The Usefulness Assessment of Attenuation Correction and Location Information in SPECT/CT (SPECT/CT에서 감쇠 보정 및 위치 정보의 유용성 평가)

  • Purpose: We make a qualitative analysis of whether Fusion SPECT/CT can find lesion's anatomical sites better than existing SPECT or not, and we want to show the usefulness of SPECT/CT through finding out effects of CT attenuation correction on SPECT images. Materials and Method: 1. The evaluation of fusion images: This study comprised patients who was tested $^{131}I$-MIBG, Bone, $^{111}In$-Octreotide, Meckel's diverticulum, Parathyroid MIBI with Precedence 16 or Symbia T2 from 2008 Jan to Aug. We compared SPECT/CT image with non fusion image and make a qualitative analysis. 2. The evaluation of attenuation correction: We classified 38 patients who was tested 201Tl myocardial exam with Symbia T2 into 5 sections by using Cedars Sinai' QPS program - Ant, Inf, Lat, Septum, Apex. And we showed each section's perfusion states by percentage. We compared the each section's perfusion-states differences between CT AC and Non AC by average${\pm}$standard deviation. Results: 1. The evaluation of fusion images : In high energy $^{131}I$ cases, it was hard to grasp exact anatomical lesions due to difference between regions and surrounding lesions' uptake level. After combining with CT, we could grabs anatomical lesion more exactly. And in meckel's diverticulum case or to find lesions around bowels or organs with $^{111}In$ cases, it demonstrates its superiority. Bone SPECT/CT images help to distinguish between disk spaces certainly and give correct results. 2. The evaluation of attenuation correction: There is no significant difference statistically in Ant and Lat (p>0.05), but there is a meaningful difference in Inferior, Apex and Septum (p<0.05). AC perfusion at inferior wall in the 5 sections of myocardium: The perfusion difference between Non AC perfusion image ($68.58{\pm}7.55$) and CT corrected perfusion image ($76.84{\pm}6.52$) was the largest by $8.26{\pm}4.95$ (p<0.01, t=10.29). Conclusion: Nuclear medicine physicians can identify not only molecular image which shows functional activity of lesions but also anatomical location information of lesions with more accuracy using the combination of SPECT and CT systems. Of course this combination helps nuclear medicine physician find out the abnormal parts. Moreover combined data sets help separate between normal group and abnormal group in complicated body part. So clinicians can carry out diagnosis and treatment planning at the same time with a single test image. In addition, when we examine a myocardium in thorax where attenuation can occur easily, we can trust perfusion more in a certain region in SPECT test because CT provides the capability for accurate attenuation correction. In these reasons, we think we can prove the justice after treatment fusion image.

A Study on the Ecological Indices for the Assessment of the Function and Maturity of Artificial Reefs (인공어초의 기능도와 성숙도 평가를 위한 생태학적 지수에 대한 연구)

  • We reviewed foreign evaluation systems based on the macrobenthic and macroalgal communities and developed a system, composed of a set of ecological indices able to evaluate the functionality (FI, Functional Index; estimation of stability and productivity) and maturity (MI, Maturity Index; comparisons with biological parameters of natural reefs) of artificial reefs by comparing the status in the adjacent natural reefs in Korean coastal waters. The evaluation system was applied to natural and artificial reefs/reef-planned areas (natural reefs), established in the 5 marine ranching areas (Bangnyeong-Daechung, Yeonpyung, Taean, Seocheon and Buan) in the west coast of Korea. The FI ranged between 31.6 (Bangnyeong-Daechung) and 72.5% (Buan) and MI did between 53.1 (Seocheon) and 76.9% (Taean) in average. The evaluation of artificial reefs by the two indices, showed the most appropriate status in Taean. The FI between the adjacent artificial and natural reefs were in significant linear relationship ($r^2=0.83$, p=0.01). This indicated the local status of biological community may be critical in determining the functionality of the artificial reefs. We have suggested an integrative but preliminary evaluation system of artificial reefs in this study. The output from the evaluation system may be utilized as a tool for environment/resource managers or policy makers, responsible for effective use of funds and decision making. Given the importance, we need to use the options to enhance and improve the accuracy as follows: (1) continuous validation of the evaluation system and rescaling the criteria of indicators, (2) vigorous utilization of observation and experience through the application and data accumulation and (3) development and testing of brand-new indicators.

Environmentally Associated Spatial Distribution of a Macrozoobenthic Community in the Continental Shelf off the Southern Area of the East Sea, Korea (한국 동해 남부해역 대륙붕에 서식하는 대형저서동물군집 공간분포를 결정하는 환경요인)

  • This study aims to understand environmental factors that determine spatial distribution of macrozoobenthic community in the southern area (ca 100-500 m depth) of East Sea, Korea, known as a candidate site for carbon storage under the seabed. From sixteen locations sampled in the summer of 2012, a total of 158 species were identified, showing density of $843indiv/m^2$ and biomass of $26.2g\;WW/m^2$, with increasing faunal density towards biologically higher diverse locations. Principal component analysis showed that a total of 33 environmental parameters were reduced to three principal components (PC), indicating sediment, bottom water, and depth, respectively. As sand content was increasing, number of species increased but biomass decreased. Six dominant species including two bivalve species favored high concentrations of ${\Omega}$ aragonite and ${\Omega}$ calcite, indicating that the corresponding species can be severely damaged by ocean acidification or $CO_2$ effluent. Cluaster analysis based on more than 1% density dominant species classified the entire study area into four faunal assemblage (location groups), which were delineated by characteristic species, including (A) Ampelisca miharaensis, (B) Edwardsioides japonica, (C) Maldane cristata, (D) Spiophanes kroeyeri, and clearly separated in terms of geography, bottom water and sediment environment. Overall, a discriminant function model was developed to predict four faunal assemblages from five simply-measured environmental variables (depth, sand content in sediment, temperature, salinity and pH in bottom water) with 100% accuracy, implying that benthic faunal assemablages are closed linked to certain combinations of abiotic factors.

Construction and estimation of soil moisture site with FDR and COSMIC-ray (SM-FC) sensors for calibration/validation of satellite-based and COSMIC-ray soil moisture products in Sungkyunkwan university, South Korea (위성 토양수분 데이터 및 COSMIC-ray 데이터 보정/검증을 위한 성균관대학교 내 FDR 센서 토양수분 측정 연구(SM-FC) 및 데이터 분석)

  • In this study, Frequency Domain Reflectometry (FDR) and COSMIC-ray soil moisture (SM) stations were installed at Sungkyunkwan University in Suwon, South Korea. To provide reliable information about SM, soil property test, time series analysis of measured soil moisture, and comparison of measured SM with satellite-based SM product are conducted. In 2014, six FDR stations were set up for obtaining SM. Each of the stations had four FDR sensors with soil depth from 5 cm to 40 cm at 5~10 cm different intervals. The result showed that study region had heterogeneous soil layer properties such as sand and loamy sand. The measured SM data showed strong coupling with precipitation. Furthermore, they had a high correlation coefficient and a low root mean square deviation (RMSD) as compared to the satellite-based SM products. After verifying the accuracy of the data in 2014, four FDR stations and one COSMIC-ray station were additionally installed to establish the Soil Moisture site with FDR and COSMIC-ray, called SM-FC. COSMIC-ray-based SM had a high correlation coefficient of 0.95 compared with mean SM of FDR stations. From these results, the SM-FC will give a valuable insight for researchers into investigate satellite- and model-based SM validation study in South Korea.

Functional analysis of seaR protein identified from Saccharopolyspora erythraea (희소방선균의 seaR 단백질 발현을 통한 기능 분석)

  • Secondary metabolism in actinomycetes has been known to be controlled by a small molecule, ${\gamma}$-butyrolactone autoregulator, the binding of which to each corresponding receptor leads to the regulation of the transcriptional expression of the secondary metabolites. We expected that expression of an autoregulator receptor or a pleiotropic regulator in a non-host was to be gained insight of effective production of new metabolic materials. In order to study the function of the receptor protein (seaR), which is isolated from Saccharopolyspora erythraea, we introduced the seaR gene to Streptomyces coelicolor A3(2) as host strains. An effective transformation procedure for S. coelicolor A3(2) was established based on transconjugation by Escherichia coli ET12567/pUZ8002 with a ${\varphi}C31$-derived integration vector, pSET152, which contained int, oriT, attP and $ermEp^*$ (erythromycin promotor). Therefore, the pEV615 was introduced into S. coelicolor A3(2) by conjugation and integrated at the attB locus in the chromosome of the recipients by the ${\varphi}C31$ integrase (int) function. Exconjugant of S. coelicolor A3(2) containing the seaR gene was confirmed by PCR and transcriptional expression of the seaR gene in the transformant was analyzed by RT-PCR. In case of S. coelicolor A3(2), a phenotype microarray was used to analyze the phenotype of transformant compared with wild type by seaR expression. After that, in order to confirm the accuracy of the results obtained from the phenotype microarray, an antimicrobial susceptibility test was carried out. This test indicated that sensitivity of the transformant was higher than wild type in tetracycline case. These results indicated that some biosynthesis genes or resistance genes for tetracycline biosynthesis in transformant might be repressed by seaR expression. Therefore, subsequent experiments, analysis of transcriptional pattern of genes for tetracycline production or resistance, are needed to confirm whether biosynthesis genes or resistance genes for tetracycline are repressed or not.

The PRISM-based Rainfall Mapping at an Enhanced Grid Cell Resolution in Complex Terrain (복잡지형 고해상도 격자망에서의 PRISM 기반 강수추정법)

  • The demand for rainfall data in gridded digital formats has increased in recent years due to the close linkage between hydrological models and decision support systems using the geographic information system. One of the most widely used tools for digital rainfall mapping is the PRISM (parameter-elevation regressions on independent slopes model) which uses point data (rain gauge stations), a digital elevation model (DEM), and other spatial datasets to generate repeatable estimates of monthly and annual precipitation. In the PRISM, rain gauge stations are assigned with weights that account for other climatically important factors besides elevation, and aspects and the topographic exposure are simulated by dividing the terrain into topographic facets. The size of facet or grid cell resolution is determined by the density of rain gauge stations and a $5{\times}5km$ grid cell is considered as the lowest limit under the situation in Korea. The PRISM algorithms using a 270m DEM for South Korea were implemented in a script language environment (Python) and relevant weights for each 270m grid cell were derived from the monthly data from 432 official rain gauge stations. Weighted monthly precipitation data from at least 5 nearby stations for each grid cell were regressed to the elevation and the selected linear regression equations with the 270m DEM were used to generate a digital precipitation map of South Korea at 270m resolution. Among 1.25 million grid cells, precipitation estimates at 166 cells, where the measurements were made by the Korea Water Corporation rain gauge network, were extracted and the monthly estimation errors were evaluated. An average of 10% reduction in the root mean square error (RMSE) was found for any months with more than 100mm monthly precipitation compared to the RMSE associated with the original 5km PRISM estimates. This modified PRISM may be used for rainfall mapping in rainy season (May to September) at much higher spatial resolution than the original PRISM without losing the data accuracy.

The Application of Computer Program for Determination of Fluid Properties and P-T Condition from Microthermometric Data on Fluid Inclusions (유체포유물의 생성시 온도-압력 조건과 유체포유물의 물리화학적 특성연구에 있어서의 컴퓨터 프로그램이용)

  • Fluid inclusion has been widely used to study the origin and physiochemical conditions of ore deposits. However, it is difficult to get the accurate physiochemical data from fluid inclusion study due to the error of microthermometric data and the complexity of calculation of density and isochore of fluid inclusion. The computer programs HALWAT, $CO_2$, and CHNACL written by Nicholls and Crowford (1985) partly contributed to improve the accuracy of physiochemical data by using complicated equations. These programs are applied to determine the densities and isochores of fluid inclusions for the Cretaceous Keumhak mine using Choi and So's data (1992) and for the Jurassic Samhwanghak mine using Yun's data (1990). The estimated PoT for Keumhak mine from calculated isochores of coexisting fluid inclusions are $230^{\circ}{\sim}290^{\circ}C$ and 500~800 bar which matche well to the poT estimated by Choi and So ($280^{\circ}{\sim}360^{\circ}C$ and 500~800 bar, 1992). However, the poT for Samwhanghak mine estimated in this study by combining the calculated isochores and sulfur isotope geothermometer data by Yun (1990) are about 4~7 kb at $329{\pm}50^{\circ}{\sim}344{\pm}55^{\circ}C$ which are quite different form the P-T estimates by Yun ($255^{\circ}{\sim}294^{\circ}C$ and 1.2~1.9kb, 1990). This discrepancy caused by misinterpretation of homogenization temperature (Th) of fluid inclusion and by application of inappropriate isochores. The application of homogenization temperature and/or inappropriately selected isochore to determine the trapping PoT condition of ore-deposits should be avoided, particularly for ore-deposits formed at pressures higher than 1~2 kb.

Water Quality in a Drainage System Discharging Groundwater from Sangdae-ri Water Curtain Cultivation Area near Musimcheon Stream, Cheongju, Korea (무심천 인근 상대리 수막재배지에서 지하수 사용 후 배출되는 최종 배수로 물의 수질 특성)

  • The Sangdae-ri riverside around Musimcheon stream, flowing through Gadeok-myon of Cheongju City, is one of the representative strawberry fields employing water curtain cultivation (WCC) in Korea. In this area, annual groundwater use for WCC has been calculated by a few methods. On the assumption that all the water flowing through the final ditch may be mostly composed of groundwater, the discharge rate in it can be used as a good proxy for assessing the groundwater use. However, in the study area, the final ditch was set up in an unpaved state near and parallel to Musimcheon stream. Under such circumstances, the drainwater is likely to be influenced by infiltration and/or inflow of nearby stream. Hence, we examined whether or not stream water has influenced water flowing out through the final ditch in respect of ion concentrations or field parameters such as T, pH and electrical conductivity (EC) values. The period of measuring field parameters and sample collection was from February 2012 through February 2015. The drainwater in the final ditch did not show the average quality of groundwater, but similar quality of stream water in respect of pH, EC, ion contents and water type. From this, it is suggested that measuring the flow rate of the final ditch should not be directly used for assessing groundwater use in the study area. In addition, because of its sensitivity to ambient temperature, water temperature proved not to be appropriate for estimating the interaction between ditch and stream. For accuracy, additional methods will be needed to calculate mixing ratios between stream and ground water within drainage system.

  • Sexing from bovine embryos fertilized in vitro implicates a possibility of the sex controlled cattle production. This study was carried out to produce the sex determined cattle through the embryonic sexing by fluorescence in situ hybridization (FISH) technique. FISH was achieved in in vitro fertilized bovine embryos using a bovine Y-specific DNA probe constructed from the btDYZ-1 sequence. Using this probe, a male-specific signal was detected on 100% of Y-chromosome bearing metaphase specimens. The analyzable rate of embryonic sexing by FISH technique was about 93% (365/393) regardless of embryonic stages. As tested single blastomere by FISH and then karyotype with their biopsied embryos, the accuracy of sex determination with FISH was 97.6%. We tried the embryo transfer with sex determined embryos on 15 cattle. Among them, the 5 cattle delivered calf with expected sex last year.

A Study on the Forest Vegetation of Deogyusan National Park (덕유산 국립공원 삼림식생에 관한 연구)

  • From March 2012 to January 2013, this study was conducted as a part of the project for making a precise electronic ecological zoning map of vegetation on a highly reduced scale of 1 to 5,000 with a view to improving management efficiency of national parks and enlarging the availability of the data produced from the basic research monitoring the resources of national parks. For the research accuracy and rapidity, a vegetation map was specially created for the on-the-site-vegetation research. To make the map more meticulous, we categorized the vegetation database into five groups: broadleaved forest, coniferous forest, mixed forest, rock vegetation and miscellaneous one. After comparing the results of the data built for the vegetation research and the actual research findings, it was made clear that vegetation of both categories was almost the same in case of broad-leaved forest with 72.20% and 78.45% respectively, and also equivalent in other groups like, for example, coniferous forest (16.70%, 13.41%), mixed forest (9.50%, 7.49%) and rock vegetation (0.60%, 0.15%). According to the precise vegetation map produced from the research, the deciduous broad-leaved forest was the most widely prevalent type in the correlated hierarchical classification of vegetation, occupying 65.78% of the overall vegetation. It was followed by mountain valley forest (15.17%), coniferous forest (10.90%), and plantation forest (7.00%) in order. It is particularly noteworthy that Mt. Deogyusan national park has retained a very stable and versatile forest vegetation in the outstanding state since approximately 20% of the mountain turns out to belong to the I grade vegetation conservation classification which contains climax forests, unique vegetation, subalpine vegetation, matured stands which are older than 50 years and etc.
