HPLC/DAD를 이용한 6종(種) 우슬(牛膝)의 분류기준 연구;우슬(牛膝)(쇠무릎, Achyranthes japonica $N_{AKAI}$)로부터 20-hydroxyecdysone 분리.동정 및 산지별 우슬의 HPLC 패턴 비교 (A Study on Discriminative Criteria of 6 Kinds of Achyranthis Radix Using HPLC/DAD;Isolation and Identification of 20-hydroxyecdysone from Aclryranthes japonica $N-{AKAI}$ and Comparison of Patterns of Achyranthis Radix from Different Locations by HPLC)

  • Objectives : This study was performed to investigate the discriminative criteria of 6 kinds of Achyranthis Radix by HPLC/DAD. Methods : 20-hydroxyecdysone is isolated by silica gel column chromatography ($CHCl_3$:MeOH, 7:1-1:1 v/v) and identified by nuclear magnetic resonance, A high-performance liquid chromatographic method with diode array detection was used to identify 20-hydroxyecdysone in A. japonica. The analysis was performed using $C_{18}$ column with isocratic elution consisted of 18% acetonitrile and 82% water and the detection was carried out by DAD at 254 nm. 6 kinds of Achyranthis Radix from different locations were extracted in MeOH. Each extracts was analyzed by HPLC in same condition as used in analysis of 20-hydroxyecdysone. The identities of each extracts were determined by comparing the retention time and UV spectrum with that of reference compound. Results : 1. A. japonica and A. bidentata showed the similar patterns of HPLC chromatogram and 20-hydroxycedysone was present in both of them because the peaks having the same retention time and UV spectrum as 20-hydroxyecdysone were shown in the HPLC chromatograms of A. japonica and A. bidentata 2. Cyathula officinalis and C. capitata showed the similar patterns of HPLC chromatogram. The peak having the same retention time and UV spectrum as 20-hydroxyecdysone was shown in the HPLC chromatogram of C. capitata but not shown in the HPLC chromatogram of C. officinalis. 3. Two species of medicinal drugs from Sacheon province showed similar patterns of HPLC chromatogram. Achyranthis Radix from Sacheon(wild) did not have 20-hydroxycedysone but Achyranthis Radix from Sacheon(cultivated) showed the peak having the same retention time as 20-hydroxyecdysone but UV spectrum of the peak was different from that of 20-hydroxyecdysone. Conclusions : These results suggested that 20-hydroxyecdysone could be the discriminative criteria for Achyranthis Radix contain 20-hydroxyecdysone though they belong to different genus and species. And the patterns of HPLC chromatogram also could be the discriminative criteria as the different species of Achyranthis Radix belonging to the same genus showed similar patterns of HPLC chromatogram.

우슬(牛膝)의 콜라겐 유도 관절염 생쥐에 대한 개선 효과 (The therapeutic effect of Achyranthis Radix on the collagen-induced arthritis in mice)

  • Objectives : The present study was undertaken to determine whether the water extract of Achyranthis Radix, which is the roots of Achyranthes japonica (Achyranthis Radix, AR), is efficacious against collagen-induced arthritis (CIA) in DBA/1J mice. Methods : Mice were immunized with bovine type II collagen and orally treated with AR-W (50 and 100 mg/kg/bw) from days 21 to 42 after immunization. Arthritis was evaluated by arthritic score, histological examination of knee joint and serological markers such as TNF-${\alpha}$, $PGE_2$ and anti-type II collagen (C II)IgG. Results : The results showed that comparing with untreated CIA mice, treated with AR-W significantly suppressed the clinical score and joint tissue pathological damages, reduced the serum levels of TNF-${\alpha}$, $PGE_2$ and anti-C II IgG in CIA-mice. These results suggest that AR-W can effectively alleviate inflammatory response on CIA, and anti-inflammatoy of AR-W can be attributed, at least partially, to the inhibition of inflamamtory mediators, $PGE_2$ and TNF-${\alpha}$, in CIA. Conclusions : This study suggests that AR-W has a therapeutic potential in inflammatory joint diseases such as rheumatoid arthritis.

양릉천(陽陵泉) 우슬(牛膝) 약침이 흰쥐의 고지혈증에 미치는 영향 (The Effect of Achyranthis Radix Herbal-acupuncture on Hyperlipidemia in Rats)

  • Objective & Methods : The purpose of this study is to observe the effects of herbal- acupuncture with Achyranthis Radix(AR-HA) at GB34(Yangleungchean) on hyperlipidemia in rats. The author performed several experimental items to analyze the levels of various components and enzymes in serum and liver, as well as the histological changes of liver and aorta. Results : 1. Achyranthis Radix herbal acupuncture solution increased DPPH radical scavenging activity and HMG-CoA reductase inhibition rate in rat liver cells. 2. AR-HA at GB34 decreased the level of serum total cholesterol, and increased the ratios of HDL-cholesterol to total cholesterol, phospholipid to total cholesterol in hyperlipidemic rat. 3. AR-HA at GB34 decreased the hepatic HMG-CoA reductase activity in hyperlipidemic rat. 4. AR-HA at GB34 increased the hepatic GSH level in hyperlipidemic rat. Conclusion : From the above results, it is suggested that AR-HA at GB34 has a therapeutic effect on hyperlipidemia.

우슬 추출물이 골아세포 증식과 분화에 미치는 효과 (Effects of Radix Achyranthis Bidentatae Extract on Proliferation and Differentiation in Human Osteoblast-like Cells)

  • In order to investigate the effects of Radix Achyranthis Bidentatae (RAB) on the growth and differentiation of human osteoblast-like cells, we supplemented the culture medium of MG-63 cells with various concentrations of RAB water extracts. RAB extracts significantly stimulated cell growth, as confirmed by the colorimetric MTT (3-dimethylthiazol-2,5-diphenyltetrazolium bromide) assay. RAB extracts also increased the alkaline phosphatase (ALP) activity, which is a osteoblast differentiation marker. These results suggest that RAB can stimulate osteoblastic activity and may represent new pharmacological tools for the treatment of osteoporosis.

우슬 메탄올 추출물이 흰쥐의 카드뮴 흡입독성완화에 미치는 영향 (Effects of Methanol Extract of Radix Achyranthis Bidentatae on Cadmium Inhalation Toxicity in Rat)

  • To study the effects between Cd inhalation toxicity and methanol extract of Radix Achyranthis Bidentatae, 4 rat groups were exposed to Cd aerosol by whole-body inhalation exposure for 6 hours/day, 5 days/week, and 4 weeks. Cd concentration in air was 0.98㎎/㎥ and mass median diameter(MMD) was 1.78㎛. 3 different dose intraperitoneal injections of methanol extract of Radix Achyranthis Bidentatae to 3 inhalation exposure groups applied for 4 weeks and the results were as follows: The highest body weight gain for 4 weeks and food intake per day were from inhalation exposure group Ⅲ(p<0.05). The highest lung weight was from inhalation exposure group Ⅲ and the highest liver and kidney weight were from inhalation exposure group Ⅱ(p<0.05). The lowest Cd content in lung was 22.77㎍/g from inhalation exposure group Ⅲ(p<0.05). The highest Cd concentration in blood was 11.71㎍/㎗ from inhalation exposure group Ⅰ(p<0.05). Cd concentrations of 14.87㎍/g in liver and 17.91㎍/g in kidney were the highest from inhalation exposure group Ⅰ(p<0.05). The lowest Cd concentration in liver and kidney were 5.71㎍/g and 3.17㎍/g from the control(p<0.05). For weekly Cd concentration in urine, the highest value was 0.48㎍/㎖ from inhalation exposure group Ⅲ of the 3rd week and inhalation exposure group Ⅰ, Ⅱ of the 4th week. For weekly Cd concentration in feces, the highest value was 0.32㎍/g from inhalation exposure group Ⅰ, Ⅱ, Ⅲ. The highest metallothionein concentration in lung was 89.02㎍/g from inhalation exposure group Ⅲ(p<0.05). The highest metallothionein concentrations in liver and kidney were 265.47㎍/g and 214.21㎍/g from inhalation exposure group Ⅲ, respectively(p<0.05). The highest Hct, Hb, and WBC values were from inhalation exposure group Ⅱ and the highest RBC value was from inhalation exposure group Ⅲ(p<0.05). Mostly damaged part in liver tissue was hepatic lobule and the degrees of damage were lessened by the intraperitoneal injection of methanol extract of Radix Achyranthis Bidentatae. Proximal, distal convoluted tubules and glomerulus in kidney tissue were mostly damaged part. Degeneration and swelling were partially observed but the degrees of kidney tissue damage were lessened more or less by the intraperitoneal injection of methanol extract of Radix Achyranthis Bidentatae.

우슬약침이 Rat의 Adjuvant 관절염에 미치는 영향 (Effects of Achyranthis Radix Aqua-acupuncture on Adjuvant Athritis in Rats)

  • Objectives : To investigate effects of Achyranthis Radix aqua-acupuncture on adjuvant arthritis in rats. Methods : After eliciting arthritis in Sprague dawley(SD) rats by injection of Freund's complete adjuvant for 2 weeks, normal saline was injected for the Exp. I group and Achyranthis Radix aqua-acupuncture was injected for the Exp. II group during 30days. The selected point was Dokbi (ST35) in both groups. Then the edema rate, the number of WBC, the quantity of total protein, albumin and globuline in the blood serum, and histological test of the muscular tissue were measured in the arthritis stricken areas. Results : I) The volume of the paws were checked. The volume of the paws was $0.84{\pm}0.l4mm$ in the Exp. I group and $0.47{\pm}0.14mm$ in the Exp. IIgroup. The swelling of the paws was restricted significantly in the Exp. II group(P<0.05). 2) The number of WBC was $10.34{\pm}0.14(l03/ml)$ in the normal group and $37.47{\pm}5.46(103/ml)$ in the Exp. I group. It was $21.24{\pm}2.58(103/ml)$ in the Exp. II group. This fact showed that treatment with Achyranthis Radix aqua-acupuncture adminstered to the Exp. II group was more effective than the Exp. I group in the treatment of arthritis(P<0.05). 3) The content of the total protein in the blood serum was $6.14{\pm}43g/dl$ in the normal group, $7.95{\pm}0.94g/dl$ in the Exp. I group, and $6.41{\pm}0.68g/dl$ in the Exp. II group. There was no significance in total protein between the Exp. II group and the Exp. I group from the statistical analysis. 4) The content of albumin in the blood serum was $2.94{\pm}0.l3g/dl$ in the normal group, $2.01{\pm}0.48g/dl$ in the Exp. I group and $3.15{\pm}0.27g/dl$ in the Exp. II group, which revealed a significant increase in the serum albumin from the statistical analysis of the Exp. II group compared with the Exp. I group. 5) The content of the globulin in the blood serum was $3.19{\pm}$0.48g/dl in the normal group, $4.70{\pm}1.26g/dl$ in the Exp. I group and $3.26{\pm}0.57g/dl$ in the Exp. II group. There was no significance in the serum globulin between the Exp. II group and Exp. I group from the statistical analysis. 6) In histological findings, because of severe inflammatory reaction, remarkably irregular tissue and large amount of inflammatory cells were found in the Exp. I group. But the Exp. II group showed a small amount of inflammatory cells, the refrained inflammatory state and even recovering state. Conclusions : These results show that Achyranthis Radix aqua-acupuncture reduces the inflammatory reaction and muscular tissue necrosis in SD rat's paws induced by Freund's complete adjuvant.

생약복합제 GCSB-5의 품질 표준화를 위한 오가피 및 우슬의 함량 분석 (Quantitative Analysis of Acanthopanacis Cortex and Achyranthis Radix for Standardization of GCSB-5 Preparation)

  • In this study, we have investigated the HPLC analysis methods and quantitative analysis of standard compounds for quality standardization of a medicinal crude drug GCSB-5, a herbal formulation consisting of 6 medicinal plants (Acanthopanacis Cortex, Achyranthis Radix, Ledebouriellae Radix, Cibotii Rhizoma, Glycine Semen, Eucommiae Cortex) which are used in traditional medicine to treat various bone disorders. HPLC analysis methods of acanthoside D(Acanthopanacis Cortex), 20-hydroxyecdysone(Achyranthis Radix) which were known standard compounds among 6 medicinal plants were developed on crude material and product. And validation of HPLC analysis methods were conformed for verification of HPLC methods by check to specificity, linearity, intra-day precision, inter-day precision and accuracy following ICH guideline. Content of acanthoside D and 20-hydroxyecdysone on raw material of GCSB-5 were decided at 0.577-0.578 mg/g and 0.311-0.312 mg/g. And we confirmed that content of acanthoside D and 20-hydroxyecdysone on GCSB-5 preparation were 0.302-0.303 mg/capsule and 0.113-0.115 mg/capsule.

생약재 추출물의 Staphylococcus aureus 와 Escherichia coli O157:H7에 대한 항균효과 (Antimicrobial Effects of Herbal Medicine Extracts on Staphylococcus aureus and Escherichia coli O157:H7)

  • 50가지 생약재를 95% 에탄올과 물추출하여 식중독 세균인 S. aureus와 E. coli O157:H7에 대한 항균활성을 조사하였다. 한천배지확산법에 따른 생육저해효과를 측한 결과, S. aureus 에 대하여 우슬>목단피>지실>황백, 건강, 민들레 순으로, E. coli O157:H7에 대하여 목단피>우슬>건강>지실, 황백 순으로 활성을 나타내었다. MIC는 S. aureus에 대하여 우슬이 156.25 $\mu$g/mL, 목단피와 황백이 625 $\mu$g/mL로 나타났으며, E. coli O157:H7에 대하여 우슬과 목단피의 MIC는 각각 625, 312.5 $\mu$g/mL로 높은 항균력을 나타내었다. 가장 높은 활성을 보인 우슬과 목단피를 용매분획(hexane, CHC1$_3$, ethyl acetate, butanol)하여 한천배지 확산법에 의한 두 균에 대한 생장저해 효과를 측정하였다. 우슬은 ethyl acetate 분획물에서, 목단피는 CHC1$_3$과 ethyl acetate 분획물에서 각각 높은 활성을 나타내었다.

우슬 중 ${\beta}-Ecdysone$의 초임계추출법(SFE)과 용매추출법의 비교 (A Comparative Study on the Solvent Extraction and Supercritical Fluid Extration Method of ${\beta}-Ecdysone$ in Achyranthis radix)

  • 우슬(Achyranthis radix)중 ${\beta}-ecdysone$의 초임계추출법과 용매추출법의 비교를 위해 초임계유체추출(SFE) 및 용매추출 후 초임계유체크로마토그래피(SFC)에 의한 우슬 중 ${\beta}-ecdysone$ 분석을 실시하였다. SFE에 있어서 추출 최적압력은 300 atm이었으며, 온도는 $80^{\circ}C$, 추출량은 100 ml, 보조제인 methanol 함량비율은 20%였다. 우슬에서 ${\beta}-ecdysone$의 분석시 SFC의 최소검출량은 5 ng이었고, SFE의 회수율은 줄기에서는 $90.7{\sim}93.5%$였으며, 뿌리에서는 $77.8{\sim}81.9%$였다. 용매추출의 회수율은 줄기에서는 $76.4{\sim}93.0%$였고 뿌리에서는 $80.6{\sim}93.1%$였다. 우슬의 생육기별 조사에서 ${\beta}-ecdysone$은 주로 뿌리에서 많이 검출되었고 1년생이면서 8월에 수확한 것이 654.9 ppm으로 가장 많이 함유한 것으로 나타났다.
