ACUTE TOXICITY STUDY OF RECOMBINANT GRANULOCYTE-MACROPHAGE COLONY STIMULATING FACTOR (LBD-005) IN RATS

  • The actue toxicity of a recombinant granulocyte macrophage colony-stimulating factor (code name: LBD-005) was evaluated in both sexes of Sprague-Dawley rats, 4 weeks old, by the oral, subcutaneous and intravenous routes of administration. LBD-005 in the acute toxicity study in the rats was not considerde to induce any toxicological effect on the rats in mortalities, clinical findings, body weights and gross findings. It is suggested that $LD_{50}$ values in rats would be >48 mg/kg in the oral route and >12 mg/kg in the subcutaneous or intravenous route.

Single Oral Toxicity Study on the Standardized Extract of Salvia miltiorrhiza (단삼 표준화시료의 급성독성에 관한 연구)

  • Acute toxicity of standardized extract of Salvia miltiorrhiza Bunge was examined using male and female ICR mice. Mice were treated with standardized extract the intragastrically at 5 mg/kg, 50 mg/kg, 500 mg/kg or 2,000 mg/kg and observed for two weeks. At the doses used, no mortality or abnormal clinical signs in animals were shown during at the observation period. Also there was no difference in net body weight gain, gross pathological findings at the terminal sacrifice among the groups mice treated with different doses of the test substance. The results suggested that acute oral toxicity of standardized extract of S. miltiorrhiza in mice is very low at the conditions employed in this study.

Toxicity Study of a New PEG-hemoglobin SB1, a Red Blood Cell Substitute: Single Intravenous Administration in Beagle dogs (새로운 혈액대용제제 PEG-헤모글로빈 SB1의 개에 대한 단회정맥투여 독성시험)

  • This study was performed to investigate the acute toxicity of PEG-hemoglobin SB1, a blood substitute, in beagle dogs. The male and female dogs were administered intravenously at the doses of 0.4375, 0.875 and 1.75 g/kg body weight, respectively. After a single intravenous administration of SB1 to dogs, we observed them daily for 2 weeks. SB1 did not induce any toxic signs in the mortalities, clinical signs, body weight changes, and gross necropsy findings of dogs. Based on these results, acute toxicity, dogs SB1 may have no side effect and its $LD_{50}$ value may be over 1.75 g/kg (25 ml/kg) of body weight in dogs.

  • The estuarine Gobiidae $Tridentiger$ $trigonocephalus$, $Chasmichthys$ $dolichognathus$ and $Favonigobius$ $gymnauchen$ were exposed to various heavy metals. As a measure of acute toxicity, the 96h-$LC_{50}$ in the three species exposed to copper, lead, zinc and chromium was 740-990, 1,660-1,980, 6,180-8,490 and 8,830-11,296 ${\mu}gL^{-1}$, respectively. In order of toxicity, the heavy metals were ranked copper>lead>zinc>chromium. The Gobiidae $C.$ $dolichognathus$ was more sensitive to heavy metals than $T.$ $trigonocephalus$ and $F.$ $gymnauchen$.

Acute Toxicity Study on SsangHwaTang in Mice (쌍화탕 급성독성에 대한 안전성 연구)

  • SsangHwaTang has been traditionally prescribed a medicine as a restorative. In this study, We investigated the acute toxicity about water-extracted SsangHwaTang. Twenty-five mice completed 14 days of oral SsangHwaTang at the respective doses of 0(control group), 2560, 3200, 4000 and 5000mg/kg. And then we observed survival rates, general toxicity, change of body weight, and autopsy. Compared with the control group, we could not find any toxic alteration in all treated groups (2560, 3200, 4000 and 5000mg/kg). In conclusion, LD50 of SsangHwaTang was over 5000mg/kg and it is very safe to ICR mice.

Study on Acute Subcutaneous Toxicity of Hydroxyapatite Sinter Produced from Tuna Bone in Sprague-Dawley Rats (참치뼈로부터 제조한 Hydroxyapatite 소결체의 랫드에 대한 급성피하독성시험)

  • This study was performed to evaluate the actue sbucutaneous toxicity of hydroxyapatite sinter produced from tuna bone in Sprague-Dawley(SD) rats. Hydroxyapatite sinter was administrated at dose levels of 5000, 2500, 1250, 625, 312.5 and 0 mg/kg. After single subcutaneous adiminstration to both sexes to both sexes SD rats, we observed rats for 14 days. Hydroxyapatite sinter did not induce any toxic signs inmortalities, clinical findings, body weights and gross findings of the rats. In view of result, it was impossible to estimate LD/ sub 50/ values in SD rats. In conclusion, these results suggest that hydroxyapatite sinter produced from tuna bone has no effect on acute subcutaneous toxicity in SD rats.

  • The acute toxicity of recombinant human epidermal growth factor, DWP-401 was evaluated in ICR mice of both sexes. Six groups of mice were administered orally or subcutaneously with 0, 0.125, 0.25, 0.5, 1 and 2 mg/kg of DWP-401. Abnormal clinical signs related to the compound were not observed, and no deaths occurred. Gross findings of necropsy revealed no evidence of specific toxicity related to DWP-401. LD$_{50}$ values for both male and female mice were evaluated to be over 2 mg/kg, which is approximately 2, 000 fold of presumed clinical dose.e.

  • This experiment was carried out to study on the safety assessment of Pinus densiflora Siebold et Zuccarini for Hebal-acupuncture. SD rats and ICR mice were used for acute toxicity test, the results were summerized as follows; 1. In rats and mice, $LD_{50}$ value could not be measured. 2. There were no abnormal finding in acute toxicity test treated Pinus densiflora Siebold et Zuccarini for Hebal-acupuncture.

Acute Toxicity Study on Scopoliae Rhizoma in Mice (낭탕근의 급성독성 연구)

  • Scopoliae rhizoma is a perennial herb which has a similar effect with atropine on the cardiovascular system. It is also known to have a seditive and anticonvulsant activity on the central nerve system. In order to evaluate an acute toxicity of Scopoliae rhizoma, the present study was performed after administration the Scopoliae rhizoma prepared by both decoctional and frozen dried extract through three different routes (oral; 5,000 mg/kg, intraperitoneal; 2,000 mg/kg, subcutaneous; 5,000 mg/kg) to the female ICR mice. In the group treated intraperitoneally with a frozen dried extract, abnormal clinical signs such as decreased activity, crouch, potosis and abnormal walking were observed for 40 rain after administration. With regard to WBC, decreased number of lymphocyte and increased number of monocyte and granulocyte were also observed in the animals received intraperitoneally with Scopoliae rhizoma extract. Taken together, what toxicity of Scopoliae rhizoma was shown differently depending on its type for administration may be resulted in the differency of administered dose. The results provided here support a pharmacological and toxicological consideration for its clinical use in the regard of oriental medicine.

Preventive Effect of Ginseng Butanol Fraction against Acetaldehyde - Induced Acute Toxicity (아세트알데히드로 유도된 급성독성에 대한 인삼부탄올 분획의 방어작용)

  • The objective of this study was to investigate the preventive effect of ginseng on acetaldehyde-induced acute toxicity in mice . Compared to the control group, treatment with acetaldehyde inhibited the hepatic cytosolic xanthine oxidise activity with increase in dose. The inhibition of enzyme activity was not changed after dialysis. Pretreatment with ginseng butanol fraction prevented the inhibition of enzyme activity by acetaldehyde. In conjunction with the our previous results (Yakhak Hoeji, 29, 18 (1985)), these results suggest that the most likely mechanism for the observed preventive effects of ginseng against the acetaldehyde-induced acute toxicity may be the decrease hepatic acetaldehyde level.
