Improved Cell Viability and Anti-Candida Activity of Probiotic Lactobacillus salivarius MG242 by Heat Adaptation (Lactobacillus salivarius MG242의 열 전처리시 생존율 증진 및 항 캔디다 효과)

  • Vulvovaginal candidiasis is a major urogenital infection in women. Lactobacilli are important in maintaining vaginal health. In the present study, the effect of heat adaptation at $47{\sim}52^{\circ}C$ prior to heat stress at $60^{\circ}C$ in improving the viability of Lactobacillus salivarius MG242 was examined. L. salivarius MG242 has antifungal effects against Candida albicans. Heat-adapted cells had a higher survival rate than non-adapted cells during the subsequent heat stress. When chloramphenicol was added during the adaptation process, heat tolerance was abolished, suggesting the involvement of de novo protein synthesis with the heat adaptation of L. salivarius MG242 strain. Exopolysaccharide quantification and scanning election microscopy did not reveal any appreciable changes during heat adaptation. The antifungal activity of L. salivarius MG242 against C. albicans was maintained during the heat adaptation. These results suggest that heat adaptation can be applied for the development of probiotic products using L. salivarius MG242 to improve its stress tolerance during processing.

The Relationship between University life Adaptation, Sex and Gender Role Identity (대학 생활적응과 성별 및 성 역할 정체감의 관련성 연구)

  • The purpose of this study was to examine the effects of sex and gender role identity on college life adaptation. The participants were 142 male and 146 female students. The university life adaptation scale and gender role identity scale were carried out, and we conducted a parallel multiple mediator model using Process macro. As a result, while the effect of sex on adaptation level and resources was significant, the explanatory power increased to a meaningful level when effect of gender role identity was considered. In the adaptation level, the effects of masculinity and femininity are similar in magnitude, but in the resources, masculinity has a more positive effect. Finally, male students were mediated through masculinity and female students were mediated through femininity, which has a positive effect on adaptation. These results suggest a differentiated guidance direction according to sex and identity, and suggest that enterprising and challenging spirit is important.

Effect of Cold Adaptation on the Improved Viability of Lactobacillus crispatus KLB46 (Lactobacillus crispatus KLB46의 생균제제화를 위한 저온 전처리시 증지의 효과)

  • Lactobacilli have been considered to play important roles in the health of human vagina. They secrete inhibitory substances to prevent vaginal infection by pathogenic organisms. In a previous study, we have isolated several lactobacilli from Korean woman and one of them (KLB46) was selected and indentified as Lactobacillu crispatus which showed high antimicrobial activity. In this study. cold adaptation prior to subsequent stresses exposure was examined whether L. crispatus KLB46 maintain the viability better than the non-adapted calls under stresses. For pharmaceutical formulation, the lyophilization process is required where stresses such as freezing/thawing and dehydration are routinely applied. Formulated L. crispatus KLB46 can be used for ecological treatment of bacterial vaginosis. The response of cold-adapted cells to other environmental stresses such as acid, heat, ethanol, NaCl, and H$_2$O$_2$ was also examined. The results showed that cold-adapted cells maintained higher survival rate compared with the non-adapted cells (freezing-thawing. 3-folds; dehydration: 3-folds; acid, 3-folds; heat, 10-folds). However, we did net observe any positive effect of cold adaptation on other stresses such as ethanol, NaCl and H$_2$O$_2$. When chloramphenicol was added during cold adaptation, adaptation effect was abolished. This confirms that de novo protein synthesis is necessary during the adaptation process. Moreover, we have identified cold shock protein homolog that codes for a major cold shock protein by PCR amplification using degenerate primers.

Adaptation Experience among Hemodialysis of Women with End-Stage Renal Disease (여성 말기신부전 환자의 혈액투석 적응경험)

  • Purpose: This study was a qualitative study to explore and understand the adaptation experiences of hemodialysis among women with End-Stage Renal Disease (ESRD) and to develop a substantive theory using the grounded theory method. Methods: Participants were 15 female patients who underwent hemodialysis for ESRD treatment from three general hospitals. The data were collected through in-depth individual interviews. Results: The adaptation experience of participants was emerged as a process of taking care and enduring. There were four adaptation stages as a negative, despair, receptive, and maintenance period in reference to hemodialysis. The causal conditions were a vague expectations of recovery and refusal to undergo hemodialysis. The core phenomenon was that of confinement to dialysis machine. The contextual conditions for this phenomenon were the loss of femininity. They used action/interaction strategies such as transition their life with a focus on hemodialysis, seeking information, and learning how to take care of their body. Through this process, they had a strong will to live or had sustained their life. Conclusion: These results indicate that there is a need for nurses to understand the different steps of adaptation experiences of the given patient population. It is necessary for nurses to support them to lead their life as much normal as possible and improve the adaptation experience of ESRD.

Improved Viability and Proteome Analysis of Lactobacillus fermentum KLB12 upon Heat Stress (Lactobacillus fermentum KLB12의 열 전처리에 따른 열 스트레스 내성 증진 및 프로테옴 변화)

  • In the previous study, we have isolated several vaginal lactobacilli from Korean woman and selected one of them (KLB12) for further study, which was indentified as Lactobacillus fermentum by sequence analysis of 16S rRNA gene. Formulated L. fermentum KLB12 can be used for ecological treatment of bacterial vaginosis. For pharmaceutical formulation, the spray-drying process is required where stress such as high temperature is routinely applied. In this study, we found that heat stress at 60$^{\circ}C$ for 20∼30min reduced the viable cell population of KLB12 by 10$\sub$6/~10$\sub$9/. However, adaptation of KLB12 cells at 52$^{\circ}C$ made them more thermotolerant upon exposure to 60$^{\circ}C$. The level of thermal protection in RSM (reconstituted skim milk) by adaptation in acid (pH 5), cold (4$^{\circ}C$), ethanol (3%), NaCI (0.3M) was also examined. Although not as efficient as the homologous stress, adaptations in both cold and NaCI gave considerable cross protection against heat stress. When chloramphenicol was added during heat adaptation, adaptation effect was abolished. This suggests that de novo protein synthesis is necessary during the adaptation process. Important changes in proteome during heat adaptation was examined with two-dimensional gel electrophoresis.

Korean Cultural Adaptation of WHODAS 2.0 (36-Item Version): Reliability and Linking to ICF

  • Purpose: This study was to conduct a Korean cultural adaptation of the WHO disability assessment schedule (WHODAS) 2.0: 36-item version. Methods: An internationally standardized process of translation and cultural adaptation of an instrument was used to develop a Korean version of WHODAS 2.0: 36-item version. Linking each item into the International Classification of Functioning, Disability and Health (ICF) was also conducted in order to ensure the concepts in the translated instrument were compatible with ICF. All translated versions of the instrument, linking results and feedback from participants were used for the final adaptation of the Korean version of the instrument. The Korean version of the instrument was assessed twice on different occasions to examine Inter- and intra-rater reliability, and the intra-class correlation coefficient (ICC) was calculated. Results: Twelve participants were involved in the translation and linking process. Ninety-five volunteers were invited to participate to examine the reliability of the instrument. Fifty participants completed the self-rated version of the instrument and 45 finished the interviewer version. The Korean WHODAS 2.0: 36-item version was found to have excellent reliability: self-rated version and interviewer version reliability coefficients were ICC=0.92 and ICC=0.94, respectively. Thirty-four items of the translated instrument were to be linked to ICF categories. Some adaptation was made; details and a familiar example were added to help respondents answer the questions. Conclusion: The study results show that the adaptation of the 36-item version of WHODAS 2.0 to Korean was successful and the instrument is ready for use in testing its psychometric properties.

  • Lactobacilli, the dominant species of microorganisms in the vaginal flora of healthy women, play important roles to prevent bacterial vaginosis and other sexually transmitted diseases. In this study, we carried out studies on stress adaptation prior to various stress treatment. We found that heat or salt adapted KLB46 showed higher cell viability than non adapted upon heat stress at $60^{\circ}C$ for 20 min. When chloramphenicol was added during the adaptation process, heat tolerance was abolished. This result suggested that de novo protein synthesis was essential during adaptation.

A Metadata-enabled Approach for Scalable Video Streaming in Heterogeneous Networks

  • In today's pervasive computing environments, multimedia content should be adapted to meet various conditions of network connections, terminals, and user characteristics. Scalable Video Coding (SVC) is a key solution for video communication over heterogeneous networks, where user terminals have different capabilities. This paper presents a standard-compliant approach that adapts an SVC bitstream to support multiple users. The adaptation problem is formulated as an optimization problem, focusing on the tradeoff between qualities of different spatial layers of an SVC video. Then the adaptation process is represented by standard metadata of MPEG-21, which can be solved by universal processing to enable interoperable and automatic operation. Our approach provides the users with optimal quality, a wide flexibility, and seamless adaptation. To the best of our knowledge, this is the first study that shows the adaptation tradeoff between spatial layers of a conforming SVC bitstream.

Error elimination for systems with periodic disturbances using adaptive neural-network technique (주기적 외란을 수반하는 시스템의 적응 신경망 회로 기법에 의한 오차 제거)

  • A control structure is introduced for the purpose of rejecting periodic (or repetitive) disturbances on a tracking system. The objective of the proposed structure is to drive the output of the system to the reference input that will result in perfect following without any changing the inner configuration of the system. The structure includes an adaptation block which learns the dynamics of the periodic disturbance and forces the interferences, caused by disturbances, on the output of the system to be reduced. Since the control structure acquires the dynamics of the disturbance by on-line adaptation, it is possible to generate control signals that reject any slowly varying time-periodic disturbance provided that its amplitude is bounded. The artificial neural network is adopted as the adaptation block. The adaptation is done at an on-line process. For this , the real-time recurrent learning (RTRL) algoritnm is applied to the training of the artificial neural network.
