The Glycosides of Araliaceaus Drugs and their Biological Activities

  • Among the Araliaceae plants indigenous to Korea, those whose medicinal usage are comparatively high have been selected in this serial studies. Chiisanoside and acanthoside D were isolated and identified from the Acanthopanax chinensis leaves and root bark. Chiisanoside and acanthoside D have been found to have the lowering S-GPT, S-GOT value and BSP-retention rate and survival rate, anti-histaminic effect in the toxic state through the bio-pharmacological experiments. ${\alpha}-hederin$, hederagenin pentaglycoside were isolated both stem bark of Kalopanax pictum Nakai var. magnificum and Kalopanax pictum Nakai var. Max. respectively. Syringoside, acanthoside D were also isolated from the root bark of Acanthopanax koreanum. The biological activity of ginsenoside $Rb_1$, $Rg_1$, Re were examined. Ginsenoside $Rb_1$, $Rg_1$, Re promotes the antileaking effect in X-ray (Co 60) irradiated toxic state.

  • Two glycosides, assumed to be one of the potential active principles, is isolate from the root bark of Acanthopanax koreanu Nakai, identified to be acanthoside D, $C_{34}H_{46}O_{18}$, mp $242^{\circ}C$, and syringo side, $C_{17}H_{24}O_{9}$ mp $192^{\circ}C$. Acanthoside D has been found to have s-GPT, s-GOT lowering effect, BSP-retention rate and survival rate in the toxic state through the bio-pharmacological experiments.

Inhibition of Lung Inflammation by Acanthopanax divaricatus var. Albeofructus and Its Constituents

  • In order to find potential therapeutic agents on lung inflammatory conditions, the extracts of Acanthopanax divaricatus var. albeofructus were prepared and its constituents were isolated. They include lignans such as (+)-syringaresinol (1), acanthoside B (2), salvadoraside (3) and acanthoside D (4), lariciresinol-9-O-${\beta}$-D-glucopyranoside (5) and phenylpropanoids such as 4-[(1E)-3-methoxy-1-propenyl]phenol (6), coniferin (7), and methyl caffeate (8). The extracts and several constituents such as compound 1, 6 and 8 inhibited the production of inflammatory markers, IL-6 and nitric oxide, from IL-$1{\beta}$-treated lung epithelial cells and lipopolysaccharide (LPS)-treated alveolar macrophages. Furthermore, the extracts and compound 4 significantly inhibited lung inflammation in lipolysaccharide-treated acute lung injury in mice by oral administration. Thus it is suggested that A. divaricatus var. albeofructus and its several constituents may be effective against lung inflammation.

  • Five constituents were isolated from the stem of Acanthopanax senticosus. Their structures were elucidated as (-)-sesamin (1), iso-fraxidin (2), 5-hydroxymethylfurfural (3), syringin (4) and acanthoside D (5) by spectral analysis. Among these compounds, 5-hydroxymethylfurfural (3) was isolated for the first time from this plant.

생물전환을 통한 음나무발효물의 지표성분 설정 및 동시분석법 검증 (Validation of Simultaneous Analysis Method of Standard Compounds in Fermented Kalopanax pictus Nakai by Bioconversion)

  • The aim of this study was to select compounds for the standardization of fermented Kalopanax pictus Nakai (KP-F), to develop the analysis method using HPLC-PDA and to perform method validation. KP-F is a fermented powder developed to improve the original physiological activities and create a new functionality. Eleutheroside E, Acanthoside B, and Syringaresinol were selected as the standard compounds and developed our own method for simultaneous analysis. The analyte was isolated using C18 column with a gradient elution of 0.05 M phosphoric acid in water and methanol as the mobile phase at a flow rate of 1 mL/min and detected at 210 nm. As a result, all standard compounds showed good linearity with an $R^2$ (coefficient of correlation) of 1.000 and for the limit of detection range of $0.710{\sim}0.831{\mu}g/mL$, and the limit of quantification as $2.150{\sim}2.520{\mu}g/mL$. The precision was RSD (%) of less than 4.80%, while the accuracy was 4.70%>RSD (%) for the range 102.44~110.48%. In conclusion, the developed analysis method is suitable for the detection of Eleutheroside E, Acanthoside B, and Syringaresinol in KP-F.

지리오갈피를 첨가한 전통 발효주의 개발 (The Development of Traditional Korean Fermented Liquor Made from Acanthopanax chilsanensis)

  • 지리오갈피의 소비를 촉진시키고 지역 특산품을 개발할 목적으로 지리오갈피 발효주의 제조하여 그 특성 등을 검토하였다. 지리오갈피의 뿌리, 줄기 및 열매를 원료미의 10%(W/W)씩 첨가하여 $25^{\circ}C$, 13일 동안 발효시킨 결과 각각 알코올 함량이 $14.7{\pm}0.7%$, $15.3{\pm}0.9%$$15.5{\pm}0.7%$의 발효주를 얻었다. 발효주의 pH는 pH 3.9~pH 4.4, 총산은 0.72~0.75%를 나타내어 3시험구간의 차이는 거의 없었다. 총당 함량은 발효 24시간부터 급격히 떨어져 발효 13일에는 7.1~7.5%를 나타내었다. 총 유리아미노산은 지리오갈피의 뿌리와 열매를 첨가한 시험구 보다 줄기를 첨가한 시험구에서 7,045.01 ppm으로 가장 높게 검출되었다. 발효주 중의 acanthoside-D의 함량을 측정한 결과 10%의 뿌리 및 열매로 제조한 발효주에서는 35.42 ppm 및 18.91 ppm이 검출되었으나 줄기로 제조한 발효주에서 57.06 ppm의 높은 함량을 나타내었다.

지리오갈피의 이화학적 분석 (Physicochemical Composition of the Acanthopanax Chilsanensis)

  • 지리오갈피(Acanthopanax chilsanensis)를 이용한 지역특산품을 개발할 목적으로 먼저 지리오갈피 뿌리, 줄기 및 열매 자체에 대한 이화학적 성분분석을 행하였다. 지리오갈피의 일반성분을 분석한 결과 뿌리, 줄기 및 열매의 조지방은 각각 2.09, 2.51 및 7.94%이었으며, 조단백질은 11.50, 7.18 및 10.17%로 나타났다. 그리고 조회분은 각각 11.07, 6.85 및 6.38%로 뿌리 중의 조회분이 줄기와 열매 중의 조회분 보다 약간 높게 나타났다. 환원당은 각각 18.90, 10.70 및 24.05 g/100 g으로 측정되었다. 색도를 측정한 결과 명도(L)는 줄기가, 적색도(a)는 열매가, 그리고 황색도(b)는 뿌리가 높은 것으로 나타났다. 유리당은 뿌리, 줄기, 열매 모두 fructose, glucose 및 sucrose의 순으로 높게 나타났다. Acanthoside-D 함량은 줄기(18.95 mg/100 g), 뿌리(8.10 mg/100 g) 및 열매(2.85 mg/100 g)의 순으로 높게 나타났다. 유리아미노산은 줄기가 955.26 mg/100 g으로 뿌리보다는 4.5배, 열매보다는 8.5배 높게 검출되었다. 향기성분 중에서는 $\alpha$-pinene, $\beta$-pinene, 3-carene 및 D-limonene 등과 같은 천연향기 성분이 동정되었다.

Two Cyanidin compound from the Fruits of Acanthopanax divaricatus var. albeofructus

  • Acanthopanax divaricatus var. albeofructus is one of the indigenous medicinal plant and the fruits of Acanthopanax spp. used as a remedial for "wipe out evil wind". Two anthocyanin were isolated from the fruits of Acanthopanax divaricatus var. albeofructus. Their structures were elucidated as cyanidin 3-lathyroside (1) and cyanidin 3-galactoside (2) by chemicophysical and spectroscopic analysis. And also, four chemical, syringin, chlorogenic acid, caffeic acid and acanthoside D were identified. Both anthocyanide were isolated for the first time from Acanthopanax species. cyanidin 3-lathyroside is one of the rare anthocyanin in natural resources.

  • Acanthopanacis Cortex (Acanthopanax sessiliflorum, Araliaceae, KP VIII), an important Korean medicinal herbal drug, has been widely used as tonic, anti-stress and immuno-enhancing drugs. To monitor the contents of active ingredients (acanthoside D[=eleutheroside E], eleutheroside B, isofraxidin, chlorogenic acid, and caffeic acid) in Acanthopanax sp., we developed the HPLC analysis method and validated. The simultaneous determination of five active ingredients was achieved in a C$\sub$18/ column with an acetonitrile water (containing 1 % phosphoric acid) (15 : 85) mobile phase. (omitted)
