Promotion of Bone Nodule Formation and Inhibition of Growth and Invasion of Streptococcus mutans by Weissella kimchii PL9001

  • Lactic acid-producing bacteria (LABs) are known to have various beneficial properties for health. However, they are generally considered to have an adverse effect on teeth, since they produce acid. Nonetheless, milk and cheese containing specific LAB strains were recently found to have an inhibitory effect on dental caries in children, with an inhibitory activity towards the growth of Streptococcus mutans suggested as the responsible mechanism. Accordingly, the current study selected a probiotic candidate for oral health and studied its inhibitory mechanism against dental caries. Twenty-two LAB species belonging to eleven genuses were screened for promoting bone nodule formation using direct microscopic examination. Only one isolate, Weissella kimchii strain PL9001, increased the bone nodule formation significantly. The addition of W. kimchii strain PL9001 to bone cells prepared from mouse calvaria increased the bone nodule formation, calcium accumulation, and activity of alkaline phosphatase (the osteoblastic marker). Moreover, W. kimchii strain PL9001 inhibited the invasion of Streptococcus mutans into bone cells, and an organic extract of the culture supernatant of W. kimchii strain PL9001 inhibited the growth of Strep. mutans. Therefore, the results suggest that W. kimchii strain PL9001 can be used as a preventive measure against dental caries. This is the first time that a LAB has been shown to promote bone nodule formation and prevent the invasion of Strep. mutans into bone cells.

  • Recently, numerous government institutions and information service organizations have shown interests in employing ISO 9001 Quality Management System(QMS) to increase their organizational performances. However, due to the gap between the system requirements and the process oriented goals in reality, they have not been able to merit from the ISO 9001 QMS. So, Korea Institute of Science and Technology Information(KISTI) has been designed a process oriented system by incorporating the process analysis results into the existing ISO 9001 system requirements for enhancing organizational competitiveness and promoting productivity through a process standardization. As a result, KISTI has been ISO 9001:2000 certificate in November 11, 2003. And we obtain a improvement of customer satisfaction and efficiency of work, reduction of work processing time using KQMS.

  • The purpose of this study is to analyze the management performance of the companies in the auto parts production requirements of new management techniques ISO9001 quality management system requirements for management innovation perspective in convergence age. For this purpose, we conducted a survey visiting the auto parts manufacturers. The results were as follows. First, among the details of the four key requirements of the ISO 9001 quality management systems in small and medium component suppliers, managers and responsible resource management showed a positive impact on business performance. In addition, based on the results of the investigation of the differences by grade, the higher rank group executive responsibilities, sub-grade group showed a low awareness of the quality management according to ISO 9001. This will contribute to establishing the guidelines and Improvement Consulting Strategies to improve productivity in small and medium automotive rubber bellows automotive parts production.

  • ISO 9001 Quality Management Systems-Requirements has been revised in 2015. It has been updated four times since its publication in 1987. It is the most widely used International Standard in the world. There are over one million companies and organizations in over 170 countries certified to ISO 9001 from an ISO survey. Organizations are supposed to be certified to this new version by late 2018. The key changes in ISO 9001:2015 are to establish a High Level Structure (HLS) and focus on Risk-Based Thinking (RBT). Risk-Based Thinking means the process approach to decide how risk is addressed in establishing the processes to improve process outputs and prevent undesirable results. It pursues process planning and controls based on risks so that organizations can improve the effectiveness of the quality management system. It maintains and manages a Quality Management System that inherently addresses risks and meets objectives. In this article we firstly attempt to explain how to understand the fundamental concept of Risk-Based Thinking which is a systematic approach to consider risks rather than treating prevention as a separate component of a Quality Management System. We comment on the detailed requirements that contain risks in ISO 9001:2015 clauses. We also summarize recent advances on the risk assessment and management in line with ISO 31000:2009 Risk Management-Principles and Guidelines. We finally propose the practical risk management procedures for implementing ISO 9001:2015 with an emphasis on RBT. This article would contribute to help quality managers and practitioners convert to ISO 9001:2015.

Weissella confusa Strain PL9001 Inhibits Growth and Adherence of Genitourinary Pathogens

  • The capability of lactic acid bacteria (LABs) to adhere to intestinal epithelial cells and vaginal epithelial cells is an important factor in the formation of a barrier to prevent the colonization of pathogenic bacteria. In addition, the ability to coaggregate with pathogens and production of antimicrobial agents also allow LABs to fight against pathogens. In this work, Weissella confusa PL9001 was tested for its ability to inhibit the growth and adherence of genitourinary pathogens, including Candida albicans, Escherichia coli, Staphylococcus aureus, and vancomycin-resistant Enterococcus faecium (VRE), isolated from the urine of hospitalized female patients. W. confusa PL9001 was found to coaggregate with the four pathogens, as observed with a light microscope and scanning electron microscope. In competition, exclusion, and displacement tests, the adherence of the pathogens to T24 bladder epithelial cells was also inhibited by W. confusa PL9001. Accordingly, these results suggest that W. confusa PL9001 is potentially useful for both preventive and therapeutic treatment of genitourinary infections.

  • This paper investigates the relative effect of each ISO 9001 : 2000 quality management system's requirement and business performance. The empirical tests are conducted in ISO 9001 certification companies. As a result, the $1^{st}$, it shows the requirements positively affect performance of financial. The $2^{nd}$, also it shows the requirements positively affect non-financial performance. The $3^{rd}$ also it show the non-financial performance positively effect performance of financial performance.

  • This Study aims to analyze the management performances of the rubber products from the perspectives of ISO9001 Quality Management System, which is a new management technique for management innovation, in order to secure the competitiveness of the small and medium manufacturers of rubber products for automobile as automobile application components. The results of the analysis can be summarized as follows: First, as the results of the analysis of effect of the (1) responsibility of the management, (2) resources management, (3) realization of the products, and (4) measurement, analysis and improvement, which are the 4 core requirements of the ISO9001 Quality Management System, on the management performances of the small and medium sized manufacturers of the rubber components for automobile, it was found that only the responsibility of the management and resources management affect the management performance. The above research findings imply that the company must absolutely rely on the upper ranked groups rather than the lower ranked groups, and on the responsibilities of the management rather than the upper ranked groups in order to elevate the management performance of the small and medium sized manufacturers of the rubber components for automobile.

  • The ISO (International Organization for Standardization) 9000 QMS (Quality Management System) standards specify requirements that need to demonstrate an organization's ability to consistently provide products or services. Korean companies have been implementing ISO 9000 QMS to compete in global markets. ISO 9000 QMS standards have been revised in 1994, 2000, 2008, and 2015 since the first release in 1987. The key change in ISO 9001:2015 is focused on risk-based thinking. It means that risk inherent within an organization is early identified and proactively controlled by use of all aspects of QMS rather than taking preventive actions. In this article we did literature review mainly on empirical study related to motivation and operation performance of ISO 9000 implementation. We present the necessity of changeover, features of new version, and comparison between new and old version. In order to see how well organizations can implement the new ISO 9001:2015 we focus on manufacturing companies that are going to convert to ISO 9001:2015 registration. We collect their key characteristics such as yearly sales, the number of MPI (Management Performance Indicators), the number of KPI (Key Performance Indicators), the number of quality control engineers, quality control engineer work experience, and the degree of adaptiveness between new ISO 9001:2015 QMS and QMS currently operating in manufacturing companies. Especially the degree of adaptiveness is analyzed by correlation analysis, regression analysis, ANOVA, and t-tests to discover the relationships among their characteristics. We found out that the degree of adaptiveness is linearly related to quality control engineer work experience and the number of MPI and KPI that is practically utilized in manufacturing companies. We summarize the implications discovered to help manufacturing industry convert to ISO 9001:2015 registration.

This study is to discuss particular requirements for the application of ISO 9001 : 2000 for automotive production and relevant service part organization , and guidelines on the application of ISO 9001 : 2000 for the food and drink industry.

Biological Control of Apple Ring Rot on Fruit by Bacillus amyloliquefaciens 9001

  • Apple ring rot disease, caused by Botryosphaeria dothidea (Moug. ex. Fr) Ces. et de Not., is one of the most important diseases on apple fruits. In this study, strain 9001 isolated from healthy apple fruits from an infested orchard was evaluated for its biocontrol activity against apple ring rot in vitro and in vivo. Strain 9001 showed obvious antagonistic activity to B. dothidea YL-1 when plated on potato dextrose agar. Soaking healthy apples in the bacterial suspensions of strain 9001 prior to artificial inoculation of fungal pathogen resulted in a dramatic decrease in disease incidence when compared to the control. Moreover, either field application in the growth season or postharvest treatment of apples from infected orchards with bacterial suspensions of strain 9001 resulted in significantly reduced disease incidence within the storage period for 4 months at room temperature. Based on the phylogenetic analysis of 16S rRNA and the gyrA gene, strain 9001 was identified as Bacillus amyloliquefaciens. These results indicated that B. amyloliquefaciens 9001 could be a promising agent in biocontrol of apple ring rot on fruit, which might help to minimize the yield loss of apple fruit during the long postharvest period.
