Using tracer technique of Zn-65, a pot experiment has been carried out in order to evaluate the efficiency of zinc fertilization on two paddy soils; an acidic from Kimpo and an alkaline from Yeongweol. The sources of zinc were zinc sulfate, zinc carbonate, zinc chloride and zinc oxide. Two rates of zinc were applied to the soils and control treatment was also included for this study. The methods of zinc application were uniform mixing throughout the soil, applying to the soil surface and irrigation water, and root dipping with zinc oxide at transplanting. In general, Yeongweol soil had higher efficiency of zinc fertilizers than Kimpo soil. The results showed that zinc fertilizer application should be required to improve the rice growing conditions in Yeongweol soil especially at early stage of growth after transplanting. As to the application method of zinc fertilizers, mixing treatment appeared to be most superior to any other methods in both soils. In addition, it is found that root dipping in the zinc oxide suspension would be a rather effective method of zinc application. In aspect of fertilizer efficiency there was no superiority or inferiority among the zinc sources used in this experiment.

Nitrate nitrogen is common contaminant in groundwater aquifers, its concentration is regulated many countries below 10 mg/L as N (As per WHO standards) in drinking water. An attempt was made to get optimal results for the treatment of nitrate nitrogen in groundwater by conducting various experiments by changing the experimental conditions for ZVI bipolar packed bed electrolytic cell. From the experimental results it is evident that the nitrate nitrogen removal is more effective when the reactor conditions are maintained in acidic range but when the acidic environment changes to alkaline due to the hydroxide formed during the process of ammonia nitrogen there by increasing the pH reducing the hydrogen ions required for reduction which leads to low effectiveness of the system. In the ZVI bipolar packed bed electrolytic cell, the packing ratio of 0.5~1:1 was found to be most effective for the treatment of nitrate nitrogen because ZVI particles are isolated and individual particle act like small electrode with low packing ratio. It is seen that formation of precipitate and acceleration of clogging incrementally for packing ratio more than 2:1, decreasing the nitrate nitrogen removal rate. When the voltage is increased it is seen that kinetics and current also increases but at the same time more electric power is consumed. In this experiment, the optimum voltage was determined to be 50V. At that time, nitrate nitrogen was removed by 94.9%.

The purpose of this study was to investigate how to modify the physical properties of cellulose after thermal treatment. Cellulose was treated between $225^{\circ}C$ and $325^{\circ}C$ for 3 hrs under air flow, and then the thermally treated cellulose was measured to specific surface area, constitute elements, consumption ofacid and base, as well as the adsorption capacity of ethylamine vapor. The higher was the treating temperature from $225^{\circ}C$ to $325^{\circ}C$, the lower was the total yield of cellulose. Elemental analysis revealed that carbon content in thermally treated cellulose was gradually increased in proportion to temperature increment. The amount of acidic functional groups tended to increase up to $300^{\circ}C$, after then to be lowered slightly. In principle, no alkaline functional groups were found in thermally treated cellulose. In case of treatment with $325^{\circ}C$, only a few amount of alkaline functional groups were detectable. Specific surface area of thermally treated cellulose are determined to $1.9m^2/g$, which value can become higher when the treated temperature rises. The thermally treated cellulose at $275^{\circ}C$ shows the highest adsorption capacity of ethylamine at $40^{\circ}C$ for 4 hrs. Solubility of those two celluloses with WPG (Weight Percent Gain) value of 113% and 108%, respectively, was determined to almost 100%. X-ray diffractogram of thermally treated cellulose suggested that the crystalline structure of cellulose began to be destroyed at the temperature of $275^{\circ}C$. As a conclusion, changes of such a physical properties make it possible to weaken inter and/or intra hydrogen bond in crystal region of cellulose macromolecules. When thermally treated cellulose adsorbs ethylamine, it turns to be well soluble to water.

RNase activity of Saccharomyces cerevisiae ATCC 7754 was investigated to obtain strains with high ribonucleic acid (RNA) content. The yeast strain contained two RNase activities; an acidic RNase with a optima of pH $3{\sim}4$ and an alkaline RNase with a optima pH 9. The acidic RNase activity was inhibited by $0.08\;M\;HgCl_{2}$ most drastically. The alkaline RNase activity was inhibited by 2.0 M NaCl or KCl, while enhanced by addition of $0.05\;M\;CaCl_{2},\;0.02\;M\;ZnSO_{4},\;or\;0.008\;M\;HgCl_{2}$. Various mutants of Saccharomyces cerevisiae ATCC 7754 were isolated by ethylmethane sulfonate (EMS) treatment or $\gamma$-ray/ultra violet irradiation. Among the mutants that were sensitive to high concentration of KCl which inhibits alkaline RNase, B24 was selected for high RNA content per culture volume. Growth characteristics of the mutant were comparable to those of the mother strain with optimum growth at pH $4.5{\sim}5.5$. The mutant accumulated higher content of RNA than the mother strain when glucose was used as the carbon source. However, both growth rate and total RNA content of the mutant were higher in molasses medium than in glucose medium. RNA content of the mutant increased rapidly during the early stage of growth, and then decreased gradually until the culture reached stationary phase by a fed-batch culture in a 5 L jar fermenter. Maximal cell harvest and the final RNA content using the mutant B24 were 69.6 g/L culture broth and 19.8 g/100 g of the dry cell while those using the mother strain were 68 g/L culture broth and 16.1 g/100 g of dry cell, respectively.
In this study, the dyeing conditions(temperature, time, concentration) are changed from various conditions on wool fabrics by using corydalis tuber extract in order to develop new natural dyes. The purpose of this study is to improve the dyeability, color fastness, and functionality and to derive optimal dyeing conditions by comparing and analyzing the changes of K/S values and surface color by dyeing pH changes, mordant method, and mordant type. As a result of the experiment, the optimum dyeing condition of the wool fabrics is shown dyeing temperature:$80^{\circ}C$, dyeing time:90min, dyeing concentration:100%. The dyeability by pH variants of corydalis tuber extract indicates that K/S values is higher alkaline than acidic. The mordant method of corydalis tuber extract showed pre-mordant has high K/S values. In terms of color fastness, marked improvement has not been shown despite of mordant treatment on wool fabrics. In particular, color change of color fastness to washing, color fastness to light indicates the low fastness. In addition, the functionality such as antibacterial activities and deodorization can be given at dyeing with corydalis tuber extract thus it is expected to be applied to underwear or apparel products for the elderly and infirm and children with weak skin that required high functionality.

The purposes of this research were to evaluate the character of laboratory wastewater, and to examine the utilization of waste egg shells for neutralization and removal of heavy metals. Waste egg shells are excellent at neutralizing acidic wastewater, because they have alkaline minerals such as calcium. It must be seemed that removal rate of heavy metals were very influenced by adsobent dosage and adsorbate concentrations, because waste egg shells acted as precipitation and adsorption. If we reflected the adsorption capacity(k) and adsorption(1/n) of Freundlich isotherm, we couldn't consider waste egg shells as a good adsorbent. In view of these results, it showed that wastes containing the similar compositions as waste egg shells could utilize the neutralization, precipitation and adsorption of heavy metals in laboratory wastewater.
The separation of nicotine and tobacco-specific N-nitrosamines is a tough problem in tobacco industry. In this study, separation of nicotine from 4-(methylnitrosamino) -1-(3-pyridyl)-1-butanone (NNK) mixtures was investigated using electrodialysis by taking the principle of the protonation status difference between these two components. The results indicated that the solution pH has a dominant impact on the separation process. In a pH range of 5-7, nicotine molecules are existed as mono- and di-protonated ions and can be separated from the uncharged NNK molecules. The acidic electrolyte is conducive to the separation process from the point of flux and energy consumption; while the alkaline electrolyte has negative impact on the separation process. A current density of $10mA/cm^2$ is an appropriate value for the separation process. The lowest energy consumption of the separation process is 0.58 kWh/kg nicotine with the process cost to be estimated at only $0.208 /kg nicotine. Naturally, electrodialysis is a high-efficiency, cost-effective, and environmentally friendly process to separate and purify nicotine from tobacco juice.
Rheum palmatum has traditionally been used as a preventive agent and medication against fever and infection. The aim of this study was to isolate and characterize an antibacterial substance from R. palmatum that is effective against bacterial vaginosis. A methanol extract from R. palmatum showed antibacterial activity against Lactobacillus vaginalis KC TC 3515, Chryseobacterium gleum KCTC 2904, and Sphingomonas paucimobilis KCTC 2834, which cause bacterial vaginosis. After extraction and pH control of the methanol extract from R. palmatum, we found that acidic and alkaline extracts did not show antibacterial activity. A neutral extract (50 mg/mL) displayed an inhibitory zone of 18 mm on a nutrient agar plate with C. gleum KCTC 2904. Fractions No. 11 and 12 among 41 fractions obtained by silica gel column chromatography produced inhibitory zones of 10 mm on nutrient agar plates with C. gleum KCTC 2904. $R_f0.15$ and $R_f0.17$ spots produced by TLC of fraction No. 11 showed antibacterial activity against C. gleum KCTC 2904. Isolation and purification of the peak at a retention time (Rt) of 9.427 min was achieved by HPLC of $R_f0.29spots$. The peak at Rt 9.427 min showed antibacterial activity against C. gleum KCTC 2904.
Due to wide abuse of antibiotics both in human and livestock use, the advent and spread of multidrug resistant (MDR) pathogens becomes a serious health problem all over the world. Since the development of new antibiotics is at a standstill in pharmaceutical industry, the choice of therapeutic antibiotics is getting narrower. In this study, in an effort to search new antibiotics, the antimicrobial activity of Paenibacillus DY1 isolated from Korean soil was characterized on its growth inhibition spectrum against various health threatening MDR strains, with its stability and chemical structure. Extracellular culture filtrate of Paenibacillus DY1 effectively inhibits the growth of all the tested MDR enteropathogenic Eshcherichia coli, enterohemolytic E. coli, and enterotoxigenic E. coli strains, at a similar level to that on the nonresistant control E. coli strains. It showed significant growth inhibition effect against the causative agents of class one legal communicable disease, MDR Salmonella typhi, MDR Salmonella paratyphi A, food poisoning bacteria, MDR Salmonella typhimurium, and other MDR Salmonella spp. The growth of all of 10 different MDR Shigella spp. strains and 6 different Vibrio spp. strains tested was also inhibited. The antimicrobial activity of Paenibacillus DY1 was well preserved after heat treatment, and was also stable in both alkaline and acidic environment. The antimicrobial activity was partially purified with Diaion HP20 column and TLC. By NMR study, the putative structure of the activity was postulated as an alkane having hydroxyl groups.

During cleaning steps, ultrafiltration membranes are mechanically and chemically stressed. This may result in membrane degradations and failures. In this paper, polysulfone membranes were used to evaluate membrane deteriorations by commercial detergents in static conditions. Ageing of the membrane was simulated by immersing samples in solutions containing commercial detergents with various concentrations, temperatures and times defined by experimental designs. Indeed, an innovative approach in the chemical membranes ageing researches, based on methodological tools, was used in order to achieve significant ageing experiments without using an accelerated ageing protocol. The macroscopic changes were monitored by permeability measurements and mechanical strength tests coupled with a microscopic characterization by ATR-FTIR and HRSEM. The present work details results obtained for three commercial detergents: an alkaline, an acidic and an enzymatic detergent. It was found that the detergents used in the industrial advised conditions (concentration, temperature and time of contact) were not detrimental for membrane properties (permeability and elongation at break) and so for the quality of the produced water. Over the industrial cumulated time of contact, different ageing effects can be observed and compared with the ones induced by NaOCl.
