Applicability Evaluation after Treated with Consolidant of 0.8T0.2E1G_3wt0.08% and 1T1G_7wt0.08% on Marble Surface (강화제 0.8T0.2E1G_3wt0.08%와 1T1G_7wt0.08%의 대리암에 대한 적용성 평가)

  • In this study, marble that consist mainly of calcite were used for applicability evaluation after treated with consolidant such as alkoxysilane series of 0.8T0.2E1G_3wt0.08% and 1T1G_7wt0.08%. The position and intensity of X-ray diffraction peaks are no change and increasing trend after treated with two consolidants. The results of scanning electron microscopic analysis, morphology of rope type and homogeneous surface of platy or leaflike form observed on the marble after treated with 0.8T0.2E1G_3wt0.08% and 1T1G_7wt0.08%, respectively. Brightness of surface after treated with consolidant are changing slightly dark. Shore hardness and ultrasonic velocity values show increasing after treated on the marble surface with two consolidants.

  • Due to the inconsistent fluctuation of blood supply for transfusion, much attention has been paid to the development of artificial blood using other animals. Although mini-pigs are candidate animals, contamination of mini-pig T cells in artificial blood may cause a major safety concern. Therefore, it is important to analyze the cross-reactivity of IL-7, the major survival factor for T lymphocytes, between human, mouse, and mini-pig. Thus, we compared the protein sequences of IL-7 and found that porcine IL-7 was evolutionarily different from human IL-7. We also observed that when porcine T cells were cultured with either human or mouse IL-7, these cells did not increase the survival or proliferation compared to negative controls. These results suggest that porcine T cells do not recognize human or mouse IL-7 as their survival factor.

Motukoreaite and Quintinite-3T from Sinyangri Formation, Jeju Island, Korea (제주도 신양리층에서 산출하는 Motukoreaite와 Quintinite)

  • Motukoreaite and quintinite-3T, Mg-Al layered double hydroxides, were found in the Sinyangri Formation of Jeju Island. They fill the pores of basaltic volcaniclastic sediments in globular and botryoidal aggregates of fine platy particles. Globular aggregates of quintinite-3T were crusted with the parallel overgrowth of motukoreaite plates. X-ray diffraction data and chemical composition are consistent with those reported in literature, while the Mg/Al ratio of motukoreaite is higher. Structural formula of motukoreaite and quintinite-3T derived from electron microprobe analysis are $Na_{1.6}Ca_{0.1}Mg_{40.7}Al_{20.7}Si_{0.9}(CO_3)_{13.6}(SO_4)_{7.4}(OH)_{108}56H_2O$, and $Mg_{3.7}Al_{1.9}Si_{0.2}(OH)_{12}(CO_3)_{0.8}(SO_4)_{0.2}3H_2O$, respectively. Motukoreaite and quintinite-3T were formed by reaction between seawater and basaltic glass, and contributed to the cementation and lithification of the volcaniclastic sediments.

  • Efficient in vitro RNA synthesis can be easily accomplished from cloned DNA using bactrio-phage SP6, T7 or T3 RNA polymerase. Despite its popularity as in vitro transcription system, molecular mechanisms of bacteriophage transcription has not been studied, although physical and catalytic properties of several phage RNA polymerases have well been documented (1). Only recently the T7 promoter has been physically mapped by footprinting of the T7 RNA polymerase (2,3). These simple phage systems, however, could be useful for detailed molecular studies of transcription.

  • The aero-industry is union industry which includes a research development type, a knowledge accumulation type and a developed country type. The aero-industry of Korea is in semi-developed type stage but departed later than that of other country such as Taiwan, Indonesia etc. Therefore, the necessity of domestic airplane material is required. This study on 7050Al extruded alloy aims to suggest an adequate heat treatment conditions of T73, T74 and T76. The results of this study show that; 1. The optimum conditions of T7x heat treatment in extruded 7050Al alloy show this; $$T73:121^{\circ}C{\times}7hr+177^{\circ}C{\times}14hr$$. $$T74:121^{\circ}C{\times}7hr+177^{\circ}C{\times}10hr$$. $$T76:121^{\circ}C{\times}7hr+163^{\circ}C{\times}21hr$$. 2. The 2nd step aging heat treatment such as T73, T74 and T76 etc. is efective in 7050Al alloy but the variation otf microstructure and mechanical property with dispersive inclusions produced for extrusion process causes some troubles. Accordingly, in order to produce a good 7050Al alloy, a careful attention is needed in manufacturing process.

Selection of Optimum Species of Tetraselmis for Mass Culture (대량배양에 적합한 Tetraselmis종의 선택)

  • Tetraselmis is widely used as a live food because of its easy handling, high nutrient, large size and wide tolerant range of temperature and salinity. In order to find the optimum Tetraselmis species for mass culture in Korea, five species of this microalgae were examined on size, optimum culture condition ${\textperthousand}$s, $^{\circ}C.$) and nutrient composition. The results obtained were as follows: Among five species of Tetraselmis, T. sp.(Haeundae) was the largest(major axis $17.6{\pm}1.87^{\mu}$m, mean cell volume 727${\mu}$m), and T. sp. (China) the smallest (major axis $14.6{\pm}1.46^{\mu}$m, mean cell volume 625m). Tetraselmis was very eurythermal and euryhaline species. But optimum temperature and salinity for growth were 24~$30^{\circ}C.$ and 27~30${\textperthousand}$, respectively. Among five species of Tetraselmis, T. sp. (China) seemed to be the most tolerant of high temperature over $30^{\circ}C.$, and T. tetrathele of low temperature below $6^{\circ}C.$. In culture density, T. suecica showed the highest growth rate among the among the five species. The cell density of this microalgae attained to $141{\times}10^4$cells/ml at $24^{\circ}C.$ and 30${\textperthousand}$ within 7 days. In chemical composition, crude protein amount was the highest in T. suecica (44.50%), and crude lipid amount it T. sp. (Haeundae, 7.13%). Total essential amino acid amount was the highest in T. sp. (Haeundae, 50.4%) and total polyunsaturated amount in T. sp. (China, 11.7%) The results on growth and chemical composition of five species of Tetraselmis indicated that T. suecica seemed to be the most suitable species for mass culture in Korea.

Optimal Pre-Plating Method of Chicken Satellite Cells for Cultured Meat Production

  • To establish a pre-plating method of chicken satellite cells with high purity, pre-plating was performed under culture conditions of 37℃ and 41℃, and the pre-plating time was set from a total of 3 hours to 6 hours in consideration of the cell attachment time. The purity of the cells was confirmed by staining paired box protein 7 (Pax7) after proliferation, and Pax7 expression was the highest in culture flasks shaken for 2 hours after incubation at 41℃ for 2 hours to prevent the attachment of satellite cells (p<0.05). Also, when pre-plating and proliferation were performed at 37℃ and 41℃, the Pax7 expression rate was higher at 41℃. The differentiation capabilities of the three groups (T3, T6, and T7) with high Pax7 expression were compared and the fusion index (%) and myotube formation area (%) determined by myosin heavy chain (MHC) staining was calculated. The T6 and T7 groups, which were cultured at 41℃, showed significantly higher values than the T3 group (p<0.05). There was no significant difference in the expression of Pax7 and MHC between the T6 and T7 groups (p>0.05). These results suggest that pre-plating at 41℃ for a total of 4 hours was the most efficient in terms of cost and time for purifying chicken satellite cells for cultured meat.

Isolation and Degradation Activity of a TBTCl (Tributyltin Chloride) Resistant Bacteriain Gwangyang Bay (광양만에서 TBTCl (Tributyltin Chloride) 내성세균의 분리 및 분해활성)

  • BACKGROUND: Tributyltin chloride is among the most toxic compounds known for aquatic ecosystems. Microorganisms are responsible for removal of TBTCl. Nevertheless, only a limited number of marine bacteria were investigated for biodegradation of TBTCl in Korea. METHODS AND RESULTS: The number of TBTCl resistant bacteria ranged from $2.5{\times}10^3$ to $3.8{\times}10^3$ cfu/mL in the seawater, and ranged from $3.2{\times}10^5$ to $9.1{\times}10^5$ cfu/g in the surface sediment, respectively. The morphological, physiological, and biochemical characteristics of TBTCl resistant bacteria were investigated by API 20NE and other tests. The most abundant species of TBTCl resistant bacteria were Vibrio spp. (19.2%), Bacillus spp. (16.2%), Aeromonas spp. (15.2%), and Pseudomonas spp. (13.1%), etc. Eleven TBTCl resistant isolates also had a resistance to heavy metals (Cd, Cu, Hg, and Zn). Among them, isolate T7 showing the strong TBTCl-resistance was selected. This isolate was identified as the genus Pantoea by 16S rRNA gene sequencing and designated as Pantoea sp. T7. In addition, this bacterium was cultivated up to the growth of 50.7% after 60 hrs at TBTCl concentration of $500{\mu}M$. TBTCl-degrading activity of Pantoea sp. T7 was measured by GC-FPD analysis. As a result of biological TBTCl-degradation at TBTCl concentration of $100{\mu}M$, TBTCl-removal efficiency of Pantoeasp. T7 was 62.7% after 40 hrs. CONCLUSION(S): These results suggest that Pantoea sp. T7 is potentially useful for the bioremediation of TBT contamination.

A Study of Functional Evaluation in Stroke Patients (뇌졸중 환자의 기능평가에 대한 연구 -Modified Barthel Index 및 PULSES Profile에 의한 평가성적을 중심으로-)

  • This study was conducted from August, 1980 to March, 9991 to the 40 subjects who were admitted to Kosin Medical Center and received rehabilitation treatment and discharged under the impression of stroke. The objectives are to evaluate the function of the activites of daily living and comprehensive function and find the status of rehabilitation treatment by Modified Barthel Index and PULSES Profile when first requested or rehabilitation treatment(T1), at 2 weeks after rehabilitation treatment(T2), at discharge(T3) and at the time of ambulatory treatment after 2 weeks(T4). The study materials were clinical charts and functional evaluation sheets, and the results are as follows : 21 subject$(52.5\%)$ were male, 19 subjects$(47.5\%)$ were female, and the age distribution was from 19 to 70 in age, the average age was 52.7. By the classification of diagnosis, 21 subjects$(52.5\%)$ were cerebral hemorrhage, 8 subjects$(20.2\%)$ were cerebral thrombosis, 6 subjects$(15.0\%)$ were cerebral embolism, and 5 subjects$(12.5\%)$ were cerebral infarction. The Barthel Index scores were 35.7, 54.5, 71.8, 88.7 on the average at T1, T2, T3, T4 respectively. The PULSES scores were 16.4, 13.7, 11.4, 8.7 on the average at T1, T2, T3, T4 respectively. Regarding the Pearson's correlation coefficient between the Barthel Index scores and the PULSES scores, it was -0.7991(P>0.001) at T1 and -0.8986(P>0.001) at T3, then beth of correlations were very high.

Reasonable Split Application Method of Nitrogen Fertilizer for Early Dry Seeding Culture of Rice in Honam Plain Area (호남평야지에서 벼 건답직파 조기파종재배에 알맞은 질소 분시방법)

  • This study was carried out to elucidate the proper split application method of nitrogen fertilizer for early dry seeding culture of rice in Honam plain area from 1997 to 1998 in Korea. Dongjinbyeo was selected as rice variety for this experiment. The rate of $160kg\;ha^{-1}$ of nitrogen was split as 40-30-30% of total nitrogen at three different application time combination ; T1) basal-5th leaf-panicle formation(PF) stage, T2) 3rd leaf-5th leaf-PF, and T3) 3rd leaf-7th leaf-PF. The content of $NH_4-N$ in soil at 5th leaf stage was higher in top dressing plots(T2, T3) compared with basal application(T1), at 7th leaf stage it was most in top dressed at 3rd leaf and 5th leaf stage, but there was no difference at heading stage. Amount of nitrogen uptake and nitrogen use efficiency was higher in the order of T3, T2 and T1(basal application). Spikelet number per unit area was more in the order of T3, T2 and T1, but rate of ripened grain and 1,000 grain weight were not significantly different among three nitrogen split application methods. Milled rice yields were higher in top dressed plots compared with basal nitrogen application plots. From the results of this experiment, reasonable nitrogen split application method for early dry seeding culture of rice could be 40-30-30% of total nitrogen at 3rd leaf, 7th leaf and panicle formation stage.
