The $\beta$-lactamase gene was cloned into E. coli DH5$\alpha$ from Bacillus sp. J105 with strong resistance against $\beta$-lactam antibiotics. The chromosomal DNA was partially digested with Sau3AI and ligated to BamHI digested pLAFR3. $\beta$-Lactamase positive clones were obtained by using in vitro packaging kit. The pKL11-${\Delta}4.6$ with $\beta$-lactamase activity was obtained by subcloning of the recombinant plasmid ($\beta$-lac +). The 6.5 kb fragment in the subcloned plasmid was sequenced. The DNA fragment that contains the $\beta$-lactamase gene encodes 309 amino acids. The 0.17 kb upstream region was similar to those of B. thuringinesis and B. cereus with 97% identity. The deduced amino acids sequence was also similar to those of $\beta$-lactamase from B. thuringinesis and B. cereus with 97% and 94% identity, respectively. The phylogenetic tree also showed the relationships of the $\beta$-lactamase gene of Bacillus sp. J105 to genetically related that of other Bacillus strains. Analysis of expression pattern of the pKL11-${\Delta}4.6$ in E. coli, revealed that the secretion efficiency of $\beta$-lactamase was $4{\sim}5%$ and the molecular weight was as same as that of original $\beta$-lactamase (31 kDa) from Bacillus sp. J105.
Antibacterial effects of six volatile essential oils against Vibrio sp. were evaluated. Volatile components of essential oil were analyzed by gas chromatography and gas chromatography mass spectrometry. Ginger oil treatment inhibited growth of V. parahaemolyticus by 22.5-85.7%. Main volatile compounds of ginger oil were ${\beta}-bisabolene$ (35.19%, peak area) and ${\beta}-sesquiphellandrene$ (12.22%). V. parahaemolyticus was completely inhibited at 1,000 ppm by treatment with mustard oil. Tolerances of V. vulnificus 01 and 02 were twice higher than that of V. parahaemolyticus. Main volatile compound of mustard oil was allyl isothiocyanate (92.55%). Garlic oil treatment of 1,000 ppm inhibited growths of V. parahaemolyticus, V. vulnificus 01, and V. vulnificus 02 by 22.8, 14.6, and 32.9%, respectively. Main volatile compounds of garlic oil were dimethyl sulfide (49.39%) and methyl 2-propenyl disulfide (10.09%). Growth of V. vulnificus 02 was inhibited by 60.6-80.3% via treatment with bud, leaf, and whole oil of clove. Antibacterial activity of whole clove oil on V. vulnificus 02 was stronger than those of ginger, mustard, and garlic oil. Main volatile compounds were eugenol (83.33%) and ${\beta}-caryophyllene$ (7.47%) in clove bud, eugenol (87.46%) and ${\beta}-caryophyllene$ (10.03%) in clove leaf, and eugenol (86.04%) and ${\beta}-caryophyllene$ (9.71%) in whole clove. These results revealed essential oils from spices could be used as potential agents to inhibit Vibrio sp.

PAHs(Polycyclic Aromatic Hydrocarbons) in coastal area and estuary adjacent to urban area and industrial activities region are mainly introduced into marine environment via atmosphere and river. This study which is evaluated the distribution characteristics of PAHs discharge from rivers and sewage disposal water which flowing into Ulsan bay, and were carried out in Jun of dry season and in Auguest of wet season, 2008. The water samples from eight main rivers and Youngyeon WWTP(Wastewater Treatment Plant) flowed into Ulsan bay were taken for analysis of dissolved PAHs. The range and mean values of dissolved PAHs concentrations showed 10.30 to 87.88 ng/L, its mean 36.94 ng/L in dry season and 10.30 to 69.57 ng/L, its mean 24.37 ng/L in wet season. The distribution of dissolved PAHs showd the high concentrations in the Gungcheoncheon which is flowed from urban and industrial area. The ranges and means values of the loading fluxes were estimated with 0.04 to 8.27 g/day, its mean 2.05 g/day in dry season, and 0.03 to 4.77 g/day, its mean 1.61 g/day, in wet season. The loading flux showed the highest value in Taewha river due to the high flow rate and the urban activity. The composition patterns of dissolved PAHs compound showed most of the trend occupying low molecuar weight PAHs than high molecular weight PAHs. These results were due to physical and chemical characteristics of PAHs compound, and were similar to those of other studies. The concentrations of dissolved PAHs in this study are lower than those of other studies, and the level of PAHs pollution appeared to be not serious in reverine waters of Ulsan coast.

Three sex pheromone components (cis-8-dodecenyl acetate (Z12Ac), trans-8-dodecenyl acetate (E12Ac), cis-8-dodecenol (Z12OH)) of the Oriental fruit moth, Grapholita molesta, were chemically synthesized. Especially to increase the composition of cis-stereoisomer, a triple bond intermediate was hydrogenated at $-20^{\circ}C$ with catalytic $Pd/BaSO_{4}$. The resulting product consisted of the acetates with a stereoisomer ratio in 92:8 (Z:E). The biological activity of the synthesized pheromone compounds was analyzed both in male responses and orientation disruption. The indoor pheromone effect was determined by male flight behavior showing wing movement in response to lure. Different mixtures of the synthetic pheromone components were prepared by mixing acetate and alcohol components in 100:0, 99:1, and 90:10 (g/g) and tested with a comparison of a standard commercial pheromone lure. The highest pheromone effect was observed in only acetate mixture (100:0) and the effect was reduced with the addition of the alcohol component. This indoor pheromone effect could be observed in field monitoring trial, in which 100:0 mixture showed the highest trap catches. Orientation disruption assay was conducted indoor by using a cage, in which the center had a commercial lure on sticky plate and the four candidates were placed at 6 cm away from the central lure on each of four directions. Test males were released to the arena during overnight (12 h) and then the caught males on the sticky plate were counted. The synthesized pheromone as well as the commercial pheromone showed 100% orientation disruption. However, the orientation disruption effect was reduced with decrease in the number of the surrounding disrupting pheromone baits. These results clearly suggest that the synthesized sex pheromone of G. molesta is biologically active and can be used for field mating disruption.
The Shinri area near the Yedang Lake, the eastern part of the Hongseong area in SW Gyeonggi Massif, consists of the Neoproterozoic Duckjeongri granodiorite-tonalite, mylonitized amphibole-bearing orthogneiss and impure marble with lens-shaped garnet-bearing metabasites. In this paper, we report mineralogical and geochemical data of Neoproterozoic lens-shaped garnet-bearing metabasites within marble of the Shinri area. The $SiO_2$ contents of garnet-bearing metabasites in marble vary between ~46.98 and 51.17 wt%, and the $Na_2O$ + $K_2O$ contents fall between ~1.95 and 2.85 wt%, similar to the tholeiitic sub-alkaline basaltic rocks. In the Zr/Y vs. Zr diagram, garnet-bearing metabasites also plot in the subalkaline basaltic rocks. The chondrite-normalized REE patterns for Shinri garnet-bearing metabasites show relatively flat patterns to that of chondrite. They show slight LREE-enriched and depleted patterns. The major and trace element data from lens-shaped garnet-bearing metabasites in marble of the Shinri area suggest that these rocks were formed in within plate. In contrast, previous major and trace element data of high pressure type garnet-bearing metabasites from the mafic-ultramafic complex in the Baekdong and Bibong areas suggest that these rocks were formed in a nascent arc to backarc spreading center within subduction zone setting. Based on mineral assemblage and mineral chemistry, P-T estimates for Shinri garnet-bearing metabasites are 9.6-12.7 kb, $695-840^{\circ}C$ for inclusions in the core, and 9.6-13.6 kb, $630-755^{\circ}C$ for those in the rim. These P-T estimates are distinct from those of the Baekdong and Bibong garnet-bearing metabasites with isothermal decompressional retrograde P-T path. In addition to Triassic tectonic activity previously reported in the Shinri area of Hongseong, the details of metamorphic history such as protolith age and Neo-Proterozoic metamorphic episode need to be solved.
Our country security industry 1960's service expense of the beginning U.S. army unit it accomplishes the growth which is quick with start, currently about 2,500 triumph the security enterprises which it goes over are being active. This study was designed to develop a conceptual framework for the curriculum and develop the details of the learning content for the education of Community Health Practitioners (CHPs). Since education programs for CHPs started 10 years ago, concepts related to CHP services have changed because of changes in society. The objectives of the study were as follows : 1) to analyse the usefulness of the present education program for CHPs, 2) to analyse the Job performance and self -confidence of the CHPs, 3) to identify the health needs of the clients served by the CHPs and the community problems related to health. 4) to develop a conceptual framework for the curriculum, for the education of CHPs, 5) to develops details for the learning content of the education program for CHPs. Phase I of the study was conducted by questionnaires to 150 CHPs who have worked in remote rural areas for more than 2 years. Among them, 147 responded. Data was collected from August 16, to August 25, 1990. In order to identify the health needs of the community people, research within the last five years was reviewed and analyzed. The data on 1, 842 communities gathered by the WHO Nursing Collaborations Center of the College of Nursing, Yonsei University was utilized to identify community problems related to health and the self - confidence in job performance of the CHPs. Psase II of the study consisted of a workshop with 13 professionals including Community Health Practitioners to evaluate the existing education program and a conceptual framework of the curriculum for the job education of CHPs. The results of the study are Summariged below : 1. The only 26 among 45 content items of the education program related to job skills was used by 80% of the responding CHPs. The knowledge of $\ulcorner$Networking community organization$\lrcorner$ was used by only 53.7% of the respondents. Educational content about $\ulcorner$Mental disease$\lrcorner$ was used by less than 50% of CHPs because of a knowledge deficit. 2. The CHPs reported that their activities concentrated on clinical services during the last six months. The survey showed that they seemed to neglect the activities for health promotion and disease prevention. Thus, $\ulcorner$Education for community loaders$\lrcorner$(15.9%), $\ulcorner$Activity for eavironmental health$\lrcorner$(16.3%) and $\ulcorner$Social work for needey people$\lrcorner$(23.3%) were done by less than 30% of CHPs. 3. More than 90% of CHPs reported being self - confident for the activities of $\ulcorner$Health education and counselling$\lrcorner$, $\ulcorner$Medicine prescription$\lrcorner$ and $\ulcorner$Immunization$\lrcorner$. But 50% of CHPs reported that they were not have self - confident in $\ulcorner$Management of water and environmental health$\lrcorner$ and only 25.6% of CHPs could insert an IUD independently. 4. It was identified that respiratory diseases and the gastrointestinal diseases were most common problems for the community people, followed by musculoskeletal and skin problems. 5. The community problems were classified into eight categories : physical environmental problems, environmental hygiene, health problems, health behavior, social problem, lack of resources, financial problem and the problems of the cultural and value system. 6. The conceptual framework consisted of the target population and their health status, nursing process working site and primary health care services such as health promotion, disease prevention, treatment and rehabilitation. 7. The contents of curriculum of education program for CHPs were formulated from the results of this study.
Disease forecasting in Korea was first studied in the Department of Fundamental Research, in the Central Agricultural Technology Institute in Suwon in 1947, where the dispersal of air-borne conidia of blast and brown spot pathogens in rice was examined. Disease forecasting system in Korea is operated based on information obtained from 200 main forecasting plots scattered around country (rice 150, economic crops 50) and 1,403 supplementary observational plots (rice 1,050, others 353) maintained by Korean government. Total number of target crops and diseases in both forecasting plots amount to 30 crops and 104 diseases. Disease development in the forecasting plots is examined by two extension agents specialized in disease forecasting, working in the national Agricul-tural Technology Service Center(ATSC) founded in each city and prefecture. The data obtained by the extension agents are transferred to a central organization, Rural Development Administration (RDA) through an internet-web system for analysis in a nation-wide forecasting program, and forwarded far the Central Forecasting Council consisted of 12 members from administration, university, research institution, meteorology station, and mass media to discuss present situation of disease development and subsequent progress. The council issues a forecasting information message, as a result of analysis, that is announced in public via mass media to 245 agencies including ATSC, who informs to local administration, the related agencies and farmers for implementation of disease control activity. However, in future successful performance of plant disease forecasting system is thought to be securing of excellent extension agents specialized in disease forecasting, elevation of their forecasting ability through continuous trainings, and furnishing of prominent forecasting equipments. Researches in plant disease forecasting in Korea have been concentrated on rice blast, where much information is available, but are substan-tially limited in other diseases. Most of the forecasting researches failed to achieve the continuity of researches on specialized topic, ignoring steady improvement towards practical use. Since disease forecasting loses its value without practicality, more efforts are needed to improve the practicality of the forecasting method in both spatial and temporal aspects. Since significance of disease forecasting is directly related to economic profit, further fore-casting researches should be planned and propelled in relation to fungicide spray scheduling or decision-making of control activities.
The principal objective of this investigation was to identify adequate species for the harvest of Perilla frutescens leaves, which provide profound antioxidant activities, and harbor abundant caffeic acid and rosmarinic acid. Namchun, Donggeul-2, Bora, Sae-bora and Neul-bora variants of the plant were assessed herein. In this study, we evaluated the antioxidant effects of these plants, and utilized an HPLC system to verify their caffeic acid and rosmarinic acid contents. Dried Perilla frutescens leaves were boiled at a temperature of $100^{\circ}C$ for three hours, and were lyopholized in a freezedryer. The extracts were then processed in order to confirm their antioxidant activities via 1,1-diphenyl-2-picrylhydrazyl (DPPH) free radical-scavenging activity assay, ferric-reducing antioxidant power (FRAP) assay, total flavonoid content and total polyphenol content assays. According to the observed antioxidant results of the five tested species of Perilla frutescens leaves, Bora and Donggeul-2 were shown to have more potent antioxidant activities than the other tested variants. The scavenge 50% of DPPH radical ($SC_50$ of DPPH) values were 241 ${\mu}g$ DM/mL in Donggeul-2 and 261 ${\mu}g$ DM/mL in Bora. Based on the results of the FRAP assay, the Bora variant showed a value of 796 mM $FeSO_4$$7H_2O/g$ DM, and Donggeul-2 exhibited a value of 748 mM $FeSO_4$$7H_2O/g$ DM, of which the total polyphenol contents were measured as 69.4 g GAE/kg DM and 61.8 g GAE/kg DM, respectively. Moreover, the Bora variant had the highest level of caffeic acid, and Donggeul-2 showed the highest rosmarinic acid content among the tested samples (0.87 mg/100 g wet base and 121 mg/100 g wet base, respectively). According to the results of this experiment, we selected two species, Bora and Donggeul-2, which were both verified to contain adequate and favorable antioxidant activities.

The suppression of neuroinflammatory responses in microglial cells can be considered a key target for improving the progression of neurodegenerative diseases such as Alzheimer's disease (AD), Parkinson's disease (PD), and Huntington's disease (HD). Asparagus cochinchinensis has traditionally been used as a medicine to treat fever, cough, kidney disease, breast cancer, inflammatory diseases, and brain diseases. In this study, we investigated the neuroprotective mechanism of an aqueous extract from A. cochinchinensis root (AEAC), particularly its anti-inflammatory effects on lipopolysaccharide (LPS)-activated BV-2 microglial cells. BV-2 cells were treated with four different concentrations of AEAC. No significant toxicity was detected in BV-2 cells treated with AEAC. Nitric oxide (NO), cyclooxygenase-2 (COX-2) mRNA, and inducible nitric oxide synthase (iNOS) mRNA levels were 21% lower in the AEAC+LPS group than in the Vehicle+LPS group. Lower proinflammatory (TNF-α and IL-1β) and anti-inflammatory cytokine (IL-6 and IL-10) levels were also detected in the AEAC+LPS group than in the Vehicle+LPS group, albeit at varying rates. Moreover, the phosphorylation of mitogen-activated protein kinase (MAPK) members after LPS treatment was significantly recovered in the AEAC-pretreated group compared to the Vehicle+LPS group, enhancement of the phosphorylation of mitogen-activated protein kinase (MAPK) members after LPS treatment was significantly recovered in the AEAC-pretreated group, while cell cycle arrest at the G2/M phase caused by LPS treatment was less severe in the AEAC+LPS group. The increase in reactive oxygen species (ROS) generation induced by LPS treatment was also lower in the AEAC-pretreated group than in the Vehicle+LPS group. This is the first study to show that AEAC exerts anti-neuroinflammatory activity against LPS stimulation by regulating the MAPK signaling pathway, the cell cycle, and ROS production.
