Effects of Poly-Gamma Glutamate Contents Cheonggukjang on Osteoblast Differentiation (폴리감마글루탐산(PGA) 함유량이 증가된 청국장이 조골세포 분화에 미치는 영향)

  • Cheonggukjang (CKJ) is a Korean traditional food made of fermented soybeans. In comparison to normal intake of soybeans, Cheonggukjang has high digestibility with bioactive, antioxidant substances, and thrombolytic enzymes. Recent studies have reported anti-oxidant, anti-cancer, anti-inflammatory, anti-obesity activities as well as inhibitory activities against osteoporosis for CKJ. In this study, we identified the effects of CKJ on osteoblast differentiation by increasing the polyglutamic acid (PGA) content of CKJ. Alkaline phosphatase (ALP) activity and mineralization significantly increased in response to treatment with both natural CKJ (CKJ A) and PGA-increased CKJ (CKJ B). However, CKJ B exhibited higher ALP activity and mineralization than CKJ A. Real-time reverse transcription PCR demonstrated that mRNA expression of osteoblastic-associated genes such as type I collagen, alkaline phosphatase, osteocalcin, and osteopontin in C2C12 cells was significantly up-regulated by CKJ A or B treatment. These results indicate that treatment with CKJ has an anabolic effect on bone by increasing osteoblastic differentiation and ALP activity. Increasing PGA content in CKJ had a greater effect than CKJ A on up-regulation of osteoblastic gene expression in osteoblast cells.

Influences of Sodium Fluoride Contents on Hepatic Functional Enzyme Activities in Rats (Sodium Fluoride 함량이 흰쥐의 간 기능 효소활성에 미치는 영향)

  • The purpose of this study was to probe the influences of NaF oral administration on a dose-effect relationship between fluoride levels of serum enzyme activity such as alkaline phosphatase (ALP), aspartate aminotransferase (AST), alanine aminotransferase (ALT), and lactate dehydrogenase (LDH) in rats fed experimental diets for 5 weeks. All groups increased the activity of serum ALP, AST, ALT, and LDH levels with increasing NaF. In addition the fluoride levels of serum and organ tissues (liver, brain, heart, lung, kidney) in oral NaF groups (NF3~NF50) were significantly increased by adding sodium fluoride in comparison with normal diet group (ND) (p<0.05). These results, a high concentration of sodium fluoride was determined that the toxicity to various organ tissues.

Effects of Extracts from Sarcocarp, Peels, and Seeds of Avocado on Osteoblast Differentiation and Osteoclast Formation (아보카도 과육, 과피 및 씨 추출물이 조골세포 분화 및 파골세포 형성에 미치는 영향)

  • Avocado (Persea americana Mill., Family Lauraceae) is an important subtropical crop in the Americas where it has been cultivated for several thousand years. To investigate the bioactivities of avocado, which acts on bone formation, we prepared methanol extracts from the sarcocarp, peels, and seeds of avocado. The methanol extracts of peels and seeds showed higher bone-forming activity than avocado sarcocarp extracts accompanied by MC3T3-E1 osteoblast proliferation and alkaline phosphatase (ALP) activity. Additionally, the extracts of sarcocarp and peel from avocado also decreased tartrate-resistant acid phosphatase (TRAP) activity against differentiation of osteoclasts, derived from mouse bone marrow macrophages. The hexane fraction from avocado peels showed strong bone-forming activity accompanied by osteoblast proliferation and ALP activity (170.7${\pm}$8.4%), and the ethyl acetate fraction from avocado peel decreased TRAP activity (5.2${\pm}$0.3%) and differentiated osteoclasts at 50 ${\mu}g$/mL. Therefore, avocado is expected to be a natural source for developing medicinal agents to prevent bone-related diseases, such as osteoporosis, by increasing osteoblast differentiation and reducing osteoclast activity.

Effects of Sachunghwan Extract on Liver Damage in Thioacetamide Induced Rats (사청환(瀉靑丸)이 thiaocetamide에 의한 백서(白鼠) 간손상(肝損傷)에 미치는 영향)

  • Objectives The purpose of this study is to examine of the effect of Sachunghwan on the liver disease. Methods It was induced liver damage rats by thioacetamide and dosed the extract orally and measured the activity rate of GOT, GPT, ALP and LDH. Results 1. GOT activity was more decreased in the experimental goup than in the control goup. Group I showed continuous significance after 72 hour, Group II was significance after 96 hours. 2. GPT activity was more decreased in the experimental group than in the control group. Group I was effective after 72 hours, but group II was effective after 96 hours. 3. The significance of ALP activity in the Group I and group II was revealed after 72 hours. 4. The significance of LDH activity in the Group I and group II was revealed after 96 hours. Conclusions It was showded that Sachunghwan extract was effective on liver disease.

  • Fluoride is one of the most potent stimulators of bone formation in vivo. But its direct effects on osteoblast is not yet clear This study was to investigate the effects of Sodium fluoride on alkaline phosphatase(ALP) activity, cAMP formation responsive to parathormone(PTH) and type I $\alpha$ 2 collagen ribonucleic acid (mRNA) level in Murin osteoblast-like (MC3T3-E1) cells. The cells were cultured in $\alpha-Minimal$ essential medium $(\alpha-MEM)$ supplemente with $10\%$ fetal bovine serum (FBS) and then changed to $0.1\%$ FBS with various concentration of Sodium fluoride. The ALP activity was assayed by the method of Lowry with disodium phenyl phosphated as substrate. cAMP formation was measured by Radioimmuno Assay(RIA). Type I $\alpha$ 2 collagen ribonucleic acid(mRNA) expression was studied by Nothern blot analysis. The results were as follows: 1. cAMP level was increased by PTH in MC3T3-E1 cells. 2. Sodium fluoride showed the tendency of inhibitory effects on cAMP responsiveness to PTH in MC3T3-E1 cells. 3. Sodium fluoride increased ALP activity at cocentration of $2{\mu}M,\;4{\mu}M,\;and\;10{\mu}M$ significantly different from control at the 0.001 level. ALP activity revealed maximum value at $10{\mu}M$ in this study. 4. Nothern blot analysis of Sodium fluoride treated cells, using Type I $\alpha$ 2 collagen prove, revealed significant increase at $10{\mu}M$ in MC3T3-E1 cells.

Evaluation of Milk Trace Elements, Lactate Dehydrogenase, Alkaline Phosphatase and Aspartate Aminotransferase Activity of Subclinical Mastitis as and Indicator of Subclinical Mastitis in Riverine Buffalo (Bubalus bubalis)

  • Mastitis is a highly morbid disease that requires detection at the subclinical stage. Tropical countries like India mainly depend on milch buffaloes for milk. The present study was conducted to investigate whether the trace minerals viz. copper (Cu), iron (Fe), zinc (Zn), cobalt (Co) and manganese (Mn) and enzyme activity of lactate dehydrogenase (LDH), alkaline phosphatase (ALP) and aspartate aminotransferase (AST) in riverine buffalo milk can be used as an indicator of subclinical mastitis (SCM) with the aim of developing suitable diagnostic kit for SCM. Trace elements and enzyme activity in milk were estimated with Atomic absorption Spectrophotometer, GBC 932 plus and biochemical methods, respectively. Somatic cell count (SCC) was done microscopically. The cultural examination revealed Gram positive bacteria as the most prevalent etiological agent. A statistically significant (p<0.01) increase in SCC, Fe, Zn, Co and LDH occurred in SCM milk containing gram positive bacterial agents only. ALP was found to be elevated in milk infected by both gram positive and negative bacteria. The percent sensitivity, specificity and accuracy, predictive values and likelihood ratios were calculated taking bacterial culture examination and $SCC\geq2{\times}10^5$ cells/ml of milk as the benchmark. Only ALP and Zn, the former being superior, were found to be suitable for diagnosis of SCM irrespective of etiological agents. LDH, Co and Fe can be introduced in the screening programs where Gram positive bacteria are omnipresent. It is recommended that both ALP and Zn be measured together in milk to diagnose buffalo SCM, irrespective of etiology.

Changes of Biochemical Composition and Activity of Enzyme in the Serum Exposed to Carbon Tetrachloride in Mice (사염화탄소를 투여한 MICE 혈청내 효소 활성 및 생화학적 조성의 변화)

  • Carbon tetrachloride ($CCl_4$) has been used to treat a variety of parasitic infection in both large and small animals, including Fasciola hepatica. Recently, we can easily contest with carbon tetrachloride in air, food, water, rain and Industry area etc. and it is using in order to induce liver injury in laboratory. In this study. we investigated activitis of LDH, ALP, AST and ALT and amount of cholesterol, triacylglycerol, glucose and BUN in mice serum exposed to $CCl_4.$ The mice divided Into a Naive control(A), corn oil control(B) and experimental group(C, D). Naive control group(A) was given feed and water only. Corn oil control group(B) was given corn oil 1ml /100g of body weight(B.W). Experimental group(C) was given carbon tetrachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:20(V /V) ), Experimental group(D) was given carbon teterachloride 1ml /100g B.W. ($CCl_4$: corn oil=1:10(V /V) ). The results obtained were summarized as follows : 1) The body weight was declined after the l0th day In mice exposed to carbon tetra-chloride. 2) The total protein level in serum was significantly in mice exposed to carbon tetra-chloride($P{\leq}0.05$). The albumin and A /G ratio was decresed significantly in mice exposed to carbon tetrachloride($P{\leq}0.01$) 3) All of the activity of LDH, AST, ALT and ALP in mice serum exposed to carbon tetra chloride inclosed significantly activity of LDH ($P{\leq}0.05$), inclosed significantly activity of AST($P{\leq}0.05, \;p {\leq}0.01$), inclosed significantly activityof ALT 3nd ALP($P {\leq}0.05,\;p {\leq}0.01$). 4) The amount of cholesterol and triacylglycerol, lipid metabolite products in serum was inclosed in case of cholesterol but did not change in case of triacylglycerol.

  • Chicken calvarial bone is known to contain various cell types, but their exact composition is unknown. By characterizing the chicken calvarial bone biochemically, it can be used to study biochemical, histochemical actions of bone cells in general. Calvaria of 18-day-old white leg horn embryo was aseptically dissected and bone cell populations were isolated by sequential enzymatic digestion. Histochemical study for osteoclast-like bone cell. population was performed with tartrate resistant acid phosphatase(TRAP) stain and for osteoblast-like bone cell population, alkaline phosphatase(ALP) stain was performed. Biochemical study for osteoblast-like bone cell population was performed using alkaline phosphatase(ALP) assay. Following conclusions were obtained from this study. 1. TRAP positive multi and mononuclear cells were mostly observed in group I and II, indicating that osteoclast-like bone cell population is mostly found in these groups. 2. All the cultured groups showed almost equal ALP activities and were positive for ALP stain, indicating that osteoblast-like bone cell population is evenly dispersed in all culture groups. 3. Experimental group treated with $1,25(OH)_{2}D_3$ showed increase in ALP activity in contrast to the control group, confirming previous studies that $1,25(OH)_{2}D_3$ increases ALP activities in in vitro bone cultures. 4. Results from von Kossa's stain indicated that in vitro bone formation had occured after 3 weeks of culture with beta-glycero phosphate.

THE CHANGE OF EXTRACELLUAR ALKALINE PHOSPHATASE EXPRESSION IN HYPERTROPHIC SCAR IN RABBITS (가토의 비후성 반흔에서 세포외 알칼리성 인산효소 발현의 변화)

  • ALP (alkaline phosphatase) is a membrane-bound metalloenzyme that is expressed in osteoblasts, hepatocytes, lung, kidney, endothelial cells, leukocytes and other cells. Normal soft tissue and skin show little tissue nonspecific ALP (TN-AP), However, scar tissue contains high levels of TN-AP activity, and in fact, TN-AP is expressed intensely in regenerating connective tissue after the wounding. The purpose of this study was to evaluate the change of ALP expression in hypertrophic scar model in rabbits and the effect of triamcinonolone on ALP expression. Adult male New Zealand white rabbits, weighing about 2.5 kg, were used. After full-thickeness wounding over the ventral surface of each ear, either saline (control ear) or triamcinolone (contralateral ear) was injected on day 16. Rabbits were sacrificed on day 3, 7, 15, 17, 19, 23, and the specimens were retrieved en bloc. Histologic and immunohistochemical examinations of tissue samples were done. The results obtained were as follows: On day 3, ALP reaction was observed on fibroblasts and inflammatory cells in wound margin. On day 7, ALP reaction was more intense than day p in capillaries, inflammtory cells, and fibroblasts behind newly formed epithelium. On day 15, ALP reaction was lessened in both groups and appeared mainly in subepidermal capillary network, Since day 17, ALP reaction was lessened in both groups and weaker in triamcinolone-injected group than in saline-injected group. These results suggest that ALP reaction isn't increased in triamcinolone-injected scar and triamcinolone reduces scar not by increasing TN-AP expression but other mechanism.

Effect of Fermented Benincasa hispida cong. Extract on Promotion of Osteoblast Differentiation and Inhibition of Osteoclast Generation (동과 발효물의 조골세포 분화 촉진 및 파골세포 생성 억제 효과)

  • The bones of the human body support the structures of the body and provide protection for a person's internal organs. Bone metabolic diseases are on the rise due to a significant increase in life expectancy over a short period of time. Therefore, we investigated the osteoblast differentiation promoting and osteoclastogenesis inhibitory activities of fermented Benincasa hispida cong. (HR1901-BS, HR1901-BSaf). We evaluated the alkaline phosphatase (ALP) activity of MC3T3-E1 mouse calvarial-derived osteoblasts. We also evaluated expression of ALP, osteocalcin (OCN), and runt-related transcription factor 2 (Runx2), which regulate osteoblast differentiation. To assess effects on osteoclast formation, tartrate-resistant acid phosphatase (TRAP) activity in RAW264.7 cells was analyzed. ALP activity increased by 121-136% and 140-156%, respectively in the presence of HR1901-BS and HR1901-BSaf. Expression of osteoblast differentiation factor also increased significantly. We also confirmed that HR1901-BS and HR1901-BSaf decreased TRAP activity in osteoclasts by 35-47% and 23-39%, respectively. Our results showed that fermented Benincasa hispida cong. (HR1901-BS, HR1901-BSaf) increase bone mineralization and osteoblast differentiation activity in MC3T3-E1 cells, and inhibit bone resorption activity in RAW264.7 cells. In conclusion, fermented Benincasa hispida cong. (HR1901-BS, HR1901-BSaf) can be used as an effective natural resource for preventing and treating bone-related diseases.
