베타-글루칸의 고지방 식이 유도 비만쥐에서 체지방 및 혈청지질 개선효과 (The Improvement Effects of β-Glucan on Adiposity and Serum Lipids Levels in High Fat Diet-Induced Obese Rats)

  • 본 연구는 미생물 유래 베타-글루칸의 항비만 효과를 검증하기 위하여 동물실험을 통해 식이 베타-글루칸이 체지방 형성과 혈중 지질에 미치는 영향을 조사하였다. Sprague-Dawley종 흰쥐를 6주간 고지방 식이로 사육하여 비만을 유도한 후 베타-글루칸을 각각 식이 무게의 0%(고지방 대조군), 0.1%, 0.5%로 함유한 식이로 5주간 사육하였다. 정상 대조군은 AIN-76A 식이로 사육하였다. 고지방식이군에서 단위체중당 체지방이 많았으며, 0.1% 베타-글루칸 공급은 고지방 식이에 의해 유도된 복막지방, 내장지방 축적을 각각 16%와 19% 억제하였고, 혈중 렙틴을 28% 감소시켰다(P<0.05). 정상 대조군과 비교하여 고지방 대조군의 평균 지방세포 크기는 1.7배 높았으며, 베타-글루칸 섭취군에서는 감소하여 정상대조군 수준으로 낮아졌다(P<0.05). 0.1% 베타-글루칸 섭취에 의해 혈청 유리지방산 농도는 19% 감소하였고, 반면 HDL 콜레스테롤 50% 증가하였다(P<0.05). 결론적으로 베타-글루칸 공급은 렙틴과 같은 호르몬 분비와 지단백질 대사 변화로 지질 대사 개선을 유도하고 체지방 형성과 축적을 억제하는 것으로 보인다. 본 연구 결과 베타-글루칸은 고지방 식사에 기인하는 비만과 이상지혈증 개선에 도움이 되는 기능성 식품 소재로 사용될 수 있을 것으로 사료된다.

  • The effects of dietary levan, high-molecular-weight $\beta$-2,6-linked fructose polymer, on adiposity, serum leptin and UCP expression in rats fed high fat diet were studied. The adipose tissue hormone, leptin has been proposed to be involved in the regulation of food intake and energy expenditure. Uncoupling protein (UCP), a mitochondrial protein that uncouples the respiratory chain from oxidative phosphorylation, generates heat instead of ATP, thereby increase energy expenditure. To determine whether the dietary levan may have the anti-obesity effect, 4 wk old Sprague Dawley male rats fed high fat diet for 6 wks to induce obesity, and subsequently fed one of three diets for further 6 wks: 1) high fat (40% of calories) diet without levan 2) with 3% (w/w) levan 3) with 5% levan. For the comparison, control group fed AIN-76A diet. Visceral and peritoneal fat masses were lower in high fat diet with levan groups compared to high fat diet group. The effect of levan was dose-dependent. Adipocyte size was significantly reduced in the levan diet groups compared to the no levan diet group. Serum cholesterol level was not affected by levan containing diet, while the serum HDL cholesterol level was higher in leven diet groups. In addition, serum triglyceride level was markedly reduced by levan containing diet, thus lower than that of control group. Serum leptin was reduced by levan containing diet and lower in 5% levan group compared to 3% levan group (p < 0.001), as a result, serum leptin and insulin levels of 5% levan group were reduced to level of control group. Futhermore, the serum leptin level reflected the adiposity. The expression of UCP 1, and UCP 2 in brown adipose tissue was up-regulated by levan containing diet. In conclusions, levan containing diet reduced adiposity and serum triglyceride but increased UCP expression in the obese rats fed high fat diet. (Korean J Nutrition 35(9) : 903~911, 2002)

Effect of Diets Supplemented with Pharbitis Seed Powder on Serum and Hepatic Lipid Levels, and Enzyme Activities of Rats Administered with Ethanol Chronically

  • The levels of $\gamma$-aminobutyric acid (GAGA) have been analyzed from pharbitis seeds by an AccQ-Tag amino acid analysis procedure. The GABA level of the pharbitis seeds was 125 nmole per gram fresh weight. To investigate the effects of pharbitis seed diets on serum and hepatic lipid levels, as well as enzyme activities of rats administered with ethanol chronically, Sprague-Dawley male rats were fed with either a AIN-76 diet (control), a control diet plus ethanol, a control plus pharbitis seed diet, or a control plus pharbitis seed diet plus ethanol for 30 days. Pharbitis seed diets decreased the serum total cholesterol, triglyceride, LDL-cholesterol, and $\gamma$-GTP levels that were increased by the chronic ethanol administration. In addition, pharbitis seed diets decreased the liver triglyceride and total lipid levels that were increased by the ethanol administration. However, ethanol metabolism was not retarded by the pharbitis seed supplemented diets. The present Endings, plus previous data showing the differences in the effects of cabbage diets having a high or a low level of GABA on the lipid levels and the enzyme activities of rats (Cha and Oh [2000] J. Korean Soc. Food Sci. Nutr. 29, 500-505), raise the possibility that GABA in plants could have a nutraceutical role in the recovery of chronic alcohol-related diseases.

식이 칼슘량이 난소 절제한 흰쥐에서 골대사 지료에 미치는 영향 (The Effect of Dietary Calcium Level on Biochemical Variables of Bone Metabolism in Ovariectomized Female Rats)

  • This study was done to evaluate the effectiveness of dietary calcium level(a diet which met 100% or twice the calcium level in AIN-76 diet) on preventing bone loss in ovariectomized rats. Forty female Sprauge-Dawley rats(body weight 200$\pm$5g) were divided into two groups. One group were ovariectomized(Ovx) while the others received sham operation(Sham). Thereafter, each rat group was further divided into normal calcium diet(NCD, 0.52%) and high calcium diet(HCD, 1.04%) sub-groups. All rats were fed on experimental diet and deionized water ad libitum for 8 weeks. Urinary pyridinoline & creatinine and serum estradiol, luteinizing hormone, calcium, phosphate, total protein, albumin, alkaline phosphatase and osteocalcin were determined. There were no significant differences in serum calcium. total protein and albumin in the two groups(Ovx vs Sham) of rats. Ovariectomized rats had significantly lower estradiol than sham operated rats. There was a highly significant correlation between total bone mineral density(TBMD) and overall level of esteradiol(r=0.59, p<0.05). Total bone mineral density did not correlate significantly with ALP or osteocalcin, although a negative trend was evident. However, the rats fed high calcium diet had a lower crosslinks value and osteocalcine than the rats fed normal calcium diet. An increased rate of bone turnover is usually associated with a decrease in bone mass bexause bone formation at each remodeling site is never as great as resorption. Ovariectomized rats fed high calcium diet had a lower crosslink value and osteocalcin; it means high cacium diet decreased bone turnover rate. The findings from the present study demonstrated that bone loss due to ovarian hormonal deficiency can be partially prevented by a high calcium diet. Futhermore, these findings support the strategy of the use of a high calcium diet in the prevention of estrogen depletion bone loss (postmenopausal osteoporosis).

Suppressed Fat Accumulation in Rats Fed a Histidine-Enriched Diet

  • The effect on body fat accumulation on male Wistar rats undergoing continuous feeding with a histidine-enriched diet was investigated. Five-week-age rats were assigned to two groups and were fed either the control diet (purified diet AIN-$76^{TM}$) or the histidine-enriched diet containing 3% histidine for 28 days. It was observed that both adipose tissue masses in retroperitoneal and epididymal areas of rats fed histidine-enriched diet significantly decreased (p<0.05) compared to those of control rats, while there was no significant difference in the food efficiency ratio between them. The blood levels of histidine derivatives of 3-methylhistidine and carnosine were significantly (p<0.05) increased in the rats fed a histidine-enriched diet, whereas there were no significant different between the histidine-enriched diet and control groups in the general amino acid distribution. Our results demonstrate that a histidine-enriched diet suppresses body fat accumulation in rats.

흰쥐에서 고지방식이가 혈중 렙틴 및 인슐린과 갈색지방조직의 UCP 1 발현에 미치는 영향 (Effects of High Fat Diet on Serum Leptin and Insulin Level and Brown Adipose Tissue UCP 1 Expression in Rats)

  • The adipose tissue hormone leptin has been proposed to be involved in the regulation of flood intake and energy expenditure via thermogenesis by uncoupling protein(UCP) in brown adipose tissue(BAT). The objective of the study was to examine the effects of high fat diet on the serum leptin levels, BAT UCPl expression and the body fat mass in rats after weaning. During experimental period of 12 weeks, 4 male Sprague-Dawley rats were killed for the baseline experiment at 4 weeks of age while the remaining rats were fed the two different diets: the control diet AIN-76A(n = 20), high fat(beef tallow) diet(n = 20) ad libitum, which provided 11.7% or 40% of calories as fat, respectively. At 16 weeks of age, the increase in the food efficiency ratio(FER) was related to fat mass in rats on high fat diet. Serum leptin level was increased by age and dietary high fat. There was no difference in serum insulin level between groups until 10 weeks of age, but rats fed high fat diet for 12 weeks showed hyperinsulinemia. The amount of body fat pads was increased significantly in high fat group compared to normal diet group. Visceral fat mass affected acutely by high fat diet, as a result, it was higher in rats fed high fat diet for 2 weeks than normal diet. At 16 weeks of age, BAT and visceral fat mass were significantly high in high fat group. Also, the serum leptin levels reflected the amount of body fat mass. BAT UCPI mRNA expression increased with age and dietary high fat. This study demonstrates that dietary high fat increased serum leptin levels, BAT UCPI expression and body fat mass. Futhermore, in rats fed high fat diets, the increases in leptin and UCPI expression counteracts only in part the excess adiposity and obesity.

곽향정기산가미방(藿香正氣散加味方)이 고지방식이(高脂肪食餌)로 유식(誘導)된 흰쥐의 체지방에 미치는 영향(影響) (Effects of Kwakhyangjungkisangamibang Extract on the Adipose Tissues Induced by a High Fat Diet in Rats)

  • Objetives This experimental study is designed to investigate the effects of kwakhyangjungkisangamibang extract on the changes of adipose tissues, serum insulin and leptin levels in rats induced by a high fat diet. The leptin has been proposed to be involved in the role of food intake and energy expenditure. Methods During the 8 weeks of experimental period, male Sprague-Dawley rats were fed on the 2 different diets: the normal diet AIN-76A and high fat(beef tallow) diet ad libitum, of which 40% of the calories intake was fat. 9 rats in the control and high fat diet group were killed for the baseline experiment at 4 weeks of age. The high fat diet group was divided into three groups: control group, experimental group I(high fat diet plus kwakhyangjungkisangamibang extract), and experimental group II(high fat diet plus mahuang). The body weight, hypodermal fat tissue, serum lipid profile, insulin and leptin levels were measured after the administration of the high fat diet and extract. Results 1. The body weight was decreased in experimental group II, but there was not a significance difference compared to the control group. 2. The peritoneal, visceral, and hypodermal adipose tissue was decreased in experimental group I, but there was not a significance difference compared to the control group. 3. The lipid profile was decreased in experimental group I, but not in group II. 4. The insulin concentration was increased in experimental group I and II, but there was not a significance difference compared to the control group. 5. The leptin level was increased significantly in experimental group I(p<0.05) and II(p<0.01) compared to the control group. Conclusion The results of this study indicate that kwakhyangjungkisangamibang has an effect on the regulation of obesity.

Anti-Obesity and Hypolipidemic Effects of Dietary Levan in High Fat Diet-Induced Obese Rats

  • We found previously that dietary high fat caused obesity, and levan supplementation to the regular diet reduced adiposity and serum lipids. In the present study, we examined the effects of levan [high-molecular-mass $\beta$-(2,6)-linked fructose polymer] supplement on the development of obesity and lipid metabolism in rats fed with high-fat diet. Thus, to determine whether the dietary levan may have the anti-obesity and hypolipidemic effects, 4-wk-old Sprague Dawley male rats were fed with high-fat diet for 6 wk to induce obesity, and subsequently fed with 0, 1, 5, or 10% levan supplemented high-fat diets (w/w) for another 4 wk. For the comparison, a normal control group was fed with AIN-76A diet. Supplementation with levan resulted in a significant reduction of high-fat-induced body weight gain, white fat (i.e., epididymal, visceral, and peritoneal fat) development, adipocyte hypertrophy, and the development of hyperinsulinemia and hyperlipidemia in a dose-dependent manner. Serum triglyceride and free fatty acid levels were greatly reduced by levan supplementation. Serum total cholesterol level was reduced, whereas the HDL cholesterol level was increased by dietary levan. The expression of uncoupling protein (UCP) was increased by dietary high fat, and was further induced by levan supplementation. The mRNA level of UCP1, 2, and 3 in brown adipose tissue (BAT) and UCP3 in skeletal muscle was upregulated in rats fed with dietary levan. In conclusion, upregulated UCP mRNA expression may contribute to suppression of development of obesity through increased energy expenditure. The present results suggest that levan supplementation to the diet is beneficial in suppressing diet-induced obesity and hyperlipidemia.

주요 Phytoestrogen들이 제2형 당뇨 마우스의 당질대사에 미치는 효과 (Effects of Phytoestrogens on Glucose Metabolism in C57BL/KsOlaHsd-db/db Mice)

  • 본 실험은 주요 phytoestrogen에 속하는 genistein, coumestrol, enterolactone의 식이보충이 제2형 당뇨동물모델에서 당질대사 개선에 미치는 효과를 알아보고자 C57BL/KsOlaHsd-db/db 마우스를 이용하여 내당능, 당화헤모글로빈 농도, 당대사 관련 효소활성, 조직 중 글리코겐과 최종당화산물 수준 등을 측정하였다. 그 결과, phytoestrogen의 보충(3.75 mg/100 g diet)이 당뇨동물의 체중변화, 식이 및 수분 섭취량 그리고 장기무게에는 유의적인 영향을 미치지 않았으나, 모든 phytoestrogen의 보충은 당뇨동물의 공복 혈당, 경구 내당능 검사 시 혈당반응곡선 아래면적 및 혈중 HbA1c 수준을 유의적으로 낮추었다. 또한 모든 phytoestrogen의 보충은 당뇨동물의 혈장 글루카곤 수준을 유의적으로 낮추었으며, coumestrol과 enterolactone의 보충은 간 조직 중 글리코겐 수준을 유의적으로 증가시켰다. 이상의 결과들은 genistein, coumestrol, enterolactone의 3종 phytoestrogen의 식이보충이 제2형 당뇨동물에서 내당능을 개선시킬 수 있음을 시사하였으나 그 기전에 대해서는 향후 추가 연구가 필요하다고 본다.

한국산 홍화씨분말 보충식이의 급여가 골절된 흰쥐의 골대사지표에 미치는 영향 (The Effects of Korean Safflower(Carthamus tinctorious L.) Seed Powder Supplementation Diet on Bone Metabolism Indices in Rats during the Recovery of Rib Fracture)

  • This study was carried out to investigate the effects of Korean safflower(Carthamus. tinctorious L.) seed powder supplementation on bone metabolism during the recovery of rib-fracture induced by surgical operation in rats. Male Sprague-Dawley rats of 10 weeks old, weighing 370$\pm$5g, were divided into two groups after arrival : the control group(C group, AIN-76 semifurified diet) and safflower seed group(S group, AIN-76 semifurified diet+10% safflower seed powder). They were fed experimental diets for 12 days before the rib fracture operation and for 30 more days after the operation. A number 9 rib was fractured surgically and sham-operation was also performed. After the rib-fracture operation, rats were sacrificed on the 8th, 11th, 16th, 21st, and 30th day, respectively. The body weights were decreased on the 8th day after operation in both groups but recovered gradually thereafter. The fractured sections of the S group were more strongly adhered and more rapidly repaired than those of C group. Effects of safflower seed powder supplementation on indices of bone metabolism during the repairing of rib-fracture can be summarized as following. There were no differences between C and S group. In the levels of PTH and calcitonin, which regulate the concentration of plasma calcium. However, PTH level on the 8th day was significantly higher than that on the 16th day in control group. There were also no differences between groups or within groups in the levels of calcitonin. Osteocalcin levels, one of the bone-formation indices, were similar in two groups. Activities of total-ALP in plasma were highest at the 8th day in both groups and decreased thereafter, except on the 30th day in control group. The activities of bone-specific ALP that is directly related to bone calcification, showed higher levels in the S group than in the C group throughout the experimental period. They were significantly increased in the S group on the 11th, 16th, and 21st days. In comparisons of plasma calcium and phosphate levels, Ca levels were tended to be lower in the S group than in the C group and P levels showed the opposite trend. It seems that the fractured ribs were more rapidly repaired in rats supplemented with safflower seed powder than in control rats, possibly due to stimulation of bone calcification from increased activity of bone-specific ALP.
