Effect of Mobile Phase Additive on Enantiomer Resolution for Chiral Amines on Polysaccharide-derived Chiral Stationary Phases by High Performance Liquid Chromatography (고성능 액체크로마토그래피의 다당유도체를 기초로 한 키랄 고정상에서 이동상 첨가제가 키랄 아민의 광학분리에 미치는 영향)

  • Chromatographic enantiomer resolution of chiral amines was performed on several covalently immobilized and coated chiral stationary phases (CSPs) based on polysaccharide derivatives under the mobile phase conditions containing base or acid or acid/base additive. The chromatographic parameters including separation factors and capacity factors were greatly influenced by the nature of the mobile phase containing base or acid or salt additive as well as the used CSPs. When 0.05% triethylamine/0.05% trifluoroacetic acid as an additive in the mobile phase was used on all CSPs in this study, the greatest enantiomer resolution was observed except for Chiralpak AD. Also, it was shown that the change of base additive into acid or salt in the mobile phase may directly affect chiral recognition mechanisms between the chiral selectors and analytes occurring during enantiomer separation, resulting in the change of elution orders.

A Comparative Study of Acidemia-induced Hyperkalemia and Hyperkalemia-induced Acidemia (산혈증 유발 고칼륨혈증과 고칼륨혈증 유발 산혈증의 비교 연구)

  • A comparative study of acid-base balance has been made between acidemia-induced hyperkalemia and hyperkalemia-induced acidemia. A group of rabbits was infused 0.1 N hydrochloric acid solution and metabolic acidosis was induced. Another group was administered 20 mM potassium chloride solution and hyperkalemia was induced. The third group was infused 0.1 N hydrochloric acid and 20 mM potassium chloride solution, simultaneously. Acid-base data and plasma potassium ion concentration were monitored every thirty minutes in these three groups of rabbits. Following results were obtained: 1 ) Along with the infusion of hydrochloric acid, acute metabolic acidosis was induced in the rabbits. Plasma bicarbonate ion concentration decreased primarily in this group. As a respiratory compensation, there was a tendency of reduction of arterial $Pco_{2}$. The alteration of data became larger along with the amount of administration and the time elapsed. However, hyperkalemia was not so severe compared with the second group. 2) In potassium chloride infused group, plasma potassium ion concentration increased along with the time elapsed and the amount of infusion. And the alteration of acid-base data was parrallel to the level of potassium ion concentration, above all depression of pH was prominent. 3) Above data suggest that when acute metabolic acidosis was induced, exchange of intracellular potassium ion with extracellular hydrogen ion seems significant for the regulation of extracellular acid-base balance. And when hyperkalemia was induced with the infusion of potassium chloride solution, the exchange of intracellular hydrogen ion with extracellular potassium ion also seems significant for the regulation of extracellular potassium balance. 4) In the group of rabbits infused hydrochloric acid and potassium simultaneously, disturbances of acid-base balance and potassium balance were much more severe than two other groups. In these mixed disturbances, the process of compensatory mechanism might be inhibited and one disturbance might aggregate each other. 5) Through above data it has been postulated that in acid-base disturbance potassium balance can be sacrificed as a compensatory mechanism, and vice versa in disturbance of potassium balance. And our data also suggest that hydrogen ion and potassium ion are compensatory pair, one another.

A Novel Therapeutic Measure for Metabolic Acidosis with Amino Acids

  • In hypoxic tissue conditions, pyruvate can not enter the Krebs cycle and lactic acid, produced from pyruvate, accumulates to induce lactic acidosis. Pyruvate, However, can also be converted to alanine by glutamate pyruvate transaminase, that could be enhanced by glutamate. Therefore, it would be a fundamental measure to treat the lactic acidosis in tissue hypoxic conditions when one can convert the accumulated lactic acid, through pyruvate, to alanine. To test the above hypothesis, we induced a lactic acidosis in cats and the effect of glutamate on recovery of acid base state and removal of the lactic acid from blood were assessed and the results were compared with those of bicarbonate administration, which is one of the most frequently used conventional measure for correction of the acid base state during lactic acidosis. The results were that glutamate and combined glutamate bicarbonate solutions not only restored the acid base status completely from the lactic acidosis in an hour or two, but also restored the blood level of lactate partially. We concluded that administration of glutamate solution to convert pyruvate into alanine is effective in preventing lactic acid accumulation and treating lactic acidosis.

Biosynthesis of Nucleic Acid in Chloroplast Isolated from Chlorella Cells. I. (Chlorella제포에서 분리한 엽록체의 핵산합성 1)

  • For the purpose of investigating the effect of nalidixic acid on the nucleic acid synthesis in chloroplast isolated from Chlorella ellipsoidea, cells were cultured in the media treated with nalidixic acid(20ppm) for 5 days. Aliquots cells were taken out at the inoculation and at intervals during the culture and growth rate of Chlorella cells measured. After extraction of nucleic acids in chloroplast isolated from these cells, their contents were analyzed by the base composition and the effect of nalidixic acid on the nucleic acid synthesis interpreted to compare with those of the control. 1. It was showed that the inhibitory concentration affected by nalidixic acid on the growth of Chlorella cells were 20ppm. 2. Because nalidixic acid had depressed the DNA replication in isolated chloroplast as well as whole cell system, these contents were markedly decreased in comparison with those of the control. 3. In the isolated chloroplast as well as in the whole cell system, nalidixic acid was decreased contents of base in the RNA by preventing RNA transcription.

  • Effect of catalysts, which was catalyzed by acid(HCl and HNO3) and base(NH4OH), on characteristics of the mullite powders prepared by sol-gel methdo wa sinvestigated by XRD, TGA, SEm AND BET. As a result, weight loss as a function of catalysts was in order of HCl=32.6%>HNO3=25.44%>Non=24.0%>NH4OH=22.5%. The mullite powder dried at 100$^{\circ}C$ appeared spherical shape in acid catalyst and different shape in base catalyst, but sintering powder at 1400$^{\circ}C$ appeared very fine particle of 0.05∼0.1$\mu\textrm{m}$ regardless of catalysts. In all cae, the pore quantity, which was capable to adsoprtion, was decreased with increasing temperature. In base catalyst, no change of special surface area in mullite appeared.

  • We propose a research for detection of the fluoride ion using borane compounds. Based on the Lewis acid-base reaction, we discussed the fundamental of sensing for fluoride ion. One of the important aspects in the chemistry of organoboranes is their behaviors as Lewis acids, which is a result of the vacant $2p_{\pi}$ orbital on the tricoordinate boron center. The electronic interaction between boron atoms and ${\pi}$-orbitals of donor molecules, constructed from the carbon 2p orbitals, is generally strong. Boron atoms can reach the desired octet configuration either through ${\pi}$-overlap with a suitable X or through formation of Lewis acid-Lewis base complexes.

  • A series of new Iron(II) Schiff base amino acid complexes derived from the condensation of amino acid and sodium 2-hydroxybenzaldehyde-5-sulfonate have been synthesized. The complexes were characterized by elemental, electronic, IR spectral analyses and conductance measurements. The stability and solubility of the prepared complexes were determined. Two spectral methods used to determine the stoichiometry of the prepared complexes which exhibited divalent tridentate coordination and formed chelates of octahedral structures. The antibacterial activity of the prepared complexes has been tested against Bacillus cereus, Pseudomonas aeruginosa and Micrococcus bacteria. The effect of HCl on investigated complexes studied spectrophotometrically.

Diagnosis of Acid-Base Disorders in Canine Parvoviral Enteritis (개 파보바이러스성 장염에서 산-염기 장애의 진단)

  • Abstract: This study was aimed to diagnose acid-base disorders of dogs with canine parvoviral enteritis (CPE) and data to establish a rational fluid therapy regimen for patients with CPE. A total of 43 dogs which had clinical signs of CPE and had detected canine parvovirus by polymerase chain reaction, were bled anaerobically from jugular vein at the time of admission. Blood chemical test, determination of electrolytes and blood gas analysis were conducted, and calculated values were obtained from each measured items. The values of blood chemical and electrolytes of dogs with CPE were various depending on the degree of clinical signs, and these tests were not specific to diagnose for CPE. Hypochloremia (20.9%), hyperchloremia (11.6%), hypokalemia (7.0%), hyperkalemia (11.6%), hyponatremia (9.3%) and hypernatremia (18.6%) were diagnosed as abnormalities of electrolytes from 43 dogs with CPE. The 29 out of 43 dogs (67.4%) were metabolic acidosis and 3 dogs (7.0%) were metabolic alkalosis. The acid-base status of 11 dogs out of 43 dogs (25.6%) was normal.

The Acid-Base-Concepts of Korean Secondary Students (우리나라 중.고등학교 학생이 갖고 있는 '산과 염기'의 개념에 관한 연구 -서울 일부지역의 여학생을 중심으로-)

  • The purpose of the study is to compare the learning contents of theme acids and bases that are commonly dealed with in elementary, middle and high schools m Korea and to investigate the acid-base-concepts of secondary students. The results of the study were as follows: 1) Many students did not have an interest in the chemistry and they pointed out the necessity of many experiments and practices and its application to the practical life. 2) It is found that most of textbooks used in the elementary school are written to make the students have an interest in the science through the experiments and pratices. However the textbooks used in the secondary school are too theoretically written so that many students are likely to lose the interests in the chemistry. 3) Although students had already learned Arrhenius's acid-base-concept, many students tried to solve the question with Liebig's theory as a acid-base-concept. This misconception might be caused by learning by cramming and teaching too theoretically. Accordingly, it is necessary that the education of science is carried out to make the students have an interest in the science by providing the students with the much more experiments and practices and also applying it to the practical life.

The Chemistry Teachers' Perceptions and Interpretations about Three Acid-Base Models (세 가지 산-염기 모델에 대한 화학교사들의 인식과 해석)

  • This study investigated the perceptions of the relationship among the three acid-base models of chemistry teachers. In addition, we examined how the perception of the relationship between models affected on the interpretation of concepts in each model. To investigate teachers' perceptions and interpretations, a questionnaire and interviews were conducted for 24 chemistry teachers. As results, most of the chemistry teachers recognized the three models as cumulative extension relationships. The perceptions were related to the contents of textbooks. The perception of the relationship of these models influenced on interpretations of the models' acid-base concept. In this study, we suggested that science teachers need to be aware of diverse models' roles.
