Evaluation of Mulberry Germplasm (Morns spp.) for Leaf Yield and Quality through Bioassay

  • Twenty - four elite mulberry germplasm each of indigenous and exotics were studied for their leaf yield and compared with commercial check ($V_1$ and Kosen). Accession MI-0416 and ME-0169 out yielded the check accession in leaf yield/plant. The other few mulberry germplasm were also performed at par with the checks. For quality test and bioassay were conducted with the leaves of selected mulberry germplasm. Among the selected twelve mulberry accessions used for bioassay, MI-0376 and ME-033 performed better than check ($V_1$, Kosen). Other mulberry accessions i.e., MI-0310 and MI-0437 are on par with the check as far as the bioassay is concerned. MI-0376 and ME-0033 out yielded in rearing parameters and qualified for 11 and 10 rearing and related traits. Other mulberry accessions i.e., MI-0310 and MI-0437 were also qualified for eight rearing traits along with check ($V_1$). The mulberry accessions tested after selection from the preliminary characterization seems to be better and equally good in rearing and leaf yield compared to check ($V_1$, Kosen), which provides scope for selection and further evaluation. The selected mulberry accession may be included in crop improvement programme.

  • With agreement from the US, EU, and other countries, China joined in WTO(World Trade Organization) on December 1, 2001. So it is expected that Korea will not expand bilateral trade but also face severe competition with China for world and Chinese market. After Chinese accession to WTO, the aspect of Korean companies may be considered to be reduced the aspect of opportunity and risk by half. Accordingly, Korean domestic companies have to search countermeasures optimized to get global competition by considering the aspect of opportunity and risk. Therefore, the purpose of this study is to review the potential possibility of China as a world market and to provide countermeasures for major Korean domestic industries.

Occurrence of Chrysanthemum chlorotic mottle viroid in Chrysanthemum(Dendranthema grandiflorum) in Korea

  • Chrysanthemum chlorotic mottle viroid(CChMVd) isolates have been identified from chrysanthemum showing yellow spots or infected without symptom. They were 399-400 nucleotides length of RNA. CChMVd-SSHA6(GenBank accession no. DQ450682) revealed a GAAA to DUUC substitution in positions 82-85 of CChMVd-MSIN34(GenBank accession no. DQ402041). In vitro RNA transcripts with the complete CChMVd sequence were infectious and induced the typical CChMVd infection symptom of yellow spots in chrysanthemum cv. Sharotte. CChMVd caused reduction in growth in some cultivars, whereas some cultivars were not affected. This is the first report on the occurrence of CChMVd in chrysanthemum in Korea.

Monitering of Ostreid herpesvirus in Pacific oyster, Crassostrea gigas (굴의 Ostreid Herpesvirus 모니터링)

  • Monthly Screening of Pacific oyster, Crassostrea gigas, was surveyed by PCR for Ostreid herpesvirus(OsHV-1) infections from 2012 to 2013 in Korea. It was showed positive results in one hatchery in April and May 2012 and in oyster farming areas from August to November in 2013. The obtained sequences showed 99.3~99.5% homology with OsHV-1 ${\mu}var$(Accession Number: HQ842610) and 96.2~96.3% homology with OsHV isolated from Korea(Accession Number: AY509253).

Cloning and Characterization of hydroxypyruvate isomerase (EC 5.3.1.22) gene in silkworm Bombyx mori

  • The sequence of hydroxypyruvate isomerase gene was obtained in NCBI. In this study, the hydroxypyruvate isomerase gene of Bombyx.mori was identified and annotated with bioinformatics tools. The result was confirmed by RT-PCR, prokaryotic expression, mass spectrographic analysis and sub-cellular localization. The hydroxypyruvate isomerase cDNA comtains a 783bp ORF, and has 4 exons. The deduced protein has 260 amino acid residues with the predicted molecular weight of 29169.30 Da, isoelectric point of 6.10, and contains conserved PRK09997 and Hfi domains. The hydroxypyruvate isomerases of Nasonia vitripennis and Bombyx mori have a high homology. Through RTPCR analysis, we found that this transcript was present in testis, ovary, blood-lymph, fat body, midgut, silk gland and tuba Malpighii. This protein was located in cytoplasm through immunohistochemistry. We submitted the cloned gene under the accession number EU344910. The enzyme has been classified under accession number EC 5.3.1.22.

Incidence of Coleus blumei viroid 1 in Seeds of Commercial Coleus in Korea

  • A viroid was detected from symptomless Coleus blumei cultivar 'Kong Scarlet'. It consisted of 249 nucleotides(GenBank accession no. EU410620), which was 100% identical to a Coleus blumei viroid 1(CbVd 1) reported from China(GenBank accession no. DQI78399), indicating that the viroid was an isolate of CbVd1. Attempts were made to determine if commercial Coleus seeds were infected with CbVd. Infection rates in seedlings of the 14 commercial cultivars of Coleus ranged from 0 to 100%. CbVd1 caused discoloration and growth retardation in some cultivars, but is symptomless in others. These results indicated that Coleus in commercial markets in Korea is highly infected with CbVd.

  • Strategic aim of Poland is an accession to The European Union. This accession means consolidation of democratic transformations and acceleration of economical development. Possibly full and quick execution of preparing works at building domain is one of conditions of Polish membership in the European Union. Poland has to adopt rules and standards, which are required at EU. It will permit to make polish companies more attactive at international market. We should know, that economical, legal whether political reforms, this is not all what consists on membership at the Union European. We must change internally, become open to the world, to represent dynamism, enterprise, knowledge and culture. We must become more tolerant and be able to communicate with people with different culture whether political orientation, we have to accept this dissimilarity and will find way of dialogue and cooperation.

Genetic Diversity among Tea (Camellia sinensis) Accessions Based on Random Amplified Polymorphic DNA (RAPD) Patterns

  • Genetic diversity of 45 tea accessions from Korea, Japan, China and Taiwan was investigated by using RAPD analysis. Out of the eighty primers screened, twenty primers generated 99 polymorphic bands with a polymorphic rate 87.0%. The size of the amplified fragments ranged from about 3,138 bp to 520 bp. By cluster analysis, all of the 45 accessions can be grouped into five groups. Over 90% of the 32 Korean accessions belonged to group II, III, IV and V. Moreover, newly developed Korean cultivars (accession no. 13, 14 and 15) belonged to very different group compared with any other Korean accessions. Among the Korean accessions, the minimum genetic similarity 0.500 was obtained between accession no. 17 and 37 and the largest genetic similarity 0.912 between no. 20 and 21.

Growth Characteristics, Main Constituents and Antioxidant Activities in Local Accessions of Sophora flavescens AIT. (고삼 수집종의 생육특성, 주요성분 및 항산화 활성)

  • Background: Sophora flavescens AIT root is used as a herbal medicine in Asian culture. This study was aimed at selecting the best cultivars, by comparing growth characteristics, active ingredients, and antioxidant activities. Methods and Results: A total twenty nine accessions of S. flavescens were collected from five different regions in Korea. Plant height, leaf length, peduncle length, fresh root weight, and seed weight were the growth characteristics selected. Antioxidant activity was determined by measuring the total polyphenol (TPC) and total flavonoid contents (TFC) as well as the radical scavenging activity of DPPH, ABTS and FRAP. Oxymatrine and matrine, active ingredients in S. flavescens were analyzed by HPLC. Results indicated that accession YS-11 had the highest plant height (152.5 cm) and peduncle length (54.0 cm). The fresh root weight was highest in JA-01 at 4.9 kg, while the highest total seed weight was recorded in accession BH-04 at 77.7 g. The YS-03 accession contained the highest oxymatrine and matrine total contents (0.9616% and 0.9638%, respectively). Accessions BH-02, YS-11, YS-05, and MJ-03 had the highest levels of antioxidant activity (p < 0.05). Oxymatrine showed a high correlation with TPC and FRAP. TPC showed high correlations with TFC, DPPH, ABTS and FRAP. TFC showed a high correlation with DPPH. Conclusions: The superior lines can be selected for use as standard variety breeding material by comparing the growth characteristics of the accessions.
