Isolation and Biocontrol Potential of Bacillus amyloliquefaciens Y1 against Fungal Plant Pathogens

  • This study was performed to investigate thermophilic bacteria from soil having broad antifungal spectrum against Rhizoctonia solani, Colletotrichum gloeosporioides, Phytophthora capsici, Fusarium oxysporum f.sp. lycopersici, and Botrytis cinerea. One isolate selected could resist heat shock of $60^{\circ}C$ for one hour, and had broad antifungal activity in dual culture assay against all tested fungal pathogens and was identified as Bacillus amyloliquefaciens Y1 using 16S rRNA gene sequence. Further investigation for antifungal activity of bacterial culture filtrate (BCF) and butanol crude extract (BCE) of various concentrations showed broad spectrum antifungal activity and fungal growth inhibition significantly increased with increasing concentration with highest growth inhibition of 100% against R. solani with 50% BCF and 11 mm of zone of inhibition against R. solani with 4 mg BCE concentration. Treatment of butanol crude extract resulted in deformation, lysis or degradation of C. gloeosporioides and P. capsici hyphae. Furthermore, B. amyloliquefaciens Y1 produced volatile compounds inhibiting growth of R. solani (70%), C. gloeosporioides (65%) and P. capsici (65-70%) when tested in volatile assay. The results from the study suggest that B. amyloliquefaciens Y1 could be a biocontrol candidate to control fungal diseases in crops.

Comparative Analysis of the Biological Characteristics of Ephedrus plagiator (Nees) and Aphidius ervi Haliday according to Different Aphid Hosts (기주에 따른 목화검정진디벌(Ephedrus plagiator (Nees))과 진디벌(Aphidius ervi Haliday)의 생물학적 특성 비교)

  • To investigate the biological control of Aulacorthum solani, a comparative analysis was carried out using an indigenous natural enemy, Ephedrus plagiator, and an exotic parasitoid, Aphidius ervi. Lifespan, spawning periods, number of mummies, number of offspring, and developmental periods of the two parasitoids on Aulacorthum solani and Acyrthosiphon pisum were studied at 15, 20, 25, and $30^{\circ}C$ with a photoperiod 16 h (L): 8 h (D). The lifespan of E. plagiator (5.3 days) was relatively higher than that of A. ervi (2.3 days) when these parasitoids were reared on A. solani at $15^{\circ}C$. Similarly, the spawning period of E. plagiator (5.3 days) was longer than that of A. ervi (2.2 days). When the two aphid parasitoids were provisioned with A. solani, the numbers of E. plagiator mummies at 15, 20, 25, and $30^{\circ}C$ were 71.5, 41, 15.7, and 7.7, respectively, whereas at the same temperatures, the numbers of A. ervi mummies were 22.1 16.3, 6.2, and 0.4, respectively. In contrast, when provision with A. pisum, the numbers of E. plagiator mummies at 15, 20, 25, and $30^{\circ}C$ were 70.3, 69.8, 34.3, and 8.4, whereas the numbers of A. ervi mummies were 93.4, 71.2, 34.8, and 14.5, respectively. The numbers of E. plagiator offspring emerging at 15, 20, 25, and $30^{\circ}C$ were 42.1, 36, 11.6, and 0, whereas the numbers of A. ervi offspring emerging were relatively lower at 19.6, 13.5, 3.7, and 0.1, respectively. By comparing these results, it can be concluded that E. plagiator is a more efficient parasitoid of A. solani, whereas A. ervi is more efficient on A. pisum.

Antifungal, Nematicidal and Antioxidant Activity of the Methanol Extracts Obtained from Medicinal Plants

  • The nematicidal, antifungal and antioxidant activities of methanol extracts from six Vietnamese native medicinal plants were evaluated by various assays in vitro. Of the plant extracts tested, Terminalia nigrovenulosa was found to possess the highest activity when compared to the others. The leaves and bark of T. nigrovenulosa showed strong inhibitory activity against Meloidogyne incognita and Fusarium solani. The DPPH (1,1-Diphenyl-2-picryl-hydrazyl) radical scavenging, reducing power and total antioxidant activities of T. nigrovenulosa bark were higher than that of the remainder plant extracts. Chitinase activity of these plants was also investigated using SDS-PAGE. The results obtained in the present study indicate that T. nigrovenulosa leaf extracts are the greatest potential source as nematicides and fungicides for the control of M. incognita and F. solani. In particularly, T. nigrovenulosa bark extracts could be used as a potential source of commercially viable levels of natural antioxidant.

Effect of Seeding Epth on Severity of Damping-off Ginseng Seedlings Caused by Rhizoctonia solani (번종 깊이가 인삼의 모잘록병 발생에 미치는 영향)

  • Incidence of damping-off callsed by Rhizoctonia solani was 0.6-10.9% at "Yangjik" seedbed in Pocheon, Korea. The seedbeds where the lengths of etiolated stems (underground portion) of ginseng seedlings were 0.78-1.25 cm showed 0.8-3.2% of the disease, while 6.9-10.9% disease incidence was observed at the seedbeds with the longer etiolated stem (1.89-2.26 cm). The pathogen produced a typical girdle symptom on the etiolated portion of ginseng stems close to the soil surface. The deeper the seeds were sown, the more the disease occurred in pot soil inoculated with the pathogen, AG 2-1, showing 18.4, 27.4 and 32.9% of damping-off at the seeding depth of 1, 2 and 4 cm, respectively. Cuticle layers of colored stems (over ground portion) were well - developed to be 42.8, 58.0, and 55.0 um in thickness compared to the etiolated stems with 8.5, 15.0 and 8.0um for seedling, 2 year-old, and 3 year-old ginsengs, respectively, when the disease occurred. In the seedling and 2 year-old ginseng, the colored stems were more rigid than the etiolated. There was however, no difference in rigidness of the stem of the 3 year-old ginseng where the disease is not severe as in seedlings and 2 year-old ginseng plants.ng plants.

  • The study was carried out to confirm the changes of soil physico-chemical properties and population of Fusarium solani by soil managements at preplanning fields for ginseng cultivation. Soil porosity and aggregation had been significantly increased during the managements while exchangeable nitrogen content and bulk density of the soil had been decreased. Available phosphate and exchangeable potassium content, in addition, seemed to be slightly increased. And soil aggregation showed positive correlation with clay, organic matter, soil moisture, and Ca content in the soil but negative with K content. Decrease rate of NH4-N and NO3-N content after soil managements were 55% and 41% in average, respectively. And better decrease effect for NH4-N was obtained in sand loam soil whereas no effect for NO3-N with soil textures. The more ploughing seemed to result in less propagules of F solani in the soil, however there was non significant decrease in population of the pathogen after the soil management. Number of F. solani in soil was significantly less in the fields where gramineous and leguminous crops had been grown as a precrop than other crops tested. Meanwhile there was no correlation between soil texture and population of the pathogen in the soil.

  • New thirty derivatives of 5,6-dihydro-2-trifluoromethyl-1,4-oxathiin carboxanilide as substrate(S) were synthesized and their fungicidal activities in vivo against rice sheath blight(Rhizoctonia solani) and wheat leaf rust(Puccinia recondita) were examined. The structure activity relationships(SAR) between the activities($pI_{50}$) and a physicochemical parameters of substituents(X) at the phenylcarbamoyl group were analyzed using the adaptive regression analysis method. The 3-methoxy, 11, 3-isopropyloxy, 13 and 3-isopropyl substituent, 25 as X on the phenylcarbamoyl group exhibited the most highest fungicidal activity against the two fungi. The fungicidal potency of the (S) against Puccinia recondita was higher than Rhizoctonia solani. In case of Rhizoctonia solani, the molecular hydrophobicity(${\pi}>0$) and resonance effect(R<0) by meta-alkyl substitutents with electron donating were important factors in determining fungicidal activity. And the HOMO energy(HOMO>0), ABSQ, sum of absolute values of the atomic charges on each atom and specific polarizability(Sp.Pol<0) of (S) were significantly influential towards fungicidal activity against Puccinia recondita.. The interaction between (S) and receptor agonist from the based on SAR studies proceeds through charge-control reaction, and conditions to show higher activity has been also discussed.

Isolation, Physico-chemical Properties, and Biological Activity of New Thiopeptide Antibiotics, Kimorexins

  • An isolate 90-GT-302, identified as Kitasatosporia kimorexae, was found to produce antibiotics that induce mycelial swelling in Magnaporthe grisea, and Fusarium solani. The strain produced at least 5 antibiotics. Among them, the main active compound designated as kimorexin A was isolated and its physico-chemical properties and biological activities were examined, and as a result was found to be of the thiopeptide antibiotic. A comparison between the properties of kimorexin A and those of the known thiopeptide antibiotics led us to conclude that kimorexin A was a new thiopeptide polythiazolyl antibiotic. Kimorexin A showed a narrow antimicrobial spectrum against very limited genus of phytopathogenic fungi. It prevented host plants from infections of Rhizoctonia solani and absolute parasitic fungi, such as Sphaerotheca fuliginea and Puccinia recondita, almost completely at the treatment concentration of approximately 20 ppm.

Development of a Selective Medium for the Fungal Pathogen Cylindrocarpon destructans Using Radicicol

  • The soil-borne ascomycete fungus Cylindrocarpon destructans causes ginseng root rot disease and produces various secondary metabolites such as brefeldin A and radicicol. The slow growth of this fungus compared with other plant pathogenic and saprophytic fungi in soil disturbs isolation of this fungus from soil and infected ginseng. In this study, we developed a selective medium for C. destructans using radicicol produced by this fungus. Supplementing 50 mg/L of radicicol to medium inhibited the mycelia growth of other fungi including Botrytis cinerea, Rhizoctonia solani and Alternaria panax, but did not affect the growth of C. destructans. In addition, conidia germination of other fungal species except for C. destructans was inhibited in submerged culture supplemented with radicicol. This medium provides a very efficient tool for isolating C. destructans and also can be used as an enrichment medium for this fungus.

Numerical Identification of Streptomyces fIaveus Producing Antibiotic Substances Inhibitory to Plant Pathogenic Fungi

  • The actinomycete strain A 11 was antagonistic to plant pathogenic fungi Phytophthora capsid and Magnaporthe grisea. Based on the diaminopimelic acid (DAP) type and morphological characteristics examined by scanning electron microscopy, the strain A 11 was confirmed to belong to the genus Streptomyces. Based on Willcox probability and similarity level, the strain A 11 was numerically identified as Streptomyces flaveus using TAXON program of Ward and Goodfellow. Antibiotic production of S. flaveus strain A 11 was most favorable when cultured on glycerol yeast extract peptone (GYP) agar for 20 days at $28^{\circ}C$. The crude antibiotics from solid GYP agar cultures of the strain A 11 were most effective against Phytophthora capsici and Sclerotinia sclerotiorum among the fungi tested. Antifungal activity of the antibiotics against Alternaria solani, Botryosphaeria dothidea, Cercospora capsici, Magnaporthe grisea, and Rhizoctonia solani was somewhat high, whereas Colletotrichum gloeosporioides and Fusarium oxysporum f. sp. cucumerinum were rarely inhibited even at high concentrations.

Effect of a Microbial Product on the Control of Soilborne Diseases of Turfgrasses (미생물제에 의한 잔디의 토양전염병 방제 효과)

  • A microbial product composed of three antagonistic fungal isolates (Aspergillus sp., Penicillium sp. and Trichoderma sp.) and three bacterial isolates (Arthrobacter sp., Bacillus sp., and Pseudomonas sp.) was tested for the control of Pythium blight caused by Pythium sp., brown patch by Rhizoctonia solani (anastomosis group(AG) 1-1) and large patch by R. solani (AG 2-2) of turfgrasses. Cultures of the antagonistic fungi and bacteria varied in the effectiveness in reducing disease severity of Pytium blight and brown patch on bentgrass. The antagonistic fungal and bacterial isolates were mixed and cultured at 20-$25^{\circ}C$ for 3 days in a growth medium, and the diluted solution of the microbial culture was applied under the field conditions after inoculation of the above turfgrass pathogens. The treated turfgrass was incubated at 28$^{\circ}C$ in a growth chamber. In this experiment, Pythium blight was almost completely controlled and brown patch was slightly decreased by the microbial product, while no control was observed in large patch of zoysiagrass. In zoysiagrass treated with the microbial culture, thatch accumulation was notably reduced.
