Piper nigrum Linne has been used spice as well as herbal medicine in worldwide and has function of anti-oxidant, anti-inflammation, anti-cancer, bioavailability of drugs and etc. In this study, anti-adipogenic activity of Piper nigrum Linne extract and its constituent piperine were investigated in 3T3-L1 differentiation. Adipogenic effects and lipid accumulation in 3T3-L1 was measured by RT-PCR and Oil Red O staining assays respectively in stimulation of Piper nigrum Linne extract and piperine. Piper nigrum Linne and piperine reduced lipid accumulation in 3T3-L1 differentiation and expression of genes associated with adipogenesis such as PPAR${\gamma}$, adipsin, SERBP-1c and LPL. All of taken, anti-adipogenic mechanism of Piper nigrum Linne and piperine were related with regulation of SREBP-1c and PPAR${\gamma}$ expression.
본 연구는 배암차즈기 에탄올 추출물(SPE)을 유효성분으로 함유하는 지방 생성 및 축적 저해 효능을 조사하였다. 배암 차즈기 에탄올 추출물은 마우스 배아 섬유아세포(mouse embryo fibroblast) 유래 지방세포인 3T3-L1에서 지방세포 분화를 억제하는 효능을 보유하고 있었으며, 지방세포 내 중성지방의 농도를 감소시키는 효능을 보유하고 있었다. 또한, $PPAR{\gamma}$, $C/EBP{\alpha}$, SREBP-1c, pACC, pAMPK, CPT-1, 지방산 합성효소(FAS, fatty acid synthase) 발현 억제, 호르몬자극지방분해효소(HSL, hormone sensitivity lipase) 활성화 등 지방합성 관련인자들의 발현을 조절하는 효능을 보유하고 있는 것으로 확인되었다. 이상의 결과로 SPE가 지방세포 분화 및 지방대사에 관련된 인자들의 발현을 조절함으로써 지방 생성 및 지방 축적을 저해하는 효능을 보유하기 때문에 배암차즈기를 활용한 비만 개선을 위한 소재로서의 활용이 가능할 것으로 보인다.
본 연구에서는 괭생이모자반 에탄올 추출물의 항비만 효과에 대해 알아보고자 3T3-L1 preadipocyte에서 Oil Red O staining과 triglyceride content, adipogenesis의 발현에 대해 조사하였다. MTT assay를 통해 괭생이모자반 에탄올 추출물의 세포 독성을 확인 해 본 결과 $200{\sim}1,000{\mu}g/ml$ 농도에서 90% 이상의 세포 생존율을 확인 할 수 있었다. 또한 insulin, dexamethasone과 IBMX 복합물을 처리하여 분화를 유도하였을 때는 분화유도제를 처리하지 않은 군에 비해 활발하게 지방구를 형성하였으나, 괭생이모자반 에탄올 추출물을 250, 500, $1,000{\mu}g/ml$ 농도로 처리하였을 때는 농도 의존적으로 지방구 형성을 억제하였다. 이를 정량하여 triglyceride content (%)로 나타낸 결과 농도별로 13%, 16%, 23% 억제 효과를 나타내었다. 또한 Western blot assay를 통해 adipogenic transcription factor의 발현양상을 비교해 본 결과, 분화유도제 처리에 의해 $C/EBP{\alpha}$, $C/EBP{\beta}$와 $PPAR{\gamma}$의 발현이 증가하였으나, 괭생이모자반 에탄올 추출물을 처리하였을 때는 이러한 전사인자들의 발현이 농도 의존적으로 감소됨을 확인 할 수 있었다. 이상의 결과를 통해 괭생이모자반 에탄올 추출물은 adipogenic transcription factor의 발현을 조절하여 지방세포의 분화를 억제하였으며, 지방구와 triglyceride 생성을 감소시켜 최종적으로 비만억제 효과를 나타낸다는 것을 알 수 있었다.
유근피(Cortex ulmi pumilae)는 항산화, 항염증, 항균, 항암 등 다양한 생리활성이 보고되어 있으며, 뛰어난 소염효과로 예로부터 약재로 사용되어 왔다. 본 연구에서는 3T3-L1 지방전구세포를 이용하여 유근피 추출물의 지방세포 분화에 미치는 영향과 그 기전을 검토하였다. 유근피 추출물을 분화유도제와 동시에 처리한 결과, 3T3-L1 지방전구세포의 세포질 내 triglyceride 생성을 농도의존적으로 감소시켰으며, 지방세포분화 관련 전사인자인 $C/EBP{\alpha}$, $C/EBP{\beta}$ 및 $PPAR{\gamma}$의 발현을 유의적으로 저하시켰다. 다음으로 유근피 추출물을 지방세포 분화 유도 단계별로 처리한 결과, 분화유도제와 동시에 처리한 경우 즉 분화 초기에 처리한 경우가 가장 효과적인 지방세포 분화 억제효능을 보였으며, 이 때 분화초기에 발현되는 $C/EBP{\beta}$의 발현량은 현저하게 감소되었다. 또한 유근피 추출물에 의해 mitotic clonal expansion 단계의 세포증식이 농도의존적으로 억제되는 것을 확인하였으며, 세포주기분석을 통해 G1 arrest를 유발함으로써 증식이 억제 된다는 것을 확인하였다. 또한, G1 arrest는 G1/S 전이 관련 단백질인 p21의 발현 증가, cyclin E의 발현 감소 및 Rb의 인산화 감소에 의한 것임을 확인하였다. 이러한 연구결과들로부터 유근피 추출물은 지방세포 분화 초기 단계에 작용하여 지방세포 분화 관련 전사인자의 발현을 억제하고 mitotic clonal expansion을 저해하여, 지방전구세포의 지방세포로의 분화를 억제하는 항비만 효능을 가지는 천연소재임을 확인하였으며, 따라서 본 연구는 향후 이를 활용한 지속적인 연구를 위한 기초자료로 그 가치가 매우 높을 것이라 생각된다.
Mesenchymal stem cells (MSCs) are multipotent stem cells capable of differentiating into adipocytes, osteoblasts, or chondrocytes. A mutually inhibitory relationship exists between osteogenic and adipogenic lineage commitment and differentiation. Such cell fate decision is regulated by several signaling pathways, including Wnt and bone morphogenetic protein (BMP). Accumulating evidence indicates that microRNAs (miRNAs) act as switches for MSCs to differentiate into either osteogenic or adipogenic lineage. Different miRNAs have been reported to regulate a master transcription factor for osteogenesis, such as Runx2, as well as molecules in the Wnt or BMP signaling pathway, and control the balance between osteoblast and adipocyte differentiation. Histone modifications on major transcription factor target genes are one of the major regulatory mechanisms controlling adipogenesis. Plant homeodomain finger 2 (PHF2) is a Jumonji domain-containing protein and is known to demethylate the histone H3K9, a repressive gene marker. To better understand the function of PHF2 in adipocyte differentiation, we constructed stable PHF2 knock-down cells by using the mouse pre-adipocyte cell line 3T3-L1. When induced with adipogenic media, PHF2 knock-down cells showed reduced lipid accumulation compared to control cells. Differential expression using a cDNA microarray revealed significant reduction of metabolic pathway genes in the PHF2 knock-down cell line after differentiation. The reduced expression of major transcription factors and adipokines was confirmed with reverse transcription- quantitative polymerase chain reaction and Western blotting. We further performed co-immunoprecipitation analysis of PHF2 with four major adipogenic transcription factors, and we found that CCATT/enhancer binding protein (C/EBP)${\alpha}$ and C/EBP${\delta}$ physically interact with PHF2. In addition, PHF2 binding to target gene promoters was confirmed with a chromatin immunoprecipitation experiment. Finally, histone H3K9 methylation markers on the PHF2-binding sequences were increased in PHF2 knock-down cells after differentiation. Together, these results demonstrate that PHF2 histone demethylase controls adipogenic gene expression during differentiation.

Hormonal and nutrient signals regulate leptin synthesis and secretion. In rodents, leptin is stored in cytosolic pools of adipocytes. However, not much information is available regarding the regulation of intracellular leptin in ruminants. Recently, we demonstrated that leptin mRNA was expressed in bovine intramuscular preadipocyte cells (BIP cells) and that a cytoplasmic leptin pool may be present in preadipocytes. In the present study, we investigated the expression of cytoplasmic leptin protein in BIP cells during differentiation as well as the effects of various factors added to the differentiation medium on its expression in BIP cells. Leptin mRNA expression was observed only at 6 and 8 days after adipogenic induction, whereas the cytoplasmic leptin concentration was the highest on day 0 and decreased gradually thereafter. Cytoplasmic leptin was detected at 6 and 8 days after adipogenic induction, but not at 4 days after adipogenic induction. The cytoplasmic leptin concentration was reduced in BIP cells at 4 days after treatment with dexamethasone, whereas cytoplasmic leptin was not observed at 8 days after treatment. In contrast, acetate significantly enhanced the cytoplasmic leptin concentration in BIP cells at 8 days after treatment, although acetate alone did not induce adipocyte differentiation in BIP cells. These results suggest that dexamethasone and acetate modulate the cytoplasmic leptin concentration in bovine preadipocytes.
In addition to spice, black pepper (Piper nigrum Linne : PnL) has been used as herbal medicine because of its function in anti-oxidation, anti-inflammation, and anti-carcinogenesis. Recently, it has been reported that piperine, a component of PnL, inhibits adipocyte differentiation by repressing various adipogenic gene expressions. In this study, we determined whether piperine is a major constituent of PnL that confers the anti-adipogenic activity at whole genome level. Differentiation of 3T3-L1 pre-adipocytes was induced in presence of PnL extract or piperine. To compare genes that are regulated by PnL extract or piperine, we performed expression profiling using microarrays (Agilent Mouse 44k 4plex). RNA samples were labeled with Cy3 and Cy5, respectively. Labeled samples were hybridized to the microarrays. Results were filtered and cut off set p<0.05. Genes exhibiting significant differences in expression level were classified into Gene Ontology (GO)-based functional categories (http://www.geneontology.org) and KEGG (http://www.genome.jp/kegg/). Extract of PnL and its component piperine reduced lipid accumulation in 3T3-L1 cells during adipogenesis. Such anti-adipogenic activity appears to result from down-regulation of transcription factor genes involved in adipogenesis, and other genes involved in fatty acid synthesis, transport, triglyceride synthesis, and carbohydrate metabolism. These genome-wide studies lead to conclude that piperine, as a critical component of PnL, plays common role with PnL in anti-adipogenesis.
In our previous study, we have shown that berberine has both anti-adipogenic and anti-inflammatory effects on 3T3-L1 adipocytes, and the anti-adipogenic effect is due to the down-regulation of adipogenic enzymes and transcription factors. Here we focused more on anti-inflammatory effect of berberine using real time RT-PCR and found it changes expressions of adipokines. We hypothesized that anti-adipogenicity of berberine mediates anti-inflammtory effect and explored leptin as a candidate mediator of this signaling. We studied this hypothesis by western blot analysis, but our results showed that berberine has no effect on the phosphorylations of STAT-3 and ERK which have important roles on leptin signaling. These results led us to conclude that the anti-inflammatory effect of berberine is not mediated by the inhibition of leptin signal transduction. Moreover, we have found that berberine down-regulates NF-${\kappa}B$ signaling, one of the inflammation-related signaling pathway, through western blot analysis. Taken together, the anti-inflammatory effect of berberine is not mediated by leptin, and berberine induces anti-inflammatory effect independent of leptin signaling.
