A Study on the Manufacture of Activated Carbon using Indonesian Coal (인도네시아 석탄을 이용한 활성탄 제조에 관한 연구)

  • Indonesian coal-based activated carbon was manufactured with steam-reaction method. Also effects of carbonization temperature and steam amount on the process yield and quality of the product were investigated at the activation temperature of $900^{\circ}C$. The rotary kiln type furnace was used for both carbonization and activation and the optimum operation conditions were carbonization temperature of $700^{\circ}C$, steam amount of 2.7g steam/g char and activation temperature of $900^{\circ}C$. At this condition, the iodine value of activated carbon was 1,010 mg/g. Methylene Blue Adsorption Number was 230mg/g and B.E.T. surface area was $1,020m^2/g$ with the hardness about 97.

Induction of Apoptosis by Gagamhwanglyeonhaedog-tang through Activation of Caspase-3 in Human Leukemia Cell Line HL-60 Cells

  • Gagamhwanglyeonhaedog-tang(GHH), a Korean genuine medicine, is a newly designed herbal drug formula based on the traditional oriental pharmacological knowledge for the purpose of treating tumorous diseases. Apoptosis is an evolutionarily conserved suicide program residing in cells. It leads to cell death through a tightly regulated process resulting in the removal of damaged or unwanted tissue. In the present study, the apoptosis inducing activities of the decocted water extract of GHH were studied. Results of the 3- [4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazolium bromide assay showed that GHH had a strong cytotoxic effect on HL-60 cells. The number of live cells was less than 20% after exposure to 1㎎/㎖ GHH for 48 hr. GHH increased cytotoxicity of HL-60 cells in a dose- and time­dependent manner. Cell apoptosis by GHH was confirmed by flow cytometric analysis of the DNA-stained cells. The percentage of apoptotic cells increased to 28%, 31% and 37% 24 hr and 37%, 44% and 81% 48 hr after treatment with 0.01, 0.1 and 1㎎/㎖ GHH, respectively. Flow cytometric analysis of GHH treated HL-60 cells showed increase of hypodiploid apoptotic cells in a dose- and time- dependent manner. DNA fragmentation also occurred in apoptosis and was characterized by a ladder pattern on agarose gel. In addition, GHH (0.01 and 0.1㎎/㎖) increased the secretion of tumor necrosis factor-alpha in 24 and 48 hr. The author showed that GHH-induced apoptosis was accompanied by activation of caspase-3. These results suggest that GHH induces activation of caspase-3 and eventually leads to apoptosis.

Influence of Gungguitang-gamibang on the Regulation of Melanogenesis through JNK Signaling Pathway in B16 Melanoma Cells

  • Gunggui-tang has been used for the therapy of blood disorders in Hangbang medicine for long time. Also, Glycyrrhiza uralensis has been used for deficientblood patterns with an irregular pulse or palpitations, coughing and wheezing, and heat or cold in the lungs. Melanogenesis is a physiological process resulting in the synthesis of melanin pigments. We investigated whether the water extract of Gunggui-tang plus G. uralensis inhibited melanogenesis in B16 melanoma cells. Because the molecular events connecting the regulation in tyrosinase activity remain to be elucidated, we also aimed to determine whether Gunggui-tang gamibang(GTG) affects tyrosinase at the gene activation level in the cells. First, we showed that GTG inhibited the tyrosinase promoter activity and further, down-regulated the tyrosinase protein activity in ${\alpha}-melanocyte-stimulating$ hormone $({\alpha}-MSH)-treated$ B16 melanoma cells. GTG also resulted in a decrease of melanin content in MSH-induced melanogenesis, indicating that GTG may be a useful drug in studying the regulation of melanogenesis. The pretreatment of GTG significantly prevented phosphotransferase activity of c-Jun N-terminal kinase (JNK1) and transcriptional activation of activating protein-1 (AP-1) in MSH-treated B16 melanoma cells. These findings indicate that GTG inhibits melanogenesis of B16 melanoma cells via suppression of phosphotransferase activity of JNK1 and transcriptional activation of AP-1.

Dielectric Passivation and Geometry Effects on the Electromigration Characteristics in Al-1%Si Thin Film Interconnections

  • Dielectric passivation effects on the EM(electromigration) have been a great interest with recent ULSI and multilevel structure tends in thin film interconnections of a microelectronic device. SiO$_2$, PSG(phosphosilicate glass), and Si$_3$N$_4$ passivation materials effects on the EM resistance were investigated by utilizing widely used Al-1%Si thin film interconnections. A standard photolithography process was applied for the fabrication of 0.7㎛ thick 3㎛ wide, and 200㎛ ~1600㎛ long Al-1%Si EM test patterns. SiO$_2$, PSG, and Si$_3$N$_4$ dielectric passivation with the thickness of 300 nm were singly deposited onto the Al-1%Si thin film interconnections by using an APCVD(atmospheric pressure chemical vapor deposition) and a PECVD(plasma enhanced chemical vapor deposition) in order to investigate the passivation materials effects on the EM characteristics. EM tests were performed at the direct current densities of 3.2 $\times$ 10$\^$6/∼4.5 $\times$ 10$\^$6/ A/cm$^2$ and at the temperatures of 180 $\^{C}$, 210$\^{C}$, 240$\^{C}$, and 270$\^{C}$ for measuring the activation energies(Q) and for accelerated test conditions. Activation energies were calculated from the measured MTF(mean-time-to-failure) values. The calculated activation energies for the electromigration were 0.44 eV, 0.45 eV, and 0.50 eV, and 0.66 eV for the case of nonpassivated-, Si$_3$N$_4$passivated-, PSG passivated-, and SiO$_2$ passivated Al-1%Si thin film interconnections, respectively. Thus SiO$_2$ passivation showed the best characteristics on the EM resistance followed by the order of PSG, Si$_3$N$_4$ and nonpassivation. It is believed that the passivation sequences as well as the passivation materials also influence on the EM characteristics in multilevel passivation structures.

Recrystallization Behavior of Aluminum Plates Depending on Their Purities (순도에 따른 Al 판재의 재결정 거동)

  • Recrystallization behavior has been investigated for commercial purity AA1050 (99.5wt%Al) and high purity 3N Al (99.9wt% Al). Samples were cold rolled with 90% of thickness reduction and were annealed isothermally at 290, 315, and 350o C for various times until complete recrystallization was achieved. Hardness measurement and Electron Backscatter Diffraction(EBSD) analyses, combined with Grain Orientation Spread(GOS), were employed to investigate the recrystallization behavior. EBSD analysis combined with GOS were distinctly revealed to be a more useful method to determine the recrystallization fraction and to characterize the recrystallization kinetics. As the annealing temperature increased, recrystallization in AA1050 accelerated more than that process did in Al 3N. Both AA1050 and Al 3N showed the same temperature dependence of the n value of the Johnson-Mehl-Avrami-Kolmogorov equation(JMAK equation), i.e., n values increased as annealing temperature increased. Activation energy of recrystallization in AA1050 is about 176 kJ/mol, which is comparable with the activation energy of grain boundary migration in cold-rolled AA1050. This value is somewhat higher than the activation energy of recrystallization in Al 3N.

Anti-inflammatory Effects of Omisodokeum (오미소독음(五味消毒飮)의 항염효과(抗炎效果) 및 기전(機轉)에 관(關)한 실험적연구(實驗的硏究))

  • Purpose: The purpose of this study was to investigate the anti-inflammatory effects of the water extract of Omisodokeum (OMSDE) on peritoneal macrophages, Methods: To verify the anti-inflammatory mechanism of OMSDE, the activation of nuclear $factor-{\kappa}B$ $(NF-{\kappa}B)$ and the phosphorylation of MAPK were examined. Results: The extract of OMSDE suppressed the production of LPS-induced nitric oxide (NO), tumor necrosis factor $(TNF)-{\alpha}$, interleukin $(IL)-1{\beta}$, IL-6 and IL-12 in the macrophages. OMSDE inhibited the degradation of inhibitory ${\kappa}B-{\alpha}$ $(I{\kappa}B-{\alpha})$ and it suppressed the activation of extracellular signal-regulated kinase (ERK 1/2) but didn't inhibit c-Jun N-terminal kinase (JNK) and p38, indicating that OMSDE may inhibit the pro-inflammatory cytokine production process by inhibiting the activation of $NF-{\kappa}B$ and ERK 1/2. Furthermore, OMSDE inhibited the production of interferon $(IFN)-{\beta}$ but didn't inhibit of $IFN-{\alpha}$ in the LPS-stimulated macrophages through the down-regulation of interferon regulatory factor (IRF)-1 and IRF-7. The Oral administration of OMSDE inhibited LPS-induced endotoxin shock and the production of $TNF-{\alpha}$ in serum but didn't inhibit of $IL-1{\beta}$ and IL-6. Conclusion: These results suggest that OMSDE may be effective in the prevention and treatment of inflammatory diseases.

Berberine Chloride Inhibits Receptor Activator of $NF-{\kappa}B$ Ligand-induced Osteoclastogenesis via Preventing ERK Activation

  • An imbalance in bone remodeling that is caused by increased bone resorption over bone formation leads to most adult skeletal diseases including osteoporosis. Since the development of anti-resorptive agents from natural substances has recently gained more interest in the treatment of osteoporosis, we evaluated the effects of 222 natural compounds on receptor activator of $NF-{\kappa}B$ ligand (RANKL)-induced of tartrate-resistance acid phosphatase (TRAP) activity in RAW264.7 murine macrophage cell, and found that berberine chloride is one of compounds inhibiting RANKL-induced TRAP activity. Berberine chloride significantly inhibited the RANKL-induced TRAP activity and the formation of multinucleated osteoclasts in a dose-dependent manner. In addition, berberine chloride prevented the RANKL-induced mRNA expression of TRAP, matrix metalloproteinase 9 and c-Src, which have been known to be highly expressed in the process of osteoclastogenesis. Interestingly, berberine chloride prevented the RANKL-induced activation of extracellular signal-regulated kinase (ERK) which is one of mitogen-activated protein (MAP) kinases. In conclusion, berberine chloride could inhibit the osteoclastogenesis via preventing the activation of ERK/MAP kinase signaling pathway.

A Study on the Residents' Participation in the Development Process of Rural Village - Focused on the Namsa Traditional Theme Village - (농촌마을개발사업의 전개와 주민참여에 관한 연구 -산청 남사 젼통테마마을을 대상으로 -)

  • After the 2000's, the rural village development projects are designated as participatory rural village development programs and projects. The purpose of this study is to draw a scheme to facilitate resident's participation in rural village development project by exploring the factors influencing resident's participation. For this, in Namsa rural theme village in Dansung-myon, Sanchung-gun, Kyongnam Province, the data were collected through field surveys and interviews residents residing in this village. In addition, observations studies and an opinion survey of the persons concerned in the presentation meetings were also conducted. I investigated and analysed the activation factors and the characteristics of residents' participation, and amenity resources which were essential for community planning including the planning-establishment, theme-searching and development of experience program. Through this study, I drew findings as follows; 1) The leader's leadership and residents' positive participation are very important activation factors in the first stage of rural viability. 2) And then, the external support and concern, the establishment of short-and long-term community planning, and the rural-income increase through the development of amenity resources play important roles as activation factors in the successive stage of rural viability. Based on the results of this study, the following recommendations were made. In order to improve the level of resident's participation in rural village development programs and projects, it is necessary to evaluate their successes, failure, barriers and the positive correlation between their success more delicately. In addition to this, it must be suggested the better model for participatory rural village development programs for the further enhancement and improvement of rural village development projects.

Effect of Methyl-Beta-Cyclodextrin (MBCD) on Cryopreservation of Bull Spermatozoa (소 정액의 동결 보존시 Methyl-Beta-Cyclodextrin (MBCD)의 영향)

  • The objective of this study was to evaluate efficiency of methyl-beta-cyclodextrin (MBCD) in the sperm preservation of bull. For this study, the freezing of diluted semen were added with Triladyl containing 20% egg-yolk and/or 0, 1, 5, 10 and 20 mM MBCD before freezing process. Analysis of viability in frozen-thawed sperm was estimated by SYBR14/PI double stain, hypoosmotic swelling test(HOST) and acrosome damage with FITC-PNA, and mitochondria activation with Rhodamin123 by flow-cytometry. The sperm viability was significantly in 0 mM and 5 mM concentrations of MBCD than other groups (p<0.05). However, the HOST was significantly lower at 20 mM concentration of MBCD than other concentrations (p<0.05). In addition, acrosome damage and mitochondria activation rates were significantly lower at 20 mM concentration of MBCD than other groups (p<0.05). In conclusion, the viability of sperm was not significantly different among concentrations of MBCD 0, 5 and 10 mM, but MBCD 20 mM was significantly lower than other groups. In addition, as concentrations of MBCD was high, HOST, acrosome damage and mitochondria activation rates had a negative effect in bull sperm.

The Provenance and Characteristic Classification of the White Porcelain in the Gyeongsangnam-do by Neutron Activation Analysis (중성자방사화분석을 활용한 경상남도 백자의 산지 및 특성 분류)

  • This study analyze concentration of minor and trace elements on 47 white porcelains excavated from Dudong-ri, Baekryeon-ri, Sachon-ri kilns in Gyeonsangnam-do by NAA(neutron activation analysis) and try to classify the provenance and characteristics according to the analytical result. Each kilns are divided into the group by PCA(principal component analysis) and LDA(linear discrimination analysis) using 17 elements; Ba Ce, Co, Cr, Cs, Dy, Eu, Hf, La Lu, Rb, Sc, Sm, Ta, Th, V, Yb. The contribution elements are Dy, Sm, La, Ce, Lu, Sc. And soft and hard white porcelains are similar with the chemical composition of the use materials therefore the difference of the chemical composition not confirmed a cause. The analytical results of the fine(I) and poor(II) quality white porcelains presume the difference of the povenance of clay materials or the poduction process such as difference purify and additive materials.
