  • We investigated the toxicity of zinc and aluminum hot-dip galvanized sheet steel to abalone Haliotis discus hannai via changes in the gill and hepatopancreas using histological and transmission electron microscopy analysis. Experimental groups were composed of one control and four exposure conditions (direct or indirect exposure to zinc and aluminum hot-dip galvanized sheet steel). In the control group, aluminum exposure groups (direct and indirect), and indirect zinc exposure group, abalone mortality was not observed until the end of the experiment, and no histopathological changes were observed in the gill and hepatopancreas. However, the direct zinc exposure group exhibited 100% mortality. Ultrastructural analysis of the cytoplasm of ciliated and microvilli-bearing epithelial cells from gill filaments revealed electron-dense vesicles near the cell membrane and disruption of the nuclear membrane. We also observed swollen mitochondria and a loss of mitochondrial cristae. The hepatopancreas showed similar changes, and we detected highly electron-dense particles within the vesicles. These results suggest that abalone exposed directly to zinc hot-dip galvanized sheet steel experience acute toxicity, causing damage to cell organelles in the gill and hepatopancreas and, finally, inducing mortality.

  • The increasing of atmospheric $CO_2$ are changing the pH (ocean acidification) and temperature of the sea. Although the effects of ocean acidification on calcifying organisms have well-documented, only a few studies have examined the combined effects of ocean acidification and elevated temperature. This study investigated the effects of ocean acidification and elevated temperature for 2100 on survival and growth of juvenile abalone, Haliotis discus hannai. Ocean acidification was simulated by bubbling $CO_2$ into seawater at concentrations of 1,000 and 1,500 ppm, and temperature was set at room temperature $+2^{\circ}C$. Neither $CO_2$ nor temperature had a significant effect on survival of abalone, while both significantly affected growth. There was no significant interaction between the two factors. Shell length can be used as a growth index of abalone to access the impacts of ocean acidification and elevated temperature.

  • A feeding trial was conducted to investigate the growth and body composition of juvenile sea cucumber Apostichopus japonicus in integrated culture with abalone Haliotis discus hannai or rockfish Sebastes schlegeli. Triplicate groups of sea cucumber averaging $1.2{\pm}0.05g$ were cultured alone or with abalone or rockfish for 12 weeks. Survival of sea cucumber was not affected by co-culturing (P>0.05). Weight gain of sea cucumber cultured with rockfish was significantly higher than that of sea cucumber cultured alone (P<0.05), and did not differ from that of those cultured with abalone (P>0.05). These findings indicate that co-culturing sea cucumber with rockfish effectively improves the growth of sea cucumber.

  • Cochtodinium votykrikoides -related red tide is the most notorious tidal bloom, resulting in mass mortality to marineanimals. This study aimed to test the effect of C. polyknkoides on the lethality to Haliotis discus hannai under con-trolled conditions. The oxygen demand of C. polykrikoides increases to reach its peak duhng the night, while the oxy-gen usage by H. discus hannai was continuously decreased with a threshold of 2 mg L U. The addition of C.polykrikoides did not effect the respiration of the H. discus hannai. However, the usage of oxygen by C. polykrikoidesduhng the night may lead to anoxia in the animal. With aeration, the level of dissolved oxygen (D.O.) was between6.06 and 7.28 mg LU; 90% of abalones survived even with a high concentration of C. potykrikoides (9000 cells mL U).Without aeration (3 mg LU of D.O.), however, the H. discus hannai suffocated immediately. Once 20 hours hadelapsed, all of the abalones were dead. The density of the H. discus hannai population contributed to their mortality.Therefore, aeration during the night and maintaining lower abalone densities is the best way to promote the sur-vivorship of H. discus hannai during a C. polykrikoides red tide.

Initial Culture Conditions for Primary Cell Populations Derived from Radula Tissue in Abalone Haliotis discus hannai

  • Abalone immortal cell lines can be used to study the physiological properties and disease mechanisms of abalone at the cellular and molecular level. As a first step for the final goal to establish abalone immortal cell lines, we examined various initial culture conditions for primary cell populations derived from Haliotis discus hannai radula tissue. The survival rate after cell isolation procedures using the enzymatic method was as low as $9.95{\pm}2.37%$. Based on three different experimental conditions for H. discus hannai radula-derived cell culture, we found that the salinity of the media and the presence of growth-promoting factors were important to support radula-derived primary cell populations during the initial culture. The growth factor-containing media adjusted to 35 psu salinity could induce 100% (8 out of 8 trials) initial cell attachment, and the rate of cell attachment reached 50-70%. The data obtained from this study will provide useful information for developing immortal cell lines from abalone species.

  • Genetic parameters for growth-related traits were estimated in 9-month old of two Korean abalone subspecies, Haliotis discus hannai and H. discus discus, using multiple traits of animal model. The data were collected from the records of 3,504 individuals produced from 16 sires and 17 dams in H. discus hannai and 821 individuals produced from 3 sires and 4 dams in H. discus discus, which was evaluated at the Bukjeju branch, NFRDI, from May 20, 2004 to February 14, 2005. The heritability estimates obtained from restricted maximum likelihood (REML) method range from 0.29 to 0.31 for three growth traits (shell length, shell width and body weight) in H. discus hannai and from 0.22 to 0.28 in H. discus discus, respectively. The heritabilities for shell shape and condition factor were lower than others of growth traits such as ranging from 0.03 to 0.24 in H. discus hannai and from 0.06 to 0.11 in H. discus discus, respectively. Genetic and phenotypic correlations were >0.91 between shell parameters and weight in two abalone subspecies, respectively, indicating that breeding for weight gains could be successfully achieved by selecting for shell length.

참전복, Haliotis discus hannai 유생발생 및 채묘에 미치는 아연독성 (Effects of Zinc Toxicity on Larval Development and Seed Collection of Abalone, haliotis discus hannai)

  • in the present study, the zinc toxicity to larval development and seed attachment of the abalone, Haliotis discus hannai was obtained under continuous flow through system. The zinc concentration melted from zinc coating pipe for 7 months ranged from $89.00\pm2.55 \mu\textrm{g}/\ell to 15.23\pm2.58\mu\textrm{g}/\ell(Y=0.85M^2-19.71+109.96)$. Treatments were carried out with zinc concentration $0~160 \mu\textrm{g}/\ell$. The maximum and minimum of fertilization rate were $87.7\pm5.3%$ in control, $83.7\pm7.6%$ in zinc concentration $160\mu\textrm{g}/\ell$, respectively. The maximum and minimum of hatching rate were $87.5\pm4.5%$ in zinc concentration $10\mu\textrm{g}/\ell$, $79.3\pm5.6%$ in zinc concentration $160\mu\textrm{g}/\ell$, respectively. Both of the results were not significantly different (P>0.05). But the normality rate, setting rate and survival rate of abalone larvae at over zinc concentration TEX>$20\mu\textrm{g}/\ell$ decreased rapidly and showed significantly different from those of the other group(P<0.05).

유동세포분석에 의한 참전복(Haliotis discus hannai) 세포내 DNA 함량 분석 최적화 (A flowcytometric determination of DNA content in Pacific abalone, Haliotis discus hannai cell)

  • DNA 함량 조사는 진화의 유전적 기작을 예측하여 종분화를 파악하게 한다. 본 연구의 목적은 참전복(Haliotis discus hannai)의 DNA 함량을 측정하고 유세포분석기(flowcytometry)로 고정(fixation) 없이 DNA 함량 측정에 최적인 조직들을 파악하는 것이다. DNA 함량 (pg nucleus-1)에 있어, 원생생물에 오염된 아가미조직(2.5±0.08)은 3.2±0.02 pg nucleus-1의 DNA 함량을 보인 근육조직과 외투막조직에 비하여 유의하게 낮았고(p<0.05) standard reference보다도 낮은 반면, 근육조직과 외투막조직은 standard reference보다 높았다. 본 연구 결과들을 고려 시, 참전복에서 고정 없이 유동세포분석에 적절한 조직은 근육과 외투막이며 본 연구에 적용된 고정 과정이 없는 유세포분석법은 참전복 DNA 함량 분석 시 정확하고, 신속한 방법임이 판명되었다.

  • 이미 연구된 참전복용 경제적인 실험 배합사료로 까막전복과 시볼트전복에 대한 효과를 평가하기 위해 기존의 상품사료 및 미역과 비교하였다. 3 종류의 전복 모두 평균 체중 0.31g 인 치패를 각 사료마다 3반복으로 9주간 사육한 결과 ,체중 및 패각성장은 동일 전복종류내에서는 배합사료를 공급한 실험구들이 미역 공급구보다 성장이 좋았고(P<0.05), 동일 사료내에서는 시볼트전복, 까막전복, 참전복 순으로 성장이 좋았다(P<0.05). 생존율은 시볼트전복과 까막전복이 97~99%로 참전복의 74~77%에 비해 높았으며(P<0.05), 각 전복 종내에서의 사료 종류별로는 그 유의차가 인정되지 않는다(P>0.05). 전복 가식부의 일반 성분은 전복종류 및 사료종류에 따라 차이를 보였다. 이상의 결과로부터 본 실험에서 사용된 배합사료들은 3종류 전복 모두에 실용적인 사료가 될 것으로 기대된다.
