단위발생 유래 생쥐 배아줄기세포의 기능성 심근세포 형성 (Functional Cardiomyocytes Formation Derived from Parthenogenetic Mouse Embryonic Stem Cells)

  • Objective : This study was to establish a reproducible differentiation system from the parthenogenetic mouse embryonic stem (P-mES02) cells into functional cardiomyocytes like as in vitro fertilization mouse embryonic stem (mES01) cells. Materials and Methods: To induce differentiation, P-mES02 cells were dissociated and aggregated in suspension culture environment for embryoid body (EB) formation. For differentiation into cardiomyocytes, day 4 EBs were treated with 0.75% dimethyl sulfoxide (DMSO) for another 4 days (4-/4+) and then were plated onto gelatin-coated dish. Cultured cells were observed daily using an inverted light microscope to determine the day of contraction onset and total duration of continuous contractile activity for each contracting focus. This frequency was compared with the results of DMSO not treated P-mES02 group (4-/4-) and mES01 groups (4-/4+ or 4-/4-). For confirm the generation of cardiomyocytes, beating cell masses were treated with trypsin-EDTA, dispersed cells were plated onto glass coverslips and incubated for 48 h. Attached cells were fixed using 4% paraformaldehyde and incubated with specific antibodies (Abs) to detect cardiomyocytes (anti-sarcomeric ? -actinin Ab, 1 : 100; anti-cardiac troponin I Ab, 1 : 2000) for 1 h. And the cells were finally treated with FITC or TRITC labelled 2nd Abs, respectively, then they were examined under fluorescence microscopy. Results: Rhythmically contracting areas in mES01 or P-mES02 cells were firstly appeared at 9 or 10 days after EBs plating, respectively. The highest cumulative frequency of beating EBs was not different in both treatment groups (mES01 and P-mES02, 4-/4+) with the results of 61.3 % at 13 days and 69.8% at 15 days, respectively. Also, the contracting duration of individual beating EBs was different from minimal 7 days to maximal 53 days. However, DMSO not treated groups (mES01 and P-mES02, 4-/4-) also had contracting characteristics although their frequency was a few compared to those of DMSO treated groups (6.0% and 4.0%). Cells recovered from the spontaneously contracting areas within EBs in both treated groups were stained positively with muscle specific anti-sarcomeric ? -actinin Ab and cardiac specific anti-cardiac troponin I Ab. Conclusion: This study demonstrated that the P-mES02 cell-derived cardiomyocytes displayed similarly structural properties to mES01 cell-derived cardiomyocytes and that the DMSO treatment enhanced the cardiomyocytes differentiation in vitro.

제지슬러지 퇴비화를 위한 요소 첨가효과 (Composting of Paper Mill Sludge by the Addition of Urea)

  • 판지슬러지는 유기물 함량이 높은 반면 중금속 함량은 낮아 농업적 재활용이 유망한 것으로 판단되나 농업적으로 이용하기 전에 농업환경에 대한 위해성을 줄이기 위한 수단으로서 퇴비화가 필요하다. 본 연구에서는 제지슬러지를 효율적으로 퇴비화하기 위한 방안으로서 요소의 첨가 효과를 검토하기 위하여 우선적으로 제지슬러지에 대하여 0~6% 범위의 요소를 첨가한 다음 정체식 시설에서 약 80일 동안 퇴비화를 실시하였고, 정체식 시설에서 우수한 것으로 판명된 처리구는 공장규모의 교반식 시설을 이용하여 현장시험을 실시하였다. 정체식 시설을 이용한 제지슬러지 퇴비화 동안 부숙온도, 탄질율, 양이온치환용량, 식물독성 등을 조사한 결과 요소 3% 이하 처리구가 퇴비화하기에 적절하였다. 이들 처리구중 요소 0%와 3% 처리구들을 대상으로 교반식 시설에서 퇴비화한 결과 부숙과정중 물리적, 화학적, 생물학적 특성의 변화는 정체식 시설과 유사하였다. 비록 교반식의 경우 80일 이후에도 $50^{\circ}C$ 부근의 고온이 유지되었으나, 이화학적 특성의 변화를 고려해 볼 때 정체식 시설에서는 약 50~60일, 그리고 교반식시설에서는 약 30~40일 사이에 안정화되는 것으로 판단된다. 그러나 최종적으로 안정화된 제지슬러지 퇴비의 색깔이 전반적으로 밝아서 퇴비제조시 암색화를 촉진하기 위한 부재료의 첨가가 필요한 것으로 판단되었다.

유리섬유로 보강된 수지에서 제품설계 및 성형조건에 따른 휨의 연구: Part 1. 비결정성 수지 (A Study on the Warpage of Glass Fiber Reinforced Plastics for Part Design and Operation Condition: Part 1. Amorphous Plastics)

  • 사출 성형품에서 발생되는 뒤틀림은 성형공정 중 제품의 불균일한 수축과 잔류응력의 이완으로 인하여 나타나는 현상이다. 휨이나 뒤틀림 현상을 방지하기 위해서 성형품이 강성이 있도록 설계하거나 수지에 유리섬유 등으로 보강하여 사용한다. 본 연구에서는 플라스틱의 성형품에서 성형품의 설계에 따른 뒤틀림 현상을 성형조건 별로 알아보았다. 성형품의 설계는 리브로 보강된 두 종류와 보강 리브가 없는 시편을 사용하였다. 수지는 유리섬유로 보강된 비결정성 수지인 PC와 ABS를 사용하였다. 실험결과 휨은 리브의 설계에 따라 큰 변화를 보였다. 게이트 주변, 게이트 반대쪽, 그리고 흐름방향쪽 등 측정위치에 따라서 휨은 다양하게 나타났다. 이러한 휨의 결과는 유리섬유의 배향과도 크게 관련이 있음을 컴퓨터 해석을 통해 확인할 수 있었다. 리브가 없는 단순 평판에서 휨이 가장 작게 나타났으며, 리브가 흐름방향으로 놓여있는 경우가 휨에 대한 큰 저항을 보였다. 시편에 나타난 휨은 성형조건보다는 제품의 설계에 따라 크게 변하였다. 리브의 설계와 그에 따른 게이트의 위치 설정은 유리섬유의 배향과 직접적인 관련이 있으므로 성형품의 휨 조절에 매우 중요함을 보여주었다.

  • 최근 해양 레저문화 확산에 따라 요트와 같은 레저선박의 설계기술이 요구되고 있다. 현재 까지 요트 설계 및 생산을 위한 국내 기술개발은 해양 레저 선진국에 비해 활발하지 않으나 최근에 요트 설계 및 생산기술기술의 발전으로 인하여, 관련 요소 기술의 개발이 필요시 되고 있다. 본 연구에서는 해양레저 선박으로서 쌍동선 요트에 대한 기본 구조 설계 및 구조 해석을 위한 기법을 제시한다. 요트와 같이 길이 50미터 이하 길이와 폭/비가 12보다 작은 소형 고속 선박의 강도 해석에 대한 규정이나 방법론은 아직 까지 선급 등에서 공식적으로 제안되지 않고 있다. 따라서 본 논문에서 부재의 스캔틀링은 DNV(Yachts, Design Principles, Design Loads, Hull Structural Design)기준과 KR(FRP선 규칙적용 지침)을 사용하였다. 또한, 구조해석 평가에 있어서는 ABS가이드 라인을 적금하여, 구조안정성을 평가하였다. 본 연구를 통하여, 38피트 보급형 쌍동선 요트의 실선 제작에 피드백 역할이 가능하게 되었다. 또한, 연구 결과는 앞으로 고속 쌍동형 요트의 구조설계 및 구조해석에 관련된 기본적인 자료로 유용하게 활용될 것으로 예상된다.

고속철도차량기술개발 표준 모니터링 절차에 대한 고찰 (A Study on Standard Monitoring Process in Development of High Speed Railway Vehicle)

  • According to railway safety regulations for securing the railway vehicle's quality and performance, it can be partly exempted that the performance tests and production inspections on the railway vehicles which are produced and assembled for the purpose of test and research. The reasons of this exemption are for ensuring maximum autonomy and for saving time considered the development period of railway vehicle. However, it was continuously demanded the necessity of supplementation on the confirmation about railway vehicle's quality & performance. Therefore, we added "The monitoring step" which is for securing the quality and performance in high speed railway vehicle's subsystem & compartment to supplement those tests & inspections in development. In this paper, based on a standard ABS(Activity Breakdown Structure) of railway vehicle and a WBS(Work Breakdown Structure) of the railway vehicle system, we investigate the necessity of monitoring in the High speed railway vehicle development. And by comparing and analyzing the essential requirements of international standard, IRIS(International Railway Industry Standard) & TSI(Technical Specification for Interoperability), we try to examine a standard monitoring procedure.

플라스틱 V-벨트 풀리 설계 및 시작품 제작에 관한 연구 (A Study on the Product Design and Prototype Manufacturing of a Plastic V-Belt Pulley)

  • In this study, product design and prototype manufacturing of a plastic water-pump pulley has been tried. The designed model is supposed to be made of 33 % glass reinforced resin of which the tensile strength is 180 MPa, and has 24 ribs on each side to increase its structural strength. Structural analysis under a static load of 300 kgf acting on both edges of the belt has been carried out using a commercial finite element code, MARC. The analysis result showed the maximum effective stress near a rib of designed model would be at most 35 MPa (less than 20% of the tensile strength), therefore, the plastic product would be sufficiently safe under that loading condition. On the basis of the structural analysis, a prototype of the designed model has been manufactured by using the fused deposition modelling (FDM) method which is one of the rapid prototyping (RP) methods, using ABS resin and support materials. The CAD data exported to the RP system in STL format was prepared by a commercial solid modeling software, SolidWorks. It has been proved that the plastic pulley can successfully replace the existing flow-formed steel product.

경쟁적 효소면역 측정법을 이용한 Treponema pallidum 항체 진단시약의 개발 및 평가 (Development and Evaluation of a Competitive Enzyme Immunoassay for the Detection of Antibodies to Treponema pallidum)

  • A competitive enzyme-linked immunosorbent assay(ELISA) for the detection of antibodies to Treponema pallidum(T.pallidum) was developed and evaluated. T.apllidum lysate was immobilized on the surface of microplate wells and horseradish peroxidase labeled human anti-T.pallidum lysate was immobilized on the surface of microplate wells and horseradish peroxidase labeled human anti-T.pallidum was prepared and used as a tracer. The performance of the competitive ELISA was evaluated by using different specimens. The competitive ELISA showed a sensitivity of 100% in a performance panel consisting of serum and plasma with anti-T.pallidum reactivity ranging from negative to strong positive by FTA-ABS test system and 120 plasma samples positive by TPHA. The specificity of the competitive ELISA was 100% in 1,200 plasma samples collected from healthy seronegative blood donors. These results suggest that the competitive ELISA provides an excellent assay method for the detection of antibodies to T.pallidum, and may be particularly useful for serological blood screening of syphilis.

휠 슬립 제어기 및 최적 슬립 결정 알고리즘을 이용한 차량의 최대 제동력 제어 (Maximum Braking Force Control Using Wheel Slip Controller and Optimal Target Slip Assignment Algorithm in Vehicles)

  • The wheel slip control systems are able to control the braking force more accurately and can be adapted to different vehicles more easily than conventional ABS systems. In order to achieve the superior braking performance through the wheel-slip control, real-time information such as the tire braking force at each wheel is required. In addition, the optimal target slip values need to be determined depending on the braking objectives such as minimum braking distance, stability enhancement, etc. In this paper, a robust wheel slip controller is developed based on the adaptive sliding mode control method and an optimal target slip assignment algorithm. An adaptive law is formulated to estimate the longitudinal braking force in real-time. The wheel slip controller is designed using the Lyapunov stability theory and considering the error bounds in estimating the braking force and the brake disk-pad friction coefficient. The target slip assignment algorithm is developed for the maximum braking force and searches the optimal target slip value based on the estimated braking force. The performance of the proposed wheel-slip control system is verified In simulations and demonstrates the effectiveness of the wheel slip control in various road conditions.

  • In this present study, the correlation between subjective preference of essential odors and EEG response were investigated quantitatively. EEG signals were measured from 19 electrodes according to the International 10-20 system (Fpl, Fp2, F2/4, F7/8, Fz, C3/4, Cz, P3/4, Pz, T3/4, T5/6, Ol/2) from 8 healthy males subjects in four odor conditions. Four odor conditions (Rose oil bulgarian, Lemon oil misitano, Jasmin abs, Laverder iol france (KIMEX co. Ltd)) were applied for each subject in the experiment. Through the subjective evaluation, the most pleasant odor for each subject was determined. The power spectrum of ${\alpha}$/${\beta}$ of EEG signals from the most pleasant odor was compared with those from the control condition, which has no odor at all. It was observed that the power spectrum of ${\alpha}$/${\beta}$ of EEG from the most pleasant odor was increased significantly on F3, F3, F4,74 comparing to the control condition. This result indicates that the power spectrum of ai${\beta}$ could be a new index for measuring the levels of pleasantness of odors.

Induction of Immunity Against Hepatitis B Virus Surface Antigen by Intranasal DNA Vaccination Using a Cationic Emulsion as a Mucosal Gene Carrier

  • Delivery of DNA vaccines to airway mucosa would be an ideal method for mucosal immunization. However, there have been few reports of a suitable gene delivery system. In this study we used a cationic emulsion to immunize mice via the intranasal route with pCMV-S coding for Hepatitis B virus surface antigen (HBsAg). Complexing pCMV-S with a cationic emulsion dramatically enhanced HBsAg expression in both nasal tissue and lung, and was associated with increases in the levels of HBs-specific Abs in serum and mucosal fluids, of cytotoxic T lymphocytes (CTL) in the spleen and cervical and iliac lymph nodes, and of delayed-type hypersensitivity (DTH) against HBsAg. In contrast, very weak humoral and cellular immunities were observed following immunization with naked DNA. In support of these observations, a higher proliferative response of spleenocytes was detected in the group immunized with the emulsion/pCMV-S complex than in the group immunized with naked pCMV-S. These findings may facilitate development of an emulsion-mediated gene vaccination technique for use against intracellular pathogens that invade mucosal surfaces.
