Adsorption performance of odorous compounds such as geosmin and 2-MIB on granular activated carbon were evaluated in this study. The coal-based activated carbon was found more effective than other carbons in adsorption of geosmin and 2-MIB. The wood-based virgin activated carbon was less effective than coconut- and coal-based carbon in adsorption nevertheless having larger pore volume and specific surface area than others carbons. The maximum adsorption capacity(X/M) of coal-based activated carbon for geosmin and 2-MIB was $1.2\sim1.9$ and $2.1\sim2.6$ times larger than coconut- and wood-based virgin activated carbon, respectively. Carbon usage rate (CUR) of coal-, coconut- and wood-based virgin activated carbons for geosmin and 2-MIB were 1.72 and 1.44 g/day, 1.72 and 2.05 g/day and 2.12 and 1.90 g/day, respectively. In the evaluation of adsorption isotherm of geosmin and 2-MIB for coal-, coconut- and wood-based virgin activated carbons, k value of 2-MIB was lower than geosmin, It menas 2-MIB is more difficult to remove by activated carbon adsorption than geosmin. The relationship of max. adsorption versus total pore volume of coconut- and wood-based virgin and used activated carbon for geosmin and 2-MIB were $y=264,459\times-79,047(R^2=0.95)$, $y=319,650\times-101,762(R^2=0.93)$.
An automated on-line portable preconcentration-short column gas chromatograph was developed, which used preconcentrator using adsorption tube with Tenax-GR and Curie-point heating. The developed system operated with 3 steps of processing, preconcentration, thermal desorption, and analysis and cleaning, and could continued operating within 1~2 min cycle. The recoveries of preconcentrator for toluene was ranged between $94.7{\pm}6.6%$ and $103.8{\pm}3.1%$ with less than 7% of RSD. For benzene, toluene and xylene(BTX) standard gas test, IDL was 41, 49, $472ng/m^3$ benzene, toluene and o-xylene, respectively. The BTX mixture was analyzed within 30 sec with baseline separation by the system equipped with 4 m long capillary column. The deficiency of separation power caused by short column was solved by the control of sample injection volume and inlet/outlet pressure ratio. The automated portable preconcentration-short column gas chromatograph system was found to be useful for the continuous air monitoring of BTX at ppb levels in ambient air.
Isolation and identification of pathogens from slaughter and meat processing plant were investigated. Antimicrobial activity of Amaranthus lividus against isolated pathogens such as Aeromonas sobria, Escherichia coli, Escherichia coli O157, Listeria monocytogenes, Salmonella spp., and Staphylococcus aureus was investigated. Among the chloroform, ethyl acetate and buthanol fraction of amaranthus lividus showed inhibitory effect against Aeromonas sobria CLFM1 and Escherichia coli CLFM2. Antimicrobial substance in chloroform fraction was isolated by silica gel adsorption column chromatography, sephadex LH-20 column chromatography and silica gel partition column chromatography. The antimicrobial compound of amaranthus lividus was identified as diethyl phtalate by HPLC, GC-MS, H-NMR and C-NMR.
In the our country, especially in Yeongil and Wolsung area, abundant authigenic zeolites are found from the tuffaceous sediments and volcanic rocks of Miocene age showing wide variation in their mineralogy and abundance from horizon to horizon. The principal zeolite species identified are clinopti-lolite. mordenite. heulandite. ferrierite, and erionite. etc. Zeolite minerals are widely used in many countries in the following applications; (a) in air separation adsorption processes; (b)as desiccants; (c)in inorganic building materials; (d)in papermaking; (e)in fertilizers; (f)as soilconditioners-this application is based upon the ability of the zeolite to ion exchange with soil nutrients; (g)in the treatment of radioactive wastes; and (h)as adsorbents for toxic gases, etc. In the present paper, using natural zeolite mordenite treated with IN hydrochloric acid or IN sodium chloride solution as column packings, separation characteristics of argon, nitrogen, carbon monoxide, and methane gases have been studied by gas chromatography. By the use of mordenite treated with hydrochloric acid solution, the tailing peak of methane showed from untreated mordenite was satisfactorily reduced, although it was difficult to separate it from carbon monoxide with a column activated at $300^{\circ}C$. Using a column activated at $350^{\circ}C$, methane could be separated from carbon monoxide easily but only carbon monoxide eluted as a bad defined peak. Mordenite treated with sodium chloride solution was generally similar to chromatograms obtained by using the untreated mordenite. Both the above chemical treatments of mordenite had little effect on the separations of argon and nitrogen. The separations and the HETP values obtained from natural zeolite mordenite treated with continuously hydrochloric acid and sodium chloride solutions were almost identical with those obtained with synthetic molecular sieve 5A zeolite. On the other hand, the efficiency of column was good in the range 20~3Oml/min of the carrier helium gas rate.
Applying enhanced coagulation is not uncommon not only to improve water quality but also to obtain economical advantages in water treatment. In this study, the economical evaluation was investigated with considering dosage of coagulant, produced sludges and usage of activated carbon with two different water sources. To simulate the granular activated carbon(GAC) adsorber mini-column test was adapted. Even if there were some variation according to the source waters, the life time of GAC was extended 60 to 110days with adding 2.5 to 3 times of coagulant in enhanced coagulation. With assumption of changing GAC adsorber at 50% of breakthrough the total treatment cost can be reduced to range from 25% to 44% with adapting the enhanced coagulation. The obtained value can not be generalized because the value can be varied by water quality and cost of the GAC or coagulant. However, the evaluation can be basis starting point to find optimum operation methods for the drinking water treatment which has plan to adapt GAC adsorber.
There are growing needs to produce relatively high purity(99.0% or higher) oxygen at low cost. For small scale production, both pressure swing adsorption(PSA) and membrane process are competitive and less expensive or more convenient than well known cryogenic fractionation technology. A continuous membrane column(CMC) combined with a PSA oxygen generator can be employed to produce high purity oxygen continuously. The oxygen enriched gas generated by a PSA unit, with a concentration of 93~94%, is fed to the CMC that consism of three modules of poly(imide) hollow fibers. Several experiments were conducted by varying parameters, such as feed flow rate, transmembrane pressure drop, stage cut, and feed location in order to obtain a high oxygen concentration above 99.0%. A two-series unit mode was also employed with CMC operation to optimize the given membrane area.
Molecularly imprinted microsphere for chloramphenicol (CAP) with high adsorption capacity and excellent selectivity is prepared by aqueous suspension polymerization, in which chloramphenicol is used as template molecule and ethyl acetate as porogen. The CAP-imprinted microspheres are used as high performance liquid chromatography (HPLC) stationary phase and packed into stainless steel column ($150mm{\times}4.6mm$ i.d.) for selective separation of chloramphenicol. HPLC analysis suggests that chloramphenicol can be distinguished from not only its structural analogs but also other broad-spectrum antibiotic such as erythromycin and tetracycline. In addition, the binding experiments of CAP-imprinted microspheres are carried out in ethanol/water (1:4, V:V), the results indicate that the maximum apparent static binding capacity of molecularly imprinted microspheres is up to 66.64 mg $g^{-1}$ according to scatchard model.
A strain of Streptomyces sp. (YS-221-B) extracellularly produced an inhibitory substance for $\alpha$-D-Glucosidase. The substance was purified 96-fold from culture filtrate by dialysis, heat treatment, adsorption on active carbon, Bio-Gel P-10 and Sephadex G-75 column chromatography with yield of 9.2%. The substance was stable in pH range from 7.0 to 11.0 at 37$^{\circ}C$, and a treatment at 10$0^{\circ}C$ for 20 min diminished only 15% of the original activity. The inhibitor was not inactivated by the treatment of $\alpha$-, $\beta$-amylases, glucoamylases, trypsin and chymotrypsin but inactivated by pyoteases from Streptomyces griseus and Tritirachium album.
Glycopeptide antibiotics, teicoplanin was purified from a mutant strain of Actinoplanes teichomyceticus ATCC31121, A. teichomyceticus MSL2211. We developed a simple procedure to separate and purify the teicoplanin from the fermentation broth. Teicoplanin was purified by two-step purification system, hydrophobic adsorption and sugar affinity chromatography in combination with HPLC analysis based on the properties of hydrophobic acyl chain and sugar moiety in teicoplanin. Teicoplanin was separated from the culture broth by Diaion HP-20 and further purified by concanavalin A affinity column chromatography. As an adsorbent resin, Diaion HP-20 in broth eliminated toxic effects on growth, reduced feedback repression of teicoplanin production, and assisted In rapid recovery of teicoplanin. The teicoplanin displayed the final yield of 80% and 95% of purity.
The performance of one-column isothermal PSA process is assessed by dynamic simulator, gPROMS. The four-step and five-step processes are compared. A five-step process is employed in order to show the effect of the addtional cocurrent depressurization step on the four-step PSA process. Two processes parameters, purity and recovery of SO$_{2}$ are used for the performance comparison. The results of dynamic simulation show that four-step process is superior to five-step process in recovery, but not in purity.
