  • The object of this stduy was to investigate the pollution by Pb in rice plants which were cultivated in paddy field soils located near roadside areas and to discover the relationship between lead content in rice plants and soils. Samples of soils and rice plants were collected directly from paddy fields at various distances from highways and expressways located in Kyungpook province. The results obtained were as follows : 1. The average content of Pb in each part of the rice plant was 12.8ppm in roots, 4.8ppm in leaves, 4.3ppm in stems, and 0.4ppm in brown rice. 2. The content of Pb in roots showed highly positive correlation with the Pb content of the soil. 3. The Pb content in stems, leaves, and brown rice showed positive correlation with the Pb content in roots. 4. The content of organically bounded, carbonate, and sulfide Pb in soils showed highly positive correlation with the Pb content in roots. 5. The absorption ratio showed positive correlation with soil pH and negative correlation with the content of Fe in soils.

Protective Effect of Monascus pilosus Mycelial Extract on Hepatic Damage in High-Fat Diet Induced-obese Rats (고지방식이로 유도한 비만 흰쥐에서 Monascus pilosus 균사체 추출물의 간 손상 예방효과)

  • Hepatoprotective effects of Monascus pilosus mycelial ethanol extract (MPME) were examined in high-fat diet induced-obese rats. The rats were randomly divided into 2 groups; normal control (NC) and a high-fat and high cholesterol diet group (HFC). The HFC diet group was fed a 5L79 diet supplemented with 15% lard and 1% cholesterol for 3 weeks for induction of obesity. And then, the rats were divided into 4 groups (n=5); the NC, a HFC diet obesity control group (HF), 0.5% MPME supplemented HFC diet group (MPM), and 2% conjugated linoleic acid (CLA) supplemented HFC diet group for 7 weeks. Whereas the daily weight gain of NC and HFC groups were 3.48 g and 4.48 g, respectively, those of MPM and CLA were 3.09 g and 4.38 g, respectively. Furthermore, activity of serum alanine and aspartic aminotransferase in HF was markedly higher than those of NC group, but, the activity in MPM and CLA was significantly lower than HF. Hepatic reduced glutathione content in MPM and CLA was higher than HF. On the contrary, hepatic lipid peroxide content in MPM and CLA was significantly lower than HF. In conclusion, although the precise mechanisms of the hepatoprotective effects of the MPME in this study are unknown, our study provides experimental evidence that MPME may prevent obesity and hepatic damage by high-fat and high cholesterol diet via inhibition of lipid absorption and induction of reactive oxygen spices scavenging enzyme such as superoxide dismutase.

The Uptake of $Sr^{90}$ by Paddy Rice from Soil and its Distribution in the Plant (답토양(畓土壤)에서 수도(水稻)의 Strontium-90 흡수(吸收)와 수도체내(水稻體內) 분포(分布))

  • Because of the drastic development of nuclear industries, the contamination of natural environments by the disposal of radioactive materials which are released from nuclear facilities have aroused a considerable concern in relation to agricultural practices. Therefore the present investigation, through pot experiment, was performed to find out the aspect of the uptake of $Sr^{90}$ by rice plants and its distribution in them in five different types(physicochemical and minerallogical properties) of paddy soils. The results obtained were as follows; 1) Visual toxic symptoms on the growth of rice plant due to treatment of $Sr^{90}$ up to $40{\mu}Ci/10㎏$ in a pot were not observed even though uptake of $Sr^{90}$ by rice plant was proportionally increased with the $Sr^{90}$ treatment. 2) Distribution of $Sr^{90}$ in the rice plant was the highest in the leaves (84.5%) followed in the order by stems (13.5%) and rough grain (2.0%). The ratio of $Sr^{90}$ to Ca was higher in the leaves (872) and stems (667) than in the rice grain (89). 3) $Sr^{90}$ absorption in the rice plant ranged $0.15{\sim}0.30%$ at harvesting time. Uptake of $Sr^{90}$ by rice plants decreased by the increase of soil pH and exchangeable canons in the soils, but $Sr^{90}$ uptake increased when nitrogen, organic matter and clay content in soil was high, and uptake of this nuclide in the rice plant was higher with low Illite and Vermiculite content in the soils.

Characteristic Properties of Fucoidan Sulfate Purified from Gompi, Ecklonia stolonifera (곰피에서 정제한 Fucoidan Sulfate의 특성)

  • The fucoidan purified from Korean brown seaweed, Ecklonia stolonifera was characterized on molecular structure and blood anticoagulant activities. Extraction was conducted at $100^{\circ}C$ with water and repeated twice. The crude fucodian was 151.1g out of 20.0 kg of Ecklonia stolonifera. The Fucoidan-1, which was purified from crude fucoidan using calcium chloride and cetyl pyridium chloride (CPC), was 35.2% against crude fucoidan. Fucoidan-5 was obtained approximately 28.1% from Fucoidan-1 through DEAE-Toyopearl 650 M ion-exchange column chromatography and showed one band by cellulose acetate electrophoresis. The molecular weight of Fucoidan-5 was estimated to be about 21,000∼23,000 dalton by Sephacryl S-300 gel filtration chromatography. Fucoidan-5 consists of 35.7% of fucose and 4.3% of galactose and the molar ratio of fucose and sulfate was about one to one. IR spectrum of Fucoidan-5 showed absorption at $1240\;cm^{-1}\;and\;850\;cm^{-1}$ and specific rotation value, $[\alpha]$, was $[\alpha]$. These results suggests that the sulfate maybe bind at $C_{4}$ carbon on ${\alpha}-L-fucose$. Gas chromatograph of methyl alditol acetate revealed that Fucoidan-5 is a fucose containing sulfated polysaccharide with $({\alpha}l-2)\;or\;({\alpha}l-2)$ glycosidic linkage. Anti-thrombin activity of the Fucoidan-5 was estimated as 1.4 time stronger than heparin. From above results, the purification methods using CPC and ion exchange chromatography is effective tools for obtaining highly purified fucoidan from Gompi, Ecklonia stolonifera.

Phytochemical compounds and quality characteristics of spray-dried powders with the blanching condition and selected forming agents from pressed extracts of Ligularia fischeri leaves (블랜칭 처리 및 부형제 종류에 따른 곰취 착즙액 분무건조 분말의 phytochemical 성분 및 품질특성)

  • This study was performed to determine the effects of the blanching condition (immersion ratio 1:15 (w/v) for 3 min at $95^{\circ}C$, and solution containing 1% sodium chloride) and selected forming agents (dextrin DE=10, dextrin DE=20, ${\beta}$-cyclodextrin; each forming agents added 5%) on the phytochemical compounds and quality characteristics of Ligularia fischeri leaves. The moisture was not affected by the forming agent. The color of a, b and chroma values were low in the blanching treatment groups and were significantly lowest with ${\beta}$-cyclodextrin (CD). The polyphenol and flavonoid contents in the blanching treatment groups were higher than those in the non-blanching-treatment group. The ascorbic acid content was higher in the non-blanching-treatment group and was significantly highest in the group treated with dextrin (DE=10) whereas the blanching treatment groups showed lower dehydroascorbic acid content than the non-blanching-treatment group. The water absorption was higher in the non-blanching-treatment group and was significantly highest in the group treated with CD. The water solubility in the blanching treatment groups treated with dextrin (DE=20) and CD was higher than that in the blanching treatment group treated with DE=10. The total chlorophyll and chlorophyll a and b contents were high in the blanching treatment group treated with CD, and for the total carotenoid contents, the same tendency as that seen with the chlorophyll content was observed. With regard to the particle diameter, those in the blanching treatment groups were lower than that in the non-blanching-treatment group and was significantly lowest in the blanching treatment groups treated with DE=20 and CD. The result of SEM observation showed that the spray-dried powders in blanching treatment groups treated with the DE=20 and CD forming agents had uniform particle distribution.

  • According to Yoshihisa Kuroki's report that the total amount of carotenoid was more in the susceptible to pine bark beetle, than in the resistant ones, carotenoids were extracted from needle leaves of one year old seedlings of Pinus thunbergii, Pinus desiflora, Pinus rigida, Pinus koraiensis, Pinus rigitaeda and Pinus taeda which are all important pines in Korea, to find their resistant ratio to the insect. The carotenoids were analyzed and compared using the spectra of them by spectrophotometer. The results were as follows: 1. The visible absorption spectra of carotenoids in those pine trees were proved to be very similar. 2. The total amount of carotenoids in needles differed with the tree species and the contents were arranged in decreasing order P. koraiensis>P. rigida>P. thunbergii>P. rigitaeda>P. taeda, it seemed that there was corelation between the cartenoid content and the extent of harm caused by the pine bark beetles except P. koraiensis. 3. But carotenoids were contained in Robinia pseudoacacia, Castanea crenata, Chamaecyparis obtusa and Cedrus deodra leaves too. 4. The total amounts of carotenoids in these pines of 9 species were arranged in decreasing order Robinia pseudoacacia>Pinus koraiensis>Pinus rigida>Pinus thunbergii>Castanea crenata>Pinus rigitaeda>Pinus taeda>Chamaecyparis obtusa>Cedrus deodara. Therefore, it was proved that there was no correlation between carotenoid cotent and extent of resistance to the insect. 5. In the thin-layer chromatography of these carotenoids, 13 kinds of components in P. densiftora, P. koraiensis and P. rigida and 12 kinds of spots in other pines, were detected respectively, under ultra-violet fluorescent lamp $3,600{\AA}$ and $2,537{\AA}$. 6. The eighth spots from the bottom in P. densiflora, P. koraiensis and P. rigida were not found in other pines and other 4 species (Robinia pseudoacacia, Castanea crenata, Chamacyparis obtusa, Cedrus deodra). Especially the spot in P. densiflora fluoresced strong cobalt blue-fluorescence under ultra-violet fluorescent lamp $2,537{\AA}$.

Comparison of Feed Efficiency Between Rotifers Enriched Lipid-contents to Enrichment and Enhanced Digestive Enzymes Activity to Starch (영양강화 Rotifer와 효소활성 향상 Rotifer의 먹이효율 비교)

  • In this study, we carried out an experiment for estimation the larval digestibility in aspects which digestive enzymatic activities and nutrition of the rotifers, Brachionus rotundiformis. Thus we enhanced the digestive enzymatic activity through the addition of starch for the increase of digestibility of rotifer (starch-rotifer), and compared with the feed efficiency through rearing of the olive flounder, Paralichthys olivaceus used rotifer lipid-enriched with Algamac $2000^{(R)}$ (CE-rotifer). The digestive enzyme activities (except for TG-lipase), total protein contents, total essential amino acid, essential amino acids (methionin and phenylalanine) of starch-rotifer (the rotifer used a starch as additive, and enriched not) was assayed significantly higher than CE-rotifer (P<0.05). And total lipid, lipid classes (except for sterol) and fatty acids as DHA and EPA showed higher in CE-rotifer than starch-rotifer (P<0.05). But, sterol contents and ST/TG ratio were shown significantly higher in starch-rotifer (P<0.05). The flounder larvae supplied the two rotifers showed standard length and body weight that not significantly differed with ranges $3.72{\sim}3.79\;mm$ and $32.9{\sim}37.8\;mg$/larva on 6 days after hatching (DAH), respectively (P>0.05). However, these of 12 DAH showed the values of significantly higher to $5.94{\pm}0.249\;mm$, $144.0{\pm}23.86\;mg$/larva and $26.2{\pm}12.13%$ in standard length, body weight and survival in CE-flounder than that of starch-flounder (P<0.05). The hydrolytic enzymatic activities of flounder larvae severally supplied the two rotifers showed the significantly higher activities in acidic -amylase, neutral -amylase, TG-lipase, lysozyme and acidic phosphatase in starch-flounder on 5 DAH (P<0.05). But neutral $\alpha$-amylase, three proteases and two phosphatases of CE-flounder on 11 DAH showed the significantly higher activities than that of starch-flounder (P<0.05). Therefore, for the flounder, Paralichthys olivaceus larvae just depleted yolk was more beneficial to supply the feed, rotifer, enhanced the digestibility than to supply the feed lipid-enriched for aspect of larval digestibility up to 6 DAH, thereafter nutrition of absorption due to the development of digestive organs suggested that enrichment effect appeared with larval somatic growth. Consequently, investigation more detailed about the larval digestive physiological and nutritional requirement variations after 6 DAH will be necessary, thereafter.

Effect of Calcium and Boron Intakes on Calcium Balance Status in Ovariectomized Rats (난소절제 흰쥐에 있어 칼슘과 보론 섭취수준이 칼슘 평형상태에 미치는 영향)

  • This study was conducted to investigate the effect of calcium and boron intakes on calcium utilization in ovariectomized (OVX) rats. Rats were divided into 9 groups and fed diets containing various levels of calcium $(0.1\%,\;0.5\%,\;1.5\%)$ and boron (0.5 ppm, 50 ppm, 100 ppm) for 4 weeks. The half of rats in each group were ovariectomized and the others were sham-operated. Rats were fed same diets for 8 weeks after operation. Feed intake and weight gain were significantly increased as the dietary calcium was increased and those of OVX group were higher than in sham-operated group. Feed efficiency ratio was significantly higher in OVX group than that in sham-operated one. With boron supplementation, serum calcium level was significantly increased in low-calcium group, but decreased in adequate/high-calcium group. In calcium balance, calcium intake was significantly increased with increasing levels of calcium and boron and higher in OVX group than that in sham-operated one. With increasing calcium intake, fecal and urinary calcium excretions were significantly increased. Urinary calcium excretion was significantly decreased with increment of boron intake. Apparent calcium absorption of adequate-calcium OVX group was the highest among the groups. Daily calcium retention was significantly increased as the dietary calcium was increased and that of high-calcium OVX group was higher than high-calcium sham-operated group. According to these results, the boron supplementation increased the calcium intake and decreased the urinary calcium excretion. Therefore, it could be suggested that the boron supplementation may be complementary to calcium nutrition and useful for bone health.

Quality Evaluation of Juanbyeo as Aseptic-packaged Cooked Rice (무균포장밥 가공 적합품종 주안벼의 식미특성)

  • The objective of this study was to evaluate several varieties of aseptic-packaged cooked rice including Juanbyeo. Quality evaluations, such as by amylose, setback, toyo values and amylograms, were significantly higher in the Juanbyeo as compared to other samples. In addition, the Juanbyeo aseptic-packeged cooked rice showed significantly higher palatability characteristics and had a greater water absorption ratio than the other samples. For cold cooked rice, the relationship between palatability and texture was significantly high (p<0.01). Ilpumbyeo showed high scores for taste as a hot cooked rice, but as a cold cooked rice taste scores were lower than other samples. In the case of Juanbyeo cold cooked rice flavor was improved. When the Juanbyeo aseptic-packaged cooked rice was reheated in a microwave oven, it had better palatability than other varieties. Thus, the results of this study suggested that Juanbyeo can be effectively utilized as aseptic-packaged cooked rice.

THERMAL ANALYSIS OF THE DUAL CURED RESIN CEMENTS ACCORDING TO CURING CONDITION (중합조건에 따른 dual cured resin cement의 열분석적 연구)

  • The purposes of this investigation were to observe the reaction kinetics of five commercial dual cured resin cements (Bistite, Dual, Scotchbond, Duolink and Duo) when cured under varying thicknesses of porcelain inlays by chemical or light activation and to evaluate the effect of the porcelain disc on the rate of polymerization of dual cured resin cement during light exposure by using thermal analysis. Thermogravimetric analysis(TGA) was used to evaluate the weight change as a function of temperature during a thermal program from $25{\sim}800^{\circ}C$ at rate of $10^{\circ}C$/min and to measure inorganic filler weight %. Differential scanning calorimetry(DSC) was used to evaluate the heat of cure(${\Delta}H$), maximum rate of heat output and peak heat flow time in dual cured resin cement systems when the polymerization reaction occured by chemical cure only or by light exposure through 0mm, 1mm, 2mm and 4mm thickness of porcelain discs. In 4mm thickness of porcelain disc, the exposure time was varied from 40s to 60s to investigate the effect of the exposure time on polymerization reaction. To investigate the effect on the setting of dual cured resin cements of absorption of polymerizing light by porcelain materials used as inlays and onlays, the change of the intensity of the light attenuated by 1mm, 2mm and 4mm thickness of porcelain discs was measured using curing radiometer. The results were as follows 1. The heat of cure of resin cements was 34~60J/gm and significant differences were observed between brands (P<0.001). Inverse relationship was present between the heat of reaction and filler weight % the heat of cure decreased with increasing filler content (R=-0.967). The heat of reaction by light cure was greater than by chemical cure in Bistite, Scotchbond and Duolink(P<0.05), but there was no statistically significant difference in Dual and Duo(P>0.05). 2. The polymerization rate of chemical cure and light cure of five commercially available dual cured resin cements was found to vary greatly with brand. Setting time based on peak heat flow time was shortest in Duo during chemical cure, and shortest in Dual during light cure. Cure speed by light exposure was 5~20 times faster than by chemical cure in dual cured resin cements. The dual cured resin cements differed markedly in the ratio of light and chemical activated catalysts. 3. The peak heat flow time increased by 1.51, 1.87, and 3.24 times as light cure was done through 1mm, 2mm and 4mm thick porcelain discs. Exposure times recommended by the manufacturers were insufficient to compensate for the attenuation of light by the 4mm thick porcelain disc. 4. A strong inverse relationship was observed between peak heat flow and peak time in chemical cure(R=0.951), and a strong positive correlations hip was observed between peak heat flow and the heat of cure in light cure(R=0.928). There was no correlationship present between filler weight % or heat of cure and peak time. 5. The thermal decomposition of resin cements occured primarily between $300^{\circ}C$ and $480^{\circ}C$ with maximum decomposition rates at $335^{\circ}C$ and $440^{\circ}C$.
