Polymerization and Preparation of Functional Ophthalmic Material Containing Carbon Nanoparticles

  • This research is conducted to create a functional hydrogel ophthalmic lens containing nanoparticles. Carbon nanoparticles and PEGMEMA are used as additives for the basic combination of HEMA, MA, and MMA, and the materials are copolymerized with EGDMA as the cross-linking agent and AIBN as the thermal initiator. The hydrogel lens is produced using a cast-mold method, and the materials are thermally polymerized at $100^{\circ}C$ for an hour. The polymerized lens sample is hydrated in a 0.9 % saline solution for 24 hours before the optical and physical characteristics of the lens are measured. The refractive index, water content, contact angle, light transmittance, and tensile strength are measured to evaluate the physical and optical characteristics of the hydrogel lens. The refractive index, water content, contact angle, UV-B light transmittance, UV-A light transmittance, visible light transmittance, tensile strength and breaking strength of the hydrogel lens polymer are 1.4019~1.4281, 43.05~51.18 %, $31.95{\sim}68.61^{\circ}$, 21.69~58.11 %, 35.59~84.26 %, 45.85~88.06 %, 0.1075~0.1649 kgf and 0.1520~0.2250 kgf, respectively. The results demonstrate an increase in refractive index, tensile strength and breaking strength and a decrease in contact angle and light transmittance. Furthermore, the visible light transmissibility is significantly increased at PEG 10 %. It is clear that this material can be used for high-performance ophthalmic lenses with wettability, ultraviolet ray blocking effect, and tensile strength.

Conditions for protoplast formation and fusion of the killer yeast (Killer 효모의 원형질체 형성 및 융합조건)

  • Auxotrophic mutant were isolated from wild types by the treatment with NTG as a mutagen, and the conditions of protoplast formation for them were established. The protoplasts of killer yeast Saccharomyces cerevisiae K52 were formed to the level of above 70% when cells grown for 20 hr in PM medium were treated with 200 unit/ml Lyticase 50,000 at $30^{\circ}C$ for 60 min after pretreatment of 50 mM 2-mercaptoethanol in 10mM potassium phosphate buffer (pH 7.5) containing EDTA and 0.6 M sorbitol for 15 min. Also, the protoplast of the recipient S. cerevisiae S 29 were formed to the level of above 85% as it was cultured to the log phase of 24 hr in PM medium under the same conditions. The fusion frequency between the protoplast of killer yeast S. cerevisiae K 52 and the protoplast of recipient S. cerevisiae S 29 was reached to $8.2\times 10^{-6}$ when the hypertonic regeneration medium embeded with the fused protoplasts after mixing the parental protoplasts to 10$^{8}$ cells/ml in SP buffer containing 20 mM $CaCl_{2}$ and 30% PEG 6,000 for 15 min at $30^{\circ}C$ were incubated.

Uplift Bearing Capacity of Spiral Steel Peg for the Single Span Greenhouse (온실용 나선철항의 인발저항력 검토)

  • This study examined the uplift bearing capacity of spiral steel pegs according to the degree of soil compaction and embedded depth in a small-scaled lab test. As a result, their uplift bearing capacity increased according to the degree of soil compaction and embedded depth. The uplift bearing capacity under the ground condition of 85% compaction rate especially recorded 48.9 kgf, 57.9 kgf, 86.2 kgf and 116.6 kgf at embedded depth of 25 cm, 30 cm, 35 cm and 40 cm, respectively, being considerably higher than under other ground conditions. There were huge differences in the uplift bearing capacity of spiral steel pegs according to the compaction conditions of ground. Their maximum uplift bearing capacity was 116.6 kgf under the ground condition of 85% compaction rate and at embedded depth of 40 cm, and it is very high considering the data of spiral steel pegs. It is thus estimated that wind damage can be effectively reduced by careful maintenance of ground condition surrounding spiral steel pegs. In addition, spiral steel pegs will be able to make a contribution to greenhouse structural stability if proper installation methods are provided including the number and interval according to the types of greenhouse as well as fixation of plastic film. The findings of the study indicate that the optimal effects of spiral steel pegs for greenhouse can be achieved at embedded depth of more than 35cm and compaction degree of more than 85%. The relative density of the model ground in the test was 67% at compaction rate of 85%.

  • Ethanol-tolerant strain, S. eerevisiae BUI a26 ($alc^r thr^-$) and gJucoamylase-producing strain, S diastatieus AI5a6 (STA+ hom-) were prepared by means of genetic manipulation, Protoplast fusion was carried out to introduce STA gene from AI5a6 strain to BUla26 strain, Protoplast formation was shown at 0,8 M sorbitol and 200 Jig/ml to 400 Jig/ml zymolyase treatment for 2 hours incubation, Fusion frequency was $ 3.25 {\times} 10^{-3}$ to the regenerated protoplast number using PEG 6000 for 90 min incubation. The excellent fusants with genotype of STA- $alc^r thr^-$ hom+/STA+ ($alc^s thr^+$ hom- (2n), F7 and FIO, were selected by ethanol-tolerant, ethanol fermentation, and glucoamylase production tests, Glucoamylase production of AI5a6 showed 2,7 units, but 4.2 or 8.4 units for F7 or FIO fusant at $30^{\circ}C$, Ethanol fermentation from 32% glucose by BUla26 was 14,0%(v/v) in fermentaion medium for 5 days incubation, but 14.5% or 15,0% for F7 or FIO strain, respectively. Ethanol fermentation from 5% starch was 2,0% by F7, or 1.8% by FIO strain in fermentation medium for 5 days fermentation.

Effects of a Digital Pegboard Training Program With Visual and Auditory Feedback on Hand Function and Visual Perception in Patients With Stroke (시각, 청각 피드백을 이용한 디지털 페그보드 훈련 작업치료 프로그램이 뇌졸중 환자의 손 기능 및 시·지각에 미치는 영향)

  • Purpose : This study aimed to investigate the effects of a digital pegboard training program with visual and auditory feedback on hand function and visual perception in stroke patients. Methods : Twenty two participants were randomly assigned to an experimental or control group. The experimental group received training using a digital pegboard training program with visual and auditory feedback (n=11), while traditional occupational therapy was administered to the control group (n=11). Hand function was assessed before and after the intervention using the Nine-Hole Peg Test and manual function test (MFT), while visual perception was assessed using the Motor-Free Visual Perception Test-3rd edition (MVPT-3). Results : Following the intervention, both the experimental and control groups showed significant improvements in performance in the Nine-Hole Peg Test and MVPT-3 (p<.05). The improvement on both tests was significantly greater in the experimental group than in the control group (p<.05). Conclusion : The results suggest that digital pegboard training with visual and auditory feedback may improve hand function and visual perception in stroke patients. Therefore, this intervention can be effective in occupational therapy to aid the recovery of stroke patients.

  • Orthodontic treatment is obviously the first choice to get aligned teeth cause of conservative treatment. But it is impossible to get highly esthetic result only orthodontic treatment in case of peg lateralis, discrepancy tooth size, discolored teeth or fractured teeth. As all treatment does, especially in esthetic treatment it is always required interdisciplinary diagnosis and treatment plan; orthodontics, periodontology, prothetics, implant therapy. As also, the treatment should be maintained long-term treatment with stable and harmonious state by esthetic satisfaction and functional occlusion those who needs various dental care as interdisciplinary treatment. This case is the laminate treatment with the orthodontic treatment on 37 ages/ Female patient who has cross bite at anteriors and premolar area and abrasion on centralis at right maxilla. By orthodontic treatment aligned teeth positions, it makes functional occlusion. As laminate treatment with minimum teeth preparation is able to release anterior esthetics. Follow up check for 2 years has been performed since the final prosthetics delivery to the patient. No sign of relapse, fracture of laminate were detected.

Effect of polydeoxyribonucleotide with xenogeneic collagen matrix on gingival phenotype modification: a pilot preclinical study

  • Purpose: To investigate the effect of xenogeneic collagen matrix (XCM) with polydeoxyribonucleotide (PDRN) for gingival phenotype modification compared to autogenous connective tissue graft. Methods: Five mongrel dogs were used in this study. Box-type gingival defects were surgically created bilaterally on the maxillary canines 8 weeks before gingival augmentation. A coronally positioned flap was performed with either a subepithelial connective tissue graft (SCTG) or XCM with PDRN (2.0 mg/mL). The animals were sacrificed after 12 weeks. Intraoral scanning was performed for soft tissue analysis, and histologic and histomorphometric analyses were performed. Results: One animal exhibited wound dehiscence, leaving 4 for analysis. Superimposition of STL files revealed no significant difference in the amount of gingival thickness increase (ranging from 0.69±0.25 mm to 0.80±0.31 mm in group SCTG and from 0.48±0.25 mm to 0.85±0.44 mm in group PDRN; P>0.05). Histomorphometric analysis showed no significant differences between the groups in supracrestal gingival tissue height, keratinized tissue height, tissue thickness, and rete peg density (P>0.05). Conclusions: XCM soaked with PDRN yielded comparable gingival augmentation to SCTG.

Development of Yeast-Vector System for Eukaryotic Gene Cloning - Optimum Condition for Intact Yeast Cell Transformation and Plasmid Stability in the Transformants - (진핵생물 유전자 조작을 위한 효모 vector계 이용에 관한 기초연구 -생효모 형질전환 최적조건과 숙주별 plasmid안정성에 관하여 -)

  • In order to obtain the optimum conditions for intact yeast cell transformation in the various yeast host-vector systems, 3 yeast plasmid vectors, YRp7, YEpl3 and YIp5 were introduced into 5 yeast hosts, Saccaromyces cervisiae Dl3-1A, DKD-5D, DBY-746, MC-16 and S2022D with various transformation conditions, and plasmid stabilities in all the transformants were also observed. The highest transformation frequencies in all the host-vector system were obtained in the 16 hour Cultured cell (5.4 $\times$ 10$^6$ - 2.4 $\times$ 10$^8$cells/$m{\ell}$) treated with 0.1-0.2 M lithium chloride in 0.1 M tris-HCl (pH 7.6), 35% polyethylene glycol 4000, and heat-shocked at 42$^{\circ}C$ for 5 minutes after 60 minutes of induction. The intact cell transformation got more transformation frequency in DKD-5D (YRp7) and DBY-746 (YEpl3) than protoplast transformation, but reverse tendency was observed in DKD-5D (YEp13) and Dl3-lA (YRp7). The transformants, D13-1A (YRp7) and DKD-5D (YRp7) were very unstable in selective medium, with 80 to 85% of the transformants losing the plasmid after 70 generations, but the transformants, DKD-5D (YEpl3) and DBY-746 (YEpl3) were quite stable, with 35% of the transformants losing the plasmid.

  • Inositol 1,4,5-triphosphate($InsP_3$) is a second messenger for mobilizing intracellular $Ca^{2+}$. It can be dephosphorylated by soluble and particulate forms on $InsP_3$ 5-phosphatase, or phosphorylated to produce inositol 1,3,4,5-tetrakisphosphate($InsP_3$) by $InsP_3$ 3-kinase. These enzymes represent possible targets for the regulation of the $InsP_3/InsP_4$ signal. $InsP_3$ 3-kinase which catalyses th ATP-dependent phosphorylation of $InsP_3$ was purified from bovine brain tissue. All operation were carried out at $4^{\circ}C$. Fresh tissure was homogenized and centrifuged. The supernatant was pooled. Proteins were precipitated from 10% polyethylene glycol, and suspended solution was applied to DEAE cellulose column for chromatography. As the result of above procedure, two isozymes of $InsP_3$ 3-kinase, I and II were obtained. Each isozyme was applied to Matriz green gel, Calmodulin-Affigel 15 column and subsequent phenyl-TSK HPLC column. Specific activites(SA) and fold of puriety were observed at each purification step of chromatography. At DEAE cellulose chromatography, SA were I, 0.6 and II, 4.8 nM/min/mg, and folds were I, 17.2 and II, 16.6. At Matrix green gel chromatography, SA were I, 18 and II, 11 nM/min/mg, folds were I, 62.1 and II, 38.0. At calmodulin-Affigel 15 column chromatography, SA were I, 19 and II, 13 nM/min/mg, folds were I, 65.5 and II, 44.8. Finally $InsP_3$ kinase I and II were purified 3,103-fold and 2,310-fold, and SA were I, 900 and II, 670 nM/min/mg, respectively. SDS-polyacrylamide gel electrophoresis elucidated 3 distinct fractions of Mr of 145,000, 85,000 and 69,500 from isozyme I, and 2 distinct fractions of Mr of 79,000 and 57,000 from isozyme II.

Production of Vitamin $B_{12}$ by Using Protoplast Fusion between Bacillus natto and Bacillus megaterium (Bacillus natto 및 Bacillus megaterium의 원형질체 융합에 의한 Vitamin $B_{12}$의 생산)

  • This study was conducted to breed a high vitamin $B_{12}$ producer by the fusion of protoplasts between Bacillus natto and Bacillus megaterium. Auxotrophic mutants of Bacillus natto SH-34 ($thr^-try^-rif^r$) and Bacillus megaterium BK-13 ($arg^-ade^-lys^-str^r$) which showed high protease activity and production of vitamin $B_{12}$, respectively, were isolated for the fusion experiment. Protoplasts were induced by incubating the cells with lysis solution containing $500{\mu}/ml$ lysozyme, and the ratio of protoplast and regeneration formation were ranged from 99% and 67%, respectively. Fusion frequencies of fusants between Bacillus natto SH-34 and Bacillus megaterium BK-13 were appeared in the ranges of $1.0{\times}10^{-5}$ under the treatment of 30% PEG 6000 containing 3% PVP. The fusant, MNF-72 showed the highest product yield of $7.85{\mu}g/g-cell\;vitamin\;B_{12}$ in production medium. For the improvement of productivity, the immobilization of fusants with sodium alginate was carried out. In batch and continuous fermentation systems, the productivity were determined to be $0.58{\mu}g/ml.hr\;and\;0.80{\mu}g/ml.hr\;vitamin\;B_{12}$ under optimum condition, respectivity.
