  • The distribution ratio of hydrogen cupferrate in $H_2O-CHCl_3$ system was considered as a function of pH ($HClO_4$), ionic strength ($NaClO_4$), and cupferron concentration in perchloric acid media, respectively. The values were independent upon pH (1.50∼3.00 range) and ionic strength (0.1∼2.00 range), but they increased as increasing the cupferron concentration in the acidic media. At the infinite dilution, the thermodynamic distribution ratio between chloroform and aqueous phase was 120. 0. The activity coefficients of hydrogen cupferrate in chloroform solution were determined by the distribution ratio. This activity coefficient may be calculated by using the empirical equation, $-log\;f_{CHCl3}=0.1285C_{CHCl3}+{7.775C^2}_{CHCl3}$ which represents the experimental data quite well for the solution in 0.1 mole/l order of hydrogen cupferrate concentration.

Complete genome sequences of Azoarcus sp. TSPY31 and TSNA42 potentially having biosynthetic ability to produce indigo (인디고 생산능을 가진 Azoarcus sp. TSPY31과 TSNA42의 유전체 분석)

  • Azoarcus are known to contain bacterial strains usually found in contaminated areas. Two strains of Azoarcus sp., TSPY31 and TSNA42, were isolated from oil-contaminated marine tidal flats, and their genomic structures were analyzed. The genomes of both TSPY31 and TSNA42 were composed of a single complete chromosome of 4,572,082 bp (G + C content: 63.2%) and 4,886,934 bp (G + C content: 62.8%), respectively. Both genomes were found to contain two copies of styrene monooxygenases that are predicted to be responsible for converting indole to indigo.

Effects of an Improved Side Vent on Yield of Oriental Melon and Temperature in Single-span Plastic Greenhouse with Roof Ventilation Fans (지붕 환기팬이 설치된 단동 플라스틱 온실에서 개선된 측창형태가 하우스 내 온도 및 참외 수량에 미치는 영향)

  • In this study, the method to attach plastic film on the side vent from inside of greenhouse for the entire length was developed as the way to make crops less stressful while uniformly getting outside air into the greenhouse when ventilating using roof ventilation fans at single-span plastic greenhouse for oriental melon in a low-temperature period. The plastic film was installed from ground to 10cm below from the height where side vent is fully opened. In order to verify that the improved side vent can improve greenhouse environment and fruit yield, it was compared with the control plot of conventional side vent. Both greenhouses were not ventilated until February 25th, 2017. Air temperature in both greenhouses exceeded $40^{\circ}C$ in mid February. Therefore, it is judged that the greenhouse should be ventilated from mid February. Air temperature in the control plot exceeded $30^{\circ}C$ from late April. Therefore, it is judged that the plastic film attached to the inside of side vent should be removed in late April, or in early May at the latest. Soil temperature in the treatment plot in the mid Aril exceeded $20^{\circ}C$, which is suitable for growth, while that in the control plot was still below $20^{\circ}C$. Soil temperature in the control plot finally exceeded $20^{\circ}C$ in late April. The consumption of electricity was 47.2 kWh in the treatment plot, and 48.3 kWh in the control plot, which was no significant difference. The marketable yield of oriental melon in the treatment plot was 5,094kg, which was 23.9% more than that in the control plot, 4,113kg. The marketable fruit ratio was 73.5% in the treatment plot, and that in the control plot was 73.9%, which was no significant difference.

Effect of Incubation Time after Cooling on the Meiotic Spindle and Chromosomes of Mouse Oocytes (냉각 후 배양시간이 생쥐 난자의 방추체와 염색체에 미치는 영향)

  • This study was conducted to determine the effects of incubation time after cooling on mouse meiotic spindle and chromosome alignment and the optimal incubation time for their restoration. Oocytes at the metaphase II were obtained from superovulated mice. Control oocytes were held at 37$^{\circ}C$ during the experiment. Oocytes were rapidly cooled to $0^{\circ}C$, held for 30 minutes, warmed and incubated at 37$^{\circ}C$ for 5, 15, 30, 60 and 120 minutes, respectively. The morphological features of spindle and chromosomes in oocytes were evaluated by immunofluorescent staining. Meiotic spindle of control oocytes exhibited a normal-looking bipolar configuration(barrel-shaped) and highly fluorescent microtubles. The chromosomes were clustered in a discrete bundles at metaphase plate. Disassembly of meiotic spindle and chromosome dispersion were occurred immediately after chilling of oocyte. Fluorescence intensity index(FIS), normal chromosomes aligned and normal spindle configuration were compared according to incubation time at 37$^{\circ}C$. Restoration of a barrel-shaped spindle and normal chromosome alignment was occurring after 5 minutes incubation at 37$^{\circ}C$, improved as a incubation time increased, and decreased gradually after 120 minutes incubation(P<0.05). The optimal incubation time for restoration of meiotic spindle and chromosomes in cooled oocytes was 60 minutes.

Non-radiologic Methods for Predicting Vesicoureteral Reflux in Childhood Urinary Tract Infection (요로감염 환아에서 비방사선학적 방법에 의한 방광요관역류의 조기 예측에 관한 연구)

  • Purpose : Vesicoureteral reflux(VUR) in childhood may be the primary cause of recurrent urinary tract infection and renal scarring. Renal ultrasonography, DMSA, and voiding cystourethrogram are the standard clinical methods for detection of vesicoureteral reflux. But these methods have many disadvantages such as invasiveness and high cost. So, we studied to observe the significance of urine ${\beta}_2$-microglobulin in association with other non-radiologic methods for predictng vesicoureteral reflux. Methods : We evaluated 40 patients with urinary tract infection who were admitted to Korea university Hospital from July 1993 to June 1994. Among them, 24 patients revealed urinary tract infection and vesicoureteral reflux(group A), 16 patients revealed only urinary tract infection(group B). Both groups were compared by presence of fever, hematuria, and proteinuria, positivity of CRP, and level of BUN, Cr, GFR by 99mTc-DTPA, urine ${\beta}_2$-microglobulin, 24 hours urine albumin. Results : 1) Among 24 patients who had vesicoureteral reflux, 14 had unilateral VUR, 10 had bilateral VUR, three kidneys with grade I, nine with grade II, eleven with grade III, eleven with grade IV by classification of International Reflux Study Committee. Among them, 14 patients had renal scar, five with type A, five with type B, four with type C, none with type D by Smellie's classification. 2) The mean of GFR, BUN, Cr, 24hrs urine albumin and the presence of hematuria and proteinuria showed no significant difference between group A and group B. The mean of urine ${\beta}_2$ microglobulin in group A and group B were $283.6{\pm}195.8{\mu}g/l$ and $78.7{\pm}48.5{\mu}g/l$ respectively, showing that group A had a higher value than group B (p<0.01). In case of ${\beta}_2$ microglobulin > $120{\mu}g/l$ and CRP(+), the sensitivity was 93.3% and the specificity is 77.8% for detecting of VUR. In case of ${\beta}_2$-microglobulin>$120{\mu}g/l$ and fever(+), the sensitivity was 92.2%, and the specificity was 62.5% for detecting of VUR Conclusions : If the level of urinary ${\beta}_2$-microglobulin is more than 120ug/l in children with urinary tract infection in association with fever(+) or CRP(+), it can predict VUR. So we can use it for early detection of VUR.

A SERI technique reveals an immunosuppressive activity of a serine-rich protein encoded in Cotesia plutellae bracovirus

  • Polydnavirus genome is segmented and dispersed on host wasp chromosome. After replication, the segments form double- stranded circular DNAs and embedded in viral coat proteins. These viral particles are delivered into a parasitized host along with parasitoid eggs. A serine-rich protein (SRP) is predicted in a polydnavirus, Cotesia plutellae bracovirus (CpBV), genome in its segment no. 33 (CpBV-S33), creating CpBV-SRP1. This study explored its expression and physiological function in the diamondback moth, Plutella xylostella, larvae parasitized by C. plutellae. CpBV-SRP1 encodes 122 amino acids with 26 serines and several predicted phosphorylation sites. It is persistently expressed in all tested tissues of parasitized P. xylostella including hemocyte, fat body, and gut. Its physiological function was analyzed by injecting CpBV-S33 and inducing its expression in nonparasitized P. xylostella by a technique called SERI (segment expression and RNA interference). The expression of CpBV-SRP1 significantly impaired the spreading behavior and total cell count of hemocytes of treated larvae. Subsequent RNA interference of CpBV-SRP1 rescued the immunosuppressive response. This study reports the persistent expression of CpBV-SRP1 in a parasitized host and its parasitic role in suppressing the host immune response by altering hemocyte behavior and survival.

Distillers Dried Grain from Makgeolli By-product Is Useful as a Dietary Ingredient for Growth of Juvenile Sea Cucumber Apostichopus japonicus

  • We tested the effects of various distillers dried grains (DDGs) in a formulated diet on growth and body composition of juvenile sea cucumber Apostichopus japonicus. DDG is a solid residue obtained by filtering an aqueous mixture of fermented rice with Aspergillus oryzae and yeasts. Six isonitrogenous and isocaloric diets (DDG0, DDG-G, DDG-C, DDG-K, DDG-W, and DDG-P) were formulated to contain 20% DDG obtained from different Makgeolli factories. Juvenile sea cucumber (average weight, $2.1{\pm}0.02$ g) were randomly distributed in eighteen 50-L rectangular plastic tanks (40 L of water each) in a seawater flow-through system at a density of 50 juveniles/tank. Three replicate groups of sea cucumber were fed each of the six experimental diets at a feeding rate of 5% body weight per day for 22 weeks. At the end of the feeding experiment, survival and weight gain of the juvenile sea cucumber were not affected by the type of dietary DDG (P > 0.05), and the proximate and amino acid compositions of the whole body were not affected by dietary DDG diets (P > 0.05). These results indicate that rice-based DDG is a potential dietary ingredient that could be used at dietary concentrations of up to 20% for growth of juvenile sea cucumber.

Heterologous Microarray Hybridization Used for Differential Gene Expression Profiling in Benzo[a]pyrene-exposed Marine Medaka

  • Differential gene expression profiling was performed in the hepatic tissue of marine medaka fish (Oryzias javanicus) after exposure to benzo[a]pyrene (BaP), a polycyclic aromatic hydrocarbon (PAH), by heterologous hybridization using a medaka cDNA microarray. Thirty-eight differentially expressed candidate genes, of which 23 were induced and 15 repressed (P<0.01), were identified and found to be associated with cell cycle, development, endocrine/reproduction, immune, metabolism, nucleic acid/protein binding, signal transduction, or non-categorized. The presumptive physiological changes induced by BaP exposure were identified after considering the biological function of each gene candidate. The results obtained in this study will allow future studies to assess the molecular mechanisms of BaP toxicity and the development of a systems biology approach to the stress biology of organic chemicals.

Microstructure and Physical Properties of Porous Material Fabricated from a Glass Abrasive Sludge (유리연마슬러지를 사용한 다공성 소재의 미세구조 및 물리적 특성에 관한 연구)

  • A porous material with a surface layer was fabricated from glass abrasive sludge and expanding agents. The glass abrasive sludges were mixed with expanding agents and compacted into precursors. These precursors were sintered in the range of $700-900^{\circ}C$ for 20 min. The sintered porous materials had a surface layer with smaller pores and inner parts with larger pores. The surface layer and closed pores controlled water absorption. As the expanding agent fraction and the sintering temperature increased, the porosity and pore size increased. The porous materials with $Fe_2O_3$ and graphite as the expanding agents had a low absorption ratio of about 3% or lower while the porous material with $CaCO_3$ as the expanding agent had a higher absorption ratio and more open pores.

A study on the turning-motion of T/S SAEBADA in shallow water (실습선 새바다호의 천수역 선회운동에 관한 연구)

  • The authors has predicted the maneuvering characteristics of a fishing vessel in deep water using Kijima's empirical formula in a previous study. Since the Kijima's empirical formula was developed by a regression analysis of merchant vessels which have dimensions ($C_b$, L/B, etc.) that are different from those of fishing vessels, it was possible to make a prediction approximately even with inaccurate estimation. In this study, the authors estimated the turning-motion characteristics of a model ship of fisheries training ship in shallow water based on the results of its previous study. The turning-motion characteristics of the model ship in shallow water was found out through quantitative analysis according to the water depth to ship draft ratio (H/d). In conclusion, the turning-motion characteristics of the model ship had significant changes immediately after an H/d 1.5, and this result will be helpful for sailing in shallow water.
