  • Heavy metal release from wastewater is a serious environmental problem, and therefore, various wastewater treatment techniques have been developed. Among the techniques, sorption technique is most attractive. Considerable researches have been recently focused on finding out inexpensive sorbents, especially from various natural materials. In order to evaluate the applicability of the scoria taken from the Jeju Island, Korea to remove heavy metals (Pb, Cu, Zn, Cd) from aqueous solutions, equilibrium sorption experiments were conducted in this study. In equilibrium tests, powdered activated carbon (PAC), one of the most commonly used sorbents, was also tested to compare the effectiveness of the Jeju scoria with that of PAC. The Jeju scoria had larger adsorption capacity and affinity for metal ions (Pb(II), Cu(II), Zn(II), Cd(II)) than PAC. The sorption parameters of the two sorbents were evaluated by using both the Langmuir and Freundlich isotherms, and the sorption data were better fitted to the Freundlich isotherm. In addition, the sorption behavior of metal ions (Pb(II), Cu(II), Zn(II), Cd(II)) onto the scoria displayed a typical characteristic of the cation sorption. The removal of metal ions decreased at a lower pH condition due to competition with hydrogen ions for the sorption sites of Jeju scoria, while the removal increased at a high pH condition due to hydroxide precipitation.

  • Aptamers are functional nucleic acids that can specially bind to proteins, peptides, amino acids. nucleotides, drugs, vitamins and other organic and inorganic compounds. The aptamers are identified from random DNA or RNA libraries by a SELEX (systematic evolution of ligands by exponential amplification) process. As aptamers have the advantage, and potential ability to be released from the limitations of antibodies, they are attractive to a wide range of therapeutic and diagnostic applications. Aptamers, with a high-affinity and specificity, could fulfil molecular the recognition needs of various fields in biotechnology. In this work, we reviewed some aptamer Selection techniques, properties, medical applications of their molecules and their biotechnological applications, such as ELONA (enzyme linked oligonucleotide assay), flow cytometry, biosensors, electrophoresis, chromatography and microarrays.

  • Since the turn of the century, scientists have earnestly sought to develop a single laboratory assay or combination of laboratory assays which accurately predict the fertility of a semen sample. Most of these assays have focused on evaluating physical characteristics of sperm such as motility, viability, acrosomal integrity and morphology. In recent years new approches have been used to assess the functional aspects of a sperm that are needed to reach the oocyte, fertilize it and contribute to successful embryo development. Among these techniques are the ability of sperm to undergo a heparin induced acrosome reaction and in vitro fertilization, and the affinity of sperm to bind heparin binding protein. Intensification of research efforts in the area of control of sperm fertilizing ability should be a high priority, in view of undoubted benifits both to our basic understanding of sperm fertilizing ability and to our ability to modify it for Al industry.

  • A pseudoreceptor combines structure-based and ligand-based techniques to represent a unifying concept for both receptor mapping and ligand matching. In this molecular modeling approach, there are opportunities to construct the pseudoreceptor models using a set of small molecules. To build a reliable pseudoreceptor model, we need a set of ligand molecules with known affinity (biological activity) to generate 3D bioactive conformation for each of these ligand molecules. Several software packages are available to generate a pseudoreceptor model and this can provide an entry point for structure based drug discovery in cases where receptor structure information is not available. In this review, we presented the concept of pseudoreceptor, as well as discussed about various software packages available to generate a pseudoreceptor model.

  • Protein phosphorylation plays a key role in signal transduction in cells. Since phosphoproteins are present in low abundance, enrichment methods are required for their purification and analysis. Chemical derivatization strategies have been devised for enriching phosphoproteins and phosphopeptides. In this report, we employed a strategy that replaces the phosphate moieties on serine and threonine residues with a biotin-containing tag via a series of chemical reactions. Ribulose 1,5-bisphosphate carboxylase/oxygenase (RUBISCO)-depleted protein extracts prepared from Arabidopsis seedlings were chemically modified for 'biotin-tagging'. The biotinylated (previously phosphorylated) proteins were then selectively isolated by avidin-biotin affinity chromatography, followed by two-dimensional gel electrophoresis (2-DE) and matrix-assisted laser-desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS). This led to the identification of 31 protein spots, representing 18 different proteins, which are implicated in a variety of cellular processes. Despite its current technical limitations, with further improvements in tools and techniques this strategy may be developed into a useful approach.

  • This paper describes a personalized advertisement technique as a part of intelligent customer services in Internet shopping malls. Based on customers' initial profile, purchase history, and behaviors in an Internet shopping mall, the technique displays appropriate advertisements on Internet web pages when customers' visit to the shopping mall. Customers preference scores for product groups which are main sources to select advertisements, are stored either a preference table or preference trees. Both of the two storage methods can support selection of advertisements on real time, and the preference tree method can reflect affinity among product groups. The suggested technique selects different advertisements to reflect changes of customers preferences as time goes by. An experiment has been performed to evaluate the effectiveness of the algorithm, which revealed that the algorithm selects more customer-oriented advertisements rather than random selection.

  • Results on localization of growth hormone receptor (GHR), interaction of growth hormone (GH) with receptor in buffalo adipose tissue and identification of activated signaling molecules in the action of GH are presented. Bovine GH (bGH) was labeled with fluorescein or biotin. Fluorescein-labelled bGH was used for localization of GHRs in buffalo adipocytes. The receptors were present on the cell surface. The affinity of binding of GH to its receptor was determined by designing an experiment in which buffalo adipose tissue explants, biotinylated GH and streptavidin-peroxidase conjugate were employed. The affinity constant was calculated to be $2{\times}10^8M^{-1}$. The receptor density on adipose tissue was found to be 1 femto mole per mg of tissue. Signalling molecules generated in the action of GH were tentatively identified by employing Western blot and enhanced chemiluminescence techniques using anti-phosphotyrosine antibody. Based on molecular weights of proteins reactive to anti-phosphotyrosine antibody, three signaling molecules viz. insulin receptor substrate, Janus activated kinase (Jak) and mitogen activated protein were tentatively identified. These signaling molecules appeared in a time (incubation time of explants with growth hormone) dependent way. The activation of Jak2 was confirmed by employing anti-Jak2 antibody in a Western blot. The activation of Jak2 occurred during 5 min incubation of buffalo adipose tissue explants with GH and incubation for an additional period, viz. 30 min. or 60 min., resulted in a drastic reduction in activation. The results suggest that Jak2 activation is an early event in the action of GH in buffalo adipose tissue.

  • The request scheduling algorithms being used for the cluster web servers are mostly in two categories : load-balancing and contents-based cache affinity The goal of the load-balancing algorithms is to balance the loads between real servers. On the other hand, contents-based scheduling algorithm exploits the cache affinity in a way that the same type of requests are to be directed to a dedicated real server allowing load imbalance. So the performance comparison of the two algorithms is necessary, nevertheless the related experiment results are not much suggested. In this paper, performance evaluations have been done to compare the performance of the two scheduling algorithms. To accomplish this, we first implement a linux cluster web server, and then present the performance measurement results. The main contribution of this paper is to help the cluster web server administrators to select an algorithm fitting in with their circumstances from the two algorithms.
