Adaptation of Anisopteromalus calandrae (Hymenoptera: Pteromalidae) to an alternative host, Callosobruchus chinensis (Coleoptera: Bruchidae) (Anisopteromalus calandrae (Hymenoptera: Pteromalidae)의 대체기주 팥구미(Callosobruchus chinensis (L.))에 대한 적응)

  • Adaptation of Anisopteromalus calandrae maintained on rice weevil (Sitophilus oryzae) to an alternative host, adzuki bean weevil (Callosobruchus chinensis) was studied. A. calandrae required two generations to adapt to the adzuki been weevil. Through the adaptation, the mean($\pm$SEM) life span of th females became shorter $8.7\pm$1.3 days at the first generation to $6.9\pm$0.6 days at the second generation. Number of eggs laid per female increased from 29.3$\pm$7.7 to $68.2\pm$6.5. Consequently intrinsic rate of natural increase and net reproductive rate were increased. The generation time and population doubling time were decreased. The searching efficiency based on functional and numerical response of A. calandrae to the adzuki bean weevil were similar to each other, but the coefficient of variation decreased with the generation, suggesting that searching efficiency became stable when the parasitoid adapted to adzuki bean weevil. No time lag was required for A. calandrae adapted to the adzuki bean weevil to adapt again to th rice weevil and the searching efficiency of the parasitoid remained on a similar level for the first generation on.

  • This experiment was carried out for the analysis of the ability of callus formation and differentiation in the eight Korean local varieties of adzuki beans (Phaseolus angularis). KLA 102 has the highest ability on the formation of callus (all tissues have 90 % over). Morphological heterogeneity (color and shape) of callus was well reflected to the cell constitution and cell meristematic activities. Meristemoid tissues and rooting were derived from only creamish-white color and soft region of callus.

Antioxidant Activities and Total Phenolic Contents of Three Legumes

  • Legumes have been important components of the human diet. They contain not only protein, starch, and dietary fiber, but also various phenolic compounds such as flavonoids and phenolic acids. The importance of phenolic compounds to human health is well known due to their antioxidant activities. In this study, three legumes (adzuki beans, common beans, and black soybeans) frequently cultivated in Korea were evaluated for their total phenolic content (TPC) and antioxidant activities using DPPH (2,2-diphenyl-1-picrylhydrazyl), ABTS (2,2'-azinobis (3-ethylbenzothiazoline 6-sulfonate)), and FRAP (ferric reducing antioxidant potential) assays. In addition, correlations between agricultural traits and antioxidant activities of these three legumes were analyzed. Antioxidant activities assessed by DPPH, ABTS, and FRAP assays and TPC showed wide variations among legumes types and accessions. Among the three legumes, adzuki beans showed higher TPC and antioxidant activity than the other two legumes. In correlation analysis, seed size showed negative correlations with antioxidant activities and TPC. In principal component analysis and hierarchical clustering analysis, each of the three legumes was clearly separate. Results of this study can be used as basic information for developing functional materials for each legume. They can also help us understand the overall antioxidant activity of the three legumes.

Identification and Characterization of Protease-Resistant Proteins from Adzuki Beans (소화 효소 저항성을 지니는 팥 단백질의 성질 규명)

  • It is already known that adzuki beans (Vigna angularis) are able to control appetite. Therefore, this study tested the proteins isolated from adzuki beans for their protease resistance and interaction with the intestinal mucosa. The major proteins from adzuki beans were found to be resistant to the digestive enzymes pepsin and pancreatin, and were identified using 2D-SDS-polyacrylamide gel electrophoresis and mass spectrometry. The major adzuki proteins were easily fractionated by treating the soluble protein extract with 10mM $CaCl_2$, and were found to contain lactotransferrin, a homologous protein to the dynein light chain domain, proteinase inhibitor, and proteins with unknown functions. From a tissue binding assay using mouse intestinal tissue sections, the major protein fraction showed weak, yet significant and specific binding to the mucosa layer of the small intestine. Thus, the current results suggest that adzuki proteins are resistant to digestive enzymes, which enables them to survive protease digestion in the intestinal tract, plus they may interact with the intestinal mucosa layer. Therefore, the molecules responsible for controlling appetite in adzuki beans are presumably protease-resistant proteins that interact with the intestinal mucosa or delay digestion in the digestive tract.

Forage Yield and Quality of Summer Grain Legumes and Forage Grasses in Cheju Island

  • Soybean [Glycine max (L.) Merr.), mungbean [Vigna radiata (L.) Wilcz.], cowpea [V. unguiculata (L.) Walp.], adzuki bean [V. angularis (Willd.) Ohwi & Ohashi], maize [Zea mays L.], sorghum [Sorghum bicolor (L.) Moench], sorghum $\times$ sudangrass [So bicolor intraspecific hybrid], and Japanese millet [Echinochloa crusgalli var. frumentacea (Link) W.F. Wight] were grown at two planting dates (18 June and 15 July) at Cheju in 1997 to select the best forage legumes adapted to Cheju Island for grass-legume forage rotation. Averaged across planting dates and cultivars, dry matter (DM), crude protein (CP), and total digestible nutrient (TDN) yields were 5,646, 1,056, and 3,637 kg/ha for soybean, 4,458, 676, and 2,661 kg/ha for mungbean, 3,289, 553, and 2,055 kg/ha for cowpea, 3,931, 674, and 2,489 kg/ha for adzuki bean, 12,695, 969, and 7,642 kg/ha for maize, 17,071, 1,260, and 8,857 kg/ha for sorghum, 16,355, 1,163, and 8,543 kg/ha for sorghum $\times$ sudangrass hybrid, and 8,288, 929, and 4,091 kg/ha for Japanese millet. Soybean was higher in CP, ether extract (EE), and TON content but was lower in nitrogen free extract content compared with the three other legumes. The legumes had much higher CP (13.7 to 21.9%), EE (2.42 to 6.23%), and TDN (58.7 to 69.9%) content but lower in crude fiber (CF) content (17.3 to 25.3%) than did the grasses tested except maize which had relatively lower CF content but higher TDN content. These results suggest that soybean could be the best forage legume for grass-legume forage rotation in the Cheju region.

Antioxidant Properties of Adzuki Beans, and Quality Characteristics of Sediment according to Cultivated Methods (재배방법에 따른 팥의 항산화 특성 및 앙금의 품질 특성)

  • We evaluated the antioxidant properties of adzuki beans and the quality characteristics of sediment using various cultivation methods. There were significant differences in total polyphenol and flavonoid contents in beans grown using different methods of cultivation (p<0.05). Also, DPPH and ABTS radical-scavenging activities were significantly different depending on cultivation method (p<0.05). The sediment yield before drying of Chungju-pat, Hongeon, and Arari was 296.64~339.01, 271.36~282.24, and 268.21~292.32%, respectively, and the sediment yield after drying was 71.68~85.41, 77.90~85.19, and 74.15~78.65%, respectively. The L-value of Chungju-pat and Arari sediments revealed a significant difference given different cultivation methods (p<0.05), but Hongeon sediment did not show a significant difference. There was a significant difference in the a- and b-value of adzuki bean sediments cultivated using different methods (p<0.05). The particle size of Chungju-pat, Hongeon, and Arari sediments was 66.21~98.80, 61.62~97.07, and $82.96{\sim}106.71{\mu}m$, respectively, and all were significantly different depending on cultivation method (p<0.05). There were also significant differences in the water absorption index, water solubility index, and swelling power when different cultivation methods were used (p<0.05).

Effects of the Drainage Methods on Antioxidant Compounds and Antioxidant Activity of Ethanolic Extracts on Adzuki Bean (논 재배 배수방법이 팥 에탄올 추출물의 항산화성분 및 항산화활성에 미치는 영향)

  • This study investigated the changes of antioxidant compounds and antioxidant activity of adzuki bean by drainage methods in poorly drained sloping paddy field. The soil moisture contents of under pipe and open ditch drainage on very poorly drained paddy soil were $18.52{\pm}4.58$ and $19.01{\pm}4.25%$, and imperfectly drained paddy soil were $14.87{\pm}4.82$ and $18.64{\pm}3.85%$, respectively. Moisture, protein, fat and ash contents of adzuki bean with drainage methods were 10.10~11.60, 14.13~21.75, 0.02~0.73 and 2.81~3.45 g/100 g, respectively. The total polyphenol, flavonoid and tannin contents, and radical scavenging activity of adzuki bean showed significant differences by drainage methods. The total polyphenol, flavonoid, and tannin contents by drainage methods were 2.73~4.14 mg GAE/g, 1.07~1.43 mg CE/g, and 1.27~1.84 mg TAE/g, respectively. The DPPH and ABTS radical scavenging activities were 2.84~4.47 and 5.11~6.74 mg TE/g, respectively. The antioxidant compounds and radical scavenging activity of the adzuki bean by drainage methods were frequently affected soil water.

Yield and Antioxidant Properties of Korean Adzuki Bean (Vigna angularis L.) Cultivars Under Different Air Temperatures and Sunshine Hours (국내 팥 품종의 일조시간과 기온 차이에 따른 수량성 및 항산화 특성)

  • This study investigated the antioxidant and yield properties of eight adzuki bean (Vigna angularis L.) cultivars grown under different mean temperatures and sunshine hours in 2020 and 2021. Compared to 2021, which had a normal mean temperature, 2020 showed a lower mean temperature during the pod-extension and grain-filling periods. In addition, due to frequent rainfall during the vegetative period in 2020, there were fewer hours of sunshine in 2020 than in 2021. The adzuki bean yield was approximately 13% lower in 2020 than in 2021 due to the decreased number of pods per plant and 100-seeds weight. 'Hongda' and 'Hongjin' cultivars showed more stable yields than the 'Arari' variety, which is the most commonly cultivated variety in Korea. Moreover, the 'Hongda' and 'Hongjin' cultivars had higher yields than the 'Arari' variety in both years. Compared to 2021, in 2020, when the weather conditions were unfavorable, both antioxidant levels and activity were higher. 'Hongda' had higher antioxidant levels and activity than the other cultivars over the 2 years. These results suggest that the antioxidant levels and activity were good in 2020, with an approximately 13% decrease in yield compared to 2021, which was a normal year with respect to mean temperature and sunshine hours.
