The Effects of Peucedani Radix Herbal Acupuncture at St36 on Ovalbumin-induced Asthma in C57BL Mouse (전호약침(前胡藥鍼)이 Ovalbumin-induced Asthma Mouse Model에 미치는 영향)

  • Objectives : The aim of this study was to investigate the effect of Peucedani Radix herbal acupuncture(PR-HA) at St36(joksamni) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks(once a week) C57BL/6 mice were injected, inhaled and sprayed with OVA for 12 weeks (3times a week). One of the two experimental groups was just treated with needle-prick on St36 and the other group was treated with 1% concentrations of PR-HAS at St36, for the later 8 weeks (3times /week). Results : 1. The weight and total cells of lung of the mice group treated with PR-HA decreased significantly compared with those of control group. 2. Total Leukocytes and Eosinophils in BALF of the mice group treated with PR-HA decreased significantly compared with those of control group. 3. Eosinophils in BALF of the mice group treated with PR-HA in Photomicrographs decreased significantly compared with those of control group. 4. According to Histological analysis of lung sections, it decreased significantly adhension of collagen in PR-HA than those of control group 5. The concentration of IgE, IL-4, IL-5, in BALF and IL-4, IL-5, Il-13 in serum of the mice group treated with PR-HA decreased significantly compared with that of control group. 6. The number of Gr-1+/CD11b+, CD11b+, CD3-/CCR3+, CD4+, CD3e+/CD69+ , CD23+B220+ cells in the lungs of the mice group treated with PR-HA decreased significantly compared with those of control group. 7. The cytokine's manifestation of mRNA of the mice group treated with PR-HA with RT-PCR decreased significantly compared with that of control group. Conclusion : We conclude that PR-HA is effective on OVA-induced asthma of C57BL/6 mouse.

Single-dose Toxicity of Guseonwangdo-go Glucose 5% Intravenous Injection in a Rat Model

  • Objectives: The purpose of this study was to examine the single-dose intravenous toxicity of Guseonwangdo-go glucose 5% pharmacopuncture (GWG5). Methods: Forty Sprague-Dawley rats were divided into four groups of five males and five females per group: an intravenous (IV) injection of 1.0 mL of normal saline solution per animal was administered to the control group; IV injections of 0.1, 0.5, and 1.0 mL of GWG5 per animal were administered to the experimental groups (G: 0.1, G: 0.5, and G: 1.0). Observation of clinical signs and body weight measurements were carried out for 14 days following the injections. At the end of the observation period, hematological, biochemical, and histopathological tests, as well as necropsy examinations, were performed on the injected parts. Results: No mortalities or adverse clinical signs were observed in any of the groups. The body weights of all groups continuously increased. In the hematological and the biochemical tests, females in G-0.1 had minimal changes, but those changes were not dose dependent. On necropsy examination, no abnormalities were observed. In the histopathological test, focal inflammatory cell infiltrations were observed in two female rats, one in the control group and one in G-1.0. Also, one female rat in the control group had an epidermis crust. These changes were concluded to have been caused by the insertion of the needle into a vein. Conclusion: The above findings suggest that the lethal dose of GWG5 administered via IV injection is more than 1.0 mL per animal in both male and female rats. Further studies are needed to establish more detailed evidence of its toxicity.

The Study of Saamchimbeop's Method of Reinforcement and Reduction (사암침법(舍巖鍼法)의 보사수기법(補瀉手技法)에 관한 연구(硏究))

  • Objectives : The purpose of this study is what Saamchimbeop's method of reinforcement and reduction. Methods : 1. We reffered to the Bo-Sa method of DongeuiBo-gam(東醫寶鑑), Uihakim-mun(醫學入門), Uihakjeong-jeon(醫學正傳), Chimgugyeongheom-bang(鍼灸經驗方), Biaoyou-fu(標幽賦) in Cimgudaeseong(鍼灸大成), Nei-Jing(內經). 2. We make a conjecture that Zheng(正), Ying(迎), Sui(隨), Xie(斜) Yingzheng(迎正), Duo(奪), Zhenghuoxie(正或斜), Wen(溫), Liang(凉), JongYang-Inyin (從陽引陰) in Saamchimbeop are another expression of method of reinforcement and reduction and compared with the method of reinforcement and reduction of DongeuiBo-gam(東醫寶鑑), Uihakim-mun(醫學入門), Uihakjeong-jeon(醫學正傳), Chimgugyeongheom-bang(鍼灸經驗方), Biaoyou-fu(標幽賦) in Cimgudaeseong(鍼灸大成), Nei-Jing(內經). Results : 1. Zheng(正) and Xie(斜) are angle of acupuncture manipulation. The descending inserting of Yang-meridian is acupuncture manipulation for the Tonifying effect(補法) and the direct inserting of Yin-meridian is the Dispersing effect(瀉法). 2. JongYang-Inyin(從陽引陰) is the contralateral acupuncture. 3. Ying(迎) and Sui(隨) in the Saamchimbeop are same meaning the method of reinforcement and reduction(補瀉手技法). 4. Saamchimbeop's the final aim is the Wen-Liang(溫凉) according to the disease strong and weak in the Ohaeng-seo of Saam-acupuncture. Conclusions : Saamchimbeop's method of reinforcement and reduction is reinforcement-reduction by lifting and thrusting the needle, breathing reinforcement-reduction method, reinforcing and reducing achieved by rapid and slow insertion and withdrawing of the needles, reinforcement and reduction by opening and closing of needles with contralateral acupuncture by Yin-meridian or Yang-meridian. Saamchimbeop's the final aim is the Wen-Liang(溫凉) according to the disease strong and weak.

The Changes of NO, nNOS, Norepinephrine by Acupucture at LU7, HT5, PC6 Acupoints in Rats (수삼음경의 락혈 침자가 백서의 혈위 조직내 nNOS. NO와 조직 및 혈장 Norepinephrine의 변화에 미치는 영향)

  • Objectives : A previous study demonstrated that the connecting points of three yang meridians attenuated changes of nNOS, and Norepinephrine(NE) in rats. The current study investigated the changes in nNOS, NO and NE upon the needle insertion at varying depths at the connecting point of three yin meridians of the hand. Methods : Needles were inserted into rats, on both left and right sides of the connecting point, including the LU7, HT5 and PC6 acupoints which are three yin meridians of the hand. After insertion, needles were retained for five minutes. Each acupuncture groups were treated acupuncture at each acupoint and at the depths of superficial, middle and deep layer. After the retention, blood was drawn via cardiac puncture, and tissues of each point near meridian vessel was extracted to examine the changes in the expression of nNOS, NO and NE. Results : Compared with the superficial layer group, nNOS production significantly decreased in the middle and deep layer at LU7 acupoint group and in the deep layer at HT5, PC6 acupoint group. The tissue NE decreased in the deep layer on PC6 acupoint and the plasma NE increased at the middle layer at LU7 acupoint group but decreased at the deep layer on at LU7 acupoint group. Conclusions : Acupuncture at connecting points of three yin meridians of the hand can regulate the activities of nNOS, and NE.

Effects of Herbal-Acupuncture with Evodiae Fructus at KI10 on Osteoporosis in Ovariectomized Mice (음곡(陰谷) 오수유(吳茱萸) 약침(藥鍼)이 난소적출(卵巢摘出)생쥐의 골다공증(骨多孔症)에 미치는 영향(影響))

  • Objective : This study is aimed to evaluate the effects of Evodiae Fructus herbal-acupuncture (EF-HA) at KI10 on osteoporosis induced by ovariectomy in mice. Method : Mice underwent bilateral ovariectomy. After recovering, the ovariectomized (OVX) mice were treated by needle prick, saline injection, herbal acupuncture with Evodiae Fructus (EF-HA) at KI10 for 8 weeks. Result : 1. EF-HA at KI10 significantly inhibited the overgrowth of tibia in ovariectomized mice. 2. NP at KI10 significantly restored the tibial BMD (bone mineral density) in ovariectomized mice. 3. EF-HA at KI10 significantly restored the phosphorus and creatinine levels in ovariectomized mice serum. 4. EF-HA at KI10 significantly restored the tibial Ca and P levels in ovariectomized mice. 5. EF-HA at KI10 significantly reduced the tibial osteoclast-like cells in ovariectomized mice. 6. EF-HA at KI10 significantly inhibited the overgrowth of tibial GPL (growth plate length) in ovariectomized mice. Conclusion : EF-HA at KI10 has protective and therapeutic effect for osteoporosis in ovariectomized mice. Thus, it is suggested that EF-HA can be an useful therapeutics in clinical field after further researches.

Effects of Acupuncture at Varying Depths at the Connecting Point on the Changes of Levels of nNOS, No and Norepinephrine in Rats (수삼양경 락혈의 깊이별 침자가 백서의 nNOS, NO 및 Norepinephrine의 변화에 미치는 영향)

  • Objectives : This study was intended to observe the changes in the expression of neurotransmitters, such as nNOS, NO and NE upon the needle insertion at varying depths at the connecting point. Methods : Needles were inserted into rats, on both left and right sides of the connecting point, including the LI6, SI7 and TE5 acupoints which are three yang meridians of the hand. After insertion, needles were retained for five minutes. Each acupuncture groups were treated acupuncture at each acupoint and at the depths of superficial, middle and deep layer. After the retention, blood was drawn via cardiac puncture, and tissues of each point near meridian vessels were extracted to examine the changes in the expression of nNOS, NO and NE. Results : In terms of the effect in nNO production, there was a significant increase only in the middle and deep layer at SI7 acupoint, but there was no significant change in the expression of NO. Regarding the formation of norepinephrine within tissues, the middle layer on LI6 acupoint, the middle layer and the deep layer on TE5 acupoint showed a significant increase, while production of plasma norepinephrine was significantly decreased at the middle layer and the deep layer on LI6 acupoint and the deep layer on SI7 acupoint. Conclusions : The effect of needles applied at the connecting point of three yang meridians on the activities of nNOS, and NE could be observed, and it can be induced that the effect of needle stimulation on disrupted nervous system can be examined through additional researches based on this one.

소자약침(蘇子藥鍼)의 천식억제(喘息抑制) 및 면역조절효과(免疫調節效果)에 대(對) 실험적(實驗的) 연구(硏究)

  • Objectives : The purpose of this study was to confirm the suppressive effect against asthma and immune regulatory effect of Perillae Fractusher Herbal-acupuncture at Chok-samni(ST36) on ovalbumin-induced asthma in mice. Methods : C57BL/6 mice were sensitized and challenged with OVA(ovalbumin) for 12 weeks. The mice in the PF-HA group were treated with PF-HA at ST36 for the later 8 weeks(3 times a weeks). The mice in the OVA-Needle-Prink(NP) group were treated with single prick with an injection needle at ST36 for the later 8 weeks(3 times a weeks). Results : 1. The lung weight of the mice treated with PF-HA at St36 were decreased significantly compared with that of the control group. 2. The total leukocytes and eosinophils in BALF of the group treated with PF-HA were decreased significantly compared with those of the control group. 3. Eosinophils in BALF and the collagen accumulation in lung of the mice treated with PF-HA were decreased significantly compared with those of the control group. 4. The Concentrations of IgE, IL-4 and IL-5 in BALF, and IL-4, IL-5 and IL-13 in serum of the PF-HA group were decreased significantly compared with those of the control group. 5. The numbers of CD3-/CCR3+, Gr-1+/CD11b+,CD4+, and CD3e+/CD69+cells in lung of the mice group treated with PF-HA at St36 were decreased significantly compared with those of the control group. 6. The mRNA expressions of TNF-${\alpha}$, IL-4, IL-5, IL-13 in lung of the mice group treated with PF-HA at St36 were decreased significantly compared with those of the control group.

  • Objectives : The following study was undertaken in order to seek the acupuncture operation method of ${\ll}$Sang Han Lun(傷寒論)${\gg}$ Liu- Jing-Bian-Zheng(六經辯誇). Methods : Based on tile documents quoted in ${\ll}$Sang Hang Za Bing Lun Xu Wen(傷寒雜病論 序文)${\gg}$ of 'Zhang, Zhong-Jing(張仲景)', the relativity of the theory of Jing-Mai(經脈) and Liu-Jing-Bian-Zheng of convalescence, and from the Liu-Jing-Bing(六經病), the origin and implication that caused Jue-yin-Bing(厥陰病) to form was studied on the basis of acupuncture medicine publications and e commentary writing of ${\ll}$Sang Han Lun${\gg}$. Results : 1. ${\ll}$Sang Han Lun${\gg}$ Liu-Jing-Bian-Zheng has succeeded and was developed based on Liu-Jing-Fen-Zheng(六經分證) of ${\ll}$Su Wen Re Lun(素問 熱論)${\gg}$. In addition, the summary of Liu-Jing-Bing became the general pnnciple of Fen-Jing(分經) and Ding-Zheng(定證) that may be applicable to Fenghan(風寒), Wenre(溫熱), Lili(疫癩) and Eating(雜病). 2. Most commentators of ${\ll}$Sang Han Lun${\gg}$ in the Song, Ming and Ching Dynasties of ${\ll}$Sang Han Lun${\gg}$ interpreted the Jue-Yin-Bing in physiological and pathological aspects of Rong-Wei(榮衛) as the disease of the Sim Po and Liver meridian. 3. From the Liu-Jing-Bing of ${\ll}$Sang Han Lun${\gg}$, the region of acupuncture treaaent of Jue-Yin-Bing is treated with the needle from the point of view of Bing-fheng-Lun-fhi(辨證論治) with the basis of the important region of acupuncture of the Sim Po of meridian and Liver of meridian.

  • Objectives : Acupoints of LR1(Dadun)${\cdot}$HT9(Shaochong), KI10(Yingu)${\cdot}$HT3(Shaohai) are used as controlling the diseases from heart problems. Especially, when they are used, tonification or sedation techniques method is taken as a controlling the medication for an early stroke in the Korean medicine. So the aim of this study is to investigate the anti-apoptotic and neuroprotective effects of Acupuncture techniques of tonification or sedation at LR1${\cdot}$HT9, KI10${\cdot}$HT3 on the focal ischemia induced by intraluminal filament insertion in rats. Materials and Methods : The focal ischemia was induced by intraluminal filament insertion into middle cerebral artery. The animals were divided into five groups (n=7 in each group) : Control group, no therapy group after ischemia induced; AT-1; acupuncture therapy group at LR1${\cdot}$HT9, KI10${\cdot}$HT3 after ischemia-induced. AT-2; acupuncture therapy at right LR1, HT9(forward direction), KI10${\cdot}$HT3(opposite direction) inserting to the direction of route of the each meridian. AT3; acupuncture therapy at right LR1, HT9(twirling forward with the thumb of right hand 9 times), KI10${\cdot}$HT3(twirling forward with the forefinger of right hand 6 times) AT-4; acupuncture therapy at right LR1, HT9(forward direction, twirling forward with the thumb of right hand 9 times), KI10${\cdot}$HT3(opposite direction, twirling forward with the forefinger of right hand 6 times) inserting to the direction of route of the each meridian. The anti-apoptotic and neuroprotective effects of Acupuncture techniques of tonification or sedation at LR1, HT9, KI10${\cdot}$HT3 were observed by Bax, Bcl-2, mGluR5, Cytochrome c, Cresyl violet and ChAT-stain. Results : The intensity of Bax and Bax/Bcl-2 ratio were increased in ACU-2 and ACU-4 group but decreased in ACU-3 group. The intensity of mGluR5 was increased in ACU-1, ACU-2, ACU-3 and ACU-4 group. The intensity of cytochrome c was decreased in ACU-3 and ACU-4 group. The density of neurons stained by Cresyl violet and ChAT were increased in ACU-1, ACU-3 and ACU-4 group. Conclusions : Our study suggests that acupuncture therapy of tonification at LR1, HT9 by twirling forward with the thumb of right hand 9times, sedation at KI10${\cdot}$HT3 by twirling forward with the forefinger of right hand 6 times after perpendicularly inserting needle shows anti-apoptotic and neuroprotective effects on cholinergic neuron in focal cerebral ischemia of the stroke in rats.

  • The following study was undertaken in order to seek the acupuncture operation method of $\ll$Sang Han Lun(傷寒論)$\gg$ Liu-Jing-Bian-Zheng(六經辯證). As a result of the foregoing study, the author has written the following acupuncture operation on the basis of a summarized Tai-Yin-Bing for $\ll$Sang Han Lun$\gg$ Liu-Jing-Bian-Zheng. Results : $\ll$Sang Han Lun$\gg$ Liu-Jing-Bian-Zheng has succeeded and was developed based on Liu-Jing-Fen-Zheng(六經分證) of $\ll$Su Wen Re Lun(素問 熱論)$\gg$. In addition, the summary of Liu-Jing-Bing became the general principle of Fen-Jing(分經) and Ding-Zheng(定證) that may be applicable to Fenghan(風寒), Wenre(溫熱), Lili(疫려) and Zabing(雜病). 2. Most commentators of $\ll$Sang Han Lun$\gg$ in the Song, Ming and Ching Dynasties of $\ll$Sang Han Lun$\gg$ interpreted the Tai-Yang-Bing in physiological and pathological aspects of Rong-Wei(榮衛) as the disease of the spleen meridian. 3. From the Liu-Jing-Bing of $\ll$Sang Han Lun$\gg$, the region of acupuncture treatment of Tai-Yang-Bing is treated with the needle from the point of view of Bing-Zheng-Lun-Zhi(辨證論治) with the basis of the important region of acupuncture of the spleen meridian.
