Chemical Components and Comparison of Biological Activities on the Fruit of Natural Bogbunja (Rubus coreanus Miquel) (국내산 복분자 열매에 대한 화학적 조성 및 생리활성 비교)

  • This study was carried out to investigate biological and antioxidative activities on the fruit of bogbunja (Rubus coreanus Miquel). Total contents of phenolic compounds contents in cultivars ripened fruit and immatured wild-type fruit were 222 and 190mg/g, respectively, Polysaccharide contents of immatured wild-type fruit were the highest value of 320U. For EDA analysis, immatured wild-type fruit showed over 95% in 100$\mu\textrm{g}$/mL of sample concentration, which is the the most effective. Levels of SOD-like activities in immatured and cultivar ripened fruits were 81% and 77%, respectively, For the inhibitory effect on lipid peroxidation, all of bogbunja prepared were similar with those of $\alpha$-tocopherol as control. The inhibition of ACE activities on the water extracts of bogbunja fruit showed over 98%, especially, in immature wild-type and cultivar bogbunja.

  • Antibacterial, antifungal, anticancer, and ACE inhibitory activities of methanol, ethyl acetate, chloroform, and hexane extracts of garlic were investigated. Methanol extract showed highest yield of 7.9-8.8 brix. Ethyl acetate and chloroform extracts showed strong antibacterial activities as determined by inhibition zone (8-16mm) against B. subtilis and P. aeruginosa, and good antifungal activities as determined by inhibition zone (8-30mm) against A. niger, M. miehei, and T. reesei. Wando, Banyans, Chinese, and Seosan garlic methanol extracts showed 65.6, 60.4, 70.1, and 55.3% ACE inhibitory activities, 42.3, 53.8, 50.4, and 54.5% anticancer activities against SNU-1, and 11.3, 20.2, 36.6, and 30.5% anticancer activities against HeLa, respectively.

Antioxidant and Antihypertensive Activities of Grains Grown in South Korea in Relation to Phenolic Compound and Amino Acid Contents

  • Hypertension is characterized by excessive renin-angiotensin system activity, leading to blood vessel constriction. Several synthetic compounds have been developed to inhibit renin and angiotensin-converting enzyme (ACE). These drugs often have adverse side effects, driving the exploration of plant protein-derived peptides as alternative or supplementary treatments. This study assessed the phenolic compound and amino acid content and the antioxidant and antihypertensive activity of 5 South Korean staple crops. Sorghum had the highest phenolic compound content and exhibited the highest antioxidant activity. Millet grains, particularly finger millet (38.86%), showed higher antihypertensive activity than red beans (14.42%) and sorghum (17.16%). Finger millet was found to contain a large proportion of branched-chain, aromatic, and sulfur-containing amino acids, which are associated with ACE inhibition. In particular, cysteine content was positively correlated with ACE inhibition in the crops tested (r=0.696, p<0.01). This study confirmed that the amino acid composition was more correlated with the antihypertensive activity of grains than the phenolic compound content. Finger millet mainly contained amino acids, which have higher ACE inhibitory activity, resulting in the strongest antihypertensive activity. These findings underscore the antihypertensive potential of select crops as plant-based food ingredients, offering insight into their biological functions.

Characteristics of Angiotensin Converting Enzyme Inhibitory Peptides from Salt-fermented Squid Liver Sauce (오징어 간 액젓으로부터 분리된 Angiotensin Converting Enzyme 저해 Peptide의 특성)

  • In order to utilize squid liver by-products, which is normally discarded as industrial waste in the process of squid manufacturing, salt-fermented squid liver sauce was prepared experimentally and also tested for inhibitory activity against angiotensin converting enzyme (ACE). ACE inhibitory activity of squid liver sauce was increased with the elapse of fermentation days until 12 months, followed by a constant level of inhibitory activity thereafter. 15-month-old sauce ($IC_{50}=29.66\;{\mu}g$) was filtered through PM-10 membrane (M.W. cut-off 10,000 Da) to obtain the peptides fractions with ACE inhibition activity. Filtered fractions were applied to a Bio-gel P-2 column and three active fractions (A, B and C) were collected. Among them, fraction B applied to a SuperQ-Toyopearl 650S column chromatography lead to the isolation of active B-1 fraction. It has the ACE inhibitory activity ($IC_{50}=5.46\;{\mu}g$). The main composition of its amino acids is lysine, glycine and proline, which cover about 85% of the total amino acids.

Optimization of Whey-Based Medium for Growth and ACE-Inhibitory Activity of Lactobacillus brevis

  • A Whey-based medium was formulated with Lactobacillus brevis to investigate whether any functional peptides could derive from whey protein. The optimal concentrations of the ingredients of the medium for the growth of Lactobacillus were determined as 2% whey protein concentrate and 1% glucose and 0.5% yeast extracts. The growth of Lb. brevis was improved with the supplementation of yeast extracts than glucose. The viable cells counts of Lb. brevis reached to 2.0 × 10$^8$CFU/mL in the whey-based medium. The whey protein hydrolysates recovered from the supernatant after centrifugation at 10,000 x g for 10min induced strong inhibitory activity against ACE. When the whey protein hydrolysate were partially purified by a membrane tubing below 8,000Da, the partially purified fraction remained 64.7 ${\pm}$ 3.6% of the ACE inhibition activity of the whey protein hydrolysates and IC$_{50}$ was 38.8 ${\pm}$ 2.2mg/mL. The whey-based medium was proved to be effective in producing ACE inhibitory peptides by lactic bacteria fermented whey protein.

  • This study was carried out to identify the biophysical utility of bioflavonoids by the determination of their antioxidative activities, radical scavenging activity and inhibitory effect on the ACE activity. The results obtained were as follows; All flavonoids experimented greatly inhibited the linoleic acid oxidation from the early period of oxidation, and the radical scavenging ability was also greater in genistein and daidzein than other flavonoids, generally showing donating ability. Rutin has the metal-chelating ability with C $u^{2+}$ and $Mg^{2+}$, which means to have the inhibitory effect on the promotive oxidation of lipid by metal ion. All flavonoids experimented inhibited the angiotensin converting enzyme (ACE) activity, which was greater in genistein and daidzein than other flavonoids.s.

Quality Characteristics of Rehmannia radix Preparata with Pre-soaking Solvents (침지용매에 따른 숙지황의 품질특성)

  • This study was performed to investigate the effects of pre-soaking methods on the preparation of Rehmannia radix Preparata (R.P). The R. radix L (R.L) was soaked in distilled water and traditional Korean wine for 24 hr, then the soaked R.L was treated with a traditional nine-time steaming process. Next, catalpol, 5-hydroxymethyl-2-furaldehyde (5-HMF), polyphenols, flavonoids, antioxidant activities, and ACE inhibition activity were analyzed for the R.P produced by the different methods. The catalpol content of the R.L was 631.4 ppm, but the content decreased as steaming increased to 8-9 times. The 5-HMF, polyphenol, and flavonoid contents of the R.L were 0.12 mg/g, 5.09 mg/g, and 0.83 mg/g, respectively, and these increased gradually with increasing steaming times. As the steaming times of the distilled soaking water increased, the antioxidant activities of 1 mg/mL increased from 19.44% to 75.60% at 14 times of steaming. The ACE inhibition activities of 1 mg/mL of the distilled soaking water increased from 28.70% to 94.78% at 10 times of steaming, but decreased afterward.

  • This study was designed to investigate the angiotensin convertin enzyme (ACE) inhibitory activity of skipjack/yellowpin tuna cooking broth. The cooking broth was pretreated with membrane filter (MW cut-off 5,000) to obtain the peptide fraction with ACE inhibition. the crude peptides fractionated with Amberlite IR-120 ($H^{+}$ form and followed by Bio-gel P-2, were separated into nine fractions (T-1 to T-9). The maximum inhibitory activity was observed in the fraction T-4 ($IC_{50}$ value, 0.619mg/ml). The abundant amino acids obtained from active fraction T-4 were phenylaanine, leucine and glutamic acid.

Angiotensin Converting Enzyme Inhibitory Activity in Enzymatic Hydrolysates of Anchovy Muscle Protein (멸치육 효소 가수분해물의 Angiotensin 전환효소 저해작용)

  • To develop functional food material with angiotensin converting enzyme (ACE) inhibitory peptides, muscle protein of anchovy, Engraulis japonica was hydrolyzed during 48 hrs by digestive pretenses such as pepsin, trypsin, $\alpha$-chymotrypsin, and commercial proteases such as papain, bromelain, complex enzyme, Elavourzyme, Novozym, Neutrase, Protamex and Alcalase. The only $50\%$ ethanol soluble hydrolysates were tested for inhibitory activity against ACE and yield of $50\%$ ethanol soluble peptide-nitrogen ($ESPN_{50}$). ACE inhibition effects and yield of $ESPN_{50}$ occurred as hydrolysis time increased to 8 hrs, Among those pretenses tested, hydrolysates by Alcalase and $\alpha$-chymohypsin had greater ACE inhibitory activity (80 and $74\%$, reipectively) with eletated levels of $ESPN_{50}$ (48 and 58 mg/ml, respectively), while Protamex hydrolysates had greater ACE inhibitory activities ($73\%$) with reduced levels of $ESPN_{50}$ (7.2mg/ml) than others. Amino acid compositions of $50\%$ ethanol solubles obtained from those hydrolysates were rich in glutamic acid, aspartic acid, cysteine and leucine.

  • Effect of BR-900317 on the angiotensin I-induced pressor response in pithed rats and the effects of its single oral administration on plasma angiotensin converting enzyme (ACE) activities in normotensive rats and on the cardiovascular system in hypertensive model rats (SHR, RHR), were compared with those of captopril. BR-900317 attenuated the angiotensin I-induced pressor effects in pithed rats. In a single oral dose administration study, BR-900317 inhibited the plasma ACE activities in a dose-dependent fashion. Duration of the action of BR-900317 was similar to that of captopril. BR-900317 produced antihypertensive effect in spontaneously hypertensive rats and dose-dependent antihypertensive effect in 2-kidney Goldblatt hypertensive rats without affecting heart rate. These results suggest that the main mechanism of the antihypertensive effect of BR-900317 is the suppression of angiotensin II production due to the inhibition of the ACE.
