The neuroprotective effect of Acori graminei rhizoma extract against cerebral ischemia in rats (석창포(石菖蒲)가 뇌허혈(腦虛血)을 유발(誘發)시킨 백서(白鼠)에서의 뇌신경보호효과(腦神經保護效果))

  • Object: Acori graminei rhizoma(AGR) extract is clinically used to treat the cerebral ischemia in Korea. The present study was undertaken to study the neuroprotective effect of AGR extract in middle cerebral artery occlusion(MCAO) rats. Methods: Changes of extracellular levels of glutamate, aspartate, GABA, glycine, taurine, tyrosine, alanine in striatum were collected at 20 minutes interval by in vivo microdialysis and then analyzed by HPLC(high performance liquid chromatography) in rats subjected to permanent focal cerebral ischemia induced by 2 hours of MCAO. AGR extract was orally administrated before MCAO. Different animals were used for measurement of cerebral infarction volume induced by 24 hours of MCAO with TTC staining and image analysis. Result : The infarction volume was decreased and focal cerebral ischemia - induced increase of extracellular glutamate, aspartate, and tyrosine were inhibited after the treatment of AGR extract. On the other hand, the increase of glycine and alanine not but GABA and taurine were enhenced after the treatment of AGR extract. Conclusion: These results suggest that AGR extract can playa role in protecting against cerebral ischemia by regulating extracellular levels of excitatory and inhibitory amino acid neurotransmitters.

Significance of albumin to globulin ratio as a predictor of febrile urinary tract infection after ureteroscopic lithotripsy

  • Background: We aimed to analyze the effectiveness of albumin to globulin ratio (AGR) in predicting postoperative febrile urinary tract infection (fUTI) after ureteroscopic lithotripsy (URS) and retrograde intrarenal surgery (RIRS). Methods: From January 2013 to May 2018, 332 patients underwent URS and RIRS. The rate of postoperative fUTI and risk factors for postoperative fUTI were analyzed using logistic regression. Patients were divided into postoperative fUTI and non-postoperative fUTI (non-fUTI) groups. AGR with other demographic and perioperative data were compared between the two groups to predict the development of fUTI after URS. Results: Of the 332 patients, postoperative fUTI occurred in 41 (12.3%). Preoperative pyuria, microscopic hematuria, diabetes mellitus, hypoalbuminemia, and hyperglobulinemia were more prevalent in the fUTI group. Patients in the fUTI group had larger stone size, lower preoperative AGR, longer operation time, and longer preoperative antibiotic coverage period. In a multivariable logistic analysis, preoperative pyuria, AGR, and stone size were independently correlated with postoperative fUTI (p<0.001, p=0.008, and p=0.041, respectively). Receiver operating curve analysis showed that the cutoff value of AGR that could predict a high risk of fUTI after URS was 1.437 (sensitivity, 77.3%; specificity, 76.9%), while the cutoff value of stone size was 8.5 mm (sensitivity, 55.3%; specificity, 44.7%). Conclusion: This study demonstrated that preoperative pyuria, AGR, and stone size can serve as prognostic factors for predicting fUTI after URS.

Heat stress effects on the genetics of growth traits in Thai native chickens (Pradu Hang dum)

  • Objective: The objective of this study was to investigate the effect of heat stress on the growth traits and genetic parameters of Thai native chickens. Methods: A total of 16,487 records for growth traits of Thai native chickens between 2017 and 2022 were used in this study. Data included the body weight at birth, body weight at 4, 8, and 12 weeks of age (BW0, BW4, BW8, BW12), average daily gain during 0 to 4, 4 to 8, and 8 to 12 weeks of age (ADG0-4, ADG4-8, ADG8-12), absolute growth rate at birth, at 4, 8, and 12 weeks of age (AGR0, AGR4, AGR8, AGR12). The repeatability test day model used the reaction-norm procedure to analyze the threshold point of heat stress, rate of decline of growth traits, and genetic parameters. Results: At temperature and humidity index (THI) of 76, Thai native chickens began to lose their growth traits, which was the onset of heat stress in this study. The estimated heritability, genetic correlation between animal and heat stress effect, and correlations between the intercept and slope of the permanent environmental effects were 0.27, -0.85, and -0.83 for BW, 0.17, -0.81, and -0.95 for ADG, 0.25, -0.61, and -0.83 for AGR, respectively. Male chickens are more affected by heat stress than female chickens with a greater reduction of BW, ADG, and AGR, values equal to -9.30, -0.23, -15.21 (in males) and -6.04, -0.21, -10.10 (in females) gram per 1 level increase of THI from the THI of 76. Conclusion: The influence of thermal stress had a strong effect on the decline in growth traits and genetic parameters in Thai native chickens. This study indicated that genetic models used in conjunction with THI data are an effective method for the analysis and assessment of the effects of heat stress on the growth traits and genetics of native chickens.

  • The dietary Root Powder Mixture of Angelicae gigantis Radix and Bupleurum falcatum Linne (RPM of AGR & BFL) was feded for 6 weeks to added 0. 0, 0. 4, 1. 0, 2. 0% respectively in the broiler diets. The ,RPM of AGR & BFL was found to be palatable as birds consumed moreyof the test diets than the control diet. The BW gain of 1. 0% RPM of AGR & BFL group was higher than the control group. Feed efficiency (F/G) was improved significantly (P by feeding of 1. 0% RPM of AGR & BFL diet. The carcass weight was improved by feeding with 1. 0% added diet and the carcass ratio was increased in added diets. The amounts of chest meat and drumsticks were heaver in 0. 4% added deit than control. There was no consistent trend in various organ weight by the addition of RPM of AGR & BFL. Fat accumulation of abdominal and gizzard surrounding was highly measured in the broiler fed 0. 4% RPM of AGR & BFL diet. Total serum protein, cholesterol and triglyceride contents were tended to increased with 2. 0% RPM of AGR & BFL ration, and in the 1. 0% addled diet to enhenced BW gain total serum protein, cholesterol and triglyceride concentration were analyzed the lowest among treatments.

Effects of Angelicae Gigantis Radix on Gene Expression of Ovarian Tissue in Polycystic Ovary Syndrome Rats (당귀(當歸)가 다낭성난소증후군이 유발된 흰쥐 난소조직의 유전자 발현에 미치는 영향)

  • Objectives: This study was performed to investigate the effects of Angelicae gigantis Radix (AGR) which is one of the most useful herbal-drug to treat patients with Polycystic Ovary Syndrome (PCOS) in Oriental medicine on gene expression of ovary tissue. Methods: The effects of AGR on gene expression of ovary tissue resected from PCOS induced rats using single injection of $\ss$-Estradiol 17-valerate (EV) was measured using microarray technique, and the functional analysis on these genes was conducted. Results: Total 2,812 genes were up-regulated or down-regulated, 1,421 genes were up-regulated, 1,391 genes were down-regulated by induction of PCOS. Up-regulated genes were mainly involved in biological function such as cell signalling pathways and inflammatory response. Expression levels of 1,442 genes were restored to those of naive animals by administration of AGR. 558 genes were restored to those of naive animals, which were lowered by induction of PCOS. 884 genes were lowered to naive levels, which were elevated by induction of PCOS. The functions of restored genes were partially involved in the restoration of expression levels, which were changed by induction of PCOS. Especially, up-regulated gene by induction of PCOS were mainly involved in these changes. These results mean restorative effects of AGR on damaged functions by induction of PCOS. The network of total protein interactions was measured using cytoscape program, and some key molecules, such as IRS2, MCM10, ORC2L related in up-regulated genes, CTBP2, CD44, RHOA, related in down-regulated genes that can be used for elucidation of therapeutical mechanism of medicine in future were identified. Conclusion: Restored genes by AGR were thought to have common pathways related in regulation of gene expressions. Especially, genes in restored expression levels by AGR, which were up-regulated by induction of PCOS, were regulated by 9 of common transcription factors, genes in restored expression levels by AGR, which were down-regulated by induction of PCOS, were involved in 25 of common transcription factors.

  • The acid gas removal (AGR) system was designed and installed to remove $H_2S$ in coal syngas in the pilot-scale coal gasification system for producing chemicals like Dimethyl Ether(DME). The syngas from the coal gasification at the rate of $100{\sim120$ $Nm^3$/hr included pollutants such as fly ash. $H_2S$, COS, $NH_3$, etc. The designed temperature and pressure of the AGR system are below 50oC and 8 kg/$cm^2$. Fe-chelate was used as an absorbent. $H_2S$ was stably removed below 0.5 ppm in the AGR system when the concentration of $H_2S$ was $150{\sim}450$ ppm. The pH of Fe-chelate solution was also stably maintained between $8{\sim}9$. FeMgO absorbent was also tested to remove $H_2S$ in the lab-scale AGR system and $H_2S$ was also removed below 0.5 ppm in the initial operation.

  • Spinaches showing dark green mosaic symptoms were used for identification of broad bean wilt virus. In host reaction test, that virus caused local lesions on the inoculated leaves and mosaic symptoms on upper leaves of Chenopodium amaranticolor, Chenopodium quinoa and Vicia faba, and developed mosaic symptoms on Physalis floridana, Spinacia oleracea, Nicotiana tabacum, (White burley, Bright yellow) Nicotiana glutinusa. In agar gel-diffusion test, the virus showed positive reaction with broad bean wilt virus antiserum. Spherical virus particles with size of 25nm in diameter were observed in electron microscope.

Chemical and Antimicrobial Properties of Essential Oils from Three Coniferous Trees Abies koreana, Cryptomeria japonica, and Torreya nucifera

  • Three coniferous essential oils were extracted from Abies koreana, Cryptomeria japonica, and Torreya nucifera by hydrodistillation. The chemical composition of each oil was analyzed by GCMS, and their antimicrobial activities were tested against two bacteria and one yeast strains. Fortyseven compounds were identified from A. koreana oil, 39 from C. japonica, and 59 from T. nucifera. Main components of the essential oils were limonene (23.5%), bornyl acetate (17.9%), ${\alpha}-pinene$ (11.1%), and camphene (10.2%) in A. koreana, kaurene (26.3%), ${\gamma}-eudesmol$ (19.0%), elemol (6.9%), and sabinene (5.1%) in C. japonica, limonene (13.5%), ${\delta}-cadinene$ (10.5%), ${\alpha}-bisabolol$ (10.2%), and ${\alpha}-copaene$ (7.7%) in T. nucifera. Among the three coniferous trees tested, the essential oils of A. koreana exhibited higher and broader antimicrobial activity against the tested organisms than those of C. japonica and T. nucifera.
