Improvement of Seed Germination in Rosa rugosa (해당화의 종자 발아 연구)

  • Rose seed shows low germination percentages (about 20%) because it has a high amount of substances that inhibit germination in the pericarp. We investigated the effect of orientation of achene, the day after pollination (DAP), and cold storage with or without half-cut in achene for the germination percentage in R. rugosa. Germination percentages of intact or half-cut achenes were investigated in a 16-hour photoperiod at $25^{\circ}C$ room on basal MS medium for two weeks. In germination percentage, maximum 100% was measured within one week when half-cut achenes were cultured on an orientation that the embryos facing to the light. Half-cut achenes at 90 DAP were germinated 100% regardless of cold storage. Various LED lights (red, blue, yellow, green, and white) were illuminated over the half-cut achenes to gain the effect of light color. Germination percentage of R. rugosa seeds under blue LED reached the greatest with 90% within one week of culture and these seedlings were the best with a steady growth rate. It is concluded that half-cut achenes would be an effective method to improve seed germination in R. rugosa without stratification or scarification. This system could be applied to breeding studies in rose cultivars.

Histochemical Study on the Mucous Secreting Cells in the Gastrointestinal Mucosae of Rana rugosa according to Hibernation (동면에 따른 옴개구리(Rana rugosa) 위장관 점액분비세포에 관한 조직화학적 연구)

  • The mucosubstances in the gastrointestinal mucous secreting cells of Rana rugosa were detected histochmeically during pre-hibernating, hibernating, post-hibernating and active phases. The results of the observation were as follows: 1. The mucosubstances in the gastrointestinal mucous secreting cells of active frog were strongly PAS-active in stomach, PAS-active and alcianophilic at pH 2.5 in small intestine and alcianophilic at pH 2.5 in large intestine. 2. The PAS-active mucosubstances in the gastric surface epithelial cells were increased remarkably during hibernation. 3. The alcianophilic mucosubstances at both pH 2.5 and pH 1.0 were decreased remarkably in the goblet cells of small intestine during hibernation, but a little PAS-active ones were increased. 4. The alcianophilic mucosubstances at pH 2.5 were decreased remarkably and a lttle PAS-active ones also were done in the goblet cells of large intestine during hibernation. 5. The increases of the contained quantity of mucosubstances in the gastric surface epithelial cells during hibernationi may have theeffects of preventing cohesions of gastric mucosae and suppressing activities of gastric acid and enzymes. The mucosubstances neutral acidity in the intestine during hibernation may be secreted, because of acidity being done near neutrality in its lumen, due to remarkable decrease of intestinal juices and gastric acids.

Review of Experimental Researches on Gastrointestinal Activity of Agastache rugosa (Fisch. & C. A. Mey.) Kuntze and Pogostemon cablin (Blanco) Benth. (곽향(藿香) 및 광곽향(廣藿香)의 위장관 효능에 대한 실험연구 고찰)

  • The pharmacological rationale of Agastache rugosa (AR) or Pogostemon cablin (PC), which have been used in traditional Korean medicine to treat dampness pattern or syndrome in gastrointestinal tract, was investigated on the gastrointestinal disorders. In-vivo model studies that examined the effect on the gastrointestinal disorders of AR or PC were collected. They were classified into disease-induced in-vivo models or non-disease in vivo models. The target disease, animal species, induction method, administration, and outcomes (changes in morphological and histological parameter, or blood and fluid) of each study were analyzed. The therapeutic mechanism of AR or PC extract was evaluated by the induced diseases and the changes in outcomes. There were contradictory reports on gastrointestinal motility of AR or PC in disease non-disease in-vivo model. AR or PC inhibited gastrointestinal motility in disease model of increased gastrointestinal motility, while promoted motility in disease model of decreased gastrointestinal motility. AR or PC also inhibited inflammatory changes in gastrointestinal inflammation model. These results suggest that the bidirectional regulation of gastrointestinal motility and the improvement of gastrointestinal inflammatory disorders might underpin traditional therapeutic effect of AR or PC, that is effect to resolve dampness of gastrointestinal tract.

Analysis of Essential oil and Variation of Estragol content in Different Growth Stages of Agastache rugosa Kuntze (배향초의 생육시기에 따른 정유성분 분석 및 Estragol 함량의 변이)

  • This study was carried out to compare the essential oil and its composition, and estragol content at different growth stages in the local collections of A. rugosa. Estragol was the major constituent in essential oil of all collections tested. Estragol content in essential oil was $82.3{\sim}89.1%$ in leaves, $81.1{\sim}89.2%$ in inflorescence, $72.0{\sim}45.2%$ in stem and there was no significant difference among nine local colletions. At full bloom stage, essential oil content was significantly higher than other growth stages and the highest content of estragol in essential oil was appeared at this stage. Especially, the content of essential oil in the inflorescence of Mokpo collection was 3.3%, which was the highest among the collections. Mokpo and Jindo collections produced much more essential oil and estragol than the other collections both at full bloom and ripening stages.

A Study for the Utilization of Wild Herbaceous Species -Effects of Gibberelic Acid Treatment on Seed Germination- (야생초류의 자원화를 위한 연구 -지베렐린($GA_3$) 처리가 몇 야생초류의 종자발아에 미치는 영향-)

  • To examine the effects of gibberelin treatment on seed germination of six wild herbaceous species -Dicentra spectabilis, Aster pekinensis, Agastache rugosa, Elsoholtzia ciliata, Boehmeria tricuspis and Pulsatillia cerrma var. chaerophyiloides - in Kangwon-do, pretreated seeds of five species were sowed on plastic pots with mixed soil(sand:peat moss:perlite: vermiculite, 1:1:1:1, v/v) in 23,April 1995. Seeds were soaked in 25,50 ppm gibberelin solution and ground water during 24 hours. Pretreated seeds of Pulsatiilia cerrma var. chaerophylloides were sowed on nursery soil in 1, June 1995. Seed pretreatments in 31, May 1995 were same method. Seed germination rate was measured and compared among species and treatments. Seed germination rate of Dicentra spectabilis, Agastache rugosa and Elsoholtzia ciliata was differed higly significant among pretreatments ,but those of that of Aster pekinensis, Boehmeria tricuspis and Pulsatillia cerrma var chaerophylloides did not significant at 5% level. Gibberelin pretreatment raised seed germination rate of Dicentra spectabilis and Agastaohe rugosa and proper concentration of GA$_3$ was 25ppm and seed germination rate of Elsoholtzia ciliata was highest at GA$_3$ 50ppm plot. The utilization of wild herbaceous species were discussed.

Hepatoprotective Effect of Catechin Isolated from the Root of Rosa rugosa Thunb (해당화 뿌리에서 분리한 Catechin의 간보호효과)

  • The root of Rosa rugosa has been used in folkloric medicine as a treatment agent for diabetes. In the present study, we investigated whether (+)-catechin isolated from this plant can change the activities of hepatic drug metabolizing enzymes in rats treated with bromobenzene. Pretreatment with (+)-catechin gave no effects on the activities of aminopyrine N-demethylase and aniline hydroxylase, enzymes forming toxic bromobenzene epoxide intermediates and glutathione Stransferase, an enzyme removing toxic epoxides. However, the activity of epoxide hydrolase, an enzyme detoxifying the bromobenzene toxic intermediates was mildly recovered by (+)-catechin treatment.

Effectiveness and Ecological Implications of Anuran Defenses against Snake Predators

  • The aim of this study was to characterize antipredator tactics of anurans and to evaluate the effectiveness of these tactics for predator avoidance in real confrontations. Two types of experiments were conducted. In one experiment, one predator and one prey were placed together for one hour in a small confined space (one-to-one interaction). In another experiment, one predator and several prey were placed together for one day in a large enclosure in a field (field-based interaction). The prey consisted of three anuran species, Rana nigromaculata, R. rugosa, and Bombina orientalls: a snake species, Rhabdophis tigrinus tigrinus, was used as a predator. Results of both experiments demonstrated a range in antipredator responses of the frogs, from toxicity and warning coloration, coupled with slow responses in Bombina to little (or only slight) toxicity, crypsis, and fast take-off responses to the predator in the ranids. oth ranid species exhibited lower survival(57%) than Bombina (95%) in the field-based interaction, suggesting that motor responses of the palatable prey due to attacks of the predator ultimately limited their survival. The jumping of the ranids increased the activity of the predator, which became more likely to strike. Simple crouching(seen in R. rugosa and B. orientalis) and chemical defense (in Bombina) reduced predatory attacks.

Genetic Relationship of Genus Rosa Germplasm and Genetic Diversity of Rosa rugosa in Korea (장미속 유전자원의 유전적 관계와 국내 해당화의 유전적 다양성)

  • Fifty-nine accessions of 23 species in genus Rosa were collected, and 15 accessions of Rosa rugosa were collected throughout 10 regions of Korea. Their genetic relationship was investigated by using morphological analysis and RAPD marker. The morphological analysis was measured for 7 quantitative and quantified for 4 qualitative traits. RAPD analysis obtained a total of 959 polymorphic bands by using twenty primers. Morphological analysis classified most according to the rose section system except for several species. The cluster analysis of genus Rosa based on RAPD data could identify the subgenus $Platyrhodon$ and $Eurosa$. The subgenus $Eurosa$ separated five sections; $Gallicanae$, $Cinnamomeae$, $Pimpinellifoliae$, $Synstylae$ and $Caninae$. Correlation analysis between morphological and RAPD analysis showed low significance ($r$ = 0.35). The accessions of R. rugosa belonged to the section $Cinnamomeae$ clustered into three groups at genetic distance ranging from 0.28 on the base of RAPD analysis. In conclusion, the genetic relationship of the genus Rosa was consistent to the previously reported rose section system, and domestic collections of $R.$ $rugosa$ were separated from 3 groups on the base of RAPD marker.
