($Compositae$) is a well recognized medicinal plant in Indonesia and Malaysia. It is believed to have an anticoagulant effect and is used in treating snake-bites, inflammations and other skin afflictions. This study was carried out to evaluate the antimicrobial effects of $Gynura$$segetum$ leaves extracts and its fractions. The chemical compositions of the active extracts were also determined. The antimicrobial activities of different solvent extracts of leaves of $Gynura$$segetum$ were evaluated using the agar well-diffusion method. The Minimum Inhibitory Concentration (MIC) of the active subfractions was determined by the tube dilution method. Gas Chromatography-mass spectrometry (GC-MS) analysis was carried out to identify the chemical compositions of the active extracts. The ethyl acetate fraction and its subfraction E4 performed potent antimicrobial activities and fifteen known chemical constituents were identified by GCMS analysis as 4-vinylphenol, 1-tetradecene, phenol, 2,4-bis(1,1-dimethylethyl), 1-hexadecene, E-15-heptadecenal, hexadecanoic acid, 1,2-benzenedicarboxylic acid, dibutyl ester, 1-docosene, octadecanoic acid, 1-eicosene, cyclotetracosane, 1,2-benzenedicarboxylic acid, bis(2-ethylhexyl)ester, butanedioic acid, monomethyl ester, niacin and 4-hydroxy-benzoic acid. The results of this study suggested a connection between the antimicrobial activities and the chemical structures. The plant may be used as a potential source for antimicrobial agents.
A squid meat has not been utilized for gel products because of its lower gel forming ability. The objectives of this study were as followed; 1) the optimum heating condition on squid meat paste products and 2) the optimum added level for jelly strength of squid meat paste products. Optimum heating conditions of squid meat kamaboko were as followed; setting (pre-heating) at 15$^{\circ}C$ or 55$^{\circ}C$ for 2 hours and heating at 90$^{\circ}C$ for 60 minutes. Effect for jelly strength of starch additives wheat starch, potato starch and com starch were examined. The jelly strength of heat induced gels differed from the levels of additives. In case of adding starch, potato starch was resulted in the superior jelly strength than the other starchs, wheat starch and corn starch, at any levels. Optimum concentration was 10%(w/w) at every additives. Folding test value was B at added 10% and this value was mean good product. Data of jumbo and flying squid meat paste products added potato starch, corn starch and wheat starch of 10% were shown below, jelly strengths were 858${\pm}$34∼1020${\pm}$37gㆍcm and 966${\pm}$33∼l148${\pm}$45gㆍcm and moisture contents were 72.43∼73.04% and 71.61∼72.78%, respectively. To adding edible agar and sea tangle, showed the highest jelly strength (edible agar>sea tangle, flying squid>jumbo squid) at added 0.5%(w/w) concentration.

Mold is one of the important bio-aerosols affecting human health in the indoor environment. To manage mold contamination, it is necessary to use an appropriate method for its detection and enumeration. Recently, the impaction method of ISO 16000-18 has been established as one of methods to detect and enumerate molds in air. To investigate the general use of the impaction method for mold detection in domestic indoor environments, the suitability of the method was assessed using different antibiotics, media and air samplers. All of the three antibiotics tested - ampicillin, chloramphenicol and streptomycin - showed inhibitory effects on bacterial colony formation on MEA and DG-18 media, without inhibiting mold growth. Of these three antibiotics, ampicillin was the most effective. There was no statistical difference between MEA and DG-18 media in the measurement of mold concentration. The formation of discriminative colony morphology was more apparent in DG-18 media. No significant difference in the measurement of mold concentration was found between Andersen samplers and MAS-100NT samplers, which are two major samplers introduced in Korea.

This study was carried out to treatment test for bovine mastitis by the determination of adenosine triphosphate (ATP) based on firefly bioluminescence. The results obtained are followed; 1. In the susceptibility test, cephalothin which looks the most effective were sensitive to Staphylococcus sp. (72.3%), Micrococcus sp. (84.2%), Streptococcus sp. (72.7%) and Gram positive bacilli (72.7%), Gram negative bacilli were sensitive to gentamicin (92.3%) and Yeast-like-fungi was the most sensitive to clotrimazole, and nystatin in order. 2. When the number of bacteria, such as Staphylococcus aureus, Candida tropicalis isolated from the mastitis milk were counted by conventional agar plating technique, and compared with the concentration of bacterial ATP, it gave a good linear relationship. The content of ATP per Staphylococcus aureus, cell was 3.1fM and Candida tropicalis showed the high level of A TP (90fM). 3. The ATP assay was applied to the determination of minimal inhibitory concentration (MIC) of various antibiotics. When Staphylococcus aureus was incubated in the presence of different concentration of tetracycline, erythromycin, kanamycin and streptomycin sulfate and the growth was monitored by the conventional agar plating technique and ATP assay, both methods shown the same results that they were 1mcg/ml, 2mcg/ml, 6.25mcg/ml and 8mcg/ml, respectively. 4. For the determination of susceptibility of sensitive and resistant Staphylococcus au reus isolated for the milk with mastitis to tetracycline, erythromycin kanamycin and streptomycin sulfate, the minimum time required for the test was determined by the assay of ATP every 30 minutes during incubation of 3 hours at $37^{\circ}C$. ATP concentration time curve calculated on both resistant and sensitive strains incubated 3 hours as the optimum time for the determination of susceptibilities of various antibiotics exemed. The ATP concentration of each test broth (antibiotic containing), expressed as a percentage of its own control brith (antibioticfree) indicated values of 30% to be indicative of each antibiotic sensitivity. Single time point ATP assay carried out on the various sensitive and resistant of Staphylococcus aureus to antibiotics examined after 3 hours at $37^{\circ}C$ correlated exactly with disc diffusion and MIC. 5. In the cure of intramammary treatment of bovine mastitis in lactating quarters, the cure rate of Staphylococcal mastitis showed to cephalexin (80%), cloxacillin and gentamicin (70%), ampicillin and oxytetracycline (60%), and Streptococcal mastitis showed to cephalexin (85%), penicillin (80%), cloxacillin and oxytetracycline (75%), and ampicillin (70%), but intramammary antimycotic drug (clotrimazol) were only a little effect about fungal mastitis.
