낙동강 하구 생태계의 환경요인과 Aeromonas spp. 분포와의 관계 (Effects of Environmental Factors on Aeromonas spp. Population in Naktong Estuary)

  • 1986년 8월부터 12월까지 낙동간 하구의 3정점에서 Aeromonas의 분포 및 이에 영향을 미치는 환경요인을 조사하였다. 조사기간 중 Aeroamonas spp. 개체수 변화는 $4.3\times10^{2}-4.6\times 10^{4}$ MPN/100ml로 기록되었다.분산분석의 결과 Aeromonas spp. 의 분포는 정점간에 유의한 차이가 이쓴 것으로 나타났으며 정점 2에서 가장 높은 개체수를, 정점 3-B에서 가장 작은 개체수를 보였다. Aeromonas spp.의 분포에 영향을 미치는 환경요인을 알아보기 위하여 중회귀 분석 및 주요인 분석(principal component analysis)을 한 결과 Aeromonas spp. 의 분포는 담수의 유출과 무기영양염류의 유입에 의해 크게 영향을 받는 것으로 나타났으며 종속영양세균, 이용가능한 질소원, 분변성대장균 및 수온고 밀접한 상관관계를 나타내었다.

  • Aeromonas spp. and Pseudomonas spp. are opportunistic pathogens widely distributed in the aquatic environment. To test the antibiotic susceptibility, the MIC of the 18 antibiotics mainly used in aquaculture were measured. Aeromonas spp. and Pseudomonas spp. straoms had different resistance patterns against most antibiotics. The MIC of tetracycline for four Aeromonas spp. strains (10.5%) was < 0.25 ㎍/mL. However, 0.5-4 ㎍/mL tetracycline inhibited most Pseudomonas spp. strains. The tet resistance performance of 14 genes including tet(B), tet(E), and tet(M) were investigated. Investigating, the tetracycline resistance gene of 38 Aeromonas spp. strains detected tet(A) in 21 strains (55.3%). Two Pseudomonas spp. strains showed high MIC values and no inhibition zone. tet gene analysis detected tet(D) in only one strain (5%).

Novel Qnr Families as Conserved and Intrinsic Quinolone Resistance Determinants in Aeromonas spp.

  • The environment has been identified as an origin, reservoir, and transmission route of antibiotic resistance genes (ARGs). Among diverse environments, freshwater environments have been recognized as pivotal in the transmission of ARGs between opportunistic pathogens and autochthonous bacteria such as Aeromonas spp. In this study, five environmental strains of Aeromonas spp. exhibiting multidrug resistance (MDR) were selected for whole-genome sequencing to ascertain their taxonomic assignment at the species-level and to delineate their ARG repertoires. Analyses of their genomes revealed the presence of one protein almost identical to AhQnr (A. hydrophila Qnr protein) and four novel proteins similar to AhQnr. To scrutinize the classification and taxonomic distribution of these proteins, all Aeromonas genomes deposited in the NCBI RefSeq genome database (1,222 genomes) were investigated. This revealed that these Aeromonas Qnr (AQnr) proteins are conserved intrinsic resistance determinants of the genus, exhibiting species-specific diversity. Additionally, structure prediction and analysis of contribution to quinolone resistance by AQnr proteins of the isolates, confirmed their functionality as quinolone resistance determinants. Given the origin of mobile qnr genes from aquatic bacteria and the crucial role of Aeromonas spp. in ARG dissemination in aquatic environments, a thorough understanding and strict surveillance of AQnr families prior to the clinical emergence are imperative. In this study, using comparative genome analyses and functional characterization of AQnr proteins in the genus Aeromonas, novel Aeromonas ARGs requiring surveillance has suggested.

  • 1986년 8월부터 12월까지 낙동강 하구에서 161주의 운동성 Aeromona를 분리한 후 42가지의 형태학적, 생화학적 특성은 조사하여 cluster 분석을 한 결과 85% 유사도 수준에서의 3개의 주 cluster와 84% 유사도 수준에서의 2개의 소수 cluster의 5 cluster로 나눌 수 있었다. 각 cluster는 MR반응과 glucose으로부터의 가스생성 여부의 생화학석 특성에 의하여 구분이 되었다. 2개의 소수 cluster를 포함한 5새의 cluster는 교차응집 실험에 의해 크게 2개의 혈청학적 cluster로 나뉘어졌으며, 이러한 결과는 독성실험에 의한 grouping 결과와 일치하였다.

경기도 내 양식어류에서 분리한 병원성 세균의 Tetracycline 내성 유전자 분포 (Distribution of Tetracycline-Resistance Genes detected from isolates of cultured fishes in Gyeonggi-do)

  • Tetracycline (TC)은 국내에서 세균 감염 치료에 반드시 필요한 대표적인 항생제로 알려져 있다. 그러나 부적절한 사용과 남용으로 인해 Tetracycline, Erythromycin, Fluoroquinolone 등과 같은 항생제에 대한 내성이 발생하고 있으며, 이는 국내 양식 분야에서 심각한 경제적 피해를 유발한다. 본 연구에서는 2015~2018년에 걸쳐 경기도 양식장의 양식생물에서 101개 균주를 분리하였다. 분리균주는 간이적인 생화학적 방법을 통해 동정하였으며, 최소 억제농도(MIC)의 확인을 통해 Oxytetracycline (OTC), Ampicillin (AMP), Clindamycin (CLI), Enrofloxacin (ENRO), Gentamycin (GEN)에 대한 내성 여부를 확인하였다. 이중 TC에 내성을 보이는 균주는 PCR법을 통해 tet 유전자의 분포를 조사하였다. 그 결과, 총 101개 균주 중에서 Aeromonas spp.가 44개(43.5%)로 가장 우점하였고, 그 다음으로 Pseudomonas spp. 4개(4.0%), Vibrio spp. 5개(5.0%)가 확인되었다. 또한, 다중 내성을 보이는 균주(77.2%)가 단일 내성균(22.8%)보다 많음을 확인하였다. tet(A), tet(D), tet(E), tet(G), tet(M), tet(S)가 TC 내성 균주에서 검출되었으며 tet(A)가 가장 우점적으로 확인되었다. Aeromonas spp.는 분리된 균주 중에서 가장 많았으며, 경기도 내 양식현장에서의 다양한 항생제 내성 유전자의 특성에 대한 추가적인 연구가 필요하다.

수입산 관상어로부터 분리된 tetracycline 내성 Aeromonas spp.에 tet(M) 및 tet(G) 유전자의 특성 분석 (Characterization of tet(M) and tet(G) Genes among Tetracycline-resistant Aeromonas spp. Isolated from Imported Ornamental Fishes)

  • In this study, the molecular structures of tet(M) and tet(G) carried by tetracycline (Tc) resistant bacteria in intestinal microflora from the imported ornamental fish were characterized and compared with each other depend on the imported countries. Of the total isolates, approximately 8.9% of the Ent-lac+(lactose fermentative bacteria on coliform media) Tc resistant isolates in fish from three different countries, Singapore, Taiwan and Brazil, were appeared to contain tet(M). Three representative isolates of different countries, Aeromonas spp. JSM-1 (Singapore), JTM-1 (Taiwan) and JBM-1 (Brazil), were isolated and analyzed the molecular structures of tet(M) gene. Interestingly, partial sequence of tet(M) genes (1099 bp) in JBM-1 (Brazil) showed 99.5% homology with the tet(M) found in the Vibrio spp. RV16 isolate, obtained from marine fish in Korea and known to carry Tn1545 parent type of tet(M). In contrast, tet(M) gene in JSM-1 and JTM-1 showed mosaic structure of Tn1545 and Tn916, and 100% homology with each other. It may suggest the presence of various characteristics in terms of tet(M) gene structure. The determined sequence of the tet(G) from Aeromonas spp. JSG-1 and JBG-1 isolated from Singapore and Indonesia ornamental fish respectively showed similar nucleotide sequence homology but revealed a few nucleotide changes in comparison with the sequence of the prototype tet(G) gene (S52437 in GenBank).

Draft Genome Sequence of Aeromonas caviae Isolated from a Newborn with Acute Haemorrhagic Gastroenteritis

  • Aeromonas spp., are Gram-negative rods that can cause infections in healthy and immunocompromised hosts. The clinical presentation of gastroenteritis varies from mild diarrhoea to shigella-like dysentery to severe cholera-like watery diarrhoea. Here, we report a case of acute hemorrhagic gastroenteritis in a newborn infant by Aeromonas caviae and its draft genome sequence. It is important to reduce the chance of incorrect isolate identification, which could lead to the exclusion of pathogenic Aeromonas spp., from routine laboratory identification in cases of diarrheal diseases. The genome sequence of A. caviae SVJ23 represents a significant step forward in understanding the diversity and pathogenesis, virulence, and antimicrobial resistance profile.

  • Aeromonas spp. 감염은 관상어 산업뿐만 아니라 양식업에서도 상당한 경제적 손실을 초래하는 것으로 보고되고 있다. 한편 2022년 12월부터 2023년 1월 사이에 국내에서 사육중인 금붕어(Crassius auratus)에서 Aeromonas병증을 보이는 어체에 대해서 발병 원인균을 규명하기 위해서 조사 하였다. 병증을 보이는 금붕어의 피부와 내부 장기에서 그람 음성균이 분리되었다. Biochemical test와 16S rRNA gene으로는 Aeromonas 속까지 동정할 수 있었으나 종레벨까지는 구분하기 어려웠다. 이에 가장 상동성이 높았던 A.veronii를 대상으로 species-specific PCR을 수행했고 명확히 종 수준으로 동정할 수 있었다. 아울러서 장독소 유전자, 감염 실험 및 항생제 내성 분석을 수행하였다. 금붕어에 대한 균주의 생체 내 병원성 시험 결과, 실험 주사 후 일주일 이내에 감염된 숙주에서 100%의 치사율을 나타냈다. 장독소 유전자인 act 독력 유전자가 검출되었다. 균주의 항균제 감수성 분석 결과 대부분의 항균제에 감수성을 보였으나, Ampicillin, Imipenem, Meropenem 및 Clindamycin 약제에는 내성을 보였다.

Trends in Bacteria Causing Diarrheal Infection from 2010 to 2018 in Cheonan, Korea: Aeromonas spp., Campylobacter spp., and Clostridioides spp.

  • Diarrhea is one of the most common infectious diseases known worldwide. However, few studies have examined anaerobic diarrhea-causing bacteria (DB), which are difficult to culture. Recent advances in molecular biology have facilitated the detection and analysis of anaerobic DB. In this study, long-term trends in anaerobic DB were evaluated in Korea. From 2010 to 2018, symptoms of diarrhea reported were analyzed among patients hospitalized at the Dankook University Hospital in Korea. Results of multiplex polymerase chain reaction based on seasonality, age, overlapping infection, and other factors in patients were evaluated. DB were detected in 38.2% of 1716 stool specimens in the duration of the study. Of the pathogens detected using this method, 49.8% (n = 405/813) were anaerobic bacteria, including Clostridioides perfringens, Campylobacter spp., Clostridioides difficile toxin B, and Aeromonas spp. Among the four anaerobic bacteria, Clostridioides perfringens was the most commonly occurring (15.5%; n = 126/813). Detection rates of Clostridioides perfringens, Clostridioides difficile toxin B, and Aeromonas spp. were 34.1% (n = 22/55), 34.9% (n = 43/126), and 40.0% (n = 38/109), respectively. The detection rate of Campylobacter spp. (32.7%; n = 37/115) was the highest in patients between 10 and 20 years of age. The detection rate of anaerobic DB showed an increase in 2018 as compared with that in 2010, and the number of events of diarrhea caused by anaerobic DB also increased in this duration. Further studies are required to devise methods that might prevent the proliferation of anaerobic DB.

Occurrence and antibiotic susceptibility of fish bacteria isolated from Oreochromis niloticus (Nile tilapia) and Clarias gariepinus (African catfish) in Uganda

  • The intention of this study was to identify the bacterial pathogens infecting Oreochromis niloticus (Nile tilapia) and Clarias gariepinus (African catfish), and to establish the antibiotic susceptibility of fish bacteria in Uganda. A total of 288 fish samples from 40 fish farms (ponds, cages, and tanks) and 8 wild water sites were aseptically collected and bacteria isolated from the head kidney, liver, brain and spleen. The isolates were identified by their morphological characteristics, conventional biochemical tests and Analytical Profile Index test kits. Antibiotic susceptibility of selected bacteria was determined by the Kirby-Bauer disc diffusion method. The following well-known fish pathogens were identified at a farm prevalence of; Aeromonas hydrophila (43.8%), Aeromonas sobria (20.8%), Edwardsiella tarda (8.3%), Flavobacterium spp. (4.2%) and Streptococcus spp. (6.3%). Other bacteria with varying significance as fish pathogens were also identified including Plesiomonas shigelloides (25.0%), Chryseobacterium indoligenes (12.5%), Pseudomonas fluorescens (10.4%), Pseudomonas aeruginosa (4.2%), Pseudomonas stutzeri (2.1%), Vibrio cholerae (10.4%), Proteus spp. (6.3%), Citrobacter spp. (4.2%), Klebsiella spp. (4.2%) Serratia marcescens (4.2%), Burkholderia cepacia (2.1%), Comamonas testosteroni (8.3%) and Ralstonia picketti (2.1%). Aeromonas spp., Edwardsiella tarda and Streptococcus spp. were commonly isolated from diseased fish. Aeromonas spp. (n = 82) and Plesiomonas shigelloides (n = 73) were evaluated for antibiotic susceptibility. All isolates tested were susceptible to at-least ten (10) of the fourteen antibiotics evaluated. High levels of resistance were however expressed by all isolates to penicillin, oxacillin and ampicillin. This observed resistance is most probably intrinsic to those bacteria, suggesting minimal levels of acquired antibiotic resistance in fish bacteria from the study area. To our knowledge, this is the first study to establish the occurrence of several bacteria species infecting fish; and to determine antibiotic susceptibility of fish bacteria in Uganda. The current study provides baseline information for future reference and fish disease management in the country.
