Optimized pH condition of protein extraction of Gastrodia elata Blume by alkaline method (알칼리에 의한 천마 단백질 추출의 최적 pH 조건)

  • This study investigated the optimum pH condition for the efficient extraction of protein from Gastrodia elata Blume. Five extraction pH values (8, 9, 10, 11, and 12) and three precipitation pH values (2, 4, and 6) were used. The protein content, browning degree, and recovery yield of the protein obtained under each pH condition were determined. Most of the G. elata Blume was made up of carbohydrates, and its protein content was also high. The amount of the extracted protein increased according to the increase in the extraction pH, but did not significantly differ between pH 8 and pH 9. The browning degree of the protein significantly increased as the extraction pH increased. The greatest amount of protein was precipitated at pH 4, the recovery yield of which was also the highest. As a result, it was found that the combination of extraction pH 9 and precipitation pH 4, which resulted in a 38.7% recovery yield and a low browning degree, is the optimum condition for the efficient extraction of protein from G. elata Blume.

Effects of Araliaceae Water Extracts on Blood Glucose Level and Biochemical Parameters in Diabetic Rats (두릅나무과 열수추출물이 당뇨흰쥐의 혈당과 혈액 성분에 미치는 영향)

  • This study was conducted to investigate the effects of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts on blood hemoglobin, $HbA_{1c}$, levels and biomarkers in the streptozotocin (STZ)-induced diabetic rats. Male Wistar rats divided into normal and diabetic groups. The diabetic groups subdivided into the control group (DM) and Araliaceae water extracts supplemented groups: Aralia elata (AE), Acanthopanacis cortex (AC) and Ulmus davidiana (UD). The extracts were supplemented in diet base on 11.42 g of raw Araliaceae/kg diet for 7 weeks. The diabetes was induced by injecting 572 (55 mg/kg B.W., i.p.) once 2 weeks before sacrifying. Relative weights of liver were significantly lowered in the DM group compared to the normal group, whereas those of kidney and heart were significantly increased in the DM group. Supplementation of the Araliaceae water extracts improved reduced liver weights in STZ-induced diabetic rats. Blood glucose level was significantly higher in the DM group than in the normal group, whereas insulin contents were significantly lowered in the DM groups. However, these parameters were normalized in the An, AC and UD supplemented groups, respectively. Blood hemoglobin and $HbA_{1c}$ levels were significantly higher in the DM group than in the normal group. When all of Araliaceae water extracts were supplemented to the diabetic rats lowered hemoglobin and $HbA_{1c}$ levels. Red blood cell, white blood cell and Lymphocyte were significantly higher in the DM group than in the normal group. The supplementation of Araliaceae family water extracts significantly lowered these parameters compared to the DM group. MCV, MCH contents were declined in the DM group, while the supplementation of Aralia elata, Acanthopanacis cortex and Ulmus davidiana water extracts elevated of these contents in STZ-induced diabetic rats. Accordingly, these results indicate that Aralia elata, Aeanthopanacis corex and Ulmus davidiana water extracts would seem to improve the blood biomarkers in STZ-induced diabetic rats.

Mycena subpiligera sp. nov., a Symbiotic Species from China Associated with the Seed Germination of Gastrodia elata

  • Mycena subpiligera, a new taxon in sect. Fragilipedes that can strongly enhance the germination efficiency of Gastrodia elata seeds, was discovered in subtropical areas of China. As revealed by a morphological comparison with related Mycena species as well as maximum likelihood (ML) and Bayesian phylogenetic analyses based on sequences of the internal transcribed spacer (ITS) and the large subunit (LSU) regions of nuclear ribosomal RNA, the new taxon can be distinguished from phenotypically similar and phylogenetically related species. Optimal cultural conditions for M. subpiligera basidiomata are reported, and the germination rate of the new species is compared with that of M. citrinomarginata.

The ptimum temperatures during cultivation period of Gastrodia elata according to growth stages (천마 생육단계별 변온에 의한 최적온도 및 재배기간)

  • This study was carried out to investigate the optimum temperature and the cultivation period according to the different growth stages of Gastrodia elata (G. elata). The growth period for the indoor cultivation of G. elata is divided into four stages that require specific temperatures during the enlargement of the tuber. The optimum temperatures and cultivation periods during the growth stages of G. elata were observed to be $20^{\circ}C$ for 30 days during the mycelial growth stage (MGS), $25^{\circ}C$ for 120 days during the tuber formation stage (TFS), $6-24^{\circ}C$ for 60 days during the tuber enlargement stage (TES), and $5^{\circ}C$ for 30 days during the dormant stage (DS). The total cultivation period was shortened by 120 days in the indoor cultivation facilities by reduction of 30 days from the mycelial growth stage, addition of 30 days to the tuber formation stage, and reduction of 120 days from the dormancy stage as compared to the outdoor field cultivation. These results provide a basis for a growth model that permits year-round cultivation of G. elata.

Effect of Immature Rhizome Productivity according to Harvest Times-based Treatment Method for Seed Production of Gastrodia elata (천마 종자 생산을 위한 수확시기별 처리방법에 따른 자마 생산력 효과)

  • This study was investigated the effect of immature rhizome production according to harvest times-based treatment method for seed production of Gastrodia elata. The results revealed that when the tuber weight of G. elata harvested in spring (GEHS) was ${\geq}100g$, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was 90.9%-94.8%, 3.1%-5.4%, and 10.1%-15.3%, respectively. When G. elata harvested in fall (GEHF) was treated at a low temperature for 4 weeks or more, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was 70.4%-87.6%, 2.2%-2.6%, 8.7%-9.5%, respectively. Therefore, to produce seeds and immature rhizomes, GEHS must have tubers of more than 100 g, whereas GEHF requires breaking dormancy by low-temperature treatment for 4 weeks or more. Compared with those of GEHS, the rate of artificial fertilization, protocorm formation, and immature rhizome formation was lower in GEHF; however, it was higher than those in the natural germination state. Thus, it can be expected that G. elata can be produced throughout the year by ensuring that the seeds and immature rhizomes of G. elata are produced using a constant tuber weight and by breaking dormancy with low temperature treatment.

  • This study was conducted to determine the optimum culture conditions for inducing callus and regenerating plantlets from cultured leaf tissues of Aralia elata. Young leaf tissues(1cm) of A. elata plant were cultured on MS medium supplemented with 2, 4-D and Thidiazuron. Embryogenic callus was induced along the leaf veins, more efficiently on the medium containing 1.0mg /1 Thidiazuron in 4 weeks after culture initiation. Calli were subcultured to proliferate on MS media containing 2, 4-D, Dicamba, Picloram, and Thidiazuron. Callus was better proliferated on the medium containing Dicamba than on the others.. However, callus subcultured on the medium containing Thidiazuron was more embryogenic and light green-colored, of which some showed embryoid-like structure on the surface. Hormone-free medium was more efficient to regenerate plantlets than media supplemented with Kinetin, BA, and Thidiazuron.

  • Effect of Aralia elata extract on experimentally alloxan-induced diabetes ill rabbits and toxicity of mouse were studied; $LD_50$ of Aralia elata extract (A. E. E.) was 5.5g/kg intraperitoneally ill mice. A. E. E. 800mg/kg exhibited more excellent hypoglycemic effect after 6 days and 14 days in 200mg/kg, compared to the control group. Blood urea nitrogen level was significantly decreased after 6 days in 800mg/kg. and blood levels of total cholesterol and GPT activity were showed more significant decreasing effect ill 800mg/kg than 200mg/kg.

  • Crude oils in the rhizoma of fresh and dried Gastrodia elata were obtained by a simultaneous steam distillation and extraction(SDE) method using n-pentane/diethyl ether (1 : 1) as solvent, and their volatile constituents were analyzed by gas chromatography-mass spectrometry(GC/MS) A total of 39 volatile flavor constituents (11 acids, 13 alcohols,6 hydrocarbons,7 carbonyls,2 esters) and 25 constituents (6 alcohols, 13 acids, 4 hydrocarbons, 1 carbonyl, ester) were identified in the fresh and dried Gastrodia elate respectively. The major volatile components of the fresh and dried sample were hexadecanoic acid(66.78%, 50.72%), 9-hexadecenoic acid(8.07%, 9.58%), heptadecanoic acid(2.01%, 0.13%), pentadecanoic acid(6.41%, 4.94%), p-cresol(1.43%,0.52%) and cyclododecene(1.83%, 6.00%).

  • An antihyperglycemic compound in a model of alloxan induced diabetic rats was isolated from the root bark of Aralia elata. The compound identified to be oleanolic acid $28-O-{\beta}-_D-glucopyranoside$ was active at a dose of 100 mg/kg p.o. in the rats. It also has increased the contents of liver glycogen which were lowered by injection of alloxan in the rats.

Quality Characteristics of $Kalopanax$ $pictus$ and $Aralica$ $elata$ Shoot according to Their Salt Conditions (염장조건에 따른 엄나무와 두릅 순의 품질특성)

  • The quality characteristics of $kalopanax$ $pictus$ and Aralica elata shoots during their preservation were investigated according to the salt conditions and storage temperatures to improve the use of the shoots. The results were as follows: the pH of $kalopanax$ $pictus$ shoots duing their storage did not changes with 10% and 12% brine at $4^{\circ}C$ and with 12% brine at $10^{\circ}C$. The pH of $Aralica$ $elata$ shoots is at $4^{\circ}C$ tended to be similar to the $kalopanax$ $pictus$ shoot and decreased at the storage temperature of $10^{\circ}C$. The salinity of the $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots tended to increase during their storage and did not change significantly as their storage temperature changed. The L values of $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots tended to decrease gradually during their storage, and the salt concentrations appeared high. The b value of the $kalopanax$ $pictus$ shoot tended to increase and that of the $Aralica$ $elata$ shoots, to decrease gradually. The a value tended to increase gradually but did not differ with variations in the storage temperatures and salt concentrations. The strength and the hardness of the $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots tended to decreased gradually during their storage and highest in 10% brine at $4^{\circ}C$, and those of $Aralica$ $elata$ shoots were highest in 8-10% brine at $4^{\circ}C$. The Chlorophyll contents of the $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots tended to decrease during their salt storage and did not change significantly as the temperatures and salt concentrations changed. Therefore, it can be concluded that the quality of $kalopanax$ $pictus$ and $Aralica$ $elata$ shoots can be maintained when the are stored in 10% brine at $4^{\circ}C$.
