Discovery and Functional Study of a Novel Genomic Locus Homologous to Bα-Mating-Type Sublocus of Lentinula edodes

  • The interaction of mating pheromone and pheromone receptor from the B mating-type locus is the first step in the activation of the mushroom mating signal transduction pathway. The B mating-type locus of Lentinula edodes is composed of Bα and Bβ subloci, each of which contains genes for mating pheromone and pheromone receptor. Allelic variations in both subloci generate multiple B mating-types through which L. edodes maintains genetic diversity. In addition to the B mating-type locus, our genomic sequence analysis revealed the presence of a novel chromosomal locus 43.3 kb away from the B mating-type locus, containing genes for a pair of mating pheromones (PHBN1 and PHBN2) and a pheromone receptor (RCBN). The new locus (Bα-N) was homologous to the Bα sublocus, but unlike the multiallelic Bα sublocus, it was highly conserved across the wild and cultivated strains. The interactions of RcbN with various mating pheromones from the B and Bα-N mating-type loci were investigated using yeast model that replaced endogenous yeast mating pheromone receptor STE2 with RCBN. The yeast mating signal transduction pathway was only activated in the presence of PHBN1 or PHBN2 in the RcbN producing yeast, indicating that RcbN interacts with self-pheromones (PHBN1 and PHBN2), not with pheromones from the B mating-type locus. The biological function of the Bα-N locus was suggested to control the expression of A mating-type genes, as evidenced by the increased expression of two A-genes HD1 and HD2 upon the treatment of synthetic PHBN1 and PHBN2 peptides to the monokaryotic strain of L. edodes.

A Study of the Relationship Between Perceived Health Locus of Control and Quality of Life of Cancer Patients. (암환자가 지각하는 건강통제위 성격과 삶의 질에 관한 관계연구 -방사선요법을 받는 암환자를 중심으로-)

  • It has been reported that the cancer patient's quality of life is influenced by the perceived health state, self-esteem, health locus of control, social support, whether there is a pain or not, the stage of a disease, the period of a disease, etc, and however, there has been scarcely the research into the fact at home whose cultural and social backgrounds are entire1y different from those of American and European Countries. Accordingly, the author of this thesis performed this study. considering that it is necessary to know the relations between the health locus of control which make it possible to predict the object's behavior related to health and to make plans to induce the object or patient into sound behavior and the quality of life which is closely related to the cancer patient's health, emotion, society, economy, etc. on the whole. This study, a cross-sectional one, includes 135 subjects of $in{\cdot}out$ patients registered at Y University Hospital whose age are more than 20 years, From these patients, data were collected for two weeks through the questionnaire which content concerns about the quality of life and the health locus of control. All the collected data were processed and analyzed through Student's t-test, ANOVA, and the calculation of Pearson Correlation Coefficient, using the SAS program 1. It appeared that the disposition of health locus of control was mostly inclined to the disposition of powerful other health locus of control($28.37{\pm}4.24$), then inclined to internal health locus of control($27.03{\pm}4.17$), and next to chance health locus of control($19.71{\pm}4.97$) By the way. the disposition of powerful other health locus of control appeared a tittle bit higher than internal health locus of control 2. The degree of the quality of life appeared to be 137.54 points in the average of total points and 3.11 in the average evaluation mark. It appeared that the quality of life was most significantly influenced by a factor of 'relations with neighbors' and least significantly influenced by factors of physical conditions and functions. 3. It appeared that the relation between the disposition of health locus of control and the quality of life has nothing to do with the relation between internal health locus of control and the quality of life(r=.1446, P>.05) and also with the relation between the disposition of powerful other health locus of control(r=.0385, P>.05). In conclusion, in the study it has been found out that there is no correlation between the health locus of control and the quality of life, and however it is sound to induce the cancer patient to internal health locus of control. Therefore, it is necessary that under the special circumstances, the cancer patient's behavior should be predicted, thereby inducing the patient to the sound change of his or her behavior. Also in order that the patient enjoys his or her life satisfactorily while living, It is deemed that some kind of multilateral meditation in health and treatment is necessary so that the patient can feel the relief of pain, better health, etc.

  • Locus of Control of Reinforcement (LOCR)' is considered as one of the most important factors on responsible environmental behaviors(REB). It is defined as 'an individual's perception of his or her ability to bring about change through his or her behavior'. This psychological construct is divided up into two directions: the external locus and internal locus. The internal locus is important as a predictor of REB. Recently, 'The Environmental Action Internal Control Index: EAICI' has been developed and it is a valid and reliable instrument to measure the relationship of two variables. The purposes of this study were to develop a strategy to reinforce the internal locus or to converse external locus to internal locus. and to verify the effects by EAICI. A new strategy, 'Self Control Management Strategy' was based on the characteristics of internal locus and the key idea of this was to self-control and to manage the courses by themselves. EAICI scores of the control group was 99.83. All of them were internal and the LOCR of females was more internal than that of the males. This strategy was applied when the instructions were carried out on seven environmental issues by the experimental group. According to the results. the seven points of the EAICI were increased significantly. Therefore this strategy is helpful in reinforcing the internal locus or to converse external locus to the internal locus.

THE RELATIONSHIP BETWEEN LOCUS OF CONTROL AND THE DISCOMFORT OF THE PATIENTS AT THE INITIAL STAGE OF THE ORTHODONTIC TREATMENT (교정치료 초기 환자의 통제소재와 불쾌감의 관계)

  • This study was attempted to explore the relationship between locus of control and the discomfort of the patients at the initial stage of the orthodontic treatment. Locus of control was measured by 'Locus of Control(LOC) scale for children' and 'Orthodontic Locus of Control(OLOC) scale for children'. The discomfort was measured by 'discomfort index card' in 52 children and adolescent patients who initiate orthodontic treatment. In addition, locus of control of the patients' mothers was measured by 'Orthodontic Locus of Control(OLU) scale for parents'. The results were as follows : 1. The test-retest reliability of 'Locus of Control(LOC) scale for children' and 'Orthodontic Locus of Control(OLOC) scale' was in the moderate to high range. 2. Out of 52 patients, 47 showed moderate to severe discomfort following placement of an initial archwire. The patients showed the most severe discomfort on the first day, and most of the discomfort was manifested within the first 3 days, then decreased until the 7th day. There was no significant difference in the discomfort according to sex and age. 3. The discomfort of the patients was the highest in the morning session when a day was divided into 4 sessions, i.e., morning, afternoon, evening, and night. 4. In the score of 'Locus of Control(LOC) scale for children' and 'Orthodontic Locus of Control(OLOC) scale for children', the group of internal locus of control expressed more discomfort than the group of external locus of control. And there was no significant correlation between locus of control of the patients and that of their mothers. 5. There was no significant difference in the score of locus of control according to sex and me. However, the score of boys tended to be lower than that of girls and the score of primary school students higher than that of middle and high school students.

Relationship between oral health locus of control and oral health behavior promotion in the adolescents (청소년의 구강건강 통제의 소재와 구강건강증진행위와의 관계)

  • Objectives: The purpose of the study was to investigate the relationship between oral health locus of control and oral health behavior promotion in the adolescents. Methods: A self-reported questionnaire was completed by 493 high school students in Jeonju by convenience sampling from March to June, 2014. The questionnaire consisted of fourteen questions of oral health locus of control, fifteen questions of oral health behavior promotion, and seven questions of the general characteristics of the subjects. Data analysis was done by frequency analysis, t-test, ANOVA, and Duncan's multiple range test. Results: Oral health behavior promotion and oral health behavior index of internal locus of control showed a positive correlation, Higher internal locus of control showed a positive oral health behavior promotion. The higher propensity of oral health on the external locus of control showed the negative effects. Conclusions: The oral health internal locus of control was closely related to better oral health behavior in the adolescents.

  • The camshaft locus at camshaft beating in a direct acting OHC valve train system has been investigated using the transient method. Forces applied to the camsfiaft are composed of two components, one is the transfer force between the cam and the tappet, the other is the frictional force. These forces have been calculated using the lumped mass model and the elastohydrodynamic lubrication theory. The alternating direction implicit method has been used for the numerical analysis of Reynolds equation, and 4th order Runge-Kutta method has been used for the transient journal locus analysis. The effects of various load conditions are presented in the form of journal locus. As a result of the analysis, it has been found that camshaft bearings were mainly in the hydrodynamic lubrication condition.

The Relationships between Stress and Health Locus of Control in Nursing College Students (간호 대학생의 스트레스와 건강통제위)

  • Purpose: This study was to identify the relations between stress and health locus of control in nursing college students. Methods: A total of 243 subjects aged between 17 and 27 were selected through convenient sampling. Data were collected with a self reported questionnaire from March 2 to 25, 2013. Collected data were analyzed with SPSS/PC Win 15.0. Results: Differences in stress and health locus of control according to general characteristics were as follows. Stress were significantly different according to gender (t=-2.51, p<.05), grade (F=5.40, p<.01), school record (F=5.72, p<.01), stress solving methods (F=2.62, p<.05). Internal health locus of control was significantly different according to gender (t=2.30, p<.05), grade (F=14.73, p<.001), religion (F=4.63, p<.01), school record (F=5.29, p<.01), economic state (F=5.50, p<.001) and smoking (F=4.17, p<.05). Chance health locus of control was significantly different according to sibling rank (F=2.86, p<.05). Except chance health locus of control, internal heath locus of control and dependence health locus of control have a negative correlation with stress. 15.6% of variance in stress was explained by dependence health locus of control, chance health locus of control, and grade. Conclusion: The findings of this study may be useful in understanding the stress expression of nursing college students and developing more specific programs on personality and self-control.

The Causal Relations of Children′s Behavior Problems, Locus of Control and Mother′s Childrearing Behavior (아동의 또래괴롭힘과 관련변인간의 인과관계 -아동의 행동문제, 내외통제소재와 어머니의 양육행동을 중심으로 -)

  • The purpose of this study was to explore the relationships of mother's childrearing behavior, children's behavior problems, locus of control and peer victimization. The subjects were 360 children of 4th and 6th grade and their mothers. The instruments were Peer Victimization Scale, CBCL, Locus of Control Scale for Children, Childrearing Behavior Scale. The major findings of this study were as follows; (1) The type of peer victimization had a significant difference between girls and boys. That is, boys were more focused on overt victimization than girls. There was no significant gender difference in relational victimization, And there was no significant grade difference in overt and relational victimization. (2) Mother's rejection-restriction childrearing behavior, children's social withdrawal behavior problems, extemal locus of control predicted children's peer victimization. 24% of the variance of children's peer victimization was explained by these variables. (3) Mother's rejection-restriction childrearing behavior was the first contribution factor and had a direct and indirect effect through children's locus of control on children's peer victimization. And children's social withdrawal behavior problems was second contribution factor and had a direct effect on children's peer victimization. Children's locus of control had a direct and indirect effect through children's social withdrawal behavior problems on children's peer victimization. Mother's warmth-acceptance and permissiveness-nonintervention childrearing behavior had indirect effect through children's locus of control on children's peer victimization.

Development of a Sequence Characteristic Amplified Region Marker linked to the L4 Locus Conferring Broad Spectrum Resistance to Tobamoviruses in Pepper Plants

  • To develop molecular markers linked to the $L^4$ locus conferring resistance to tobamovirus pathotypes in pepper plants, we performed AFLP with 512 primer combinations for susceptible (S pool) and resistant (R pool) DNA bulks against pathotype 1.2 of pepper mild mottle virus. Each bulk was made by pooling the DNA of five homozygous individuals from a T10 population, which was a near-isogenic $BC_4F_2$ generation for the $L^4$ locus. A total of 19 primer pairs produced scorable bands in the R pool. Further screening with these primer pairs was done on DNA bulks from T102, a $BC_{10}F_2$ derived from T10 by back crossing. Three AFLP markers were finally selected and designated L4-a, L4-b and L4-c. L4-a and L4-c each underwent one recombination event, whereas no recombination for L4-b was seen in 20 individuals of each DNA bulk. Linkage analysis of these markers in 112 $F_2$ T102 individuals showed that they were each within 2.5 cM of the $L^4$ locus. L4-b was successfully converted into a simple 340-bp SCAR marker, designated L4SC340, which mapped 1.8 cM from the $L^4$ locus in T102 and 0.9 cM in another $BC_{10}F_2$ population, T101. We believe that this newly characterized marker will improve selection of tobamovirus resistance in pepper plants by reducing breeding cost and time.

Further Evidence of Linkage at the tva and tvc Loci in the Layer Lines and a Possibility of Polyallelism at the tvc Locus

  • Three lines of White Leghorn (WL) chickens (IWJ, IWG and IWC) maintained at Central Avian Research Institute, Izatnagar (UP), were used for chorioallantoic membrane (CAM) and liver tumour (LT) assay. Eleven-day-old embryos of each line were partitioned into three groups and inoculated with 0.2 ml of subgroup A, subgroup C and an equal mixture of subgroup A and C Rous sarcoma virus (RSV). Subgroup virus receptor on the cell surface membrane for subgroup A is coded for by tumour virus a (tva) locus and for subgroup C by tumour virus c (tvc) locus. The random association of the genes at the tva and tvc loci in IWJ and IWC line was assessed and the $x^2$-values for phenotypic classes were found to be significant, indicating the linkage between the tva and tvc loci. The linkage value was estimated to be 0.09 on pooled sex and pooled line basis. On the basis of four subclass tumour phenotypes a 4-allele model was proposed for tva locus having $a^{s1}$, $a^{s2}$, $a^{r1}$ and $a^{r2}$ alleles and the frequencies were calculated as 0.47, 0.13, 0.13 and 0.27 for IWJ line, 0.31, 0.33, 0.14 and 0.22 for IWG line and 0.44, 0.11, 0.21 and 0.24 for IWC line, respectively. Similarly, for tvc locus the frequencies of four alleles i.e. $c^{s1}$, $c^{s2}$, $c^{r1}$ and $c^{r2}$ were calculated as 0.42, 0.20, 0.21 and 0.17 for IWJ line, 0.42, 0.17, 0.27 and 0.14 for IWG line and 0.30, 0.21, 0.16 and 0.33 for IWC line, respectively. The $x^2$-values for all classes of observations were not significant (p>0.05), indicating a good fit to the 4-allele model for the occurrence of 4-subclass tumour phenotypes for tva and tvc loci. On the basis of the 2-allele model both tva and tvc locus carries three genotypes each. But, on the basis of the 4-allele model tva and tvc locus carries 10 genotypes each. The interaction between A-resistance and C-resistance (both CAM and LT death) was ascertained by taking the 10 genotypes of tva locus and 3 genotypes of tvc locus by pooling the lines and partitioning the observations into 3 classes. The $x^2$-values for the genotypic classes of CAM (-) LT (+) and CAM (-) LT (-) phenotypes to mixed virus (A+C) infection were found to be highly significant (p<0.01), indicating increased resistance, which indicates the joint segregation of $a^r$ and $c^r$ genes, suggesting the existence of close linkage between the tva and tvc loci. Therefore, an indirect selection approach using subgroup C viruses can be employed to generate stocks resistant to subgroup A LLV, obviating contamination with the most common agent causing LL in field condition.
