Partial Purification of Mussel Adhesive Protein for Mytilus Edulis and Preparation of Mussel Protein Hydrolysates

  • Mussel adhesive protein (MAP) was extracted from Korean Mytilus edulis and then partially purified using Sephacryl S-300 gel permeation chromatography and reversed-phase high performance liquid chromatography. As an indicator of adhesiveness, is 3,4-dihydroxyphenylalanine (DOPA) content was determined. Its DOPA/protein ratio of 0.19 was higher than those of other reports, indicating a good adhesive. The partially purified MAP was confirmed by acid-urea polyacrylamide gel electrophoresis using cetylpiridinium bromide as a cationic detergent. Sea mussel hydrolysates were prepared using three commercial proteases to provide value-added functional materials and their angiotensin converting enzyme (ACE) inhibitory activities were determined. Among hydrolysates of sea mussel, Protamex was the best and further purification would improved ACE inhibitory activity.

Isolation of an Angiotensin Converting Enzyme Inhibitory Substance from Lycium chinense Miller

  • An angiotensin converting enzyme (ACE) inhibitory substance was isolated and purified from Lycium chinense Miller. A crude water extract of Lycium chinense Miller was prepared by adding it to water shaking at $25^{\circ}C$ for 1 hr, followed by centrifugation at 8000 ${\times}$ g for 30 min. The crude extract was then filtered using YM-3 and YM-1 membranes. An ACE inhibitor was isolated using consecutive chromatographic methods including: ion exchange chromatography, gel permeation chromatography, and FPLC. The inhibitor was identified to have a molecular mass of 862 daltons by mass spectrometry.

식물자원으로부터 Angiotensin Converting Enzyme 저해활성 탐색 (Screening of 94 Plant Species Showing ACE Inhibitory Activity)

  • 94종 식물자원을 대상으로 혈압상승을 주도하는 효소인 angiotensin converting enzyme의 저해활성을 검색하였다. 그 결과 추출물의 농도가 1mg에서 60%이상의 높은 ACE 억제활성을 보인 식물은 털여뀌 (81.6%), 결명자 (64.2%) 등의 2종이었으며, 40%이상의 ACE 억제 활성을 보인 식물은 금불초(49.7%), 골담초(49.4%), 황금(48.1%), 회향(45.2%)등 4종으로 ACE 억제활성이 비교적 높게 나타났다. 또한 $30{\sim}40%$의 ACE 억제활성을 보인 식물은 까마귀머루(35.5%), 부용(32.3%), 삼립국화(32.6%), 산비장이(33.2%), 산사나무(33.9%), 율무(34.8%), 제비꽃(37.1%), 진득찰(37.1%), 짚신나무(36.1%), 참죽나무(39.7%), 창질경이(33.5%), 컴프리(33.9%), 털냉초(39.7%), 하눌타리(39.4%), 해바라기(34.2%), 호두나무(39.4%), 회양목(30.6%), 희수(35.8%), 금불초(36.8%), 도꼬마리(34.8%), 두충나무(30.3%), 둥글레(35.2%), 방풍(33.5%), 산옥잠화(34.8%), 살구나무(39%), 섬오갈피(38.4%), 원추리(39.7%), 지모(31.9%)등 28종이었다. 그 외 흰제비꽃, 황해쑥, 쥐방울덩굴, 자귀나무, 일당귀, 오크라, 애기기린초, 순비기나무, 쇠뜨기 , 석창포, 석결명, 사위질방, 산수유나무, 부추, 뽕나무, 봉선화, 보리수나무, 큰꽃삽주, 범부채, 벌등골나무, 모감주, 동과, 도라지, 땃두릅, 도꼬마리, 산옥잠화, 방풍, 둥글레, 닭의장풀, 닥나무, 노루오줌, 가시오갈피, 개오동, 귀릉나무, 꽈리, 까실쑥부쟁, 골담초, 고삼, 고비고사리, 개발나물, 결명자, 형개, 향유, 한련초, 파고지, 초과, 지골피, 마두령, 금앵자, 곽향 등 50여 종의 추출물은 $10{\sim}30%$의 낮은 ACE 억제활성을 보였다. 앞으로 ACE 억제활성이 높은 식물자원의 유효성분에 대한 물질 확인과 동물모델을 이용한 효능검증에 대하여 더 많은 연구가 있어야 할 것으로 사료된다.

Production of Antihypertensive Angiotensin I-Converting Enzyme Inhibitor-Enriched Edible Yeast Using Gugija (Lycium chinesis Mill)

  • To produce bioactive compound enriched yeast using medicinal Gugiga (Lycium chinensis Mill), several edible Saccharomyces species were cultured in Gugija extracts added yeast extract, peptone and dextrose medium (GE - YEPD medium) at $30^{\circ}C$ for 24 hr, and their growth were determined. Growth of Saccharomyces cerevisiae K-7 and Sacchromyces cerevisiae ACTC 7904 were better than those of the other yeasts. Two yeasts were selected and then determined their some physiological functionalities after cultivated the yeasts in the GE - YEPD medium and compared those grown on YEPD medium. Antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of S. cerevisiae K-7 grown on GE - YEPD medium was about 20% higher than that grown on YEPD medium. Superoxide dismutase-like activity of S. cerevisiae ACTC 7904 was also about 12% more high. However, the other physiological functionalities were almost same or lower. Optimal addition concentration of Gugija extract was 10%, and maximally growth and ACE inhibitory activity of S. cerevisiae K-7 were shown when the strain was cultured in 10% Gugija extracts containing YEPD medium at $30^{\circ}C$ for 12 hr.

The Relationship between Physiological Activity and Cell Number in Dolsan Leaf Mustard Kimchi (Brassica juncea)

  • Changes in antioxidative activity and Angiotensin converting enzyme(ACE) inhibitory activity in juice prepared from Dolsan leaf mustard kimchi (DLMK) ar various fermentation temperatures were investigated. Antioxidative activity of juice from optimally ripened DLMK at 20 and 3$0^{\circ}C$ showed 80 and 83%, respectively. Juice from 10-day fermented DLMK at 3$0^{\circ}C$ showed 62% inhibitory activity against the ACE. In the juice fermented DLMK at 2$0^{\circ}C$~3$0^{\circ}C$, physiological activity was higher than that of the 4~1$0^{\circ}C$. In particular, optimally ripened DLMK at 3$0^{\circ}C$ showed the highest physiological activity. The physiological activity in DLMK juice at the fermentation period increased significantly with an increase in the growth of microbes. Consequently, a maximum physiological activity was shown at the maximum cell number. These results suggest that the microorganisms in DLMK juice would play an important role in the physiological activity.

In vitro에 의한 녹용 추출물의 생리 활성 효과 (The Biological Activity of Deer Antler Extract in vitro)

  • Our research objective was to examine the in vitro biological activity of deer antler(Nogyong in Korean) extract, including the antioxidative, nitrite scavenging, and tyrosinase inhibitory effects, as well as the antithrombotic, and angiotensin I converting enzyme(ACE) inhibitory activities. The carbohydrate, protein, fat, and mineral contents of the deer antler were 7.6%, 65.3%, 3.2% and 23.9%, respectively. The electron donating ability(EDA) by the reduction of 2,2'-diphenyl-1-picrylhydrazyl(DPPH) was 67.1%, and the inhibition rate of lipid peroxidation by the thiocyanate method using linoleic acid was 92.1% in 100 mg/ml of extract. The nitrite scavenging effects were pH dependent, and were highest at pH 1.2 and lowest at pH 6.0. The sample inhibition rate against tyrosinase was above 64.0%. The platelet aggregation induced by ADP(adenosine-5'diphosphate) was inhibited up to 51.7%, and the inhibitory effect was dependent on the sample concentration. Lastly, the inhibition rate of ACE was 47.5% in 100 mg/ml of deer antler extract.

해조류로부터 항고혈압성분의 분리정제 (Separation and Purification of Antihypertensive Substances from Edible Seaweeds)

  • To isolate natural antihypertensive substances from edible seaweeds, we screened for and separated active compounds contained in natural Underia pinnatifida, cultural Underia pinnatifida, Laminaria japonica, Sporophylls and Agarum cribrosum. They were extracted using room temperature water, boiling water, acetone, and methanol in turn or using room temperature water, ether, acetone, methanol and boiling water in order. The in vitro antihypertensive activity was quantified as inhibitory efficacy against angiotensin-I converting enzyme (ACE), which is a factor inducing hypertension. For all of the seaweeds tested, the fractions soluble in room temperature water and in boiling water showed the strongest ACE inhibitory effect among the extracted fractions. Conversely, the methanol-extracted fractions for all of the seaweeds tested showed no antihypertensive activity. While the ether and acetone fractions had slight antihypertensive effects. The compounds in the aqueous extracts that had antihypertensive activity were presumed to be polysaccharides, such as fucoidan and alginate.

Physiological Functionalities of Vitis hybrid (Sheridan)-Rubus coreanus Red Wine Made by Saccharomyces cerevisiae

  • Vitis hybrid (Sheridan)-Robus coreanus red wine was vinified by fermentation of a mixture of Vitis hybrid and Robus coreanus must at $25^{\circ}C$ for 10 days. The Vitis hybrid-Robus coreanus red wine had ethanol contents of 10.9%. It had high antihypertensive angiotensin I-converting enzyme (ACE) inhibitory activity of 57.8% and antioxidant activity of 64.8%. Changes in the physicochemical properties and functionality of the Vitis hybrid-Robus coreanus red wine was investigated during a post-fermentation period of three months. The ACE inhibitory activity of the red wine increased as the post-fermentation period prolonged, and showed the highest ACE inhibitory activity of 70.4% 60 days post-fermentation. However, the antioxidant activity declined significantly to 47.2% during the post-fermentation period of 60 days. In terms of sensory evaluation, the Vitis hybrid-Robus coreanus red wine had the best acceptability 60 days post-fermentation.

프로바이오틱 유산균 발효조건 탐색을 위한 다반응 최적화의 활용 (Applying Multi-Response Optimization to Explore Fermentation Conditions of Probiotics)

  • This review serves two purposes: first, to promote the use of improved optimization techniques in response surface methodology (RSM); and second, to enhance the optimum conditions for the fermentation of probiotics. According to research in dairy science, Lactiplantibacillus plantarum K79 is a candidate probiotic that has beneficial health effects, such as lowering blood pressure. The optimum conditions for L. plantarumK79 to produce peptides with angiotensin-converting enzyme (ACE) inhibitory activity were proposed, through modeling each of ACE inhibitory activity and pH as a function of the four factors that are skim milk concentration (%), incubation temperature (℃), incubation time (hours), and starter added amount (%). To estimate optimum conditions, the researchers employed a desirability-based multi-response optimization approach, utilizing third-order models with a nonsignificant lack of fit. The estimated optimum fermentation conditions for L. plantarum K79 were as follows: a skim milk concentration of 10.76%, an incubation temperature of 36.9℃, an incubation time of 23.76 hours, and a starter added amount of 0.098%. Under these conditions, the predicted ACE inhibitory activity was 91.047%, and the predicted pH was 4.6. These predicted values achieved the objectives of the multi-response optimization in this study.
