Water Extract of Rosa laevigata Michx. Protects Hepatocytes from Arachidonic Acid and Iron-mediated Oxidative Stress (아라키돈산과 철 유도성 산화적 스트레스에 대한 금앵자(金櫻子) 열수 추출물의 간세포 보호 효능)

  • Objectives : Rosa laevigata Michx. has been used for the treatment of renal disease in traditional Korean medicine. In this study, we investigated cytoprotective effect of R. laevigata water extract (RLE) against oxidative stress induced by arachidonic acid (AA) + iron.Methods : To evaluate the protective effects of RLE against AA + iron-induced oxidative stress in HepG2 cell, cell viability and changes on apoptosis-related proteins were assessed by MTT and immunoblot analyses. The effects of RLE on reduced glutathione level, production of reactive oxygen species and mitochondrial membrane potential were also monitored. Furthermore, to verify underlying molecular mechanism, NF-E2-related factor 2 (Nrf2) was examined by immunoblot analysis. Additionally, Nrf2 transactivation and its downstream target genes expression were also determined by reporter gene and realtime RT-PCR analyses.Results : RLE pretreatment (30-300 μg/ml) prevented cells from AA + iron-mediated cell death in a concentration dependent manner. In addition, 100 μg/ml RLE inhibited AA + iron-induced glutathione depletion, reactive oxygen species production and mitochondrial dysfunction. RLE accumulated nuclear Nrf2 and also transactivated Nrf2, which was evidenced by antioxidant response element- and glutathione S-transferase A2-driven luciferase activities and mRNA level of glutamate-cysteine ligase catalytic subunit, NAD(P)H:quinone oxidoreductase 1 and sestrin 2. Moreover, protective effect of RLE against AA + iron was abolished in Nrf2 knockout cells.Conclusions : These results indicate that RLE has the ability to protect hepatocyte against oxidative stress through Nrf2 activation.

Determination of EMCs for Rainfall Ranges from Transportation Landuses (교통관련 토지이용에서의 강우계급별 EMC 산정)

  • The contribution of pollutant loadings from non-point source (NPS) to the four major rivers in Korea exceeded 22~37 % of the total loadings in 2004 and is expected to reach 60 % in 2020. Most of NPS loadings are coming from urban areas, especially from paved areas. Because of high imperviousness rate, many types of NPS pollutant are accumulating on the surface during dry periods. The accumulated pollutants are wash-off during a storm and highly degrading the water quality of receiving water bodies. For this reason, the Korean Ministry of Environment (MOE) developed the Total Maximum Daily Load (TMDL) program to protect the water quality by managing the point source and NPS loadings. NPS has high uncertainties during a storm because of the characteristics of rainfall and watershed areas. The rainfall characteristics can affect on event mean concentrations (EMCs), mass loadings, flow rate, etc. Therefore, this research was performed to determine EMCs for rainfall ranges from transportation landuses such as road and parking lot. Two sites were monitored over 45 storm events during the 2006/06 through 2008/10 storm seasons. Mean TSS EMCs decrease as rainfall ranges increase and highest at less than 10mm rainfall. The results of this study can be used to determine the efficient scale of BMP facility considering specific rainfall range.

Ginseng Transformation of Betaine Aldehyde Dehydrogenase Gene Relative Salt Resistant through Somatic Embryogenesis (염류내성관련 유전자 Betaine Aldehyde Dehydrogenase Gene의 인삼 체세포 배발생을 통한 형질전환)

  • Korean ginseng(Panax ginseng C.A. Meyer) is very difficult to obtain stable production of qualified ginseng roots because of variable stresses in soil environments. In transformation of ginseng with betain aldehyde dehydrogenase gene, compounds synthesized for controlling osmotic pressure such as proline, glycine, betaine, polyols and sugar were accumulated in cell for salt resistance in transgenic plants. 2 Agrobactgerium conjugants were acquired with bet A and bet B genes for solt resistant plants. A. tumefaciens MP90/pBetA and A. tumefaciens MP90/pBetB were recombined for increasing the tolerance to salt stress. To confirm the transformation of the binary vector, tobacco plant was transformed, and the transformant can grow on media containing high concentration of kanamycin. To identify NPT 11, BetA and BetB genes of the transformants, the band on the agarose was confirmed by PCR and RT-PCR techniques. The transformants of ginseng with bet A and bet B genes were acquired on the phytohormone free basic MS media containing only antibiotics and 1M mannitol used for selection of transgenic plant, but the transfomation efficiency for BetA and BetB was very low.

Pharmacokinetics of 11-Hydroxyaclacinomycin X (ID-6105), a Novel Anthracycline, after i.v. Bolus Multiple Administration in Rats

  • We investigated the pharmacokinetics of 11-hydroxyaclacinomycin X (ID-6105), a novel anthracycline, after intravenous (i.v.) bolus administration at a multiple dose every 24 h for 5 days in rats. To analyze ID-6105 levels in biological samples, we used an HPLC-based method which was validated in a pharmacokinetic study by suitable criteria. The concentrations of ID-6105 after the multiple administration for 5 days were not significantly different from the results after the single administration. The $t_{1/2\alpha}, t_{l/2\beta}, V_{dss}, and CL_{t}$ after the multiple administration were not significantly different from the values after the single administration. Moreover, the concentrations of ID-6105 1 min at day 1-5 after i.v. bolus multiple administration did not show the significant difference. Of the various tissues, ID-6105 mainly distributed to the kidney, lung, spleen, adrenal gland, and liver after i.v. bolus multiple administration. ID-6105 concentrations in the kidney or lung 2 h after i.v. bolus administration were comparable to the plasma concentration shortly after i.v. bolus administration. However, the ID-6105 concentrations in various tissues 48 h after i.v. bolus administration decreased to low levels. ID-6105 was excreted largely in the bile after i.v. bolus multiple administration at the dose of 3 mg/kg. The amounts of ID-6105 found in the bile by 12 h or in the urine by 48 h after the administration were calculated to be $14.1\% or 4.55\%$ of the initial dose, respectively, indicating that ID-6105 is mostly excreted in the bile. In conclusion, ID-6105 was rapidly cleared from the blood and transferred to tissues, suggesting that ID-6105 might not be accumulated in the blood following i.v. bolus multiple dosages of 3 mg/kg every 24 h for 5 days. By 48 h after i.v. bolus administration, ID-6105 concentrations in various tissues had decreased to very low levels. The majority of ID-6105 appears to be excreted in the bile.

The Effect of Sea Water Containing Heavy Oil on RO Membrane (유탁해수의 RO막에 대한 영향)

  • An experimental study regarding the effect of emulsions on RO is presented. Heavy oil was added to the sea water and the distilled water separately and treated for 30 minutes by a homogenizer to make emulsions. For the case of the sea water without heavy oil the permeate decreased from the beginning of the experiment. Chloride and conductivity increased with time, due to the fouling occurring as the suspended solids in the sea water accumulated on the membrane surface. Rejection rate of salt was 99.6~99.7%. As for the sea water containing heavy oil, the permeate decreased slowly from the beginning of the experiment. This result was the same for the case of the sea water only. However. chloride and conductivity increased significantly when heavy oil was added. In the second experiment with sea water containing heavy oil, the operation time of RO was reduced considerably. With addition of oil, the chloride increased greatly, while the permeate reduced comparatively. In the experiment where emulsion of $0.3{\sim}0.8mg/{\ell}$ was supplied to RO. oil concentration was about 10ppb in the permeate at the end of the experiment. In case of the distilled water containing heavy oil. the conductivity increased. However. the permeate reduced to 30% compared to the case of the sea water containing heavy oil. The case of sea water containing heavy oil showed an opposite result, but the effect of the addition of oil on RO was significant. Oil caused fouling of the RO and the contamination of the whole system, and as the result the system could not be operated properly. As a result the membrane capacity, the amount and water quality of permeate deteriorated significantly.

Dispersion Characteristics of Hydrogen Gas by the Effect of Leakage Hole Size in Enclosure Space (누출공 크기에 따른 밀폐공간 내 수소 가스의 확산 특성)

  • As a potential clean energy resource, the production and consumption of hydrogen gas are expected to gradually increase, so that hydrogen related studies are also increasing. The thermal and chemical properties of hydrogen result in its high flammability; in particular, there is a high risk if leaks occur within an enclosed space. In this study, we applied the computational fluid dynamics method to conduct a numerical study on the leakage behavior of hydrogen gas and compared these numerical study results with an experimental study. The leakage hole diameter was selected as an important parameter and the hydrogen gas dispersion behavior in an enclosed space was investigated through various analytical methods. Moreover, the flammable regions were investigated as a function of the leakage time and leakage hole size. We found that the growth rate of the flammable region increases rapidly with increasing leakage hole size. We also investigated the relation between the mass flow rate and the critical time when the hydrogen gas reaches the ceiling. The analysis of the monitoring points showed that the hydrogen gas dispersion behavior is isotropic and independent of the geometry. We found that the concentration of gas in an enclosed space is affected by both the leakage flow rate and amount of gas accumulated in the enclosure.

Physiological and Biochemical Responses of Sedum kamtschaticum and Hosta longipes to Ozone Stress (기린초와 비비추의 오존에 대한 생리·생화학적 반응)

  • In this study, the resistance to ozone and characteristics of ozone-induced damage were investigated on the perennial ground cover plant species. Sedum kamtschaticum and Hosta longipes were exposed to $200{\mu}g{\cdot}kg^{-1}$ ozone for 8 hours per day (from 08:00 to 16:00) in the naturally irradiated phytotron. The extent of ozone-induced damage was measured through the analysis of physiological parameters, such as water use efficiency (WUE), chlorophyll content (Chl. a, Chl. b, Chl. a + b, and Chl. a/b ratio), carotenoid contents, and the induction of reactive oxygen species (ROS). Ozone exposure significantly reduced the daytime WUE in both species. The contents of chlorophyll and carotenoid were also decreased and ROS, such as hydrogen peroxide ($H_2O_2$) and superoxide radical ($O_2{^-}$) were accumulated after ozone exposure. The above results of this study suggested that S. kamtschaticum is more resistant to atmospheric ozone than Hosta longipes. Considering its fast responses to ozone, it was also assumed that Hosta longipes can be used as an indicator plant of an increase in atmospheric ozone concentration.

The study on bioaccumulation of heavy metals in the cultured Pacific oyster, Crassostrea gigas, along the coast of Tongyeong, Korea (통영연안 해역의 양식 참굴 (Crassostrea gigas) 의 중금속 농축에 관한 연구)

  • In order to investigate contamination of heavy metal in seawater and cultured oyster, samples were collected November 2003 to July 2004 from 12 sites (13 sites for seawater) along the coast of Tongyeong, Korea. The mean concentrations of metal in oyster tissues were as follows: 0.09 (0.01-0.3) ${\mu}g/l$ for Cd, 0.47 (0.01-1.4) ${\mu}g/l$ for Cr, 0.59 (0.2-2.3) ${\mu}g/l$ for Ni, 1.02 (0.1-4.2) ${\mu}g/l$ for Pb and 0.48 (0.01-3.9) ${\mu}g/l$ for Hg in the seawater, whereas 2.45 (0-5.47) mg/kgDW for Cd, 3.63 (0.10-12.91) mg/kgDW for Cr, 3.2 (0.01-15.73) mg/kgDW for Ni, 3.51 (0.01-6.47) mg/kgDW for Pb and 0.39 (0.004-0.74) mg/kgDW for Hg, respectively. Most metal concentration values were below the permissible range for the related regulations. Mean bioconcentration factors (BCF) for each metal were as follows: 38,964 (1,771-207, 171) for Cd, 9,583 (1,231-80, 162) for Cr, 191 (3-20, 980) for Ni, 1,416 (245-5, 207) for Pb and 180 (5-716) for Hg, respectively. The BCF values from this study corresponded to the transitional phase from the pristine to the contaminated waters. Notably, Cd showed the highest BCF, which suggest that the Pacific oyster could be utilized as a useful biomarker for Cd contamination in sea water. The multidimensional scaling analysis suggested that the metal contaminants are mainly originated from combustion of fossil fuel and accumulated to oyster through food web.

Effect of Thermophilic Ammonium Tolerant Bacteria on Malodors Emission of Composting of Pig Manure (돈분 퇴비화 과정중 악취물질에 대한 고온성 암모니움 내성균 접종 효과)

  • In order to investigate microbiological control of malodors, particularly including ammonia, the effect of three thermophilic ammonium tolerant bacteria strains. TAT112. TAT117 and TAT119, were tested during composting of pig manure in the laboratory scale composters. The total weight, volatile solids and BOD of the pig manure compost were decreased during composting process in all treatments. The temperature in all treatments rose in first 3 days dramatically, but that in control without inoculation reached its maximum most lately among the treatments. The nitrogen content of drain water accumulated inside and outside composter, and trapped in 6N $H_2SO_4$ was lower in TAT112 inoculated composter than in control. However, it was not lower in the treatment of TAT117 and TAT119 inoculated. Ammonia concentration in the exhaust gas monitored everyday during composting also demonstrated that it was lowest at TAT112 inoculated among all treatments. It was appeared to have an effect on reducing ammonia emission at the treatment of TAT112 inoculated than the control.

Effects of Nitrogen Fertilization on Leaf Yield and Pyranocurmarine Accumulation in Angelica gigas Nakai

  • Angelica gigas Nakai is one of the most widely used herbal medicines and is known to have many pharmaceutical effects including an anti-oxidant, anti-cancer etc. This study was carried out to investigate an effect of fertilization on leaf yield, production of dry-matter and accumulation of pyranocurmarine compounds such as decursin (DE) and decursinol angelate (DA) in Angelica gigas Nakai. Effect of fertilization was determined from response surface regression equation composing of 2 by 3 factorial arrangement of urea, sodium dihydrogen phosphate and potassium chloride. Yield of leaf in Angelica gigas Nakai significantly increased until 100 days after transplanting. Production of leaf also tended to increase with increasing nitrogen fertilization. Model of regression equation showed that leaf production depended upon nitrogen ($Pr>{\mid}t{\mid}$ : 0.087, 0.256 and 0.079). Also, statistical results between nitrogen application level and production of dry-matter showed significant relationship (p<0.05) and contents of dry-matter was highest in 10 kg 10a-1 treatment on 24 Sep. Active compound isolated and purified from leaf and root of Angelica gigas Nakai was identified as DE and DA by gas chromatograph-mass spectrophotometry (GC-MS). Concentration of DA as prevalent compound in leaf was highest on 20 Aug. but decreased on 24 Sep. Amount of DE and DA accumulated in Angelica gigas Nakai significantly increased with growth stages and nitrogen level. The result of our investigation imply that nitrogen fertilization is important factor for production of leaf and accumulation of pyranocurmarine in Angelica gigas Nakai as a medicinal/food materials.
