Biological activities of decursin and decursinol, natural coumarin derivatives, on the germination, growth and adventitious root formation of several plants were observed. In 10 ppm of decursin or decursinol, the growth of Avena coleoptile sections was inhibited, and the activity of IAA-oxidase was gradually enhanced by the increase of its concentrations. Inhibition effect on seed germination was observed from 100 ppm of each chemcials, and the activity of amylase in the germinating seeds was also gradually decreased. However, in the higher concentrations of decursin, the inhibited germination rate of wheat was slightly reduced. Decursin and decursinol also promoted the adventitious root formation in seeds of Phaseolus vulgaris.
This paper reported the establishment of mass production system of adventitious roots of Codonopsis lanceolata through shake flask and bioreactor culture. Induction of adventitious roots was started from the explants of leaf, stem and root on 1/2 strength Murashing and Skoog (MS) solid medium. Stem segments were the best explants for induction of adventitious roots compared to leaf and root segments. Among the different auxins tested (NAA, IBA and IAA), number of adventitious root per explant was highest on solid medium with 1.0 mg/L IBA, and produced $9.9{\pm}1.2$ roots per explant. However, growth of adventitious roots was fast in the presence of IBA at low concentration (0.1 mg/L). In shake flask culture, maximum production of adventitious roots (fresh weight) was obtained in half-strength MS medium compared to full-strength and one-third MS medium. When the adventitious roots produced in shake-flask culture were transferred to 5 L air-lift bioreactor, 16 times of fresh weight increase was gained after one month of culture. These results indicate that this protocol for the production of C. lanceolata adventitious roots can be applied to large scale culture for practical application.

Viral infections and root growth were examined to elucidate the relationship between viral resistance and root growth in 26 Korean hulless barley cultivars. Viral resistance was estimated in experimental filed of Honam agricultural research institute for 3 years. Length and number of seminal and adventitious roots were examined for evaluation of root growth in both field and green-house conditions 30 days after seeding. Dominant viral infection occurred in Korean hulless barley by Barley yellow mosaic virus (BaYMV) in fields; however, susceptible cultivars were infected by either BaYMV, Barley mild mosaic virus (BaMMV) or both. Only four cultivars, including Donghanchalssalbori, Kwangwhalssalbori, Namhossalbori and Naehanssalbori, presented stable resistance to viral infections. Susceptible cultivars to viral infection in fields showed shorter seminal root length and fewer adventitious root number than resistant cultivars. Resistant cultivars showed better root growth and significant difference in adventitious root length in green house conditions. Increase in the number of seminal roots in resistant cultivars was derived from decreased damage of roots by the viral infection compared to the susceptible cultivars.

The study for the quantitative analysis of saikosaponin of Bupleurum falcatum root produced by tissue culture was attempted. The optimal concentration of 2,4-D was 0.1 mg/l for inducing the callus. The induction and growth of the adventitious root from the callus was obtained in the suspension culture containing MS basal medium supplemented with no plant growth regulators. The results of the quantitative analysis of saikosaponins by high performance liquid chromatography (HPLC) showed that the content of saikosaponin a and c was high in the one-year-old root from somatic embryos and that of saikosaponin d was remarkably high in three-months-old adventitious root from callus.
Three-day-old sunfower(Helianthus annuus L.) seedlings were do-rooted and incubated in the nutrient solutions that contained either deficient or sufficient boron supply, and exposure to various pH and temperatures. In the absence of boron, no adventitious roots were formed in the majority of the seedlings. Boron caused the development of numerous adventitious roots in the lower part of the hypocotyl. The low pH damage was ameliorated by the simultaneous application of calcium, implying the involvement of calcium-requiring process in overcoming proton toxicity. Borate showed a strong ameliorative effect with $Ca^{2+}$. These results suggest that the primary target of proton toxicity may be linked to a disturbance of the stability in the pectic polysaccharide network, where calcium plays a key role in plant roots. Maximum temperature for the growth of adventitious roots was $25^{\circ}C$. Although cessation of growth is the most apparent symptom of boron deficiency, exogenous ascorbate improved adventitious root growth in plants in the absence of boron. From the results it is suggested that the inhibition of adventitious root resulting from boron deficiency, low pH and temperature damage may be a consequence of disrupted ascorbate metabolism.
This study was carried out to investigate the effect of $NO_3{^-}$ and $NH_4{^+}$ on the adventitious root growth and eleuthroside synthesis of Eleutherococcus senticosus during 3 L-bioreactor culture. The change of medium component ratio was also measured during culture. The fresh weignt of adventitious root reached to the greatest level of 24.4g FW/L in the presence of 50 mM $NO_3{^-}$ and 10 mM $NH_4{^+}$, representing 3.4-fold increase compared to the 60 mM $NH_4{^+}$. However, as the increase of the portion of $NH_4{^+}$, the root growth was decreased. Maximum eleutheroside B and E1 production were $249{\mu}g/g$ and $43{\mu}g/g$, respectively, with 30 mM total nitrogen source. The maximum production of eleutheroside E were $788{\mu}g/g$ with 120 mM total nitrogen source. The greatest weight of adventitious root increased up to 6.2 fold of inoculum size within 9 weeks. The change of pH was influenced from 4.81 to 6.35 and the amounts of $NH_4{^+}$ and $K^+$ were decreased during culture periods. From these results we suggest, need further study of various treatment to increase the growth of biomass and the accumulation of useful secondary metabilites.
When the leaves, roots and stem segments of seedling of Polygonatum odoratum were cultured on Murashige and Skoog medium with 2.0mg/l BAP, stem segments were the most efficient explants for adventitious shoot inductino. To observe the efficient combination of growth regulators on the adventitious shoot formation , stem segments were cultured on MS medium with various kinds of cytokinins (BAP, kinetin, zeatin). From this experiment, cytokinin treatement was prerequisite for theadventitious shoot formatino, especially BAP was the most effective. Auxin (NAA or IBA) in combination with cyotokinin highly enhanced the adventitious shoot formation. Twenty five percents of explants produced the adventitious shoots on medium with 2.0mg/l BAP solely, while 83% of explants produced the adventitious shoots on medium with 2.0mg/l BAP and 0.1mg/l IBA. Root formationform adventitious shoot was promoted after transfer to 1/2 MS medium supplemented with 0.1mg/l IBA and 0.5mg/l zeatin, thereafter the plantlets with shoots and roots were cultured on 1/2MS medium lacking growth regulators. When the stem segments were cultured to MS medium with 1.0mg/l 2,4 NAA and IBA , yellow and nodulous cali were formed from the stem segments which were developed into adventitious roots. These roots were actively grew after transferred to MS liquid medium lacking growth regulators.

In this study, as a result of exploring the physiological activity of plants useful for agriculture on various plant resources, it was possible to confirm an activity similar to auxin that promotes plant rooting in methanol extract of Cinnamon Bark (cortex of Cinnamomum cassia J.Presl). After separating the active body by applying column chromatography and HPLC to the CHCl3 active fraction obtained by solvent extraction for each polarity from the methanol extract of cinnamon bark, cinnamyl alcohol was identified through GC/MS analysis. By bioassay using cinnamyl alcohol standard and the active fraction separated and purified from the methanol extract of cinnamon bark, the rooting rate of mung bean seedlings of the cinnamyl alcohol standard was 290% compared with the untreated control at 134.2 ㎍/mL concentration, and the adventitious root formation activity similar to the rooting rate (268.6%; 100 ㎍/mL) of the active fraction was shown. In conclusion, it is believed that cinnamyl alcohol contained in methanol extract of Cinnamon Bark is the main compound that induces adventitious root formation in mung bean.
In order to study the role of ethylene on the formation of adventitious roots, trichomes and calli, the effects of 1-aminocyclopropane-l-carboxylic acid (ACC), ethephon, $CoCl_2$ and $AgNO_3$ were investigated in the leaf segments from ecotype Columbia of Arabidopsis thaliana. When the leaf segments were cultured on the media for forming adventitious roots (0.1 mg/L NAA), trichomes (2.0 mg/L NAA) and calli (10.0 mg/L NAA), and then each cultures was treated with 1-100 mg/L of ACC and ethephon, respectively. On the adventitious root-forming medium adventitious root formation was decreased, and trichomes were induced. And on the trichome-forming medium trichome formation was decreased, and calli were induced. In order hand each culture was treated with 1-100 mg/L of $CoCl_2$ and $AgNO_3$, respectively. On the adventitious root-forming medium adventitious roots was increased without trichome formation, and on the trichome-forming medium trichome formation was decreased, and adventitious roots were induced. However on the callus-forming medium treated with ACC, ethephon, $CoCl_2$ and $AgNO_3$, respectively, callus formation was inhibited and trichomes were induced in all cultures.
This study was carried out to investigate the effect of ${NO_3}^-$ and ${NH_4}^+$ on the adventitious root growth and eleuthroside synthesis of Eleutherococcus koreanum in 5 L-bioreactor culture. The change in the medium components was also measured during culture. The fresh weignt of adventitious root reached to the highest level of 30.8 g FW/L in the presence of both 50 mM ${NO_3}^-$ and 10 mM $NH_4^+$, representing 3.6-fold increase compared to the 60 mM ${NH_4}^+$ alone. However, as the increase of the portion of ${NH_4}^+$, the root growth was decreased. However, the maximum eleutheroside B, E and E1 contents were $57.3{\mu}g/g$ DW, $188.4{\mu}g/g$ DW and $47.3{\mu}g/g$ DW, with 30 mM, 60 mM and 15 mM total nitrogen source, respectively. Fresh weight of adventitious root increased up to 6.8-fold of inoculum size within 9 weeks. The amounts of ${NH_4}^+$, $K^+$, ${NO_3}^-$ and ${PO_4}^-$ were decreased during culture periods. Based on these results, we suggest that various further studies are required to increase the biomass and the useful secondary metabilites.
