지리오갈피나무 엽 신 배당체 Chiisanoside의 생물학적 효능 (The Biological Activity of a New Glycoside, Chiisanoside from Acanthopanax chiisanensis Nakai Leaves)

  • A new glycoside was isolated from Acanthopanax chiisanensis Nakai (Araliaceae) leaves and its biological activity was investigated. The new glycoside was tentatively assigned the structure of Asecotriterpenoid glycoside, $C_{48}H_{76}O_{19}$ m.p. $208~209^{\circ}$ and named chiisanoside. Chiisanoside exhibited non-toxic effects and significant antihistaminic activity. It was found that chiisanoside showed the antidiabetic activity against epinehrine-and alloxan-induced diabetes, decreased the toxicity of $LD_{50}$ by ephedrine hydrochloride and promoted the elimination of chloramphenicol from blood. Chiisanoside also increased the survival rate in rats intoxicated by carbon tetrachloride from death and led to re-establishment of normal enzymatic function. In the histopathological studies, chiisanoside improved fatty degeneration and parenchymal cell necrosis of the liver induced by carbon tetrachloride in rats.

한국특산 섬오갈피나무 및 지리산오갈피나무의 정유성분 GC/MS 분석 (GC/MS Analysis of Volatile Constituents from Endemic Acanthopanax koreanum and A. chiisanensis in Korea)

  • 한국특산 섬오갈피나무 및 지리산오갈피나무의 잎과 줄기에서 추출한 주요 정유성분으로, 섬오갈피나무의 경우에는 ${\delta}-3-carene$ (31.34%), l-limonene (17.01%), ${\beta}-elemene$ (4.53%), trans-p-menth-2-ene-1,8-diol (3.13%), 1,8-cineole (4.73%), 1-dodecen-3-yne (2.64%), (Z)-3,7-dimethyl-1,3,6-octatriene (3.21%), 지리산오갈피나무의 경우에는 ${delta}-3-carene$ (14.78%), limonene (7.24%), t-ocimene (7.22%), ${\alpha}-terpinolene$ (8.76%), ${gamma}-elemene$ (4.32%), ${\beta}-selinene$ (7.72%), veridifloral (3.25%), dodecane (2.44%) 등의 성분함량이 높음을 알 수 있었다. 특히 ${\delta}-3-carene$는 섬오갈피나무 및 지리산오갈피나무에서 가장 높은 함량을 나타내었다.

Analysis of Flavonoid Contents in the Fruits of Acanthopanax Species using HPLC

  • Analysis of flavonoid contents in the fruits of Acanthopanax species (A. chiisanensis, A. divaricatus, A. koreanum, A. senticosus, and A. sessiliflorus) was conducted by high performance liquid chromatography. A Discovery$^{(R)}$ C18 ($4.6{\times}250$ mm, 5 ${\mu}m$) column was used with a gradient mobile phase of water and acetonitrile (90 : 10 to 60 : 40 for 60 min) and UV detection was conducted at 350 nm. The contents of rutin, hyperin, quercetin, afzelin, and kaempferol were 0.063~0.540, 0.494~7.480, 0.584~0.704, 0.388~0.567, 0.190~0.471 mg/g, respectively, in the fruits of Acanthopanax species. Total content of flavonoids in the fruits of Acanthopanax species was highest in those of A. chiisanensis. Furthermore, hyperin was the most abundant compound in the fruits of Acanthopanax species. Consequently, our results demonstrate that the fruits of Acanthopanax species containing flavonoids have promising potential as a new income source of agriculture and industry in medicinal natural products, health supplements, and beverages.

Determination of Hyperin in the Fruits of Acanthopanax Species by High Performance Liquid Chromatography

  • The content of hyperin in Acanthopanax species was determined by high performance liquid chromatography (HPLC). Hyperin was quantified by a reverse-phase column with elution program [initially gradient solvent (acetonitrile : water = 85 : 15 to 80 : 20 for 20 min), then isocratic solvent (acetonitrile : water = 80 : 20 for 20 min), and finally gradient solvent (acetonitrile : water = 80 : 20 to 65 : 35 for 20 min)]. UV detection was conducted at 210 nm. The content of hyperin in the fruits of Acanthopanax was measured in the species A. chiisanensis (2.04 mg/g), A. sessiliflorus (1.13 mg/g), A. divaricatus (0.98 mg/g), A. koreanum (0.75 mg/g) and A. senticosus (0.05 mg/g). The content of hyperin in A. chiisanensis was higher than that of other Acanthopanax species.

지리오갈피 발효주의 숙성 중 품질변화 (Developing the Quality of Traditional Fermented Liquor Using Acanthopanax chiisanensis)

  • 지리오갈피(Acanthopanax chilsanensis)를 이용한 지역 특산품 개발 및 소비촉진을 목적으로 지리오갈피 발효주를 개발하여 5~10℃, 60일 동안 숙성시키면서 발효숙성주의 품질특성 등을 검토하였다. 숙성 기간 동안 발효숙성주 중의 환원당 함량은 약간 감소하였으나 총산의 함량은 아주 완만하게 증가하였다. 지리오갈피 뿌리, 줄기 및 열매로 제조한 발효숙성주 중의 유기산 함량은 3종류의 숙성주 모두 lactic acid, formic acid, malic acid, tartaric acid 및 acetic acid의 순으로 그 농도가 높게 검출되었다. 그리고 유리당은 3 종류의 숙성주 모두 sucrose, glucose 및 mannose 등의 유리당이 검출되었으며 이들 중 glucose 함량이 모든 시험구에서 가장 높게 나타났다. Acanthoside-D 함량은 줄기 첨가 발효숙성주에서 64.12 ppm으로 가장 높게 나타났으며 발효주와 발효숙성주의 acanthoside-D 함량을 비교해 보면 발효숙성주에서 약간 높아지기는 했으나 거의 변화가 없는 것으로 나타났다. 발효숙성주 중의 총 유리아미노산 함량은 뿌리, 줄기 및 열매의 발효숙성주에서 각각 1,858.9, 2,855.1 및 2,672.2 mg/l로 나타났다. 숙성주의 색도를 검토한 결과 뿌리와 줄기를 첨가한 숙성주에서는 거의 유사한 L, a, b값을 나타내어 발효숙성주의 색깔도 비슷하게 나타났으나, 열매를 첨가한 숙성주에서는 L값이 낮고, a값이 높아 붉은 색을 많이 띄었다.

가시오갈피의 생약학적 연구 (Pharmacognostical Studies on "Ga Si O Gal Pi")

  • "Ga Si O Gal Pi" is one of the Korean crude drugs used mainly to cure arthritis, acute gastritis, and beriberi. With regard to the botanical origin of "Ga Si O Gal Pi", it has been considered to be Acanthopanax species of Araliaceae, but there was no pharmacognostical confirmation on it. To clarify the botanical origin of "Ga Si O Gal Pi", the anatomical characteristics of the branch of Acanthopanax species growing wild in Korea and Japan, A. chiisanensis, A. divaricatus, A. koreanum, A. rufinerve, A. senticosus, A. seoulense, A. sessiliflorus, A. sieboldiaum were studied. As a result, it was clarified that "Ga Si O Gal Pi" was the branch and rarely the bark of branch of Acanthopanax senticosus.

  • The content of chiisanoside in the Acanthopanax Species was determined by reversed-phase high performance liquid chromatographic method. Chiisanoside was separated from the other components in the plant extracts using Zorbax 300 SB $C_{18}$ column with gradient elution of acetonitrile. Identification of chiisanoside was carried out by comparison in the LC/MS spectrum of separated peak from extract with that of standard. By HPLC analysis in this experiment, Acanthopanax species could be classified into two groups based upon the content of chiisanoside-one with low concentration of chiisanoside, such as A. senticosus and A. koreanum, and another with high concentration of chiisanoside, such as A. senticosus f. inermis, A. Divaricatus var. albeofructus, and A. chiisanensis.

한라산 화구내 식물과 토도식물 (Plants in the crater of Halla-San (Mt. Halla) and Plants in To-do (Island))

  • (1) Plants in the crater of Halla-San (Mt. Halla), a conical volcano that originated in the Tertiary period, were studied on three different occasions; July 1937, July 1955, and August 1957, and noted 52 familites, 149 species. In the latest study undergone in 1957, the authors collected 108 species. Notable species among them are Geranium Shikokianum Matsum which has not been reported in Korea, Clematis Chiisanensis Nakai, Lactuca Chelidonifolia Makino, and Platanthera Maximowiczana Schelchter which have not been reported in Jeu-Do(Island) so far. (2) Plants in To-Do(Island), territorial island of Jeju-do, were also studied. The authors noted Canavalia lineata D.C.non-reported Species in Korea, usually distributed along Southern Honshu, Shikoku, and Kyushu of Japna, Ryukyu and Taiwan. The overfall flora of the island is composed of 30 families, 44 Species; 5 Species of wood plants and 39 species of herbs, and is mostly of littoral plants.

PSL, TL 및 ESR 분석에 의한 감마선 조사 한약재의 검지 특성 (Detection Characteristics of Gamma-Irradiated Korean Medicinal Herbs by Using PSL, TL, and ESR)

  • 한약재의 방사선 조사여부를 확인하기 위해 도라지와 오가피에 $0{\sim}10\;kGy$의 감마선을 조사하고 PSL, TL 및 ERS 특성을 비교 검토하였다. PSL을 측정한 결과 비 조사된 한약재에서 한계치($T_1$)인 700보다 낮은 photon count 값을 보이면서 negative로 판정되었고, 0.15 kGy 이상 조사구는 한계치($T_2$)인 5,000보다 높은 photon count 값을 나타내면서 positive로 표시되어 방사선 조사된 것을 확인할 수 있었다. 또한 시료로부터 mineral을 분리하여 TL의 glow curve와 TL ratio($TL_1/TL_2$)를 확인하였다. 방사선 조사가 되지 않은 도라지와 오가피는 TL ratio가 0.007 이하의 값을 나타내었으며, 조사구(1.0 kGy 이상)에서는 0.314 이상의 TL ratio를 나타내었다. 도라지와 오가피는 본 연구에서 설정한 TL ratio의 한계치 0.1보다 높은 수치를 나타내어 방사선 조사여부 확인이 가능하였다. ESR 측정을 통한 방사선 조사 한약재 검지에서는 cellulose 라디칼에서 유래된 특이한 라디칼 신호를 보여주었고, 이는 도라지 시료에서 더욱 뚜렷하게 확인할 수 있었다. 이상의 결과에서 PSL, TL 및 ESR 분석은 도라지와 오가피의 방사선 조사 여부 확인에 적합한 방법임을 확인할 수 있었다
