Purpose: We aimed to investigate the effect of combined various microgrooves and thermal oxidation on the titanium (Ti) and to evaluate various in vitro responses of human periodontal ligament cells (PLCs). Materials and methods: Grade II titanium disks were fabricated. Microgrooves were applied on titanium discs to have $0/0{{\mu}m}$, $15/3.5{{\mu}m}$, $30/10{{\mu}m}$, and $60/10{{\mu}m}$ of respective width/depth by photolithography. Thermal oxidation was performed on the microgrooves of Ti substrata for 3 h at $700^{\circ}C$ in air. The experiments were divided into 3 groups: control group (ST), thermal oxidation group (ST/TO), and combined microgrooves and thermal oxidation group (Gr15-TO, Gr30-TO, Gr60-TO). Surface characterization was performed by field-emission scanning microscopy. Cell adhesion, osteoblastic differentiation, and mineralization were analyzed using the bromodeoxyurdine (BrdU), Alkaline phosphatase (ALP) activity, and extracellular calcium deposition assays, respectively. Statistical analysis was performed using the oneway analysis of variance and Pearson's bivariate correlation analysis (SPSS Version 17.0). Results: In general, the combined microgrooves and thermal oxidation group (Gr15-TO, Gr30-TO, Gr60-TO) showed significantly higher levels compared with the control (ST) or thermal oxidation (ST-TO) groups in the BrdU expression, ALP activity, and extracellular calcium deposition. Gr60-TO group induced highest levels of cell adhesion and osteoblastic differentiation. Conclusion: Within the limitation of this study, we conclude that the Ti surface treatment using combined microgrooves and thermal oxidation is highly effective in inducing the cell adhesion andosteoblastic differentiation. The propose surface is also expected to be effective in inducing rapid and strong osseointegration of Ti oral implants.

For successful osteogenesis around the implants, interaction between implant surface and surrounding tissue is important. Biomechanical bonding and biochemical bonding are considered to influence the response of adherent cells. But the focus has shifted surface chemistry. The purpose of this study is to evaluate the MC3T3-E1 osteoblast like cell responses of magnesium (Mg) ion implanted titanium surface produced using a plasma source ion implantation method. Commercially pure titanium disc was used as substrates. The discs were prepared to produce four different surface, A: Machine turned surface, B: Mg implanted surface, C: sandblasted surface, D: sandblasted and Mg implanted surface. MC3T3 El osteoblastic like cells were cultured on the disc specimens. Cell adhesion, proliferation, differentiation, and synthesis of extracellular matrix were evaluated. The cell adhesion morphology was evaluated by SEM. RT PCR assay was used for assessment of cell adhesion, proliferation and differentiation. ALP activity was measured for cell differentiation. The results of this study were as follows: 1. SEM showed that cell on Mg ion groups was more proliferative than that of non Mg ion groups. On the machine turned surface, cell showed some degree of contact guidance in aligning with the machining grooves. 2. In RT PCR analysis, osteonectin and c-fos mRNA were more expressed on sandblasted and Mg ion implanted group. 3. ALP activity was not significantly different among all groups. Within the limitations of this study, the following conclusions were drawn: It might indicate Mg ion implanted titanium surface induce better bone response than non Mg ion groups.

The aim of this study was to evaluate the effects of Welsh onion administration on $CCl_4$-induced hepatic injury in rats. The increasing rate of the body and organ weight was not significantly different by the ad-ministration of the Welsh onion. Welsh onion (2, 4, $10\%$ w/w) was given for 4 weeks with the injections of $CCl_4$. Total serum lipid was significantly decreased in all treatment groups compared to $CCl_4$ only treatment group (p<0.05). The Welsh onion from winter season was more protective effect by lowering the serum levels of transminase (SGOT and SGPT) compared with others, the levels of transminase - phosphatase (ALP) in serum. The Welsh onion from winter at a dose of $4\%,\;10\%$ (w/w) showed significantly hepatoprotective activity which was comparable to that $CCl_4$-induced hepatic damage. Histological evaluation showed that Welsh onion partially prevented $CCl_4$-induced inflammation, necrosis and vacuolation. Pretreatment of Welsh onion reduced extent of the necrosis found 24 hr after the intraperitoneal administration of $CCl_4$. The present study shows the liver protective action of the Welsh onion against experimentally induced liver damage in rats. This suggests that the Welsh onion may be used as an effective hepatoprotective agent.
This study was carried out to investigate the effects of powders of autoclaved soy flour and doenjang fermented using Bacillus subtilis DJI on lipid profiles and antioxidative activities of rats fed a high cholesterol diet. Sprague-Dawley male rats weighing 200~210 g were divided into four groups: normal diet group (N), high cholesterol diet group (HC), autoclaved soy flour and high cholesterol diet group (SHC), and doenjang and high cholesterol diet group (DHC). The serum ALT, AST and ALP activities of the SHC and DHC groups were lower than those of the HC group, but exerted no significant change on serum LDH activity. Serum triglyceride, total cholesterol and LDL-cholesterol levels were markedly decreased by autoclaved soy flour and doenjang administration, while the serum HDL-cholesterol level was higher in groups given autoclaved soy flour and doenjang administration. The GSH-Px and catalase activities in liver elevated by a high cholesterol diet were significantly decreased by autoclaved soy flour and doenjang administration (p<0.05). Liver GSH levels of the SHC and DHC groups were significantly decreased compared to the HC group (p<0.05). Liver TBARS level was significantly decreased in the DHC group fed with doenjang powder compared with those of the HC group (p<0.05). These results suggest that soy flour and doenjang may reduce levels of serum cholesterol and prevent oxidative stress by stimulating antioxidative systems in rats fed a high cholesterol diet.
For orthodontic tooth movement, optimal orthodontic force should be maintained without periodontal breakdown and alveolar bone should be remodeled physiologically Therefore, To obtain proper occlusion through tooth movement within alveolar bone, we should know the biomechanics of teeth and supporting 4issues. The present study was performed to observe histologic changes of periodontal tissue immediately after application of orthodontic force and during the retention period in growing young adult dogs. In this study, experimental group contained between mandibular left canine and 1st molar and control group contained contralateral teeth of same animal. The .018'x.022' stainless steel closed coil spring(Dentaurum Co.) was ligated on the experimental teeth at initial 200gm-force from mandibular canine to 1st molar The animals(4 to 6 months aged young adult dogs) were sacrificed on 0, 14, 28 days after the finish of appliance activation, and then tissue samples were divided into hematoxylin-eosin(HE) staining section, ground section, alkaline phosphatase(ALP) staining section, and tartrate-resistant acid phosphatase(TRAP) staining section. Thereafter, the preparations were examined under light microscopy The following results were obtained: 1. Immediately after the finish of appliance activation, the periodontal space was increased in tension side, but decreased in pressure side compared to that of control. The hyalinized zone was also observed in the periodontium. 2. After the 14-day retention, peridontal space was decreased in tension side and slightly increased in pressure side compared to that of immediately after the finish of appliance activation. The hyalinized zone was repaired and a few osteoblasts showing slightly new bone formation were seen. Osteoblasts were scarcely observed along the alveolar bone. 3. Aftter the 28-day retention, the periodontal fibers are normally repaired. A lot of TRAP(+) osteoclasts md increased alveolar bone resorption were observed in pressure side, and AP(+) osteoblast and increased new bone formation were observed in tension side.
This study is aimed to evaluate the protective effect of Cordycepin-enriched Cordyceps militaris (CM${\alpha}$) strain on orotic acid (OA)-induced fatty liver in rats. OA treatment induced the retardation of body weight gain and enlargement of the liver. The activities of liver marker enzymes, alanine aminotransferase (ALT), aspartate aminotransferase (AST), lactate dehydrogenase (LDH), Alk. phosphatase and Cholinesterase were increased when treated with OA, but these parameters were significantly decreased in the CM${\alpha}$ group. The current study observed significant elevations of thiobarbituric acid-reactive substance (TBARS) levels. However, among the OA groups, the CM${\alpha}$ group showed significantly low TBARS levels of hepatic homogenate. The OA group resulted in a significant decrease in the levels of plasma and hepatic glutathione, but these reductions were significantly increased in the CM${\alpha}$ group. These effects were more pronounced in the CM${\alpha}$ group than in the PJ or CM groups in Orotic acid treated rats. Accordingly, Cordycepin-enriched Cordyceps militaris (CM${\alpha}$) may be an ideal candidate for hepatoprotective effects in animal models.

This study investigated the effects of green tea on the disorders of lipid metabolism, oxidative system and hepatic functions induced by 2,3,7,8-tetrachlorodibenzo-p-dioxin (TCDD), using adult male rats (SD) for 3 weeks. These 36 animals were divided into four groups. TCDD ($50{\mu}g/kg$ BW) was intraperitoneally injected at the beginning of experiment. Green tea powder was added 1% or 3% levels in basal diets respectively. Relative weights of thymus were decreased about one-third of control group, but those of liver, brain and testis were significantly increased in rats treated TCDD. Neutrophill% and lymphocyte% by TCDD treatment was improved by green tea diets. In liver functional enzyme, elevation of glutamic oxaloacetic transaminase (GOT) and alkaline phosphatase (ALP) activities due to TCDD treatment was lowered by green tea diets. The concentrations of serum and liver lipids were significantly increased by TCDD treatment, however, those of serum and liver triglyceride tended to decrease by green tea diets. Fecal lipid excretion was increased in rats fed green tea diets. Especially, fecal total cholesterol level was significantly elevated by 3% green tea diets. The activities of superoxide dismutase (SOD) were increased in rats fed 3% green tea diets. Increment of benzphetamine N-demethylase (BPND) activity by TCDD treatment was declined by 1% green tea diets. These results indicate that green tea can exert improving effects on liver lipid accumulation and unfavorable hepatic functions, and elevate antioxidation.
A strain producing ${\alpha}$-galactosidase and ${\beta}$-glucosidase was isolated from Kimchi. The isolated strain was identified as Weissella cibaria by 16S rDNA analysis and designated as Weissella cibaria K-M1-4. The enzyme activity of ${\alpha}$-galactosidase and ${\beta}$-glucosidase reached the maximum in the soy medium at $37^{\circ}C$ for 24 hr. The enzymes were purified by ethanol fractionation, DEAE sepharose fast flow, and sephacryl S-100HR column chromatography. ${\alpha}$-Galactosidase specific activity was shown by 576 Units/mg protein and the yield was 3.5% of the total activity of crude extracts. ${\beta}$-glucosidase specific activity was shown by 480 Units/mg protein and the yield was 2.9% of the total activity of crude extracts. The optimum temperature for ${\alpha}$-galactosidase was $60^{\circ}C$ and 43% of its original activity remained when it was treated at $80^{\circ}C$ for 30 min. For ${\alpha}$-galactosidase shows the optimum pH of 8.0 and is fairly stable between pH5.0 and pH9.0. The enzyme activity was increased in the presence of $Fe^{2+}$ and $Cu^{2+}$. The value of Km and Vmax for the enzyme were 0.98 mM and $1.81{\mu}$mole/min, respectively. The ${\beta}$-glucosidase has the optimum temperature of $50^{\circ}C$ and 46% of its original activity remained when it was treated at $80^{\circ}C$ for 30min. Its optimum pH of 7.0 and is fairly stable between pH5.0 and pH9.0. The enzyme activity was increased in the presence of $Fe^{2+},\;Co^{2+}$ and $Cu^{2+}$. The value of Km and Vmax for the enzyme were 1.24 mM and $6.81{\mu}$mole/min, respectively.

This study was conducted to investigate the effects of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts on plasma glucose and biomarkers in the streptozotocin (STZ)-induced diabetic rats. Male Wistar rats were divided into normal and diabetic groups. The diabetic groups subdivided into the control group (DM), Aralia elata (DM-AE), Acanthopanacis cortex (DM-AC) and Ulmus davidiana (DM-UD). The extracts were supplemented in diet base on 11.42 g of raw materials/㎏ diet for 7 weeks. The diabetes was induced by injecting STZ (55 ㎎/㎏ B.W., i.p.) once 2 weeks before sacrifying. Plasma glucose level was significantly higher in the DM group than in the normal group, whereas insulin and C-peptide concentrations were significantly lowered in the DM groups compared to the normal group. These parameters were normalized in the DM-AE, DM-AC and DM-UD supplemented groups. Plasma albumin content was significantly lowered in the DM group compared to the normal group, yet it was significantly higher in the DM-AE group than in the DM group. Bilirubin and creatinine contents were elevated in the DM group, while the supplementation of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts ameliorate the change of these contents in STZ-induced diabetic rats. Plasma AST, ALT, ALP and LDH activities were significantly higher in the DM group than in the normal groups. The supplementation of Araliaceae family water extracts significantly lowered these parameters compared to the DM group. Accordingly, these results indicate that Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts would seem to improve the glucose and biomarker in STZ-induced diabetic rats.

In this study, we investigated the hepatoprotective effects of garlic extracts fermented with Lactobacillus plantarum on alcohol-induced fatty liver damage in C57BL/6J mice. The weight of liver tissue of the lactic acid-fermented garlic extract (LAFGE) diet groups decreased in a dose-dependent manner compared to that of the normal diet group. The activities of typical serum enzymes such as ALT, AST, and ALP were low in the LAFGE 200 mg/kg administered group. The LAFGE 200 mg/kg administered group significantly decreased values of TBIL and DBIL, which are an important index of liver damage. LAFGE also ameliorated alcohol-induced hepatic lipid accumulation in histological analysis dose-dependently. In addition, total GSH and reduced GSH levels in LAFGE-treated groups were gradually recovered up to normal levels. The present study revealed that LAFGE improved unique tastes and odors of raw garlic through lactic acid fermentation technology and thus could be a commercially potential material for hepatoprotective functional foods against alcoholic fatty liver.
