Changes in the constituents and UV-photoprotective activity of Astragalus membranaceus caused by roasting (황기의 볶음 조건에 따른 성분 및 자외선 광보호 활성 변화)

  • Purpose: Astragalus membranaceus (AM) is an important traditional medicinal herb. Pharmacological research has indicated that AM has various physiological activities such as antioxidant, anti-inflammatory, immunoregulatory, anticancer, hypolipidemic, antihyperglycemic, and hepatoprotective activities. The bioactive substances responsible for the physiological activities in AM, including many antioxidant substances, change during the roasting process. This study investigated and compared the changes in the antioxidant constituents of AM caused by roasting. Methods: DPPH (1,1-diphenyl-2-picryl hydrazyl) and $ABTS^+$ (2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt) radical scavenging activities and their total phenolic content (TPC) were measured. High-performance liquid chromatography (HPLC) analysis was performed to confirm any changes in the isoflavonoids of roasted AM (R-AM),. The cell viability of UVB-induced HDF (Human dermal fibroblast) cells treated with AM and R-AM extracts was investigated. The comet assay was used to examine the inhibitory effects of R-AM extracts on DNA damage caused by oxidative stress. Results: The DPPH and $ABTS^+$ radical scavenging activities were $564.6{\pm}20.9$ and $108.2{\pm}3.1$ ($IC_{50}$ value) respectively, from the 2R-AM. The total phenol content was $47.80{\pm}1.40mg$ GAE/g from the 1R-AM. The values of calycosin and formononetin, which are the known isoflavonoid constituents of AM, were $778.58{\pm}2.72$ and $726.80{\pm}3.45{\mu}g/g$ respectively, from the 2R-AM. Treatment of the HDF cells with R-AM ($50{\sim}200{\mu}g/mL$) did not affect the cell viability. Furthermore, the R-AM extracts effectively protected against UVB-induced DNA damage. Conclusion: The findings of this study indicate that R-AM increases its isoflavonoid constituents and protects against UVB-induced DNA damage in HDF cells.

Inhibitory effects of ethanol extract of Atractylodis Rhizoma on melanogenesis in B16/F10 melanoma cells

  • The inhibitory effect of Atractylodis Rhizoma alba extract(AM) on melanogenesis was studied by using B16/F10 melanoma in culture. Cells were cultured in the presence of various concentrations of AM for 48 hrs, and the experiment of total melanin content as a final product and activity of tyrosinase, a key enzyme, in melanogenesis. AM significantly inhibited tyrosinase activity, and melanin content in a dose-dependent manner. These results show that Atractylodes macrocephala extract could be developed as skin whitening components of cosmetics.

Antimicrobial Susceptibility of Bordetella bronchiseptica Isolates from Pigs (돼지에서 분리한 Bordetella bronchiseptica의 항생제 감수성)

  • Susceptibility or resistance of the Bordetella bronchiseptica was determined by using the broth microdilution method. Each Bordetella bronchiseptica was tested fur sutraceptluility with 7 different antimicrobial agents.4 high prevalence of resistance (greater than 80 %) was found when Bordetella bronchiseptica was tested with amikacin (AK), ciprofloxacin ILrJl gentamlcin IGAgf, kanamycin (Kbf) and tobramycin (TM). However, the Bordetella bronchiseptica were sensitive to chloramphenicol (CP) and tetracycline (TC). Three different combination of drug resistance were observed : AM-CF-GM-TM (1 strain), AM-CF-GM- KM-TM (45 strains), and AM-CF-GM-KM-TM-CP (3 strains). The MICs against Bordetella bronchiseptica were 0.13 to >2.00 for TC, 4 to >648 for CP, 8 to >128 for CF 16 to >256 for GM, 16 to >256 for TM, 32 to >256 for KM and 64 to 256 for AM.

A Study on the Characteristic Micro-Climate of Myeong-Kwan Kim House and the Moisture Content Behavior of Outside Exposed Columns (김명관고택의 미기후 특성과 외진노출기둥의 함수율 양상에 관한 연구)

  • Wood is one of the main materials of wooden building. Hanok also uses wood as its main component. Recently, Hanok continues to be built. Wood is affected by the climatic environment. The growth of decay bacteria is activated at more than 80% relative humidity. The microclimate environment and moisture content were measured for architectural cultural properties that have been maintained for a long time as a wooden building. The method analyzed the measured data by distinguishing between cloudy and sunny days. In the case of the old house, Anchae moisture content was higher than that of Sarangchae. This seems to be due to the narrow front yard or the planting of trees. The microclimate environment inside the house began to decrease in humidity from 8 am. According to the survey data, the relative humidity was less than 80% from 9 am when there was wind around 4-6 am. It appeared an hour earlier than in the absence of wind. As a result, the time interval for dehumidifying of wood was widened. Therefore, the wooden building is open to the front so there is airflow under the eaves during the daytime and wind in the morning appear to be alternatives in order to lower the moisture content.

Sa-am Five-element Acupuncture and Hwangyeon-haedoktang Pharmacopuncture Treatment for an Essential Tremor: Three Case Reports

  • The purpose of this study was to report the effect of a combination of Sa-am five-element acupuncture and eight-principle pharmacopuncture (EPP) for the treatment of an essential tremor (ET). This study reviewed the medical records treated at OO Korean medical hospital for ET by using diverse types of acupuncture without herbal medicine, other types of physical therapy, and western medication related ET or Parkinson's disease and was performed after the approval of the institutional review board (IRB). The three cases that were finally selected were then extracted and reviewed. The three cases that were finally selected involved three women in their 70s to 80s. The evaluation of the progress was made by using the numeric rating scale. A combined treatment, the method of liver excess (肝乘格), from among Sa-am five-element acupuncture, and Hwangyeonhaedoktang EPP at CV23 and CV17, was applied to all cases. In all three cases, the ET was improved, and recurred ETs improved with the same treatment. The results suggest that the combined treatment of Sa-am five-element acupuncture and Hwangyeon-haedoktang EPP may be effective for treating an ET, even though this conclusion is based on only three cases.

CO-SEPARATION OF Am AND RARE EARTH ELEMENTS FROM A HIGHLY ACIDIC RADWASTE SOLUTION BY A SOLVENT EXTRACTION WITH (DIMETHYLDIBUTYL TETRADECYLMALONAMIDE-DIHEXYLOCTANAMIDE)/N-DODECANE

  • This study was carried out to investigate the high-acidity co-separation of Am and RE from a simulated radwaste solution by a solvent extraction using a mixture of Dimethyldibutyltetradecylmalonamide (DMDBTDMA, as an extractant) and dihexyl octanamide (DHOA, as a phase modifier) diluted with n-dodecane (NDD). All the experiments were conducted as a batch type. First, the environmentally friendly DMDBTDMA and DHOA composed of only CHON atoms were self-synthesized. Then, the conditions for the prevention of a third phase, generated in the organic phase were examined. In addition, the effects of the concentration of nitric acid, DHOA, oxalic acid and $H_2O_2$ on the co-extraction of Am and RE were elucidated. Consequently, the optimum condition of (0.5M DMDBTDMA+0.5M DHOA)/NDD-0.3M $C_2H_2O_4-4.5M$ $HNO_3$ and O/A=2 was obtained through experimental work. Under this condition, the extraction yields were found to be about 80% for Am, more than 70% for RE such as La, Eu, Nd, Ce, etc., 3% for Cs and Sr, 69% for Fe and less than 11% for Mo and Ru. For the co-extraction of Am and RE, Fe should be removed in advance or prevented from a co-extraction with Am by controlling the different extraction rates of Am and Fe. About 95% of the Am and RE in the organic phase were stripped using a 0.5M $HNO_3$.

  • In this paper, we had designed and fabricated the integrated circuit of AM tuner system for car. The very important characteristics of this system need very good sensitivity, selectivity and very strong automatic gain control function. In this system we had designed, maximum sensitivity is about 21dBu, selectivity above 40dB, AGC range above 80dB, etc. We had fabricated this IC using SST standard linear process.

Physicochemical components of Astragalus membranaceus fermented with mushroom mycelia (담자균 균사체가 배양된 황기의 이화학적 성분분석)

  • This study analyzed the physicochemical characteristics of Astragalus membranaceus (AM) fermented with seven different mushroom mycelia. Physicochemical characteristics, such as contents of moisture, pH, total reducing sugars, free sugar, and isoflavonoid, were investigated. The moisture content was increased in most of the samples. The pH values of AM fermented with Phellinus linteus and Flammulina velutipes were increased, while the pH of other samples were similar to that of non-fermented AM. The reducing sugar content was in the range of 211.69~391.74 mg/100 g. The extraction yield using water was higher than that when extracted with 80% ethanol. The free sugar content was increased through fermentation with mushroom mycelia. However, the glucose contents of the 80% ethanol and water extracts were decreased. Finally, the calycosin and formononetin contents in 80% ethanol and water extracts of AM fermented with Phellinus linteus were 2,549.24 mg/g, and 827.66 mg/g for calycosin, and 1,366.69 mg/g and 221.28 mg/g for formononetin, respectively. These results suggest that fermentation with mushroom mycelia could be used to increase the bioactivity of AM. The mycelium-fermented AM might be a valuable source of functional material and edible resource for industry.

Ethanol Extract of Astragalus membranaceus Bunge Induces Aquaporin-3 Expression in HaCaT Cells (HaCaT Cell에서 황기 에탄올 추출물의 Aquaporin-3 발현 효과)

  • Astragalus membranaceus Bunge is used in herbal medicine in Eastern Asian countries including Korea. In this study, we assessed the effects of A. membranaceus extract (AM) on the aquaporin-3 (AQP3) protein expression in HaCaT cells. AM did not affect viability of HaCaT cells. AQP3 expression and cell migration seem to be maximal at $100{\mu}g/mL$ concentration. Epidermal growth factor receptor (EGFR) kinase inhibitor, PD153035, blocked AM-induced AQP3 expression and cell migration. In addition, an 80% ethanol extracts of herbal prescription, SinhyoTakleesan (ST), which is composed of A. membranaceus, Angelicae gigantis, Glycyrrhiza glabra Linne, and Lonicera japonica Flos also induced AQP3 expression at $20{\mu}g/mL$ in HaCaT cells. Collectively, these results suggest that AM induce AQP3 expression via EGFR pathway.
