야생쑥 추출물을 이용한 견직물의 천연염색성에 관한 연구 (The study on Natural Dyeability of silk with Artemisia Extract)

  • The purpose of this study was to investigate the natural dyeability of silk on extract of Artemisia princeps, The experimental items were divided into the mordanting method. comonent of fabric, kind of mordant. The experimental study was done by laundering fastness, abrasion(dry/wet) fastness, perspiration(acid/alkali) fastness, light fastness test and color difference by C.C.M system. The summarized finding resulted from experiments and investigation are suggested as follows; First, in the C. C. M test on mordanting methods, color difference was significantly improved when mordants were treatmented. And the premordanting method showed the highest color difference, color was most yellow-greenish, Second, in the C.C.M test on component of fabrics, color difference of silk was higher than cotton. It is considered that silk has -$NH_2$ , -COOH, -OH than more than cotton. Third, in dyeing-fastness on mordants, laundering fastness showed that color-change was 2~3 grade, the contamination on attached fabric was 4~5 grade. perspiration fastness(acid/alkali) showed 4~5 grade nearly and those of acid was higher than alkali. abrasion fastness(dry/wet) was 4~5 grade and in Fe(3~4 grade) was lower than the other mordants. Forth, in color difference analysis on mordants, Fe(50.0) showed the highest and the order of color difference was alum(16.0), Cu(7.2), Sn(3.5), Al(3.1), Cr(2.3), The Hue was turned into yellow-greenish in alum mordant treatment, the luminocity of color was most dark in Fe(-48.9) and Cu(-7.2), chroma was the highest in alum (15.7) method.

Enterococcus faecium 19-46-4에 의한 Cholic Acid의 생산

  • A facultative anaerobe Enterococcus faecium 19-46-4 was used to study the production of an antimicrobial substance in anaerobic conditions. Major part of the antibiotic activity was found in the culture filtrate of the bacterium. The active compound was extracted by an equal volume of iso-butanol and concentrated in vacuo (at 50$\circ$C) before purification by C-18 liguid column chromatography and HPLC. A chromatographically pure compound was obtained by two passages of HPLC columns, The compound appeared as a pale-yellow powder. The yield was about 2.5 mg 1$^{-1}$ culture filtrate. The compound was named as KIST 194. KIST 194 were identified as cholic acid (3$\alpha$, 7$\alpha$, 12$\alpha$-trihydroxy-5$\beta$-cholan 24-oic acid) based on its physico-chemical properties determined by UV, IR, $^{1}H-NMR, $^{13}$C-NMR, El-MS and LC-MS.

바이오에탄올 생산을 위한 백합나무(Liriodendron tulipifera)칩의 동시당화발효 및 Response Surface Method를 이용한 옥살산 전처리 조건 탐색 (Evaluation of Oxalic Acid Pretreatment Condition Using Response Surface Method for Producing Bio-ethanol from Yellow Poplar (Liriodendron tulipifera) by Simultaneous Saccharification and Fermentation)

  • 이 연구에서는 백합나무($Liriodendron$ $tulipifera$)를 옥살산으로 전처리한 시료로부터 에탄올 생산 가능성을 조사하고, response surface methodology (RSM)를 도입하여 전처리 조건을 분석하고자 한다. 산농도, 전처리 시간, 반응 온도를 조절하여 $2^3$ factorial central composite experimental design을 바탕으로 각기 다른 15가지의 전처리 조건에서 시험하였다(central point에서 2반복). 전처리 후 고체 시료는 발효 균주인 $Pichia$ $stipitis$를 사용하여 동시당화발효로 에탄올 생산에 이용되었으며, 각각의 시료에서의 72시간에서의 에탄올 생산량(y,g/${\ell}$)이 최대값으로, 종속변수로써 RSM에 적용되었다. $180^{\circ}C$에서 40분간 0.013 g/g의 옥살산으로 처리한 시료가 가장 많은 양의 에탄올(9.7 g/${\ell}$)을 생산하였으며, response surface methodology 분석에 따르면, 전처리 조건에서 온도 인자가 ethanol에 가장 큰 영향을 미치는 것으로 제시되었으며, 결과는 수식화되어 나타내었다.

알긴산 가수분해물을 이용한 어육연제품용 천연 식품보존료의 개발 (Development of Alginic Acid Hydrolysate as a Natural Food Preservative for Fish Meat Paste Products)

  • Na-alginate에 등량의 ascorbic acid를 섞은 후 물을 가하여 알긴산 4%용액을 만든 다음 $121^{\circ}C$에서 $20{\sim}30$분간 가열하여 상대점도 1.05, pH 3.2의 whitish-yellow color의 불투명한 알긴산 가수분해물을 얻었으며, 이 알긴산 가수분해물은 nutrient broth에 0.1% 첨가(가수분해물속의 알긴산의 양을 기준하여)로 어묵의 주요 부패균인 Bacillus sp.의 증식을 억제시킬 수 있었다. 가수분해물 0.3% 첨가 제품은 pH 6.7이었고, 0.5% 첨가 제품은 pH 6.6으로서 무첨가 제품의 pH 7.4 보다는 약간 낮았다. 첨가제품은 약간 구수한 맛(고구마 맛과 유사)을 내었다. 제품의 탄력을 파단강도로 측정한 결과 가수분해물의 첨가량이 증가할수록 저하되는 것으로 나타났다. 제품의 색상은 가수분해물의 첨가량이 증가함에 따라 황색화되었으므로 0.3% 이상의 첨가는 곤란하였다. 어묵의 제조시 가수분해 물을 0.3% 첨가하고 $30^{\circ}C$에서 저장하였을 때 가수분해 물을 첨가하지 않은 제품에 비해 1일 가량 보존기간이 연장되었고, $20^{\circ}C$ 저장시에는 2일이, $15^{\circ}C$에서 저장하였을 때는 4일 가량 보존기간이 연장되는 것으로 나타났다.

고구마 품종과 국의 종류를 달리하여 제조한 고구마 소주의 특성 및 향기성분 (Effects of Sweet Potato Cultivars and Koji Types on General Properties and Volatile Flavor Compounds in Sweet Potato Soju)

  • 진홍미와 호박고구마, 흑국, 백국, 황국, 개량누룩을 사용하여 고구마 소주를 제조하였다. 최종 술덧의 성상은 진홍미의 경우 $^{\circ}Brix$는 9.2-11.4, 환원당 195.6-260.5 mg glucose/100 mL, pH 4.6-4.9, 산도 0.53-0.83%, 알코올 함량 13.2-16.2%이었고, 호박고구마의 경우 $^{\circ}Brix$는 7.0-8.4, 환원당 31.9-47.4 mg glucose/100 mL, pH 4.4-4.7, 산도 0.22-0.24%, 알코올 9.6-11.2% 이었다. 알코올 수 득량(L/ton)은 진홍미의 경우 흑국, 백국, 황국, 개량누룩에서 각각 229.2, 194.5, 238.6, 229.3 L/ton, 호박고구마의 경우 132.8, 144.4, 141.6, 167.4 L/ton이었다. 향과 맛에 대한 관능평가에 있어서 대부분의 고구마 소주가 일본산 고구마 소주인 쿠로기리시마 보다 좋은 평가를 얻었다. 특히 향은 진홍미 흑국(3.75)과 호박고구마의 개량누룩(3.78)이, 맛은 진홍미 흑국(3.0)과 호박의 흑국(3.0)이 가장 강하였다. 또한 고구마 소주의 향기성분은 decanoic acid, dodecanoic acid, tetradecanoic acid, hexadecanoic acid, 9-octadecanoic acid, octadecanoic acid ethylester류가 대부분의 피크면적율을 차지하고 있었으며, 쿠로기리시마 소주의 향기성분은 종류는 유사하였으나 대체적으로 피크 면적율이 적었다.

누에 배설물에서 Ethanol 추출한 녹색색소의 일반적 성질 (Characteristics of Greenish Pigments from Silkwarm Excrement by Ethanol Extraction)

  • A study of characteristics of greenish pigments from silkwarm excrement by ethanol extraction. Through visible absorption scanning, it showed two absorption peaks at 415 and 657nm, and it was shown to be greenish color. In the presence of light, the stability of pigments rapidly degraded, but in the presence of Al-foil, green and blue cover were very stable. It was shown to be stabilized at the temperature of 7$0^{\circ}C$ until 20days and presence of metal ions, such as Na+, K+, Ca2+, Mg2+ and Zn2+. The pigments was shown to be stabilized in 5% acetic acid solution, but in the presence of lactic acid, citric acid and tartaric acid solution were unstable. On the result of TLC analysis, pigments were shown to be composed of eight color fractions, and main color fractions were F-1, F-2 and F-3. In F-1, F-2 fractions were revealed green color and F-3 fraction were revealed yellow color.

Comparison and Sequence Analysis of the 3` - terminal Regions of RNA 1 of Barley Yellow Mosaic Virus

  • An isolate of barley yellow mosaic virus(BaYMV-HN) obtained from Haenam, Korea was compared with two BaYMV strains. BaYMV-Ⅱ-1 from Japan and BaYMV-G from Germany. The sequence of the 3'-terminal 3817nucleotides[excluding the poly (A) tail] of RNA 1 of BaYMV-HN was determined to start within a long open reading frame coding for a part of the NIa-VPg polymerase(26 amino acids). NIa-Pro polymerase (343 amino acids), NIb polymerase(528 amino acids) and the entire capsid protein(297 amino acids), which is followed by a noncoding region(NCR) of 235 nucelotides. In the partial ORFs, BaYMV-HN shows higher sequence homology with BaYMV-Ⅱ-1(99.5%) than BaYMV-G(92.7%). The 3' non-coding regions of BaYMV-HN(235nt) shows higher nucleotide sequence homology with BaYMV-G(235nt)(99.6%) than BaYMV-Ⅱ-1(231nt)(97.0%). The 3' NIa-Pro protein sequence of BaYMV-HN shows higher amino acid sequence homology with BaYMV-Ⅱ-1(95.0%) than BaYMV-G(93.6%), but, NIb protein sequence of BaYMV-HN shows same all amino acid sequence. The capsid protein sequence of BaYMV-HN(297aa) shows same with BaYMV-Ⅱ-1, and shows higher nucleotide sequence homology with BaYMV-UK (from United Kingdom)(97.3%) than BaYMV-G(96.9%) and G2(96.9%). Difference of capsid protein amino acid were 0-9 between the Japan, United Kingdom and Germany and were 2-6 between all Korean isolates. Many of the amino acid differences are located in the N-terminal regions of the capsid proteins from 1 to 74 amino acid positions.

  • 산성암(酸性岩)인 화강암(花崗岩)의 풍화잔적층(風化殘積層)을 모재(母材)로한 전남통(全南統)에 대(對)한 형태(形態), 물리적(物理的), 화학적(化學的) 및 생성(生成)과 분류(分類)에 관(關)하여 조사(調査)한 결과(結果)를 요약(要約)하면 다음과 같다. 1. 형태적특성(形態的特性)에 있어서 표토(表土)는 갈색(褐色) 내지(乃至) 암갈색(暗褐色)의 미사질양토(微砂質壤土) 내지(乃至) 토양(土壤)이나 침식(侵蝕)을 받은 곳에서는 진갈색(眞褐色) 내지(乃至) 황적색(黃赤色)의 미사질식양토(微砂質埴壤土) 또는 식양토(埴壤土)이다. 심토(心土)는 황적색(黃赤色) 내지(乃至) 적색(赤色)의 미사질식양토(微砂質埴壤土) 내지(乃至) 미사질식토(微砂質埴土)로서 구조표면(構造表面)에 엷은 점토막(粘土膜)이 형성(形成)되여 있다. 기층(基層)은 심(甚)하게 풍화(風化)된 화강암(花崗岩)의 잔적모재(殘積母材)이며 토양(土壤)의 깊이는 매우 깊다. 2. 물리적(物理的) 특성(特性)에서 토양입자(土壤粒子)의 분포비율(分布比率)은 표토(表土)에서 심토(心土)로 내려 갈수록 점토(粘土)의 함량(含量)이 증가(增加)되어 심토(心土)의 중앙부분(中央部分)이 34-45% 이며 특(特)히 미사(微砂)의 함량(含量)은 50~60%로서 표토(表土) 및 심토(心土)의 차이(差異)는 별(別)로 없다. 3. 화학적(化學的) 성질(性質)은 유기물(有機物) 함량(含量)이 낮고 토양반응(土壤反應)이 강산성(强酸性)이며 염기포화도(鹽基飽和度)가 낮다. 4. 전남통(全南統)은 온난(溫暖) 습윤(濕潤)한 기후조건하(氣候條件下)에서 생성(生成)되며 분류(分類)에 있어 미농무성(美農務省) 7차시안(次試案)에 의(依)하면 Typic Hapludults, UNESCO/FAO의 분류(分類)에 의(依)하면 Helvic Acrisols에 속(屬)한다.

Changes of Air Permeability and Moisture Absorption Capability of the Wood by Organosolv Pretreatment

  • The air permeability of yellow poplar log cross section before and after organosolv pretreatment was investigated, and the moisture absorption of control and organosolv pretreated rectangular parallelepiped specimens was investigated in this study. It was revealed that the diameters of through pores were enlarged and the number of bigger pore was increased by the organosolv pretreatment. The air permeabilities of the cross sections of yellow poplar log were changed from 1.61 darcy to 23.30 darcy, but their weights were reduced by 5 percent. The equilibrium moisture content of control wood specimen at the exposed relative humidity were 5.9 % at 32 %, 9.7 % at 58 %, 14.8 % at 80.5 %, 19.7 % at 90 %, 25.7 % at 95 % and 29.9 % at 100%. The equilibrium moisture content of the specimens pretreated with the parameter of sulfuric acid catalyst of 0.5 % (w/w) were 19.5 % at 32 %, 29.3 % at 58 %, 39.6 % at 80.5 %, 59 % at 90 %, 111.3 % at 95 % and 111.3 % at 100 %, while those pretreated with the parameter of sulfuric acid catalyst of 1.0 % (w/w) were 17.4 % at 32 %, 23.9 % at 58 %, 27.7 % at 80.5 %, 40.6 % at 90 %, 68.8 % at 95 % and 110.0 % at 100 %. The moisture absorption of organosolv pretreated rectangular parallelepiped specimens was higher than that of control specimen.

Sequence Analysis of the Coat Protein Gene of a Korean Isolate of Iris Severe Mosaic Potyvirus from Iris Plant

  • The coat protein gene of iris severe mosaic potyvirus, which was isolated in Korea, ISMV-K, from iris plant was cloned and its nucleotide sequence was determined. The coat protein of the virus contained 252 amino acid residues, including five potential N-glyxosylation site motifs. The coat protein of ISMV-K has 99.1% and 98.4% sequence identities with those of the Netherlands isolate of ISMV (ISMV-Ne) form crocus for the nucleotide and amino acids, respectively. The coat protein of ISMV-K has 50.4% to 60.3% nucleotide sequence identities and 47.3% to 55.7% amino acid identities with those of other 21 potyviruses, indicating ISMV to be a distinct species of the genus. The coat protein of ISMV-K was closely related with bean yellow mosaic virus and clover yellow vein virus in the phylogenetic tree analysis among the potyviruses analyzed. ISMV was easily and reliably detected from virus-infected iris leaves by RT-PCR with a set of the virus-specific primers.
