A Study for Effects of Automatic Transmission Fluid on Fuel Economy (자동변속기유가 연비에 미치는 영향에 대한 연구)

  • In order to improve the fuel economy by design change of automatic transmission, various technologies such as increased shift stages, slip control of lock-up clutch and compact and low-weight design have been developed. And also many OEMs have developed their own ATFs as a part of these automatic transmissions. In this study. to investigate the effects of ATF characteristics on fuel economy, we got the worldwide OEM ATFs and made some reference fluids. And physical properties, frictional characteristics and fuel economy using dynamometer test for these fluids were evaluated. From the investigation, it was found that viscosities of ATFs are correlated with fuel economy in dynamometer test and reducing the viscosities made it possible to obtain fuel economy.

  • High glucose leads to physio/pathological alterations in diabetes patients. We investigated collagen production in human gingival cells that were cultured in high concentrations of glucose. Collagen synthesis and secretion were increased when the cells were exposed to high concentrations of glucose. We examined endoplasmic reticulum (ER) stress response because glucose metabolism is related to ER functional status. An ER stress response including the expression of glucose regulated protein 78 (GRP78), C/EBP homologous protein (CHOP), inositol requiring enzyme alpha (IRE-$1{\alpha}$) and phosphoreukaryotic initiation factor alpha (p-eIF-$2{\alpha}$) was activated in the presence of high glucose. Activating transcription factor 4 (ATF-4), a downstream protein of p-eIF-$2{\alpha}$ as well as a transcription factor for collagen, was also phosphorylated and translocalized into the nucleus. The chemical chaperone 4-PBA inhibited the ER stress response and ATF-4 phosphorylation as well as nuclear translocation. Our results suggest that high concentrations of glucose-induced collagen are linked to ER stress and the associated phosphorylation and nuclear translocation of ATF-4.

  • The endoplasmic reticulum (ER) in the eukaryotic cells is the first compartment in the secretory pathway. Almost secretory proteins and membrane proteins are secreted through the ER, in which post-translational modifications occur via diverse signals from the ER lumen to the cytoplasm and nucleus. Only then are correctly-folded proteins secreted to the outside cells. Unfolded proteins that accumulate in the ER cause a kind of intracellular stress, ER stress, and activate an unfolded protein response (UPR) system. The 3 major transducers of the UPR are inositol requiring 1 (IRE1), PKR-like ER kinase (PERK) and activating transcription factor 6 (ATF6), all of which are ER transmembrane proteins. Recently, novel types of a new ATF6 family have been identified. Those commonly have an ER-transmembrane domain, a transcription-activation domain and a basic leucine zipper (bZIP) domain―Luman, OASIS, BBF2H7, CREBH and CREB4. Each factor functions by regulating the UPR in specific organs and tissues. Although the detailed molecular mechanisms of OASIS family members are unknown, in this study we comprehensively introduce these molecular signals.

Regulation of the Endoplasmic Reticulum Stress by BIP/GRP78 is involved in Meiotic Maturation of Porcine Oocytes In Vitro

  • In the present study, we investigated the role of binding immunoglobulin protein/glucose-regulated protein, 78-kDa (BIP/GRP78)-regulated endoplasmic reticulum (ER)-stress on meiotic maturation and cumulus cells expansion in porcine cumulus-oocyte complexes (COCs). Previously, it has been demonstrated that unfolded protein response (UPR)-related genes, such as molecules involved in ER-stress defense mechanisms, were expressed in matured oocytes and cumulus cells during in vitro maturation (IVM) of porcine oocytes. However, BIP/GRP78-mediated regulation of ER stress in porcine oocytes has not been reported. Firstly, we observed the effects of knockdown of BIP/GRP78 (an UPR initiation marker) using porcine-specific siRNAs (#909, #693, and #1570) on oocyte maturation. Among all siRNAs, siRNA #693 significantly reduced the protein levels of UPR marker proteins (BIP/GRP78, ATF4, and P90ATF6) in porcine COCs observed by Western blotting and immunofluorescence analysis. We also observed that the reduction of BIP/GRP78 levels by siRNA#693 significantly inhibited the meiotic maturation of oocytes (siRNA #693: $32.5{\pm}10.1%$ vs control: $77.8{\pm}5.3%$). In addition, we also checked the effect of ER-stress inhibitors, tauroursodeoxycholic acid (TUDCA, $200{\mu}M$) and melatonin ($0.1{\mu}M$), in BIP/GRP78-knockdown oocytes. TUDCA and melatonin treatment could restore the expression levels of ER-stress marker proteins (BIP/GRP78, $p-eIF2{\alpha}$, $eIF2{\alpha}$, ATF4, and P90ATF6) in siRNA #693-transfected matured COCs. In conclusion, these results demonstrated that BIP/GRP78-mediated regulation of UPR signaling and ER stress plays an important role in in vitro maturation of porcine oocytes.

Transcriptional activation of human GM3 synthase (hST3Gal V) gene by valproic acid in ARPE-19 human retinal pigment epithelial cells

  • The present study demonstrated that valproic acid (VPA) transcriptionally regulates human GM3 synthase (hST3Gal V), which catalyzes ganglioside GM3 biosynthesis in ARPE-19 human retinal pigment epithelial cells. For this, we characterized the promoter region of the hST3Gal V gene. Functional analysis of the 5'-flanking region of the hST3Gal V gene revealed that the -177 to -83 region functions as the VPA-inducible promoter and that the CREB/ATF binding site at -143 is crucial for VPA-induced expression of hST3Gal V in ARPE-19 cells. In addition, the transcriptional activity of hST3Gal V induced by VPA in ARPE-19 cells was inhibited by SP600125, a c-Jun N-terminal kinase (JNK) inhibitor. In summary, our results identified the core promoter region in the hST3Gal V promoter and for the first time demonstrated that ATF2 binding to the CREB/ATF binding site at -143 is essential for transcriptional activation of hST3Gal V in VPA-induced ARPE-19 cells.

Design and Implementation of Low Noise Amplifier for GPS Reciver (GPS수신기용 저잡음 증폭기의 설계 및 구현)

  • This papers describes two low-noise amplifiers that use the Hewlett-Packard ATF-10236 low noise GaAs FET device, The actual measured performance of the amplifiers compares favorably to that predicted by the computer simulation(ADS) the noise figure of the 1575MHz amplifier was measured at 1.78dB which is lower that 2dB as specified. Measurement gam measured 33.0075dB which is within 35dB$\pm$0.5㏈ of the GPS specification. Network Analyzer(HP8510) is used to measure all the s-parameters and Noise Figure meter(HP8970B) is used to measure noise figure. As the result of experiment, gain, input VSWR, output VSWR is within the GPS specification sufficiently.

  • In this paper, two-stage low noise amplifier(LNA) for S-band is designed and implemented using ATF54143 HEMT of HP CO. In order to get noise figure and input VSWR to be wanted, it is considered input VSWR and noise figure simultaneously in matching-circuit designing. The fabricated two-stage low noise amplifier has the gain of 27.8dB, input VSWR and output VSWR under 1.5.

Endoplasmic Reticulum Stress Response and Apoptosis via the CoCl2-Induced Hypoxia in Neuronal Cells (CoCl2 처리로 유도된 hypoxia상태에서 세포자살과 ER stress에 관련된 인자의 발현)

  • Cobalt(II) chloride, a chemical compound with the formula$CoCl_2$, has been widely used in the treatment of anemia, as a chemical agent for the induction of hypoxia in cell cultures, and is known to activate hypoxic signaling. However, excessive exposure to cobalt is associated with several clinical conditions, including asthma, pneumonia, and hematological abnormalities, and can lead to tissue and cellular toxicity. It is also known to induce apoptosis. One of the questions was that of whether $CoCl_2$ might induce apoptosis via endoplasmic reticulum (ER) stress in neurons. To address this question, first, the level of DNA fragmentation was measured for assay of apoptotic rates using $CoCl_2$ with neuron PC12 cells. After confirmation of apoptosis inductions, under the same conditions, the expression levels of ER stress associated factors [ER chaperones Bip, calnexin, ERp72, ERp29, PDI, and ER membrane kinases (IRE1, ATF6, PERK)] were examined by RT-PCR and Western blotting. These results indicated that apoptosis is induced through activation of ER membrane kinases via ER stress. In conclusion, during induction of apoptosis through $CoCl_2$-induced hypoxia in neuron PC12 cells, ER membrane kinase of IRE1 was dominantly up-expressed, and, consecutively, TRAF2, which has been suggested to be one of the links connecting apoptosis and ER stress, was strongly up-expressed.

  • This paper presents development of a LNA operating at 1.71 ∼ 1.18 GHz used for a receiver of KOREA PCS base station and transponder. Using resistive decoupling circuits, a signal at low frequency is dissipated by a resistor. This design method increases the stability of the LNA and suitable for input stage matching. The LNA consists of low noise GaAs FET ATF-10136 and internally matched VNA-25. The LNA is fabricated with both the RF circuit and the self-bias circuits in aluminum housing. As a result, the characteristics of the LNA implemented here shows 30 dB in gain and 0.85 dB in noise figure.
