Trichloroethylene 처리한 흰쥐의 간 미크로좀 Alcohol dehydrogenase와 Aldehyde dehydrogenase 활성도에 관한 연구 (Studies on Hepatic Microsomal Alcohol Dehydrogenase(ADH) and Aldehyde Dehydrogenase(ALDH) Activities in Rats Treated with Trichloroethylene)

  • Chloral hydrate(CH), an intermediate metabolite of trichloroethylene(TRI) is reduced to trichloroethanol(TCE-OH), and is oxidized to trichloroacetic acid(TCA) by the nicotinamide adenine dinucleotide(NAD)-dependent enzymes such as alcohol dehydrogenase(ADH) and aldehyde dehydrogenase(ALDH) in liver. This study was performed to find out the change of activity of ADH and ALDH with increasing amount of TRI. Intraperitoneal injection of TRI were done to the male Sprague Dawely rats(mean body weight, $170{\pm}10g$) in com oil at the dosage of 150, 300, 600 mg/kg for 2 days. The results of experiments are following : 1. The contents of xenobiotic metabolic enzymes in liver are tended to be decreased with increasing amount of, but not significantlly (p>0.05). 2. Activity of ADH in microsome is decreased(p<0.05), and activity of ALDH is increased with amount of TRI(P<0.05). 3. Total trichloro-compounds(TTC) concentration in urine are increased with amount of TRI, but the ratio of between the TCE-OH and the TCA were not shown any critical change. These results suggests that the ALDH in microsome may be related to metabolism of TRI, but ADH was nothing less than the effected to metabolism of TRI.

재조합 alcohol dehydrogenase의 발현 및 기능분석 (The Expression and Functional Analysis of Recombinant Alcohol Dehydrogenase)

  • Bacillus stearothermopilus의 염색체 DNA로부터 polymerase chain reaction 법을 이용하여 ADH의 구조유전자를 증폭시킨 후, 발현 벡터 pGEX-KC에 삽입시켜 glutathion S-tansferase와 융합 단백질로 대장균에서 대랑 발현시켰다. 재조합 ADH는 $37^{\circ}C$에서 1 mM의 isopropyl-${\beta}$-D-thiogalactopyranoside로 단백질의 발현을 유도시켰으며, 발현된 단백질은 glutathione affinity 컬럼을 이용하여 정제하였다. 재조합 ADH는 에탄올에 높은 기질특이성을 나타내었으며 최적 pH와 온도는 각각 pH 9.0과 $70^{\circ}C$이었다. 또한 이 재조합 ADH는 본래의 효소보다 열에 안정하였다. 이 효소는 알코을 측정을 위한 효소학적 방법과 알코올의 공업적 생산에 이용될 수 있다.

염건 조피볼락(Sebastes schlegeli) 제조 중 ADH 및 ALDH의 활성변화 (Changes in Alcohol Dehydrogenase (ADH) and Acetaldehyde Dehydrogenase (ALDH) Activity during the Processing of Salt-Dried Rockfish Sebastes schlegeli)

  • The objective of this study was to determine the processing conditions for salt dried rockfish Sebastes schlegeli by sun drying and cold-air drying, as measured by alcohol dehydrogenase (ADH) and acetaldehyde dehydrogenase (ALDH) activity. We processed salt dried rockfish samples. The salinity of rockfish samples was within 1% following salting with 25% salt brine for 3 h. The moisture content of salt dried rockfish was found to reduce linearly from 70.12 to 39.5 g/100 g over the same time interval. The water activities of salt dried rockfish by sun and cold-air drying were 0.94 and 0.87, respectively, after three days of drying. Acid values (AV) were 10.71 and 5.96 mg KOH/g, respectively, after the three day drying period. The ADH activity in a water extract from salt dried rockfish following sun and cold-air drying for 24 h was 228.5% and 226.1% at 13.3 mg/mL, respectively, and was higher than that when drying lasted for 48 and 72 h. The ALDH activity was not affected but both ADH and ALDH activity tended to decrease as the drying time increased from 24 to 72 h. The conditions of processing for the best quality of salt dried rockfish were determined to be drying with a cold-air system for 24 h. These results indicated that water extracts from salt dried rockfish have valuable biological attributes owing to the metabolizing of alcohol and can provide useful information for the design of drying systems for salt dried rockfish.

인삼 사포닌이 효모 Alcohol Dehydrogenase에 미치는 영향 (The Effect of Ginseng Saponin on Yeast Alcohol Dehydrogenase)

  • Yeast alcohol dehydrogenates and ginseng saponin interaction has been investigated to understand the non-specific enzyme stimulating effect of the saponin of Panax ginseng C.A. Meyer. It was confirmed that several amphiphiles such as sodium dodecyl sulfate(SDS), Triton X-100, sodium taurodeoxycholate (Na-TDC) as well as ginseng saponin mixture and purified ginseng glycosides lowered Km values of yeast alcohol dehydrogenase (ADH) for ethanol and NAD in the presence of the above amphiphiles suggesting that the surface activity of the amphiphiles might play a significant role in the ADH catalyzed reactions. Conformational change of yeast alcohol dehydrogenase in the presence of the above amphiphiles at their optimal concentration for the maximum activity was studied. Circular dichroism (C.D) spectrum of yeast ADH showed that the conformational change of the enzyme occurred in the presence of above amphiphiles. Fluorescence data also showed that the hydrophobic area increased in the presence of above amphiphiles. Examination of the interaction between ADH and ginseng saponin using radioactive saponin showed that there might be a very weak interaction between them. From the above results, it was concluded that the non-specific enzyme stimulating effect of the saponin might be due to the change of polarity of the enzyme solution in the presence of the saponin.

정상인의 혈중 알코올 탈수소효소 활성도 및 에탄올, 아세트알데히드 농도에 관한 연구 (Activity of Alcohol Dehydrogenase and Ethanol, Acetaldehyde Levels in Normal Adults Blood)

  • To identify normal levels of ethanol, acetaldehyde and alcohol dehydrogenase(ADH) activity in blood of Koreans, ethanol and acetaldehyde levels, activity of ADH in blood of Koreans, ethanol and acetaldehyde levels, activity of ADH in blood were measured in 97 subjects(male : 36, female : 61), 45 subjects(male : 21, female : 24) were not exposed to organic solvents and any other chemicals. Fifty two subjects(male : 15, female : 37) were exposed to organic solvents including toluene and xylene. The results were summerized as follows : 1. The blood ADH was not detected in exposed and non-exposed group. 2. The average blood ethanol level of non-exposed group was 0.0490 mg/dl, and exposed group was 0.0363 mg/dl. They were statistically significant(p<0.05). 3. The blood acetaldehyde levels in exposed group were significantly higher than that of non-exposed group was not statistically significant (p>0.05). 4. The average blood ethanol level of males in both groups was significantly higher than that of females, however, they were not statistically significant (p>0.05).

주상에 활용되는 수종의 한약물이 알콜올대사 및 간장해에 미치는 영향 (The Effects of Some Oriental Herbs Which Have Been Used in the Treatment of Alcoholic Diseases on Alcoholic Metabolism and Alcoholic Liver Damages)

  • Objectives : This experiment was conducted to verify the effects of some oriental herbs(Alny Cortex et Ramulus, Artemisiae Capillaris Herba, Aurantii Nobilis Pericarpium, Giseng Radix, Hoveniae Semen, Puerariae Flos, Puerariae Radix, and Xanthii Fructus) which have been used in the treatment of alcoholic diseases, on alcoholic metabolism, and on alcoholic liver damage. Methods : The effects of the herbs on the activities of alcohol dehydrogenase(ADH), aldehyde dehydrogenase(ALDH) were evaluated and their protective effects of liver function and cells from alcoholic damage were analysed. For the evaluation of the protective effects, the levels of glucose, triglyceride, BUN, AST, and ALT in serum of rats were measured. Results and Conclusions : It is concluded that Puerariae Radix interferes with the ADH activity directly, thereby reducing the toxicity of alcohol, resulting in enhancing alcohol-tolerance and protecting liver functions. Also Artemisiae Capillaris Herba interferes both ADH and ALDH activities. Isolation of the biologically active compounds from Puerariae Radix and its detailed characterization are matters for future research.

저산소 조건하에서 맥류 유묘의 근생장 및 혐기발효 효소의 반응 (Effects of Hypoxia on Root Growth and Anaerobic Fermentative Enzymes in Winter Cereal Seedlings)

  • 본 연구에서는 맥류 습해의 원인인 과습에 의한 근권의 산소부족이 맥류 뿌리의 생장 및 혐기 발효대사 효소(alcohol dehydrogenase, ADH; lactate dehydrogenase, LDH)에 미치는 영향을 조사하였다. 양액재배를 이용하여 1.5엽기 맥류 유묘에 용존산소 1.5-2 ppm 정도의 혐기처리를 1, 3, 5, 7일간 실시하였다. 혐기조건에서 뿌리의 생장은 모든 맥종에서 감소하였으며, 감소정도는 내습성이 낮은 보리가 내습성이 높은 밀과 호밀보다 낮았다. 정상조건에서 ADH 활성은 보리가 LDH 활성은 호밀이 가장 높았으나 두 효소 모두 맥종 간의 차이는 크지 않았다. 혐기처리에 의해 ADH와 LDH 활성은 모든 맥종에서 공통적으로 증가하였으며 증가 정도는 호밀과 밀에서 가장 높았고 보리에서 가장 낮았다. 혐기조건에서 이들 효소의 활성증가는 모든 맥종에서 유사하게 항시발현 동위효소의 증가 및 새로운 동위효소의 발현유도에 의해 나타났다. 혐기조건에서 ADH와 LDH 활성 증가 정도는 맥종의 재해저항성과 정의 상관관계가 인정되었다. 이러한 결과는 맥류의 내습성에 혐기 발효계 효소가 관여하고 있음을 의미하는 것으로 판단된다.

천연소재복합물의 알코올 및 아세트알데히드 분해활성 및 호기중 알코올 농도 감소 (Natural herbal extract complex induces the degradation of alcohol and acetaldehyde and reduces the breath alcohol concentration)

  • 본 연구는 천연소재 12종의 페놀성 화합물 함량, 라디칼 소거능, 알코올탈수소효소(ADH), 아세트알데히드탈수소효소(ALDH) 활성도를 측정하고 상승효과를 가지도록 배합한 복합물의 숙취해소에 미치는 효과를 규명하는데 목적이 있다. 우수한 개별 소재를 선정하고 배합비를 달리하여 효능 평가를 통해, 배합비가 효능에 미치는 상관관계를 분석하였다. 8종의 추출물 중 보이차, 소나무, 갈근 순으로 ADH 활성이 높았고, 갈근은 배합량과 효능 간의 양의 상관관계, 보이차, 소나무 등은 음의 상관관계를 가지는 것으로 나타났다. 8종의 소재로 배합한 F1~F7 을 대상으로 ADH, ALDH 활성평가를 통해 F.7에서 상승효과가 가장 높은 것을 확인하여 최적 배합비로 도출하였다. F.7를 섭취한 시험군에서 알코올 섭취 30분 후부터 호기 알코올 농도가 섭취 직후 호기중 알코올 농도 대비 58%로 유의적으로 감소되기 시작하였고, 120분 후 27%로 호기중 알코올 농도가 감소되었다. 음주 후 발생하는 숙취증상 설문 결과 대조군에 비해 시험군에서 음주 후 발생하는 피곤함과 갈증에서 유의적인 개선효과 (p<0.05)가 있었다.

차나무(Camellia sinensis) 추출물이 아급성 알코올 투여 마우스의 항산화 및 알코올 분해 효소 활성에 미치는 영향 (Effects of Camellia sinensis Extracts on the Antioxidant System and Alcohol Down-Regulation Enzymes in Sub-Acute Ethanol Treated ICR Mice)

  • Mouse에 알코올을 투여한 결과 항산화 효소의 활성이 감소하는 것을 볼 수 있으며 ADH 및 ALDH활성도 모두 감소하였다. 녹차, 홍차, 우롱차 및 보이차의 알코올 섭취 mouse에서의 항산화 및 알코올 분해효소에 미치는 영향을 측정하여 비교한 결과 네 가지 차 모두에서 투여하지 않은 군에 비하여 항산화 및 알코올 분해 촉진효과를 나타내었으며 그 중 보이차가 간에서의 SOD 및 GR활성을 크게 증가시켰고 MDA함량을 감소시켰다. 또한 우롱차는 혈액에서의 SOD 및 GSH-PX활성을 크게 증가시켰고 MDA함량을 감소시켰으며 녹차와 홍차의 항산화 효과는 이들에 비하여 비교적 낮았다. 또한 알코올 분해에 관여하는 효소인 ADH, ALDH 활성은 녹차를 투여한 군에서 매우 높게 나타났으며 녹차 다음으로는 우롱차가 ADH 활성측정에서, 보이차가 ALDH 활성측정에서 높은 수치를 보였고 홍차 투여 군에서 가장 낮았다. 따라서 네 가지 차 중 우롱차와 보이차를 섭취하였을 때 높은 항산화 효과를 기대할 수 있으며 알코올 분해에는 녹차의 섭취가 가장 효율적일 것으로 사료된다.

Lactobacillilus brevis를 이용한 다시마 발효물의 알코올 분해 활성 (Alcohol Metabolizing Activity of Fermented Sea Tangle Juice)

  • Alcohol metabolizing activity of fermented sea tangle juice (FSYJ) using Lactobacillilus brevis BJ20 were evaluated by measuring relative alcohol dehydrogenease (ADH) and aldehyde dehydrogenase (ALDH) activities. According to the results of MTI assay. the fermented sea tangle juice by Lactobacillilus brevis BJ20 appeared safe in the cytotocxity. The relative ADH activity of FSTJ showed 124% at 10 mg/mL, which increased with increasing concentration. The relative ALDH activity showed, however, insignificant difference (P>0.05) between concentrations of FSTJ up to 50 mg/mL. These results suggested that fermented sea tangle juice by L. brevis BJ20 could be used as a potential material for metabolizing alcohol.
