Cell Cycle Regulation and Antioxidant Activity of Psammaplin A, A Natural Phenolic Compound from Marine Sponge

  • Psammaplin A (PSA), a naturally occurring biophenolic compound has been demonstrated to deliver significant cytotoxicity to many cancer cell lines. In this article, we investigated the effect of PSA on cell cycle progression of lung cancer cells (A549). It was found that PSA could slightly perturb the cell cycle progression of A549 cells and lead to the cell cycle arrest at G2/M phase, indicating PSA might disturb the mitosis process of A549 cells. In addition, inspired by the two phenolic groups in the structure of PSA, the antioxidant activity of it has been evaluated. Although PSA was weak in scavenging the stable free radical 1,1-diphenyl-2-picrylhyrazyl (DPPH), it showed stronger ABTS radical scavenging activity than ascorbic acid in TEAC assay. Furthermore, it was found that PSA could effectively prevent DNA strand scission induced by oxidative stress. These results suggest that PSA have both cell cycle regulation and antioxidant activities. Herein, we suggest that PSA would be a very interesting and promising candidate to be developed as a multi-function drug.

Antioxidant Activity of (8E,13Z,20Z)-Strobilinin/(7E,13Z,20Z)-Felixinin from a Marine Sponge Psammocinia sp.

  • During the course of our screening for bioactive metabolites from marine sponges, EZZ, the inseparable 1:1 mixture of (8E,13Z,20Z)-strobilinin and (7E,13Z,20Z)-felixinin has been found to deliver significant cytotoxicity against some cancer cell lines. In this study, the antioxidant activity of EZZ was first time evaluated by a series of antioxidant assays. It was found that EZZ was weak in scavenging the stable free radical 1,1-diphenyl-2-picrylhyrazyl (DPPH), but it was comparable to ascorbic acid in scavenging ABTS and superoxide radicals. In addition, EZZ could protect DNA from hydroxyl radical-induced strand cleavage. The findings of the present study suggest that EZZ possess certain antioxidant activity, which might help to prevent occurrence of cancer by alleviating the oxidative stress in cells.

발효 천마의 기능성 물질 함량 변화 및 항산화 활성 (Antioxidant Activity and Changes in Major Functional Components of Fermented Gastrodia elata Blume)

  • This study was carried out to investigate the antioxidant activity and changes in major functional components of fermented Gastrodia elata Blume. Fermented G. elata Blume powder by Phellinus linteus repeated thrice ($3^{rd}$ FGP) showed more DPPH radical scavenging activity than a non-fermented G. elata Blume powder (NFGP), and once fermented G. elata Blume powder ($1^{st}$ FGP) at a concentration of 500 and 1,000 ppm. Free radical scavenging activity of $3^{rd}$ FGP was similar to that of BHA at a dose of 1,000 ppm. Moreover, the ABTS radical scavenging activity of the $3^{rd}$ FGP increased compared to NFGP and $1^{st}$ FGP at a concentration of 31.25 ppm. Total polyphenols and flavonoid contents were increased as fermentation progressed. Ergothioneine content was increased more than 8 times in the $1^{st}$ FGP, 3 times in the $3^{rd}$ FGP, respectively than NFGP. In conclusion, this study indicated that the antioxidant activity and functional component contents of G. elata Blume were increased depending on the fermentation number.

Orthosiphon pallidus, a Potential Treatment for Patients with Breast Cancer

  • Objective: Orthosiphon pallidus (O. pallidus), which belongs to the Lamiaceae family, is a popular garden plant that is widely used for the treatment of various diseases, such as urinary lithiasis, fever, hepatitis, cancer and jaundice. The objective of the present work was to investigate the antioxidant free-radical scavenging and the anticancer activities of O. pallidus against human breast-cancer cell lines. Methods: The antioxidant activity of Orthosiphon pallidus aqueous extract (OPAE) was investigated using different models, such as the 1,1-diphenyl-2-picrylhydrazyl (DPPH) and the 2, 2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) models, as were the $Fe^+$ chelation, the hydroxyl radical and superoxide radical scavenging, and total reducing power activities. The anticancer activities of the extract were determined by using the 3-(4, 5- dimethylthiazolyl-2)-2, 5-diphenyltetrazolium bromide (MTT) and the sulforhodamine (SRB) assays on the MCF-7 and the MDA-MB-231 cancer cell lines. Results: The aqueous Orthosiphon pallidus extract showed potent activity in in-vitro models. It significantly inhibited the scavenging of hydroxyl and superoxide radicals, but induced a remarkable $Fe^+$ chelation activity. For both cell lines, the percent cytotoxicity was found to increase steadily with increasing OPAE concentration up to $240{\mu}g/mL$. Conclusion: These results suggest that Orthosiphon pallidus has excellent antioxidant, antimicrobial, and anticancer activities against human breast-cancer cell lines.

Effects of Inhibiting Glycoprotein MUC5AC by Seaweed Ecklonia cava Extract in human Airway Epithelial Cells

  • In the present study, antioxidant and MUC5AC mucin inhibition activities were measured in Ecklonia cava (E. cava) extract. The E. cava extract showed the total polyphenol and flavonoid contents of 607.40±19.44 ㎍ GAE/mg and 13.33±5.28 ㎍ QE/mg, respectively. The free radical scavenging activity of E. cava extract was high in the DPPH radical scavenging activity (RC50 7.08 ㎍/mL) and ABTS+ radical scavenging activity (RC50 4.74 ㎍/mL). Also, we investigated whether E. cava extract affects airway MUC5AC mucin gene expression, production and secretion induced by phorbol 12-myristate 13-acetate (PMA) from NCI-H292 cells. Cells were treated with E. cava extract and then stimulated with PMA for 24 h. The E. cava extract inhibited the gene expression of MUC5AC mucin from NCI-H292 cells. This result suggests that E. cava extract can inhibit the gene expression of mucin induced by PMA through directly acting on airway epithelial cells.

Antioxidant capacity of silkworm pupa according to extraction condition, variety, pupation time, and sex

  • Silkworm pupa has been used as an edible insect with the high quality of protein and unsaturated fatty acids. In this study, antioxidant activities of pupa according to variety, pupation day, sex, and extraction solvent were analyzed. The 30% ethanol extract showed highest radical scavenging activity compared with the DW, hexane, and 70-100% ethanol extracts. In the DPPH and ABTS radical scavenging assay according to the type of pupa, the antioxidant effect was increased in female with the early stage of pupation day. In cell-based assay, reactive oxygen species (ROS) level was decreased in pupa groups by -30 to -50% followed by tert-butyl hydroperoxide (t-BHP) treatment. The ROS levels were significantly reduced in 7th day in each variety. In conclusion, the free radical and ROS scavenging effects were increased in female pupa with the early pupation day. The result could be used for development of bioactive materials using silkworm pupa.

품종을 달리한 오디 추출물의 Resveratrol 및 기능성 성분과 항산화 활성 (Composition of Resveratrol and Other Bioactive Compounds, and Antioxidant Activities in Different Mulberry Cultivars)

  • 오디의 건강기능성 자원으로서의 가치를 높여 이용성을 증가하기 위해 '대성뽕', '수원뽕', '청일뽕'의 오디 3종과 함께 블루베리, 딸기를 시료로 사용하여 기능성 성분의 함량 및 항산화 활성을 측정하고 비교해 보았다. 시료 과실의 60% 에탄올 추출물의 총 phenol성 화합물, 총 flavonoid 함량 및 trans-resveratrol 함량은 '대성뽕'이 가장 높았고, 다음은 '수원뽕'이 높았으며, 총 anthocyanin 함량은 '대성뽕' 다음으로 블루베리가 높았다. 공시된 과실 추출물의 DPPH 및 ABTS free radical 소거능, FRAP에 의한 항산화 활성은 총 phenol성 화합물, 총 flavonoid, 총 anthocyanin 함량과 비례하여 증가하는 경향이었으며, 항산화 활성과 기능성 성분간에는 높은 상관관계가 있었다. 국내산 '대성뽕' 품종 오디는 국제적 건강기능성 과실인 블루베리보다도 기능성 성분과 항산화 활성이 현저하게 높아 이의 추출물은 기능성 식품소재로 이용하여 오디 소비 증진에 기여할 수 있을 것으로 생각되었다.

광양 재래종 복분자의 항산화 활성 (Antioxidant Activities of Native Gwangyang Rubus coreanus Miq.)

  • 본 연구는 광양 재래종 복분자의 항산화 활성을 평가하였다. 주요 유리당으로는 글루코오스와 과당이 각각 $2.83{\pm}0.01%$, $3.49{\pm}0.17%$로 분석되었다. 복분자의 항산화 활성을 나타내는데 기여하는 비타민 C는 0.04 g/100 g, 비타민 E는 0.01 g/100 g으로 분석되었다. 항산화 기능을 대표하는 광양 재래종 복분자내 안토시아닌 함량은 $2.41{\pm}0.01$ g/100 g으로 분석되었다. 열수추출물과 80% 에탄올추출물의 폴리페놀 함량은 열수추출물 $5.99{\pm}0.15%$, 80% 에탄올추출물 $6.20{\pm}0.41%$로 통계적 유의 차이를 나타내지 않았다. 라디칼 소거 활성을 평가한 결과 동일한 농도에서 DPPH 라디칼 소거활성은 80% 에탄올추출물이 $89.29{\pm}0.68%$, 열수추출물이 $65.04{\pm}3.95%$로 나타났고 ABTS 라디칼 소거활성은 각각 $30.22{\pm}2.91%$, $47.65{\pm}0.45%$로 나타났다. 지질과산화 억제 활성은 80% 에탄올추출물이 $23.27{\pm}0.17%$로 열수추출물 $3.58{\pm}0.21%$보다 높게 나타났다. 안토시아닌을 다량 함유한 광양 재래종 복분자는 이를 함유하는 식품의 가공 및 천연 색소로의 이용이 가능할 것이다. 본 연구의 재래종 복분자의 이화학적 특성은 재래종 복분자 식품의 개발에 활용될 수 있고, 항산화 활성이 높은 80% 에탄올추출물은 건강기능식품의 소재로 활용할 수 있을 것으로 판단되었다.

사상 체질 처방의 항동맥경화 효능 비교 연구 -열다한소탕, 양격산화탕, 청심연자탕, 태음조위탕- (Anti-atherosclerotic Effects of Herbal Formulas for Sasang Constitutional Medicine)

  • Objectives : The purpose of this study was to investigate the anti-atherosclerotic effects of 4 herbal formulas for Sasang constitutional medicine (Yeoldahanso-tang: YDHST, Yanggyeoksanhwa-tang: YGSHT, Cheongsimyeonja-tang: CSYJT and Taeeumjowi-tang: TEJWT). Methods : The antioxidant activities of herbal formulas were studied by measuring free radical scavenging activities on ABTS and DPPH. The inhibitory effects on LDL oxidation was evaluated by the formation of TBARS, REM and fragmentation of apolipoprotein B-100 (ApoB). Effects of herbal formulas on macrophage lipid accumulation were determined in native LDL and LPS co-incubated macrophages using Oil Red O staining. Results : The scavenging activities on ABTS and DPPH of herbal formulas were increased in dose-dependent manner (YDHST>YGSHT>CSYJT>TEJWT). Herbal formulas reduced the oxidation properties of LDL induced by $CuSO_4$. YDHST, YGSHT and CSYJT showed strong suppressive effect on LDL oxidation than TEJWT. In addition, YDHST, YGSHT and CSYJT significantly inhibited foam cell formation in LDL/LPS stimulated RAW 264.7 cells. Conclusions : These results demonstrate that YDHST, YGSHT and CSYJT have potentials on anti-atherosclerosis by antioxidative effect and suppressive effect on LDL oxidation.

Effects of Leaf Maturity and Solvent Extract on the Antioxidant Activity of Litsea elliptica

  • Litsea elliptica, a Southeast Asian tree with a rich history of medicinal applications, is attracting increasing research attention. This study investigated the effects of leaf maturity and solvent selection on the extraction of bioactive compounds from L. elliptica leaves, specifically with regard to their antioxidant activity. 2,2'-Azino-bis(3-ethylbenzothiazoline)-6-sulfonic acid (ABTS) method was employed to quantify the free radical scavenging capacity of L. elliptica leaf extracts prepared using three different solvents (n-hexane, ethyl acetate, and ethanol) at three different leaf stages (tender, immature, and mature). These results highlight the significant effects of leaf maturity and solvent selection on the extraction of phenolic compounds and flavonoids from L. elliptica leaves. Ethanol is the most effective solvent for the extraction of bioactive compounds, particularly from mature leaves. The ethanol extraction of tender leaves demonstrated potential for optimizing the antioxidant content. Further research is necessary to investigate the specific factors influencing the observed differences in antioxidant activity between leaves of varying ages and the potential impacts of other bioactive compounds present in the leaves.
