Method Development for the Sample Preparation and Quantitative Analysis of Synthetic Colors in Foods (식품 중 식용타르색소의 시료별 전처리방법 확립 및 함량 분석)

  • Method for sample preparation and quantitative analysis of 19 permitted and non-permitted synthetic colors in foods was developed based on reversed-phase ion-pairing high performance liquid chromatography. For color extraction of samples, deionized water was added, and pH was appropriately adjusted with 1% ammonia water. Any undissolved matters were extracted with 50% ethanol or 70% methanol. Lipid in snacks was first removed using n-hexane with centrifugation, water was added to extract colors, followed by clean-up and concentration using Sep-Pak $C_{18}$ cartridge. Recovery efficiencies at known concentrations of 19 standard food colors spiked into foods were in 90.3-97.9% range far soft drink, 79.2-101.9% for candy, 84.1-103.4% for jelly, 86.4-100.8% for chewing gum, 83.5-103.4% for ice cream, and 78.5-95.6% for snack.

Studies on the Compositional Change of Composts During Mushroom Cultivation (양송이 재배(栽培)에 따른 재배상퇴비(栽培床堆肥)의 성분변화(成分變化)에 관(關)한 연구(硏究))

  • In order to investigate the compositional change oil composts during the growing of cultivated mushroom (Agaricus bisporus), composts and mushrooms during the period of filling to ending under commercial conditions were subjected to chemical analyses. The results are summarized as follows and the mechanism of composting for mushroom cultivation was proposed. 1) The temperature change of growing bed and room was observed and the yield of mushroom for each cropping time was recorded to get $15.6kg/m^2$ in total crops. 2) Composts after filling showed pH 8.2 which dropped to 6.4 after casing and continued so up to ending. 3) On the dry weight basis of composts, crude ash increased whereas total nitrogen, ether extract and crude fibre decreased gradually to bring about the lowering of organic matter. 4) Total nitrogen of composts decreased gradually and more insoluble nitrogen was lost than soluble nitrogen. The C/N ratio of composts was initially 21 which was gradually lowered to 16. 5) The losses of ${\alpha}-cellulose$, pentosan and lignin in composts were 87%, 75%, and 60%, respectively, in which ${\alpha}-cellulose$ decreased markedly after casing. 6) Free reducing sugars of composts increased continuously. Gradually increased free amino acids till second cropping decreased again thereafter. Composts at the filling stage contained alanine, glutamic acid, glycine and serine in which glycine decreased markedly whereas proline increased remarkably upon mushroom cultivation. 7) Among minerals of composts, phosphorus and zinc tended to decrease, potassium and copper tended to increase anti sodium showed no marked change. 8) In comparison of mushrooms from different cropping time with respect to proximate composition, minerals, free reducing sugars and amino acids, no marked difference was observed. However, a little higher values were observed in crude fat, free reducing sugars and sodium content for early crops and in free amino acids and phosphorus content for late crops. Twelve free amino acids including alanine, serine, threonine, and glutamic acid were detected in the cultivated mushroom. 9) According to above experimental results, it was possible to support the mechanism of compositing that the formation of ammonia and decomposition of carbohydrates by mesophiles are followed by protein biosynthesis, formation of microbial bodies and nitrogen-rich lignin humus complex by thermophiles, thus supplying necessary nutrients for mushroom growth, along with residual carbohydrates.

Effect of Diets Containing Ground Charcoal Powder, Wood Vinegar and Fermented Acetic Acid on the Protein and Energy Metabolism in White Leghorn Strain Layer (백색 산란계의 단백질 및 에너지 대사에 미치는 성형 목탄가루, 목초액 및 양조식초 첨가사료의 영향)

  • The investigation concerned an effect of the ground charcoal powder and organic acids on the digestibilities of protein and energy or the contents of uric acid, ammonia, creatine and urea in excreta of 113 week-old White Leghorn strain layers. Birds were fed basal (control) diet composed of mainly corn-soybean meal during a week of previous feeding and subsequent experimental diets during 12 weeks of experimental feeding . The experimental diets were the control diet(CON). diet(CPD) substituted 0.5% of the ground charcoal powder with the defatted rice bran of the CON, diet(PWV) added 0.1mM(based on the acetic acid) wood vinegar in the CPD and diet(PFA) added 0.1mM (based on the acetic acid) fermented acetic acid in the CPD. Birds fed CPD excreted significantly(P<0.05) more fecal nitrogen(FN) and lower urinary nitrogen (UN) than those of birds fed CON. Digestibility of protein was lower significantly (P< 0.05) in CPB-fed bird than in bird fed CON. while birds fed CON. PWV and PFA showed similar values. Also urinary nitrogen per nitrogen intake (UN/NI) or absorbed nitrogen (UN/AN) was significantly (P<0.05) lower in birds fed CPD compared with those in birds fed CON. And birds fed PWV tended to increase UN/NI and UN/AN, while PFA-fed birds excreted significantly (P<0.05) higher UN/Nl and UN/Ah than those of birds fed CPD diet. The uric acid nitrogen (UAN) per nitrogen intake (UAN/NI) or absorbed nitrogen (UAN/AN) were lower significantly(P<0.05) in CPD-fed birds and were tended to decrease in birds fed PWV compared with those in birds fed CON and PFA The ammoniacal nitrogen(AMN) per nitrogen intake (AMN/NI) or absorbed nitrogen (AMN/NI) was tended to increase in birds fed experimental diets and was increased significantly(P<0.05) in birds fed PFA compared with those of birds fed CON. The excretion of creatine and urea nitrogen per nitrogen intake or absorbed nitrogen was shown similar values among birds fed experimental diets Digestibility of energy (DE/GE) was not shown any significant effect of experimental diet and were in the range of 80~84%. But metabolizability (ME/GE or MEn/GE) was increased in birds fed CPD and PWV and was decreased in birds fed PFA compared with those in birds fed CON. Although birds fed PWV showed significantly(P<0.05) higher ME/GE than bird fed PFA, the MEn/GE were higher significantly (P<0.05) in birds fed CON and CPD compared with that in birds fed PFA. Fecal energy affects 10~23% in the change of metabolizability though significant effect of fecal energy on the metabolizability were not found. But the effect of urinary energy on the metabolizability of diet was lowered as 2.3~3, 0% and the effect of experimental diets on the metabolizability of diets was due to change of urinary energy which also was originated from the change of uric acid energy.

Field Bioassays On Shellfish To Assess Environmental Pollution Levels Of The Masan Bay (마산만(馬山灣)의 환경오염(環境汚染) 평가(評價)를 위한 야외(野外) 생물(生物) 오염(汚染) 시험(試驗) 연구(硏究))

  • A study on field bioassay test using four species of commercially important shellfish was carried out to assess the effect of pollutants and determine the extent of marine environmental pollution of the Masan Bay from 9 to 15 August 1978. Water quality analysis and planktological examination of sea water were made during the experiment and the examination gave the following results. The water temperature was so high at 31.7$^{\circ}C$ in maximum and rather subject to change on weather condition of the land than on the effect of the water mass from outer bay. The range of DO,COD and SS at the stations were 0.3-7.08cc/l, 0.07-3.31ppm and 5.5-117ppm, respectively with the high values of COD and SS at the stations 7 and 1. The concentrations of the dissolved inorganic nitrogen in sea water, NH$\sub$4/-N, NO$\sub$2/-N, NO$\sub$3/-N, and PO$\sub$4/-P were 18.90-99.80, 2.48- 19.60, 13.00-39.00 and 1.04-14.0$\mu\textrm{g}$ at/l, respectively with decrease of their values in the outer part of the Bay. The high values mentioned above were caused by the sewage and industrial activities. The effects of organic waste are increased oxygen demand, nutrient concentration, turbidity and a higher input of pathogens, leading to structural changes in the marine ecosystems and to a considerable hazard to public health. The percentage composition of phytoplankton standing crop between diatom and dinoflagellate was characterized by making a difference between the two groups in respect of location: a decrease of diatom and a increase of dinoflagellate in numerical abundance toward inner part from outer part of the bay. Namely phytoplankton organisms were composed of 80% of diatom and 20% of dinoflagellate in outer bay, on the contrary, only 4% of diatom and 96% of dinoflagellate occupied by 94% of prorocentrum micans known as tolerant species to polluted reaas in the inner bay. On the occurrence and composition of zooplankton, there are two significant communities in the bay:one is characterized by the predominance of Oithona nana and the other by Favella sp They were composed of a range from 84% to 90% of the total organisms and monotonously constituted of themselves only at most inner station 3 even small numbers. From the results mentioned above, Oithona nana, Favella and prorocentrum micans recommed themselves as valuable indicators for judging the extent of the marine pollution.During the period of the biossays Mytilus edulis showed the highest mortality and Tapes japonica the lowest one between the four test species. The highest death rate by stations was found at most inner stations 3'and 4near Masan Free Export Zone with the most sensitive response and the lowest one occured at outer station 13 where no death specimen of oyster and arkshell was found during the whole test period.As for mussel,85 percent death rate appeared after 72 hours and 100percent rate after 120hours at station 4. It was found that the significant high mortality of the test shellfish mentioned above was caused by severe pollution with mainly organic pollutants from domestic sewage and industrial wastes from the results of too much higher concentrations of dissolved inorganic nitrogen especially ammonia-N,COD,SS and lack of dissolved oxygen,and furthermore occurrence and abundance composition of Prorcentrum,Favella and Oithona nana by stations, valuable indicator species of coastal pollution by orgnic and boilogical pollutants.

Influence of Supplemental Dietary Yeast Culture on the Noxious Gas Emission in Broiler Houses and Performance of Broiler Chicks (효모배양물 첨가 사료가 계사내 유해가스 발생 및 육계의 생산성에 미치는 영향)

  • Two experiments were conducted to investigate the effect of dietary supplementation of yeast culture on the performance of broiler chicks and noxious gas emission in broiler houses. Two hundred forty and three hundred and twenty, one day old Cobb male broiler chicks in Expt 1 and Expt 2, respectively were alloted to four treatment levels of yeast culture (0, 0.1, 0.2, 0.4%). To each treatment, 60 birds were assigned in Expt 1 and 5 replicates of 16 birds each were assigned in Expt 2. Basal diets contained 21.5% and 19.0% CP, and 3,100kcal/kg ME for the starting and finishing periods, respectively. Ammonia and $CO_2$ gas emission were detected twice a day for seven days during the five week period of Expt 1. Weight gain, feed intake and feed efficiency were measured for five weeks in Expt 2. Intestinal microbes, blood cholesterol and ND antibody titer were examined at the end of Expt 2. In Expt 1, the concentration of $NH_3$ in the house of birds fed yeast culture tended to be lower than the control. It was significantly lower in the 0.4% yeast culture treatment than the control (P<0.05). $CO_2$ concentration was significantly lower in all yeast culture treatments regardless of its dietary supplemental level than the control (P<0.05). different from others. Feed efficiency (feed/gain), however, was significantly improved in all yeast culture treatments relative to that of the control for starting period (P<0.05) and that of 0.2% yeast culture treatment was significantly lower than those of the control and 0.4% for the overall period. Total number of E. coli in the ileum of birds fed yeast culture at 0.1 and 0.2% was significantly lower than those of the control and 0.4% in the ileum. The CFU of Lactobacillus spp. of birds fed yeast culture at 0.1% was higher in the cecum compared to other treatments (P<0.05). Total cholesterol level of chicks fed 0.1% yeast culture seemed to be lower compared to that of other treatments, whereas LDL-cholesterol level was significantly lower than those of the control and 0.4% treatment. ND antibody titer tended to be higher in the yeast culture treatments than the control, but was not significantly different. The results of these experiments indicated that 0.2% yeast culture may have a potential to reduce the noxious gas emission in broiler houses and maximize the performance of broiler chicks.

Effect of Temperature and Dissolved Oxygen on the Survival Rate and Physiological Response of the Warty Sea Squirt Styela clava (수온과 용존산소 변화에 따른 미더덕 Styela clava의 생존율 및 생리적 반응)

  • Decrease in dissolved oxygen concentrations associated with temperature fluctuation is an important criteria to evaluate the mortality rate of the species. Based on this parameter, we investigated the survival rate, physiological response and histological change of warty sea squirt. It was found that the survival rate of the warty sea squirt species was 63.3% at $23^{\circ}C$ and 56.6% at $26^{\circ}C$ respectively. However, exposure of six days at $29^{\circ}C$ caused deaths among species, which indicated the 6day-$LT^{50}$ of the tested species to be $24.58^{\circ}C$ ($19.48{\sim}35.48^{\circ}C$). Further, after 11 day of exposure, the dissolved oxygen concentration has been found to decrease, with the survival rate of 20% at $4.0mg\;L^{-1}$ and deaths at $2.0mg\;L^{-1}$, thus 11day-$LC^{50}$ calculated to be $3.88mg\;L^{-1}$ ($3.29{\sim}4.57mg\;L^{-1}$). In addition, decrease in rate of oxygen consumption and excretion of ammonia was also noted at this critical water temperature and dissolve doxygen concentration. Moreover, there has been common histopathological changes were observed in warty sea squirt's gill pouch, digestive tract, and tunic as follows such as: proliferation of epithelial cells, condensation and necrosis, permeation of phagocyte and blood cell, loss of cilium and muscular fiber degeneration. Based on our study results, we suggest that these parameters can also be useful to evaluate the survival rate and physiological response in other species.

Composition and emission characteristics of fine particulate matters at the 1100 Site of Mt. Halla during 2011-2012 (한라산 1100고지 대기 미세먼지의 조성 및 배출 특성: 2011~2012년 측정)

  • PM10 and PM2.5 samples were collected at the 1100 site of Mt. Halla in Jeju Island during 2011~2012, and their ionic and elemental species were analyzed, in order to investigate the characteristics of emission sources as well as aerosol compositions. The mass concentrations of PM10 and PM2.5 were 22.0±13.1 µg/m3 and 11.3±6.1 µg/m3, respectively, showing 2.4~2.6 times lower than those of the capital city area of Korea. The composition ratios of major secondary pollutants (nss-SO42−, NH4+, and NO3) were the highest as 85.5 % for PM10 and 91.3 % for PM2.5, and followed by the order of marine (Na+, Cl, and Mg2+), organic acid (HCOO and CH3COO), and soil (nss-Ca2+) sources. Among the elemental species in PM10, soil-originated components (Al, Fe, and Ca) were consisted of 50.9 %, which was higher proportion than marine and anthropogenic elements. The acidification of the fine particulate matters was found to be influenced mostly by sulfuric and nitric acids, and these acids were mainly neutralized by calcium carbonate in PM10 and by ammonia in PM2.5. The clustered back trajectories showed that 47 % of total air mass inflows was from the China, and the concentrations of NO3 and nss-Ca2+ were especially high corresponding to the inflows.

The Analysis of Environmental Loads and Material Recycling of the Nutrients by the Livestock Wastewater Originating from Imported Feeds (수입사료에 의한 가축분뇨 물질순환 및 환경부하 분석)

  • The nearly 75% of animal feed materials used for livestock production are imported every year in Korea. Most of imported feed ingredients are concentrated feeds such as com, wheat, soybean, soybean meal, etc. and they are used as the source materials for the production of assorted feed. The imported concentrated feeds are high in nitrogen and phosphorus. Therefore, the consistent import of feed ingredients may cause an increase of nutrient deposit in our agricultural ecosystem. In the current review, it was discussed with the situation of the feed importation and its nutritional composition to evaluate the nutrient load by the imported feeds onto agricultural ecosystem. The nutrient load caused by imported feeds in agricultural environment was compared with the nutrient demand for crop production. The amounts of N, $P_2O_5\;and\;K_2O$ introduced by the imported fteds in Korea were 371, 140 and 143 Ktons. And, the N, $P_2O_5\;and\;K_2O$ loads excreted from imported feeds in livestock were 148, 84 and 86 Ktons of N, $P_2O_5\;and\;K_2O$ and These nutrient loads by the imported feeds are at the percentage of N 52%, $P_2O_5$ 52% and $K_2O$ 42% in the comparison of total nutrient amounts excreted from livestock animals in Korea. The 82.3% of nutrients excreted from livestock was recycled to crop land as compost and liquid fertilizer, and the others were discharged to river after water treatment processing or disposed to ocean. Also, passing through the recycling process far the production of compost and liquid fertilizer, the amount of nutrients was reduced by the ammonia vaporization of livestock feces and urine. Accordingly, N 81, $P_2O_5$ 74 and $K_2O$ 76 Ktons in the nutrients excreted from livestock were estimated to be utilized in the crop land. Consequently, it was estimated that 44, 48 and 69 Ktons of N, $P_2O_5\;and\;K_2O$ were taken up with crops in the consideration of the ratio of mineralization, and the amounts of leached or deposited N, $P_2O_5\;and\;K_2O$ in crop land were estimated to be 37, 27 and 7 Ktons, respectively. It is estimated that 12%, 34% and 48% of N, $P_2O_5\;and\;K_2O$ introduced by the imported feeds were used by crops, and 10%, 34% and 5% of N, $P_2O_5\;and\;K_2O$ were leached or deposited in agricultural ecosystem. Therefore, considering the leached and deposited amounts of N, $P_2O_5\;and\;K_2O$ originated from the imported feed ingredients, the consistent import of feeds may gradually increase the nutrient load onto agricultural ecosystem.

Seasonal Variations of Chemical Composition of the Estuary Water at the Tidal Flat in Nack Dong River from Nov. 1962 to Oct. 1963 (洛東江 河口 干潟地 水質의 年間變化 (1962年 11月~1963年 10月))

  • Seasonal variations of the contents of some chemical constituents of the estuary water at two definite stations of the laver bed in Nack Dong River have been determined over one tidal cycle in spring tide from Nov. 1962 to Oct. 1963. The ranges of annual variations of the contents at station 1 and station 2 are as follows: water temp. $2.2-30.8^{\circ}C$, $3.3-28.0^{\circ}C$; pH 7.8-8.5, 7.9-8.4; chlorosity 0.025-19.66 g/l, 4.31-19.56 g/l; magnesium 0.00355-1.565 g/l, -1.524 g/l; calcium 0.00557-0.482 g/l, - -0.590 g/l; saturation % of dissolved oxygen 71.8-123.2%, 88.2-113.8%; silicate-Si 8.00-125.5 ${\mu}$g-at./l, 6.70-100.5 ${\mu}$g-at./l; phosphate-P 0.12-1.47 ${\mu}$g-at./l, 0.11-1.09 ${\mu}$g-at./l; ammonia-N 4.88-25.45 ${\mu}$g-at./l, 4.12-17.58 ${\mu}$g-at./l; nitrite-N 0.07-0.75 ${\mu}$g-at./l, 0.08-0.58 ${\mu}$g-at./l; nitrate-N 2.11-6.89 ${\mu}$g-at./l, 1.85-7.43 ${\mu}$g-at./l each. The annual tidal variations of the constituents at station 1 are more remarkable than of station 2. The chlorosity, magnesium and calcium contents are decreased nearing the slack after ebb, and increased abruptly then one hour after the slack. The contents of the other constituents are varied according to the chlorosity variety. The values of pH, chlorosity, magnesium and calcium contents are lower in summer than winter, while the difference of seasonal variations of the % saturation of dissolved oxygen is not remarkable. The phosphate-P and total nitrogen contents have a tendency of increasing within a definite range, while the silicate-Si increase proportionally, to the increasing of mixing percentage of fresh water. The average values of Si/P and N/P are several times greater than of the normal in sea water. The chemical composition considered from the value of Mg/Cl or Ca/Cl of estuarine water varies according to the variety of chlorosity, even at the high chlorosity of 19 g/l.

Changes in isoflavone content and quality characteristics of Cheonggukjang prepared with Bacillus subtilis HJ18-3 and KACC 15935 (Bacillus subtilis HJ18-3과 KACC 15935를 이용하여 제조한 청국장의 품질특성과 isoflavone 함량의 변화)

  • This study was conducted in order to investigate the change of isoflavone composition (glycoside and bio-active aglycone), and to evaluate the quality characteristics of Cheonggukjang, which was prepared by different bacillus strains. After the 48-hour fermentation, the contents of daidzein, genistein, and glycitein in the Bacillus subtilis HJ18-3 have significantly increased up to approximately $89.06{\pm}3.59$, $10.36{\pm}0.28$, and $101.37{\pm}3.67ug/g$, respectively. The contents of daidzein, genistein, and glycitein in the Bacillus subtilis KACC 15935 were $38.88{\pm}5.39$, $12.58{\pm}2.14$, and $80.13{\pm}0.71ug/g$, respectively. The original content of daidzein was 3.96 ug/g, while genistein and glycitein were not measured. However, the contents of daidzen and genistein in HJ18-3 and in KACC 15935 were decreased. The ${\alpha}$-Amylase and cellulase activities of Chungkookjang in HJ18-3 were higher than in the KACC 15935. The contents of Chungkookjang in HJ18-3 were $29.70{\pm}11.66$ and $4861.3{\pm}388.07unit/g$, respectively. The amino type nitrogen contents and ammonia type nitrogen contents of Chungkookjang in KACC 15935 were higher than in the HJ18-3. These results suggested that it could be used to increase the bioactivity via fermentation with the Bacillus subtilis possessing a ${\beta}$-glucosidase activity with a view towards the development of functional foods.
