Split-flow thin cell fractionation (SPLITT fractionation, SF) is a particle separation technique that allows continuous (and thus a preparative scale) separation into two subpopulations based on the particle size or the density. In SF, there are two basic performance parameters. One is the throughput (TP), which was defined as the amount of sample that can be processed in a unit time period. Another is the fractionation efficiency (FE), which was defined as the number % of particles that have the size predicted by theory. Full-feed depletion mode (FFD-SF) have only one inlet for the sample feed, and the channel is equipped with a flow stream splitter only at the outlet in SF mode. In conventional FFD-mode, it was difficult to extend channel due to splitter in channel. So, we use large scale splitter-less FFD-SF to increase TP from increase channel scale. In this study, a FFD-SF channel was developed for a large-scale fractionation, which has no flow stream splitters (‘splitter less’), and then was tested for optimum TP and FE by varying the sample concentration and the flow rates at the inlet and outlet of the channel. Polyurethane (PU) latex beads having two different size distribution (about 3~7 µm, and about 2~30 µm) were used for the test. The sample concentration was varied from 0.2 to 0.8% (wt/vol). The channel flow rate was varied from 70, 100, 120 and 160 mL/min. The fractionated particles were monitored by optical microscopy (OM). The sample recovery was determined by collecting the particles on a 0.1 µm membrane filter. Accumulation of relatively large micron sized particles in channel could be prevented by feeding carrier liquid. It was found that, in order to achieve effective TP, the concentration of sample should be at higher than 0.4%.
Anthocyanin synthesis in strawberry (Fragaria x ananassa cv Maehyang) begins approximately 26 days postflowering and continued throughout fruit ripening. A set of cDNA clones encoding the anthocyanin biosynthetic enzymes were isolated from strawberry. A pair of primers were designed for polymerase chain reaction (PCR) through the comparison of the nucleotide sequences of homologous genes from diverse plants. Reverse transcriptase-PCRs were performed using cDNA synthesized from ripe fruit total RNA and the primers corresponding to each gene. Eight genes of the anthocyanin pathway were cloned and confirmed by sequencing to code for phenylalanine ammonia lyase (PAL), 4-cummarate CoA ligase (4CL), chalcone synthase (CHS), chalcone isomerase (CHI), flavanone-3-hydroxylase (F3H), dihydroflavonol 4-reductase (DFR), anthocyanidine synthase (ANS), UDP-glucose:flavonoid-3-O-glucosyl-transferase (UFGT). Northern analyses showed that the corresponding genes were differentially expressed during the fruit development process. All genes except PAL were predominantly expressed in fruit. Expression of PAL, DFR and ANS was detected 10 days postflowering at the early stage of fruit development, declined for a while and sharply increased 22 days postflowering then showed a peak 34 days postflowering. The other genes, however, were not expressed up to 22 or 30 days postflowering when the initial fruit ripening events occur at the time of initiation of anthocyanin accumulation. The onset of anthocyanin synthesis in ripening strawberry coincides with a coordinated induction of the anthocyanin pathway genes, suggesting the involvement of regulatory genes. We propose that at least two different regulatory mechanisms playa role in the biosynthesis of anthocyanin during color development of strawberry.

Optimum pasture management during the summer season is an important factor to maintain good regrowth and persistence of pasture. The field experiment was carried out to investigate the effects of cutting management on growth and reserve carbohydrates in stubble. and on dry matter yield of orchardgrass dominated pasture during the mid-summer season. Three different cutting times(July 15, 25 and August 5) as a main plot and three cutting heights(3, 6 and 10cm) as a sub plot were disigned with three replications. The experiment was done at pasture of Changsung Agricultural High School in 1988. The results obtained were summarized as follows: 1. Temperature of soil surface and underground in the cutting of July 15 and July 25 was not showed significantly different, because of low intensity of radiation with a run of wet weather, and that of August 5 cutting tends to be lowed in high cutting height during the mid-summer season. 2. Amount of soil moisture against cutting height showed that high cutting had a low content. 3. Growth rate against regrowth of plant height, lear length and leaf area showed to be fast in high cutting. 4. Rapid recovery period of carbohydrate content after cutting and large amount of carbohydrate accumulation were showed in the cutting of July 15 and 25 compared with August 5 cutting in the reserve carbohydrate content against cutting of time and height during the mid-summer season. 5. Dry matter yield of the 4th and 5th cutting showed to be remarkable in July 15 cutting compared to those of July 25 and August 5. From the above results, it is suggested that the 10cm cutting height during the mid-summer season is the most effective for good regrowth, reserve carbohydrates and dry matter yield of orchardgrass.
This study was performed in order to investigate the anti-obesity effect of Polygala tenuifolia on lipid mechanism in 3T3-L1 adipocytes. The chemical composition of the P. tenuifolia was analyzed in order to assess its nutritional value. Total dietary fiber was the highest among the proximate component of the P. tenuifolia. These results showed that the P. tenuifolia may be used as a potential functional ingredient for anti-obesity effect. Intracellular lipid droplets in the adipocyte were stained with oil-red O dye and quantified. In comparison to the control, lipid accumulation was significantly decreased by 40.1% and 22.4% when treated with the water extract and 70% EtOH extract of the P. tenuifolia at the concentration of $10{\mu}g/mL$, respectively. The anti-adipogenic effect of the water extract was stronger than that of the 70% EtOH extract. The gene expression levels were measured via Western blot and real-time PCR. As a result, the water extract was found to have decrease the gene expression of SREBP-1c, PPAR, $C/EBP{\alpha}$, FAS, ACC in a dose-dependent manner. These indicate that the water extract inhibits pre-adipocyte differentiation and adipogenesis by blocking the SREBP-1c gene expression in 3T3-L1 cells. Therefore, P. tenuifolia can be used as an effective anti-obesity agent.
Chlorinated water in swimming pools contains chloroform at elevated levels compared to chlorinated drinking water Chloroform levels in four indoor swimming pools(swimming pools A, B and C in a city of Korea and swimming pool D in a city of New Jersey in the United States) were examined. The chloroform levels in the water of swimming pool C (city-managed) weve shown to be significantly(p=0.0001) different from those of private swimming pools A and B: the mean chloroform levels in the pools A, B, and C are 22.8, 17.8, and 31.1 $\mu\textrm{g}$/l, respectively. Furthermore, all of these chloroform levels are significantly(P=0.0001) different from those of New Jersey: chloroform concentration of the Korean pools ranged from 10.9 $\mu\textrm{g}$.l to 47.9 $\mu\textrm{g}$/l with a mean of 23.2 $\mu\textrm{g}$/l, while it ranged from 27 $\mu\textrm{g}$/l to 96$\mu\textrm{g}$/l with a mean of 64.4 $\mu\textrm{g}$/l in the New Jersey pool. The disinfection processes would cause part of this difference since the swimming pools in Korea applied both chlorination and ozonation method, while the swimming pool in New Jersey used chlorination method only. It was implied that swimming parameters inconsistently vary, resulting in fluctuation of and no constant accumulation of chloroform in the water with the change of time for the day. A regression analysis showed no relationships between sampling time and chloroform concentrations for the sampling day in the swimming pools of Korea. A F-test indicated no significant difference of chloroform concentrations in the morning and afternoon samples collected in the swimming pools. Ingestion dose was estimated to be 0.58$\mu\textrm{g}$ from an hour swimming in a city of Korea, taking into accounting an average of 23.2 $\mu\textrm{g}$/l in swimming pools in the city In extreme situation, the ingestion dose was estimated to be 12.0 $\mu\textrm{g}$ from an hour swimming in a city of Korea.
Korean Shamanism has long been regarded as the peripheral cult or supersition by the majoriy of the society. This fact is a result by the influence of the mass media, especially the motion pictures, which intentionally reflected the negative images of the Shamanism. The documentary films, which stress the objectivity and the reality, rather disclose the neutral position toward the Shamanism. In fact, the directors of the documentary films who have more or less close relationship with the Shamanism have tried to exhibit the mental world and the traditionality of the Shamanism. In this paper, I will explore the value of Shamanism reflected in the documentary films. I focus of two films; directed by the Korean and directed by a foreigner. In this way, I can compare the attitudes of the two directors toward the Korean Shamanism. The director of confessed that he was attracted by the aspect of a musical underlied in the shaman ritual. However, the film does not show the artistic beauty of the shaman music because the director failed to understand the essential aspect of the shaman ritual, that is, the music. In this way, the director failed to show the distinct characteristics of the shamanism to the audience. The director of , a music herself, was focused on the music of the shamanism. The story flows to the adventure to seek a "master" with a long journey to enjoy diverse genres of Korean performing arts. This story resembles the epic shaman song, the Princess Bari. In this way, the audience can easily grasp the beauty of Korean culture. Music is said to be a universal language and, at the same time, a non-universal language which reflects a special trait of a cultural community. The Korean shaman ritual music is a non-universal language that is an accumulation of the Korean culture for a long time. The Korean director fails to exhibit the essential characteristics of the Shamanism since he does not have enough knowledge toward the shaman ritual music in . However, the foreign director, who is a music herself, successively disclose the underlying beauty of Korean shaman ritual music and Korean traditional culture in .
This study was conducted to get some basic information about rice plant development rate and heading ecology in various climatic conditions, growing nine varieties at three locations, Jinbu as the mountainous cool area, Suweon and Iri as the plain area for two years from 1987 to 1988. Average daily air temperature and day length from transplanting to heading date were analyzed in relation to the heading. Heading date and development rate of each variety were estimated by the Symplex method and the fitness of the model was evaluated. The results obtained as follows: Average daily air temperatures among varieties during the period from transplanting to heading ranged from 18 to $19^{\circ}C$ in Jinbu, from 22.5 to $23.5^{\circ}C$ in Suweon, and from 23.5 to $24.5^{\circ}C$ in Iri, the late-maturing varieties requiring the higher temperatures. The average heading days were about 20 days longer in Jinbu and $3{\sim}5$ days shorter in Iri than those in Suweon in all varieties. Little differences in accumulated temperature from transplanting to heading were observed in regions and years, and also among varieties. Developmental stages could be expressed as the accumulation of daily development rate and the predicted heading dates by the Symplex method were similar to the observed ones. The development rate of each variety varies with air temperatures. The early maturing Japonica types including the Unbong variety were fast in development rate at lower temperatures, but the late-maturing varieties of the Japonica type were late. A model to predict the heading dates of rice varieties by the Symplex method using air temperatures and day lengths was feasible.
Yams (Dioscorea spp), which are edible or medicinal tuber crops, are a important crop in South Korea. Yams require a high level of soil fertility. The various cultural practices such as fertilizing and plowing were tested for marketable tuber production in Chinese yam (Dioscorea opposita). Tuber yield was also affected by organic matters in soil. Application higher level of organic matters result in increased each tuber weight and tuber yield per unit area. The nutrient absorption quantity of the plant such as nitrogen, phosphoric acid, calcium and potassium was increased from 100~120 days after planting, which time to begin tuber enlargement. The tuber yield was increased when the fertilization increased in quantity. Total yield and marketable ratio were the highest in 31~32 kg/10a of nitrogen fertilizer. From above result, income become larger with increase of marketable yield and quality improvement at 63% (27 kg/10a) level of conventional N fertilization (43 kg/10a). The tuber yield was not significantly different between with in various application level of potash fertilizer. Tuber size and weight decreased accordingly to decreased fertilizing level, so the rates of small tubers increased greatly at cultivation without chemical fertilizer. In considering the accumulation rates of allantoin in Chinese yam tubers, the apt harvest season was after October. The allantoin quantity of it was not influenced with nitrogen fertilizing. Moreover it was advantageous with decrease of chemical fertilizer and appropriate fertilizing in soil environment protection. Commercial tuber's number and yield were increased in trenching before planting with trencher compared with rotavating with tractor.
Protection against ischemia-reperfusion injury is crucial for successful transplantation of the lung. It has been known that nitric oxide has many favorable effects on the donor lungs but at the same time, has some potential side effects of cytotoxicity. In this regards, we investigated whether the administration of nitroglycerin could decrease ischemia-reperfusion injury in isolated rat lung reperfusion model for the confirmation of the effect of nitroglycerin, a donor of nitric oxide, on lung transplantation. Material and Method: 35 Sprague-Dawley species male white rats were used for this experiment. For nitroglycerin group (n=18), nitroglycerin was administered intravenously followed by mixed in flushing solution for preservation. As a control group (n=17), we used the same amount of normal saline. To evaluate the effect of nitroglycerin on the lung, heart-lung block was obtained, weighed and stored in University of Wisconsin Solution at 1$0^{\circ}C$ for 24 hours. In each group of the isolated lungs, reperfusion was carried out with Krebs-Hensleit-diluted human blood for 60 minutes. As parameters of the state of the isolated lung, peak inspiratory and pulmonary arterial pressures were continuously recorded. Oxygen and carbon dioxide tension of reperfusing blood were measured before and after 30, 60 minutes of reperfusion. After sixty minutes of reperfusion, protein content in bronchoalveolar lavage fluid was measured also for the evaluation of the degree of alveolar flooding. Lung myeloperoxidase activity was determined to verify the accumulation of neutrophils. Results: Although statistically significant differences were not noted in peak inspiratory and pulmonary arterial pressure between control and nitroglycerin group, latter group showed lowering tendency of pulmonary arterial pressure during the entire reperfusion period. Oxygen tension was higher (p<0.05) in nitroglycerin group compared with that of the control group, in contrast, there were no differences in carbon dioxide tension, protein content in bronchoalveolar lavage fluid and myeloperoxidase activity between the groups. In the examination of ultrastructural changes, nitroglycerin denoted the protective effect on the pulmonary architecture compared with that of control group. Conclusion: Collectively, on the bases of these experimental results, prior treatment of donor lung with nitroglycerin could result in better preservation of the lung. Consequently, these nitroglycerin preserved lungs are thought to be more suitable for successful transplantation of the lung.
The present experiment was performed to evaluate the osteogenic differentiation of human adipose tissue derived mesenchymal stem cells (ATMSCs) seeded in bioceramic-poly D,L-latic-co-glycolic acid (PLGA) scaffold. Osteogenic differentiation of ATMSCs were induced using the osteogenic induction (OI) medium. ATMSCs were cultured with OI medium during 28 days in well plate. The proliferation of ATMSCs in OI medium group was significantly increased for 14 days of plate culture but slowed after 21 days. On the other hand, proliferation in the control group showed constant increase for 28 days of culturing. The alkaline phosphatase (ALP) activity of ATMSCs in OI medium group increased during the 21 days of culture but decreased on 28 days. However, in control group ALP activity of ATMSCs was continuously decreased as time goes. Nodule was observed at 21 days of culture in OI medium group and confirmed accumulation of calcium in cell by alizarin red staining. ATMSCs were seeded in PLGA scaffold or in Bioceramic-PLGA scaffold, and cultured with OI medium. ALP activity of ATMSCs by osteoblast differentiation in each scaffold increased on 21 days of culture and decreased rapidly on 28 days. ALP activity of ATMSCs was increased highly in Bioceramic-PLGA scaffold compared to PLGA scaffold on 21 days of culturing. SEM-EDS analysis demonstrated that calcium and phosphate content and Ca/P ratio in Bioceramic-PLGA scaffold increased higher than in PLGA scaffold. Biodegradability of scaffold at 56 days after implantation showed that Bioceramic-PLGA scaffold was more biodegradable than PLGA scaffold. The results demonstrated that the differentiation of ATMSCs to osteoblast were more effective in scaffold culture than well plate culture. Bioceramic increased cell adhesion rate on scaffold and ALP activity by osteoblast differentiation. Also, bioceramic was considered to increase the calcium and phosphate in scaffold when ATMSCs was mineralized by osteogenic differentiation. Bioceramic-PLGA scaffold enhanced the osteogenesis of seeded ATMSCs compared to PLGA scaffold.
